Synthesis and Characterization of Hyaluronic Acid-α-Cyclodextrin Conjugate as the Potential Carrier of PEGylated Drugs

  • The hyaluronic acid (HA) conjugate bearing $\alpha$-cyclodextrin ($\alpha$-CD) was synthesized as the potential carrier of poly(ethylene glycol) (PEG)-drug conjugates. The HA conjugate was prepared by the reaction between the carboxylic acid of HA and the primary amine of $\alpha$-CD in the presence of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide and 1-hydroxybenzotriazole. The chemical structure of the conjugate was confirmed using $^1H$ NMR and FT-IR spectroscopy. The conjugate could form nano-sized particles in the presence of PEG by forming the inclusion complexes between $\alpha$-CD at the backbone of HA, which was demonstrated using electrophoretic light scattering and field emission transmission electron microscopy. It is anticipated that this novel kind of nanoparticles can serve as a useful delivery system for PEGylated drugs.

Synthesis and Characterization of Fluorinated Polyimides for Optical Waveguide Application

  • Fluorinated polyimides with high thermal stability, low optical absorption loss in the optical communication wavelengths of 1.3 and 1.55㎛, and low birefringence and water absorption have been investigated for optical waveguide applications. These polyimides were prepared from 2,2-bis(3,4-dicar-boxyphenyl) hexafluoropropane dianhydride (6FDA) with 1,4-bis-(4-amino-2-trifluoromethyl-phenoxy) tetrafluorobenzene (ATPT) , 1,4-bis-(4-amino-2-trifluoromethyl-phenoxy) benzene (ATPB) , and 1,3-bis-(4-amino-2-trifluoromethyl-phenoxy ) 4,6-dichlorobenzene (ATPD ), respectively. The polyimides obtained had glass transition temperatures that ranged from 260 to 280$\^{C}$, and low water absorption less than 0.4% and low optical loss (> 0.2 dB/cm) at 1.3 and 1.55㎛. Birefringency values of polymer films ranged from 0.0041 to 0.0066.

  • One-dimensional coordination polymer of cadmium(II) complex, $[Cd(SCN)_2(C_4H_6N_2]_n$, has been prepared and characterized by X-ray single crystallography. Structure analysis reveals that each cadmium(II) atom is six-coordinated in distorted octahedral fashion with $CdS_2N_4$ composition. $CdS_2N_4$ composition contains two S and two N atoms from four thiocyanates and tow N atoms from two 4-methylimidazole ligands. Central cadmium(II) atoms are run in parallel to the a-axis and are doubly bridged with neighboring cadmium(II) atoms by the thiocyanate and isothiocyanate ligands. Thus, this complex has a one-dimensional polymer structure in which the 4-methylimidazole is in the trans conformation.

Characterization and Modification of Low Molecular Water-Soluble Chitosan for Pharmaceutical Application

  • The low molecular water-soluble chitosan nanoparticles (LMWSC-NPs) were prepared, which was modified with hydrophilic and hydrophobic moieties to evaluate the potential for pharmaceutics application. The synthesis of LMWSC-NPs was identified by FT-IR and $^1H$-NMR spectra. Also, we measured the photon correlation spectroscopy (PCS), transmission electron microscope (TEM) and atomic force microscope (AFM) to investigate the characteristics and morphology of the LMWSC-NPs. At the PCS measurement, the more increase the number of substitutive group, the more decrease the positive charge of LMWSC-NP surface. From the results of TEM and AFM, spherical morphologies were observed, and their sizes were 30-150 nm. Resultantly, LMWSC-NPs prepared in this experiment will be expected as a suitable device for the drug targeting system.

  • A series of fluorinated aromatic poly(hydroxyamide)s (PHAs) were synthesized by direct polycondensation of diacides containing 2,6-dimethylphenoxy group and quinoxaline ring in the main chain with 2,2-bis-(3-amino-4-hydroxyphenyl) hexafluoropropane. The PHAs had relatively low inherent viscosities in the range of 0.35~0.43 dL/g at $35^{\circ}C$ in DMAc solution. All PHAs exhibited excellent solubility in aprotic solvents such as NMP, DMAc, DMF and DMSO as well as in common organic solvents such as pyridine, THF, and m-cresol at room temperature. However, the poly(benzoxazole)s (PBOs) were quite insoluble in all organic solvents except partially soluble in concentrated sulfuric acid. The PBOs showed glass transition temperatures between 233 and $284^{\circ}C$ by DSC and maximum weight loss temperatures in the range of $536-546^{\circ}C$ by TGA.

  • Pulsatile swelling behaviors and their application to drug delivery system were studied by using interpenetrating polymer networks(IPN) hydrogels constructed with poly(vinyl alcohol) and poly(acrylic acid). The PVA/PAAc IPNs hydrogels were symthesized by UV irradiation tallowed by repetitive freezing and thawing method. These hydrogels showed pH and temperature sensitive swelling behaviors. From the release experiment, the release amount of model drug incorporated into these hydrogels showed pulsatile patterns. Permeability coefficients obtained by various solutes differed in response to changes of permeation conditions.

  • Fusion and characterization of bisphenol-A diglycidyl ether based thermosetting polymer mortars containing an epoxy resin, Fly ash and Rock sand are presented here for the Experimental study. The specimens have been prepared by means of an innovative process, in mild conditions, of commercial epoxy resin, Fly ash and Rock sand based paste. In this way, thermosetting based hybrid mortars characterized by a different content of normalized Fly ash and Rock sand by a homogeneous dispersion of the resin have been obtained. Once hardened, these new composite materials show improved compressive strength and toughness in respect to both the Fly ash and the Rock sand pastes since the Resin provides a more cohesive microstructure, with a reduced amount of micro cracks. The micro structural characterization allows pointing out the presence of an Interfacial Transition Zone similar to that observed in cement based mortars. A correlation between micro-structural features and mechanical properties of the mortar has also been studied.

Copolymers of p-acryloyloxyacetophenone (AcAP) with MMA: Synthesis, Characterization and their Antifouling (AF) Efficiency

  • AcAP was prepared by reacting p-hydroxyacetophenone (HAP) with acryloyl chloride (Ac) in presence of triethylamine (TEA) in MEK at $0^{\circ}C$. The reaction was monitored by TLC and the prepared monomer was characterized by UV, IR, $^{1}H-NMR\;and\;GC-MS$. The homo- [poly (AcAP)] and copolymers [poly (AcAP-co-MMA)] were prepared by solution polymerization at $70^{\circ}C$, in which BPO as initiator. The molecular weight of the polymers was determined by GPC. In order to find out the AF activity of prepared polymers, representatives of marine microfoulers, shipfouling bacteria (B. macroides & P. aeruginosa) and microalgae (A. coffeaeformis & N. incerta) were screened. The results of antibacterial activity and diatom attachment assays revealed potential AF efficiency of these polymers.

  • A series of superabsorbents based on acrylic acid (AA), sodium acrylate, 2-acrylamido-2-methylpropane sulphonic acid, N,N'-methylene bis-acrylamide (MBA) were prepared by inverse suspension polymerization. These hydrogels were further crosslinked on the surface with polyethylene glycol-600 (PEG-600). The water absorbency or swelling behaviors for these xerogels in water and 0.9% saline solutions, both under free condition and under load were investigated. Absorption characteristics of these hydrogels were found to depend on nature and concentration of crosslinker in the system. It was also found that the saline absorption was significantly improved as the incorporation of AMPS in the polymer was increased. The surface crosslinking introduced in the polymers was found to improve the absorption under load characteristics without lowering the free water absorption capacities of the polymer to a considerable extent.
