토양으로부터 oleic acid 를 탄소원으로 이용하여 MCL-PHA의 생산능이 우수한 균주를 분리하여 Pseudomonas sp. MBEL21이라 명명하였다. Pseudomonas sp. MBEL21의 PHA함량을 증가시키기 위하여 여러 가지 필수 영양분의 제한 조건에서 배양한 결과, 인산이 제한된 조건에서 가장 높은 함량의 PHA를 축적하였다. 고농도의 MCL-PHA생산을 위한 Pseudomonas sp. MBEL21의 유가식 배양 전략을 개발하여 28.1 wt%의 함량으로 23 g/L. MCL-PHA를 생산하였다. 또한, 분리된 Pseudomonas sp. MBEL21을 olive oil을 탄소원으로 이용하여 배양한 결과 9.3 wt%의 PHA가 축적되어졌으며, MCL-hydroxyalkanoate와 함께 3-hyoxy-butyrate 의 단량체들로 이루어진 PHA가 확인되어졌다. 이러한 결과들은 분리된 신규 Pseudomonas sp. MBEL21이 저가의 olive oil로부터 생분해성 탄성체의 응용분야를 갖는 MCL-PHA를 효율적으로 생산할 수 있다는 것을 보여준다.
Enantiomerically pure hydroxycarboxylic acids have great potential as chiral building blocks in fine chemicals due to their easily modificable two functional groups. Microbial polyhydroxyalkanoates (PHAs) can have more than one hundred of hydroxycarboxylic acids as monomeric constituents by growing cells under various conditions. All of the monomeric constituents are enantiomerically pure in (R)-conformation if there is a chiral center. Therefore, efficient production of enantiomerically pure hydroxycarboxylic acids by degrading PHAs is possible. In this presentation, we report on the development of a novel method for the preparation of (R)-hydroxybutyric acid by in vivo depolymerization of Polyhydroxybutyrae.
오일 피크, 기후변화 그리고 미세 플라스틱과 관련된 석유합성 플라스틱의 생산과 사용은, 지속 가능한 인류 생활을 위협하는 범세계적 이슈로 대두되고 있다. 이러한 문제를 해결하기 위해 다양한 생분해성 친환경 바이오플라스틱 소재가 대안이 되고 있으며, 그중 주목받고 있는 소재 중 하나는 polyhydroxyalkanoates (PHA) 생분해성 바이오플라스틱이다. 본 총설은 PHA 생산공정에 이용될 수 있는 해양 미생물의 산업적 활용 이점과 현재까지 밝혀진 해양미생물의 생산성을 비교하고, 이들이 산업에서 활용되기 위해 필요한 연구개발 현황에 대해 조사하였다. 조사 결과 해양미생물로부터 생산된 PHA는 석유합성 플라스틱이 보유한 다양한 물리적 특성을 띄는 중합체 소재로의 대체 가능성과, 배지 제조 시 해수를 사용할 수 있는 장점, 특별한 멸균 과정이 필요치 않은 장점, 그리고 분리 정제 과정 등에서 비용 절감의 장점을 제공할 수 있음을 확인하였다. 하지만 해양미생물의 PHA 생산성은 육상에서 분리된 상용화 균주에 비해 다소 떨어지는 효율을 보이는 것을 확인하였다. 이러한 문제점을 해결하기 위해 최근 선진 omics 기반 합성 미생물학 기반 기술을 활용하여, 고밀도 연속배양기술 개발, PHA 효율 증진 및 다양한 시장 요구에 필요한 생분해성 플라스틱 소재 개발이 미래 플라스틱 대체 소재 개발에 필요한 연구임을 확인하였다.
식물유를 탄소원으로 사용하여 medium-chain-length polyhydroxyalkanoates (MCL-PHAs)를 생산할 수 있는 Pseudomonas chlororaphis HS21을 대상으로 대사경로의 변화를 통하여 식물유의 MCL-PHA로의 전환율을 증진시키고 MCL-PHAs의 단위체 조성의 변화를 유도하기 위한 방안을 모색하였다. P. chlororaphis HS21의 MCL-PHAs 생합성은 세포생장과 동시에 일어나는 특징을 보였으며, 팜유를 유일 탄소원으로 사용한 회분배양의 결과 2.4 g/L의 건체량과 건체량의 $40.2\;wt{\%}$에 해당하는 MCL-PHAs를 얻을 수 있었다. 또한 생합성된 MCL-PHAs의 단위체는 3-hydroxyhexanoate($7.0\;mol{\%}$, 3-hydroxyoctanoate ($45.3\;mol{\%}$. 3-hydroxydecanoate ($39.0\;mol{\%}$), 3-hydroxydodecanoate ($6.8\;mol{\%}$) 및 3-hydroxytetradecanoate ($1.9\;mol{\%}$)로 구성되어 있었다. 식물유와는 달리 포도당과 같은 탄수화물은 P. chlororaphis HS21의 생장에는 이용되지만 MCL-PHAs의 생합성에는 거의 이용되지 못하는 탄소원임을 확인하고, 식물유와 함께 포도당을 보조기질로 공급한 결과 식물유의 MCL-PHAs로의 전환율이 크게 증가함으로써, PHA 생산에 직접적으로 이용되지 못하는 보조기질의 사용을 통하여 특정 기능기를 함유하는 기질로부터 해당 기능기를 갖는 MCL-PHAs를 효율적으로 생산할 수 있음을 알 수 있었다. 또한 지방산의 ${\beta}-oxidation$ 회로를 방해하는 아크릴산을 첨가할 경우 아크릴산의 독성에 의하여 세포생장은 저해를 받지만 세포 내 MCL-PHAs의 축적율은 감소하지 않았으며, MCL-PHAs를 구성하는 단위체 중 3-hydroxydo-decanoate 및 3-hydroxytetradecanoate와 같이 탄소수가 보다 큰 단위체의 함량이 크게 증가하였다. 이러한 특징에 의해 해바라기유와 옥수수유로부터는 3-hydroxydodecenoate, 3-hydroxytetradecenoate와 같은 불포화 단위체의 함량이 크게 증가된 기능성 MCL-PHAs를 생산할 수 있었다. 이러한 결과는 아크릴산의 첨가와 같은 PHA 대사경로의 인위적 변화가 새로운 단위체 조성을 갖거나 기능기를 가짐으로써 독특한 물성을 지니는 신규의 MCL-PHAs 개발에 유용할 수 있음을 보여준다.
Fed-batch culture of Pseudomonas oleovorans was carried out for the production of medium-chain-length polyhydroxyalkanoates (MCL-PHAs) using octanoate as a carbon source. Octanoate and the salt solution containing ammounium sulfate and magnesium sulfate were intermittently fed in the course of fermentation. Cell mass and PHA concentrations of 42.8 and 16.8g/L, respectively, could be obtained in 40 h. The PHA content and the PHA productivity were 39.2% and 0.42 g PHA/L-h, respectively. The yields of cell mass and PHA were 0.71 g dry cell mass/g octanoate and 0.28g PHA/g octanoate, respectively. Therefore, octanoate can be used for the production of MCL-PHAs to a high concentration with high productivity.
나노기술과 바이오기술의 융합연구에 의해 나노바이오기술이 발전되고 있다. 나노바이오기술의 중요한 응용연구 중의 하나로서, 진단이나 바이오센서 분야에서 단백질-단백질 및 단백질-바이오물질간의 상호작용을 연구하기 위한 단백질 센서 칩이 개발되어 왔다. 본 논문에서는 단백질의 선택적 고정화를 위한 새로운 생체고분자 기질로 PHA를 이용하는 첫 번째 예로서, 단백질-단백질 및 항원-항체 반응의 구현을 나타내고자 하였다. 본 시스템은 PHA 표면 위에서 PHA depolymerase의 SBD와의 선택적 결합에 기반한 것으로, PHA depolymerase의 SBD와 융합된 단백질이 PHA가 코팅된 표면 위에 spotting 될 수 있고 미세접촉인쇄방법에 의해 PHA 위에 미세패턴이 제조되어지는 것을 알 수 있었다(52, 53). 이러한 새로운 전략이 PHA depolymerase의 SBD와 다른 단백질을 융합함으로서 미세 spotting과 미세패터닝이 가능하게 되었고 항원-항체의 생물학적 반응을 통해 많은 바이오센서 칩 연구에 응용될 수 있음을 확인하였다. 또한, PHA 마이크로 비드에도 PHA depolymerase의 SBD와 융합된 단백질을 고정시킴으로서 항원-항체 반응을 유도할 수 있음을 확인하였다(54). PHA의 구조를 변경하여 PHA 기판, PHA 필름, PHA 미세패턴, PHA 마이크로 비드 등을 이용할 수 있으며 multiplex assay를 동시에 진행할 수 있는 다양한 융합 단백질을 사용할 수 있을 것이다. 생분해성 플라스틱으로서 성공적으로 개발된 PHA를 이용한 새로운 플랫폼 기술이 PHA depolymerase의 SBD를 이용함으로서 특이적이고 선택적인 단백질의 고정화에 이용될 수 있음을 확인하였다. 본 전략이 다양한 단백질-단백질 및 단백질-바이오물질 반응을 이용한 바이오칩 및 바이오센서의 응용연구에 유용하게 사용될 것이다.

One of the major challenges in metabolic engineering for enhanced synthesis of value-added chemicals is to design and develop new strains that can be translated into well-controlled fermentation processes using bioreactors. The aim of this study was to assess the influence of various fed-batch strategies in the performance of metabolically engineered Pseudomonas putida strains, ${\Delta}gcd$ and ${\Delta}gcd-pgl$, for improving production of medium-chain-length polyhydroxyalkanoates (mcl-PHAs) using glucose as the only carbon source. First we developed a fed-batch process that comprised an initial phase of biomass accumulation based on an exponential feeding carbon-limited strategy. For the mcl-PHA accumulation stage, three induction techniques were tested under nitrogen limitation. The substrate-pulse feeding was more efficient than the constant-feeding approach to promote the accumulation of the desirable product. Nonetheless, the most efficient approach for maximum PHA synthesis was the application of a dissolved-oxygen-stat feeding strategy (DO-stat), where P. putida ${\Delta}gcd$ mutant strain showed a final PHA content and specific PHA productivity of 67% and $0.83g{\cdot}l^{-1}{\cdot}h^{-1}$, respectively. To our knowledge, this mcl-PHA titer is the highest value that has been ever reported using glucose as the sole carbon and energy source. Our results also highlighted the effect of different fed-batch strategies upon the extent of realization of the intended metabolic modification of the mutant strains.
The potential polyhydroxyalkanoates (PHA)-producing bacteria, Bacillus megaterium PP-10, was successfully isolated and studied its feasibility for utilization of pineapple peel waste (PPW) as a cheap carbon substrate. The PPW was pretreated with 1% (v/v) H2SO4 under steam sterilization and about 26.4 g/l of total reducing sugar (TRS) in pineapple peel hydrolysate (PPH) was generated and main fermentable sugars were glucose and fructose. A maximum cell growth and PHA concentration of 3.63 ± 0.07 g/l and 1.98 ± 0.09 g/l (about 54.58 ± 2.39%DCW) were received in only 12 h when grown in PPH. Interestingly, PHA productivity and biomass yield (Yx/s) in PPH was about 4 times and 1.5 times higher than in glucose. To achieve the highest DCW and PHA production, the optimal culture conditions e.g. carbon to nitrogen ratios of 40 mole/mole, incubation temperature at 35℃ and shaking speed of 200 rpm were performed and a maximum DCW up to 4.24 ± 0.04 g/l and PHA concentration of 2.68 ± 0.02 g/l (61% DCW) were obtained. The produced PHA was further examined its monomer composition and found to contain only 3-hydroxybutyrate (3HB). This finding corresponded with the presence of class IV PHA synthase gene. Finally, certain thermal properties of the produced PHA i.e. the melting temperature (Tm) and the glass transition temperature (Tg) were about 176℃ and -4℃, respectively whereas the Mw was about 1.07 KDa ; therefore, the newly isolated B. megaterium PP-10 is a promising bacterial candidate for the efficient conversion of low-cost PPH to PHA.
Five microorganisms capable of degrading an aromatic medium-chain-length polyhydroxyalkanoate ($PHA_{MCL}$), poly(3-hydroxy-5-phenylvalerate) (PHPV), were isolated from wastewater-treatment sludge. Among the isolates, JS02 showed degrading activity consistantly during several transfers. The isolate JS02 could hydrolyze another aromatic MCL copolyester, poly(3-hydroxy-5-phenoxyvalerate-co-3-hydroxy-7-phenoxyheptanoate), [P(5POHV-co-7POHH)], and other short-chain-length PHAs ($PHA_{SCL}) such as poly(3-hydroxybutyrate) [P3(HB)], poly(3-hydroxybutyrate-co-4-hydroxybutyrate) [P(3 HB-co-4 HB)], and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] with relatively low activity. The culture supernatant of JS02 showed hydrolyzing activity for the p-nitrophenyl esters of fatty acids.
