Chemical Absorption of Carbon Dioxide into Non-Newtonian Polyacrylamide Solution (Polyacrylamide 비뉴튼액체에서의 이산화탄소의 화학흡수)

  • Absorption rate of carbon dioxide was measured in the aqueous polyacrylamide (PAA) solution containing triethanolamine (TEA) of $0{\sim}2.0\;kg\;mol/m^3$ in a flat-stirred vessel with an impeller of 0.05 m and agitation speed of 50 rpm at $25^{\circ}C$ and 101.3 kPa. The chemical absorption rate of $CO_2$ was estimated by mass transfer mechanism based on the film theory using the physicochemical properties containing the liquid-side mass transfer coefficient of $CO_2$ and the kinetics of reaction between $CO_2$ and TEA to compare with the measured rate. The aqueous PAA solution acted as a reducing agent by viscoelastic property of non-Newtonian liquid based on the same viscosity of the solution.

  • We propose a method for rapid evaluation and permanent record of sodium dodecyl sulfate polyacrylamide gel electrophoretic runs. This method is based on the photocopy process, rather than on photography and requires no extensive or expensive investment. Comparison of a print obtained through this method and a 35mm photography indicated, on a balanced gel, equal sensitivity.

  • Two-dimensional electrophoresis (2-DE) was executed to separate the seed storage proteins from the buckwheat. The proteins extracted from the whole seed proteins were better separated and observed in the use of lysis buffer. Using this method, the highly reproducible isoelectric focusing (IEF) can be obtained from polyacrylamide gels, and IEF from the polyacrylamide gel at all the possible pH range (5.0-8.0) was more easily separated than IPG (immobilized pH gradient) gels. The polyacrylamide gels in the first dimension in 2-DE was used to separate and identify a number of whole seed proteins in the proteome analysis. In this new apparatus using 2-DE, 27cm in length of plate coated with polyacrylamide gel was used and the experiment was further investigated under the various conditions.

  • In this paper, a series of amphoteric polyacrylamide(PAM) with different molecular weight and charge were synthesized and used as retention and dewatering aids. Better products had been selected and tested under varied conditions. Effects of molecular weight, cationic charge density, degree of hydrolysis and other factors of PAM on retention and dewatering of papermaking were also considered. Finally, synthesized quaternary and tertiary ammonium amphoteric polyacrylamide were practised in paper mill trial.

  • The optimum culture time and initial pH, for the production of exo-maltotetraohydrolase from Pseudomonas stutzeri IAM 12097, in the trypticase medium were 36 hrs and pH 6.3, respectively. Exo-maltotetraohydrolase was purified by $(NH_4)_{2}SO_4$ and two times of column chromatography on DEAE-cellulose. Specific activity of the purified enzyme was 108.6U/mg protein and yield of the enzyme activity was 9.4%. The purified enzyme showed a single band on polyacrylamide gel electrophoresis and SDS-polyacrylamide gel electrophoresis.

  • The denature polyacrylamide gel stain silver nitrate is used for routine nucleic acid detection. More sensitive stains, such as Vistra Green, SYBR Green are available to address a broad range of DNA applications requiring lower detection limits in polyacrylamide gel formats. Gel Scanners, laser-scanning instruments, provide sensitive fluorescence detection of DNA gel stains. We established one step fluorescent impregnation enhanced sensitivity with simple, rapid and low cost. We have applied this fluorescent staining procedure for the routine analysis of DNA profiles generated by SSR amplification.

  • Porcine liver esterase was immobilized in polyacrylamide gel for the enantioselective production of levofloxacin from ofloxacin butyl ester. The initial activity of immobilized esterase was found to be significantly affected by the polyacrylamide gel composition. The optimum concentrations of monomer and crosslinker were determined to be 20% and 8.3%, respectively. The activity of immobilized esterase was 55.4% compared to a free enzyme. Enantiomeric excess was maintained at 60%, almost the same level as that of free enzyme. In addition, the immobilized esterase could be used repeatedly up to 10 times without experiencing any severe loss of activity and enantioselectivity.

  • An invertase from Korean ginseng (Panax ginseng C. A. Mayer) was purified by means of DEAE-cellulose column chromatography and gel-filtration through Sephadex G-75. The homogeneity of the purified invertase was proved by polyacrylamide gel disc electrophoresis. The enzyme was separated into two subunits by SDS-polyacrylamide gel electrophoresis, showing its molecular weights as 48,000. The enzyme preparation showed a characteristic protein UV-spectra.

Ultrasonic Velocity and Absorption Measurements for Polyacrylamide and Water Solutions

  • Both ultrasonic velocity at 3 MHz and absorption coefficient for the frequency range of 0.2-2 MHz were measured in an aqueous solution of polyacrylamide for the concentration range of 0.5% to 2.5% by weight. Pulse echo overlap method was taken for measuring the ultrasonic velocity over the temperature range of 10-90℃ and the high-Q ultrasonic resonator method was used for the absorption coefficient at 30℃. The velocity exhibited a maximum value at approximately 70℃, 71℃, 72℃, 73℃ and 74℃ in 2.5%, 2.0%, 1.5%, 1.0%, and 0.5% solutions, respectively. The velocity increased with the concentration at a given temperature. The ultrasonic absorption (a/f²) at a given temperature increased linearly with the concentration for the concentration below 1.5%, but suddenly increased for the concentration above 1.5% concentration. The value of a/f² at 1MHz was entirely due to the classical Stoke's viscous effect. The ultrasonic relaxation in polyacrylamide aqueous solutions, which may be the result of structural fluctuations of polymer molecules such as the segmental motion of the polymer chains, was observed, and at 2.5%, the value of a/f² was found to suddenly increase as frequency decreased.
