Ziyuglycoside-II ($3{\beta}-3-{\alpha}$-1-arabinopyranosyloxy-19-hydroxyurs-12-en-28-oicacid)는 오이풀(Sanguisorba officinalis L.)의 주요 활성 화합물 중 하나로 지혈작용, 항산화활성, 항염증활성 등의 활성이 보고되어 있다. 그러나 염증성 대장암에서의 ziyuglycoside-II의 활성에 관해서는 아직 보고되지 않아 본 연구에서 azoxymethane (AOM)/dextran sulfate sodium (DSS)으로 유발된 대장암 모델에서 ziyuglycoside-II 항종양활성을 조사하였다. 염증성 대장암을 유발하기 위해 BALB/c 마우스에게 AOM을 1회 복강 주사하고 AOM 투여 1주일 후 총 3 cycle의 2% 농도의 DSS를 음용수로 공급 하였다. Ziyuglycoside-II (1 또는 5 mg/kg)는 1주일에 3회 경구 투여하고, 64일에 대장을 적출하였다. 대장 조직에서의 종양 개수를 관찰한 결과 ziyuglycoside-II의 투여가 종양의 형성을 유의적으로 감소시키는 것을 확인하였다. 또한 Western blot 방법과 면역 조직학 분석의 결과, ziyuglycoside-II의 투여가 대장 조직에서 nuclear factor kappa-B 양성 세포와 염증 관련 단백질의 양을 현저히 감소시킴을 관찰하였다. 또한 ziyuglycoside-II 투여에 의해 대장조직내의 세포사멸이 촉진되었고 cleaved-caspase 3, cleaved-caspase 7과 같은 세포사멸 관련 단백질의 발현이 증가함을 확인하였다. 이러한 결과는 ziyuglycoside-II의 투여가 염증반응을 억제하고 세포 자멸을 유도하여 염증성대장암의 발생을 억제함을 시사하고 있다.
파킨슨병은 운동완서, 근육경직, 진전 및 비정상적인 자세 등을 임상적 특징으로 하는 주로 운동 신경계에 영향을 주는 진행성 신경 퇴행성 질환이다. 파킨슨병은 산화 스트레스와 세포 내 신호 전달 경로의 조절 장애에 의한 뇌 흑색치밀부에서의 도파민성 신경세포의 사멸을 특징으로 한다. Quercetin의 주요 대사산물인 Quercetin-3-O-glucuronide (Q3GA)는 신경 보호 효과가 있는 것으로 보고 되어 왔다. 본 연구에서는 SH-SY5Y 세포에서 1-methyl-4-phenyl pyridinium ($MPP^+$)에 의해 유도된 신경 독성에 대한 Q3GA의 신경 보호 효과와 그 분자 조절 기전을 조사하였다. Q3GA는 $MPP^+$에 의해 유도된 세포 사멸을 유의적으로 감소시켰으며 PARP 절단을 감소시켰다. 또한, Bax/Bcl-2 비율의 감소와 함께 $MPP^+$에 의해 증가된 세포 내 ROS를 감소시켰다. Q3GA는 $MPP^+$에 의해 감소된 Akt와 CREB의 인산화를 유의적으로 회복시켰지만, ERK에는 영향을 미치지 않았다. 이 결과는 Q3GA가 ROS 생산 억제와 Akt/CREB 신호 전달 경로를 통해 $MPP^+$ 에 의해 유도된 신경 독성을 억제시킬 수 있음을 시사한다. 본 연구는 Q3GA가 파킨슨병에 대한 예방제 또는 치료제로 개발될 수 있는 가능성을 제시한다.
고중성지방혈증은 죽상동맥경화증의 주요한 위험 요인 중 하나이다. 중성지방은 대식세포의 세포 사멸을 유도하여 죽상동맥경화증 발생에 기여하는 것으로 알려져 있다. 본 연구팀은 앞선 연구에서 대식세포의 중성지방-유도 세포 사멸이 pannexin-1 활성화에 의한 세포 외 ATP 농도 증가, caspase-2와 caspase-1 활성화, caspase-8을 포함한 apoptotic caspase 활성화 경로로 일어나는 것을 보고하였다. 한편 다른 연구들에서는 세포 내 다른 여러 기전에서 caspase-8이 caspase-1과 -2의 상위 단백질이라 보고하고 있다. 따라서 본 연구에서는 caspase-8이 중성지방-유도 대식세포 사멸 과정에서 상위단백지로 영향을 미치는지 여부를 조사하기 위해 수행되었다. 본 연구진은 caspase-8이 중성지방-유도 대식세포 사멸 과정에서 caspase-3 활성화 및 PARP 절단을 유도하였다. 다음으로 중성지방이 처리된 대식세포에서 caspase-8 억제 시, caspase-8의 상위 단백질로 보고한 caspase-1 및 -2의 활성이 감소하는 것을 확인하였다. 또한 ATP 처리 시 caspase-8 억제제 처리에 의해 감소된 caspase-2의 활성이 회복되는 것을 확인하였다. 위의 결과를 통해 caspase-8이 중성지방-유도 대식세포 사멸 과정에서 세포 외부 ATP 농도 증가에 관여하는 단백질 또는 그 상위 기전에 양성피드백 방식으로 영향을 미쳐 caspase-1과 -2를 활성화하여 중성지방-유도 대식세포 사멸을 증진시킴을 알 수 있다.
Background: 20(S)-protopanaxadiol (20(S)-PPD), one of the aglycone derivatives of major ginsenosides, has been shown to have an anticancer activity toward a variety of cancers. This study was initiated with an attempt to evaluate its anti-cancer activity toward human endometrial cancer by cell and xenograft mouse models. Methods: Human endometrial cancer (HEC)-1A cells were incubated with different 20(S)-PPD concentrations. 20(S)-PPD cytotoxicity was evaluated using MTT assay. Apoptosis was detected using the annexin V binding assay and cell cycle analysis. Cleaved poly (ADP-ribose) polymerase (PARP) and activated caspase-9 were assessed using western blotting. HEC-1A cell tumor xenografts in athymic mice were generated by inoculating HEC-1A cells into the flank of BALB/c female mice and explored to validate 20(S)-PPD anti-endometrial cancer toxicity. Results: 20(S)-PPD inhibited HEC-1A cell proliferation in a dose-dependent manner with an IC50 value of 3.5 μM at 24 h. HEC-1A cells morphologically changed after 20(S)-PPD treatment, bearing resemblance to Taxol-treated cells. Annexin V-positive cell percentages were 0%, 10.8%, and 58.1% in HEC-1A cells when treated with 0, 2.5, and 5 μM of 20(S)-PPD, respectively, for 24 h. 20(S)-PPD subcutaneously injected into the HEC-1A cell xenograft-bearing mice three times a week for 17 days manifested tumor growth inhibition by as much as 18% at a dose of 80 mg/kg, which sharply contrasted to controls that showed an approximately 2.4-fold tumor volume increase. These events paralleled caspase-9 activation and PARP cleavage. Conclusion: 20(S)-PPD inhibits endometrial cancer cell proliferation by inducing cell death via a caspase-mediated apoptosis pathway. Therefore, the 20(S)-PPD-like ginsenosides are endowed with ample structural information that could be utilized to develop other ginsenoside-based anticancer agents.

BACKGROUND/OBJECTIVES: Zanthoxylum schinifolium is traditionally used as a spice for cooking in East Asian countries. This study was undertaken to evaluate the anti-proliferative potential of ethanol extracts of Z. schinifolium leaves (EEZS) against human bladder cancer T24 cells. MATERIALS/METHODS: Subsequent to measuring the cytotoxicity of EEZS, the anti-cancer activity was measured by assessing apoptosis induction, reactive oxygen species (ROS) generation, and mitochondrial membrane potential (MMP). In addition, we determined the underlying mechanism of EEZS-induced apoptosis through various assays, including Western blot analysis. RESULTS: EEZS treatment concentration-dependently inhibited T24 cell survival, which is associated with apoptosis induction. Exposure to EEZS induced the expression of Fas and Fas-ligand, activated caspases, and subsequently resulted to cleavage of poly (ADP-ribose) polymerase. EEZS also enhanced the expression of cytochrome c in the cytoplasm by suppressing MMP, following increase in the ratio of Bax:Bcl-2 expression and truncation of Bid. However, EEZS-mediated growth inhibition and apoptosis were significantly diminished by a pan-caspase inhibitor. Moreover, EEZS inhibited activation of the phosphoinositide 3-kinase (PI3K)/Akt pathway, and the apoptosis-inducing potential of EEZS was promoted in the presence of PI3K/Akt inhibitor. In addition, EEZS enhanced the production of ROS, whereas N-acetyl cysteine (NAC), a ROS scavenger, markedly suppressed growth inhibition and inactivation of the PI3K/Akt signaling pathway induced by EEZS. Furthermore, NAC significantly attenuated the EEZS-induced apoptosis and reduction of cell viability. CONCLUSIONS: Taken together, our results indicate that exposure to EEZS exhibits anti-cancer activity in T24 bladder cancer cells through ROS-dependent induction of apoptosis and inactivation of the PI3K/Akt signaling pathway.

Sirtuins (SIRTs) belong to the nicotinamide adenine dinucleotide (NAD+)-dependent class III histone deacetylase family. They are key regulators of cellular and physiological processes, such as cell survival, senescence, differentiation, DNA damage and stress response, cellular metabolism, and aging. SIRTs also influence carcinogenesis, making them potential targets for anticancer therapeutic strategies. In this study, we investigated the anticancer properties and underlying molecular mechanisms of a novel SIRT1 inhibitor, MHY2251, in human colorectal cancer (CRC) cells. MHY2251 reduced the viability of various human CRC cell lines, especially those with wild-type TP53. MHY2251 inhibited SIRT1 activity and SIRT1/2 protein expression, while promoting p53 acetylation, which is a target of SIRT1 in HCT116 cells. MHY2251 treatment triggered apoptosis in HCT116 cells. It increased the percentage of late apoptotic cells and the sub-G1 fraction (as detected by flow cytometric analysis) and induced DNA fragmentation. In addition, MHY2251 upregulated the expression of FasL and Fas, altered the ratio of Bax/Bcl-2, downregulated the levels of pro-caspase-8, -9, and -3 proteins, and induced subsequent poly(ADP-ribose) polymerase cleavage. The induction of apoptosis by MHY2251 was related to the activation of the caspase cascade, which was significantly attenuated by pre-treatment with Z-VAD-FMK, a pan-caspase inhibitor. Furthermore, MHY2251 stimulated the phosphorylation of c-Jun N-terminal kinase (JNK), and MHY2251-triggered apoptosis was blocked by pre-treatment with SP600125, a JNK inhibitor. This finding indicated the specific involvement of JNK in MHY2251-induced apoptosis. MHY2251 shows considerable potential as a therapeutic agent for targeting human CRC via the inhibition of SIRT1 and activation of JNK/p53 pathway.
Fisetin is a bioactive flavonol molecule and has been shown to have antioxidant potential, but its efficacy has not been fully validated. The aim of the present study was to investigate the protective efficacy of fisetin on C2C12 murine myoblastjdusts under hydrogen peroxide (H2O2)-induced oxidative damage. The results revealed that fisetin significantly weakened H2O2-induced cell viability inhibition and DNA damage while blocking reactive oxygen species (ROS) generation. Fisetin also significantly alleviated cell cycle arrest by H2O2 treatment through by reversing the upregulation of p21WAF1/CIP1 expression and the downregulation of cyclin A and B levels. In addition, fisetin significantly blocked apoptosis induced by H2O2 through increasing the Bcl-2/Bax ratio and attenuating mitochondrial damage, which was accompanied by inactivation of caspase-3 and suppression of poly(ADP-ribose) polymerase cleavage. Furthermore, fisetin-induced nuclear translocation and phosphorylation of Nrf2 were related to the increased expression and activation of heme oxygenase-1 (HO-1) in H2O2-stimulated C2C12 myoblasts. However, the protective efficacy of fisetin on H2O2-mediated cytotoxicity, including cell cycle arrest, apoptosis and mitochondrial dysfunction, were greatly offset when HO-1 activity was artificially inhibited. Therefore, our results indicate that fisetin as an Nrf2 activator effectively abrogated oxidative stress-mediated damage in C2C12 myoblasts.
Mangiferin is a kind of natural xanthone glycosides and is known to have various pharmacological activities. However, since the beneficial efficacy of this compound has not been reported in retinal pigment epithelial (RPE) cells, this study aimed to evaluate whether mangiferin could protect human RPE ARPE-19 cells from oxidative injury mimicked by hydrogen peroxide (H2O2). The results showed that mangiferin attenuated H2O2-induced cell viability reduction and DNA damage, while inhibiting reactive oxygen species (ROS) production and preserving diminished glutathione (GSH). Mangiferin also antagonized H2O2-induced inhibition of the expression and activity of antioxidant enzymes such as manganese superoxide dismutase and GSH peroxidase, which was associated with inhibition of mitochondrial ROS production. In addition, mangiferin protected ARPE-19 cells from H2O2-induced apoptosis by increasing the Bcl-2/Bax ratio, decreasing caspase-3 activation, and blocking poly(ADP-ribose) polymerase cleavage. Moreover, mangiferin suppressed the release of cytochrome c into the cytosol, which was achieved by interfering with mitochondrial membrane disruption. Furthermore, mangiferin increased the expression and activity of heme oxygenase-1 (HO-1) and nuclear factor-erythroid-2 related factor 2 (Nrf2). However, the inhibition of ROS production, cytoprotective and anti-apoptotic effects of mangiferin were significantly attenuated by the HO-1 inhibitor, indicating that mangiferin promoted Nrf2-mediated HO-1 activity to prevent ARPE-19 cells from oxidative injury. The results of this study suggest that mangiferin, as an Nrf2 activator, has potent ROS scavenging activity and may have the potential to protect oxidative stress-mediated ocular diseases.
본 연구에서는 메탄올로 추출한 시금치, 양배추, 양파 추출물의 인간 위암세포 AGS와 인간 유방암세포 MDA-MB-231, SK-BR-3 암세포에서의 세포 증식 억제 효과와 apoptosis 유도 효과에 대하여 비교 조사하였다. AGS, MDA-MB-231, SK-BR-3 암세포에 시금치, 양배추, 양파 추출물을 0, 50, 100, $200{\mu}g/mL$로 24시간 동안 처리한 뒤 MTT assay를 통하여 세포 생존율을 측정한 결과 암세포 생존율이 농도 의존적으로 감소하였다. 이러한 시금치, 양배추, 양파 추출물의 암세포 증식 억제 효과가 apoptosis에 의해 유도되는지 확인하기 위해 AGS, MDA-MB-231, SK-BR-3 암세포에 각 추출물을 0, $200{\mu}g/mL$로 24시간 동안 처리한 후 DAPI staining을 수행한 결과 apoptotic body와 염색질 응축이 추출물 처리군에서 증가하였다. MTT assay와 DAPI staining 결과 암세포 증식 억제 효과 및 apoptosis 유도효과가 가장 우수했던 양파 추출물에 의한 apoptosis 관련 단백질들의 변화 양상을 확인하기 위해 western blotting을 수행하였다. AGS, MDA-MB-231, SK-BR-3 암세포에 양파 추출물을 0, $200{\mu}g/mL$로 24시간 동안 처리한 뒤 western blotting을 수행한 결과 pro-apoptotic 인자인 Bax 발현은 증가하였으며 PARP 단백질의 분절 또한 증가한 것을 확인하였다. 반면 anti-apoptotic 인자인 Bcl-2의 발현은 감소하였다. 이러한 결과들을 종합하였을 때 시금치, 양배추, 양파 추출물은 인간 위암세포인 AGS, 인간 유방암세포인 MDA-MB-231, SK-BR-3에서 apoptosis 유도를 통해 암세포 성장을 억제하는 것으로 생각한다. 그중 양파 추출물의 효과가 가장 우수했으며 유방암세포보다 위암세포에서의 apoptosis 유도 효과가 뛰어났다. 따라서 양파의 섭취는 위암의 예방 및 치료를 위한 치료제로의 개발 가능성이 우수함을 제시하며 추후 지속적인 연구를 통하여 in vivo에서의 양파 추출물의 항암 효과에 대한 연구가 이루어져야 할 것으로 생각한다.
본 연구에서는 인체 대장암세포인 HT29를 사용하여 포황 메탄올 추출물(Methanol extract of Typha orientalis, METO)의 항암 활성 및 그 분자적 기전에 관하여 분석하였다. 먼저 METO가 다양한 암세포의 증식에 미치는 영향을 분석한 결과, 인체 대장암 세포, 폐암 세포, 간암 세포 등의 세포증식을 억제하였으며 그 중에서도 대장암 세포인 HT29에 대해 강한 세포 증식 억제 효과를 나타내었다. METO에 의한 세포 증식 억제 기전을 분석하기 위하여 Flow cytometry analysis를 수행한 결과, METO 농도의존적으로 HT29 세포의 G2/M기 세포분포가 증가하고 아울러 apoptosis 유발군인 SubG1기 세포분포가 증가하는 것을 확인할 수 있었다. METO에 의한 HT29 세포의 G2/M arrest는 Cdc2의 inactive form인 phospho-Cdc2의 증가에 의한 G2/M checkpoint 관련 단백질의 활성억제에 의한 것이라 사료된다. 이러한 phospho-Cdc2의 증가는 METO에 의해 발현이 증가된 Wee1 kinase와 발현이 감소된 Cdc25C phosphatase에 의해 야기된 것임을 확인하였다. 또한 METO에 의한 HT29의 apoptosis 유도에 관한 분자적 기전 분석을 위해 Western blot analysis를 수행한 결과, METO 농도가 증가할수록 종양 억제 유전자인 p53, death receptor인 FAS, Bcl-2 family 중 pro-apoptotic 단백질인 Bax 및 cytosolic cytochrome C의 발현이 증가되고, Caspase-3가 활성화되어 단편화된 Caspase-3의 증가가 관찰되었다. 또한 활성화된 Caspase-3의 기질 단백질인 PARP의 단편화가 일어나 apoptosis가 유도되는 것을 알 수 있었다. 이상의 결과들로부터 METO는 인체 대장암세포 HT29의 G2/M arrest 및 apoptosis 유도에 의한 항암활성을 보유함을 확인하였다.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
