Fabrication of Non Viral Vector for Drug and Gene Delivery using Particle Replication In Non-Wetting Templates (PRINT) Technique (Particle Replication In Non-Wetting Templates (PRINT) 방법을 이용한 약물 및 유전자 전달체의 제작)

  • Polymeric hydrogel particles were fabricated to demonstrate the scale-up possibilities with the Particle Replication In Non-wetting Templates (PRINT) process. A permanently etched, specifically designed master was made on a silicon wafer using conventional photolithography, then reactive ion etching. The master and substrate were used repeatedly to make a large number of identical elastomeric perfluoropolyethers (PFPE) replica molds. The PFPE replica molds were used to fabricate and harvest individual, monodisperse micron-sized particles using the PRINT process. A water-soluble polymer adhesive was used as a sacrificial layer for harvesting particles. Particles were composed of biodegradable poly (ethylene glycol) diacrylate (PEG-diA), and aminoethylacrylate (AEM) and 2-acryloxyethyltrimethyl ammonium chloride (AETMAC) were added to them for improving the uptake of the cells. This study suggested PRINT used to produce the uniformed and shape specific biodegradable polymer is the effective technique for the non viral vector for the drug and the gene delivery.

  • Poly(ethylene glycol)(PEG) macromers terminated with diacrylate Iyoups and interpenetrating poly- mer networks(IPN) composed of poly(hydroxyethyl methacrylate)(PHEMA) or poly(hydroxyethyl methacrylate-co-hydronypropyl methacrylate-co- N-vinyl pyrrolidone ) [ P( HEM A-co- HPM A-co- NVP) ] and PEG macromer were synthesized with the aim of obtaining protein adsorption resistant soft contact lens. Polymerization of PEC macromer resulted in the formation of cross-linked gels due to the multifunctionality of macromer. Crosslinked P(HEMA) or P(HEMA-co-HPMA-co-WVP) chains were interpenetrated into the cross-linked three-dimensional networks of PEG. It was found that albumin adsorption onto the contact lens prepared by P(HEMA-co-HPMA-co-NVP) /PEG IPW decreases with an increase of molecular weight of PEG. Also, it was found that albumin adsorption onto the both contact lens decreases with an increase of concentration of PEC macromer in the IPN preparation. There are also more adequate in the bioinertnen for the contact lens by P(HEMA)/PEG IPN or P (HEMA-co-HPMA-co-NVP)/PEG IPN than that by P(HEMA) or P(HEMA-co-HPMA-co-NVP)

  • Poly (ethylene glycol)(PEG) - Poly (${\varepsilon}-caprolactone(CL)$) - Poly (D,L lactide(LA) (PCLA-PEG-PCLA) was synthesized by ring-opening polymerization to form temperature sensitive hydrogel triblock copolymer. The triblock copolymer was acrylated by acryloyl chloride. ${\beta}-amino$ ester was used as a pH sensitive moiety, in this study ${\beta}$- amino ester obtained from 1,4-butandiol diacrylate, and 4, 4' trimethylene dipiperidine, it have pKb around 6.6. pH/temperature sensitive penta-block copolymer (PAE-PCL-PEG-PCL-PAE) was synthesized by addition polymerization from acrylated triblock copolymer, 1,4-butandiol diacrylate, and 4, 4' trimethylene dipiperidine. Their physicochemical properties of triblock and penta-block copolymers were characterized by $^1H-NMR$ spectroscopy and gel permeation spectroscopy. Sol-gel phase transition behavior of PAE-PCL-PEG-PCL-PAE block copolymers were investigated by remains stable method. Aqueous media of the penta-block copolymer (at 20 wt%) changed from a sol phase at pH 6.4 and $10^{\circ}C$ to a gel phase at pH 7.4 and $37^{\circ}C$. The sol-gel transition properties of these block copolymers are influenced by the hydrophobic/hydrophilic balance of the copolymers, block length, hydrophobicity, stereo-regularity of the hydrophobic of the block copolymer, and the ionization of the pH function groups in the copolymer depended on the changing of environmental pH, respectively. The degradation and the stabilization at pH 7.4 and $37^{\circ}C$, and the stabilization at pH 6.4 and $10^{\circ}C,\;5^{\circ}C,\;0^{\circ}C$, of the gel were determined. The results of toxicity experiment show that the penta block copolymer can be used for injection drug delivery system. The sol?gel transition of this block copolymer also study by in vitro test ($200{\mu}l$ aqueous solution at 20wt% polymer was injected to mouse). Insulin loading and releasing by in vitro test was investigated, the results showed that insulin can loading easily into polymer matrix and release time is around 14-16days. The PAE-PCL-PEG-PCL-PAE can be used as biomaterial for drug, protein, gene loading and delivery.

  • Novel biodegradable elastic hydrogels, based on hydrophilic and hydrophobic polymer blocks, were synthesized via the radical crosslinking reaction of diacrylates of poly(ethylene glycol) (PEG) and poly(${\varepsilon}-caprolactone$) (PCL). PEG and PCL diols were diacrylated with acryloyl chloride in the presence of triethylamine, with the reaction confirmed by FT-IR and $^1H-NMR$ measurements. The diacrylate polymers were used as building-blocks for the syntheses of a series of hydro gels, with different block compositions, by simply varying the feed ratios and molecular weights of the block components. The swelling ratio of the hydrogels was controlled by the balance between the hydrophilic and hydrophobic polymer blocks. Usually, the swelling ratio increases with increasing PEG content and decreasing block length within the network structure. The hydrogels exhibited negative thermo-sensitive swelling behavior due to the coexistence of hydrophilic and hydrophobic polymer components in their network structure, and such thermo-responsive swelling/deswelling behavior could be repeated using a temperature cycle, without any significant change in the swelling ratio. In vitro degradation tests showed that degradation occurred over a 3 to 8 month period. Due to their biodegradability, biocompatibility, elasticity and functionality, these hydrogels could be utilized in various biomedical applications, such as tissue engineering and drug delivery systems.

Preparation of UV curable coating solution from multi functional acrylates and characterization of optical properties of coated layer on PET film (다관능 아크릴레이트계 자외선 경화형 코팅액의 제조 및 이를 이용한 PET 필름 도막의 광학 특성 연구)

  • Ultraviolet curable coating solution was prepared with poly(ethylene glycol) acrylate oligomer and various mono and multi-functional acrylate monomers. The optical properties of UV cured coating layer on PET film with acrylate coating solution containing metal oxides, such as fumed silica and alumina, were also investigated to reduce light reflection on films. Poly(ethylene glycol) diacrylate which has 575 of average molecular weight was used as oligomer acrylate, and pentaerythritol triacrylate and dipentaerythritolpenta-/hexa acrylate were used as multi-functional acrylate monomers. Also, butyl acrylate was used to improve the adhesion as well as to reduce glass transition temperature to give a better flexability. 1-hydroxy cyclohexyl phenyl ketone was used as photoinitiator. We found out the metal oxides in acrylate coating solution showed a homogeneous dispersion from energy dispersive spectroscopy data. Transmittance and light reflection of coated PET film was measured with UV/vis spectrometer and gloss meter, respectively. When 1.00 g of both metal oxides was added into coating solution, the transmittance and the glossiness were reduced from 90% to 30% and from 190 GU to 35 GU, respectively. However, adding up to 1.00 g of the metal oxide into coating solution did not affect on the hardness of coating layer and adhesion between coated layer and PET film. Conclusively, we can control transmittance and light reflection of coated film by adjusting the amounts of metal oxide in coating solution.

  • A disposable light exposure detector kit has been developed by UV curing of a hydrogel material. The devised light exposure detector kit consisted of light sensitive structures, bottom plate, character sheet and sticky back plate. A light exposure detector kit has a serial light sensitive structures that contain various light sensitive dyes such as rhodamine and fluorescein. The light sensitive structure composed of UV curable hydrogel polymer material as a supporing material and photosensitive dye in a certain concentration. The fabrication procedure of the ligh exposure detector kit is very simple and fast due to UV curing procedure of a photopolymerizable hydrogel material such as poly(ethylene glycol) methyl ether acrylate (PEGMEA) and poly(ethylene glycol) diacrylate (PEGDA) with a photosensitive dye. By the proposed fabrication method, various size and shape of a light exposure detector kit could be fabricated using a flexible elastomer mold. Due to a fast and inexpensive fabrication method, the light exposure detector kit could be use a single use for various industrial applications. According to light irradation, the light sensitive structure on a light exposure detector kit could be lose its color by decomposition of a photosensitive dye chemical in the structure. Thus the amount of the exposed light on a substrate could easily be recognised by changing color or transparency of the structure.

Effect of Crosslinking Agents on the Morphology of Polymer Particles Produced by One-Step Seeded Polymerization

  • One-step seeded polymerization was used to prepare $7{\sim}10{\mu}m$ of crosslinked monodisperse spheres with four crosslinking agents using $4.68{\mu}m$ poly(methyl methacrylate)(PMMA) seed particles in aqueous-alcoholic media in the absence of the swelling process. The crosslinking agents used were ethylene glycol dimethacrylate(EGDMA), allyl methacrylate(AMA), 1,6-hexanediol diacrylate(HDDA) and trimethylolpropane trimethacrylate(TMPTMA). The effects of the type and concentration of the crosslinking agents on the swelling, pore size, thermal property of the networks and morphology of the particles were studied. The chemical structures and concentrations of the crosslinking agents affected both the swelling ratio and the porosity of the networks. In addition, the chemistry of the reactive vinyl group and chain length of the crosslinking agents affected the stability of the monodisperse particles of the ultimate morphology.

  • A bio-encapsuled media was developed to apply on reducing odors produced from organic waste treatment process. The microorganism, candida tropicalis, was encapsulated in sponge media consisted of polyurethane material. Sodium alginate as a natural polymer which does not affect to hydrophilic microbes and PEGDA(poly ethylene glycol diacrylate) as a artificial polymer were used for the encapsuled media. The media was evaluated with TMEDA (N,N,N',N'-tetramethylethylenediamine, 0.02~0.1%) as a catalyst at different temperature 25 and $35^{\circ}C$. The best performance was achieved with 0.02% of TMEDA at $25^{\circ}C$. The microbes' activity in the media was examined by Live/Dead cell method.

Micro pH Sensor Using Patterned Hydrogel with pH Indicators

  • This paper presents a study into pH Indicator-Embedded hydrogel micro-particles which are encoded various shapes according to the captured indicator. We incorporate various pH indicators into a photo-curable hydrogel, PEG-DA(Poly(ethylene glycol) diacrylate). Using the latest fluidic lithography techniques, we can easily fabricate variously patterned hydrogel particles based on in-situ photopolymerization of the PEG-DA in a micro-fluidic channel. The shape of the particle is related to the pH indicator inside the particle. We demonstrate that the micro pH sensors change their colors according to the pH levels. The micro pH sensors have various characteristics that are related to the curing time, particle size, etc. By changing these conditions, we can adjust the long term stability and reaction time of the hybrid micro pH sensors.

  • Solid polymer electrolyte films are prepared by ultraviolet radiation in the mixtures of an ionic liquid salt (1-ethyl-3-methylimidazolium tetrafluoroborate, $EMIBF_4$) and solvent (acetonitrile (ACN) or propylene carbonate(PC)), and an oligomer (poly(ethylene glycol)diacrylate, PEGDA, 45-60 wt.%). Electrochemical properties of activated carbon supercapacitors adopting the solid polymer electrolyte films as a separator are also examined by cyclic voltammetry and impedance measurement techniques. As a result, the supercapacitor adopting the PEGDA as much as 45 wt.% exhibits a superior capacitance of $46Fg^{-1}$ at $20mVs^{-1}$. It seems that this is due to fast kinetics of ion conduction by sufficient film flexibility, which can be allowed by comparatively weak ultraviolet curing of small anount of the PEGDA.
