Platycodin D Induced NF-$textsc{k}$B Activation and Apoptosis in Immortalized Keratinocytes

  • In this study, we investigated the molecular pathways targeted by platycodin D, which could involve apoptosis in immortalized human keratinocytes (HaCaT). We demonstrated that platycodin D-mediated apoptosis of HaCaT cells exhibited representative features, including DNA fragmentation, caspase-3, caspase-8 activation, and upregulation of Fas and FasL expression, but not p53 activation. To investigate the events involved in activation-induced FasL upregulation, we have examined mRNA accumulation, protein expression, and NF-$\kappa$B activity to elucidate transcription level in the HaCaT cell line treated with platycodin D. 저장방법과 기간이 도라지 뿌리의 경도와 사포닌함량 미치는 영향 (Effects of Storage Methods and Periods on Root Hardness and Content of Saponin in Platycodon grandiflorum Radix)

  • 도라지를 수확 후 진공포장, 밀폐포장, 질소가스포장 후 저온과 상온에 100일 동안 저장하면서 저장 50일과 100일에 도라지 경도와 사포닌 함량을 조사한 결과를 요약하면 아래와 같다. 1. 저장온도에 따른 도라지의 경도는 저온저장이 상온저장보다 높게 나타났으며, 저장기간이 길어질수록 도라지의 경도는 낮아졌다. 2. 저장방법에 따른 도라지의 경도는 저장 50일차 지퍼팩을 이용한 밀봉에서 1,765 g으로 가장 높게 나타났으며, 저장온도와 저장방법과의 상호작용에서는 고도의 유의성($P{\leq}0.01$)이 인정되었다. 3. 저장온도에 따른 platycodin D3, polygalacin D, 그리고 deapioplatycodin D의 함량조성이 다르게 나타났다. 4. 저장기간에 따른 platycodin D3와 polygalacin D의 함량은 저장 50일 이후 감소하는 경향을 보였지만, deapioplatycodin D는 저장기간이 길어질수록 증가하는 경향을 보였다.

Compositional Analysis of Major Saponins and Anti-inflammatory Activitiy of Steam-Processed Platycodi Radix under Pressure

  • Platycosides are the saponins in Platycodi Radix and they have several beneficial effects such as antiinflammatory and anti-obesity activities. This study was designed to determine the changes in the saponin composition in Platycodi Radix (platycosides) after being processed under steam and pressure and to investigate the anti-inflammatory effects of their extracts. The change of the platycoside compositions was investigated after 1, 2, 3, 6 and 9h heat processing of Platycodi Radices by using HPLC coupled with an evaporative light scattering detection (ELSD) system. After heat treatment ($125^{\circ}C$, 1, 2, 3, 6 and 9 h), the contents of several platycosides such as platycoside E, platycodin $D_3$, platycodin D, polygalacin D, and platycodin A decreased as the processing time was longer. While the total contents of the saponins decreased, the contents of deapi-forms of deapiplatycoside E, deapi-platycodin $D_3$, and deapi-platycodin D increased relatively. These results indicate that the linkage between apiose and xylose located at C-28 is labile to heat and pressure. The LPS-induced iNOS inhibitory activities of the samples treated for 1 and 2 hours were enhanced and after then, the activities were reduced. These results suggested that heat treatment of the samples affect the content of the total saponins and the saponin content may be the important criteria representing the anti-inflammatory activity.

더덕, 길경, 감초 복합추출물에 대한 지표성분 동시분석 및 함량평가 (Simultaneous Quantitative Analysis of Natural Complex Extract of Codonopsis lanceolata (Siebold & Zucc.) Trautv., Platycodon grandiflorum A. De Candolle, Glycyrrhiza uralensis Fischer)

  • Fine dust has emerged as seriously hazardrous particles, which can cause respiratory and cardiovascular diseases. Thus, we have developed a natural complex extract consisted of Codonopsis lanceolata (Siebold & Zucc.) Trautv., Platycodon grandiflorum A. De Candolle, and Glycyrrhiza uralensis Fischer in the ratio of 3 : 5 : 2, which have a beneficial effect on respiratory system. In this study, simultaneous quantitative analysis of marker compounds, lobetyolin, platycodin D, and glycyrrhizic acid in the natural complex extract was developed and validated with high performance liquid chromatography-photodiode array detector. The marker compounds were shown in a large linearity with a correlation coefficient (R2) of 1.000. The limit of detection (LOD) of lobetyolin, platycodin D and glycyrrhizic acid were 5.40 ㎍/mL, 3.94 ㎍/mL and 5.86 ㎍/mL, respectively. The limit of quantification (LOQ) of lobetyolin, platycodin D and glycyrrhizic acid were 16.36 ㎍/mL, 11.94 ㎍/mL and 17.75 ㎍/mL, respectively. The content analysis revealed that the amounts of lobetyolin, platycodin D, and glycyrrhizic acid were 0.039±0.013 mg/g, 0.337±0.048 mg/g, and 1.171±0.003 mg/g, respectively, in the natural complex extract. Therefore, these results could be used as basic data for the standardization and quality control of the natural complex extract.

건조 온도에 따른 도라지의 사포닌과 당 함량 변화 (Effects of Drying Temperature on the Saponin and Free Sugar Contents of Platycodon grandiflorum Radix)

  • 도라지 건조 온도에 대한 기본 정보를 제공하고자 건조 온도에 따른 환원당 그리고 사포닌 함량을 조사한 결과를 요약하면 아래와 같다. Sucrose 함량은 $85^{\circ}C$에서 3,825 mg% $45^{\circ}C$에서 1,226 mg%로 건조 온도가 낮을수록 감소하였지만, glucose함량은 건조 온도가 높을수록 감소하였다. 본 시험에서 분석한 도라지 4종의 사포닌 중에 platycodin D 함량은 $85^{\circ}C$에서 197.5 mg%, $65^{\circ}C$에서는 334.7 mg%로 $45^{\circ}C$에서 956.0 mg%로 가장 높게 나타나 건조 온도가 낮을수록 증가하였으며, 고온(85) 건조에서 사포닌 중 polygalacin D의 함량이 가장 많았다. Sucrose와 사포닌 함량 간의 상관계수는 각각$-0.987^{**}$로 고도의 음의 상관을 보였다(p<0.01). 건조 온도 $45^{\circ}C$가 다른 건조 온도에 비해 색이 밝게 나타남을 확인할 수 있었다. 이상의 결과를 통하여 도라지의 건조 온도가 도라지의 사포닌 함량과 당 함량에 영향을 미치는 것을 확인할 수 있었으며, 도라지의 이용 용도에 따라 건조 온도를 달리 할 필요성이 있을 것으로 사료된다.

HPLC를 이용한 길경(桔梗) 사포닌 분석법(分析法) (HPLC Analysis of Saponins in Platycodi Radix)

  • A rapid and practical HPLC assay was developed for quantitative analysis of saponins in Platycodi Radix. Seven saponin components in Platycodi Radix, i.e., deapioplatycoside E, platycoside E, deapioplatycodin D$_3$, platycodin D$_3$, polygalacin D$_2$, platycodin D$_2$ and platycodin D were successfully resolved on C18 column and detected by ELSD. Standard curves were linear over the concentration range 1 ${\sim}$2,000 ${\mu}$g/ml (r$^2$>0.992). Intra- and inter-day coefficients for variation of seven saponin components were < 10% and limit of quantification of them were around 0.7${\sim}$1.5 ${\mu}$g/ml, respectively. Using this method, contents of seven saponins in various plant materials under different cultivating conditions were estimated.

이부프로펜에 의해 유발된 급성 위궤양에 있어 Platycodin D의 보호효과 (Protective Effect of Platycodin D in the Acute Gastric Ulcer Induced by Ibuprofen in Rats)

  • 급성 위궤양은 위점막에서 세포증식과 세포사멸의 불균형으로 발병되어진다. 현재 Platycodin D (PD)는 항산화 및 항염증 등의 다양한 약리효능을 가진다고 보고되고 있다. 본 실험은 ibuprofen에 의해 유발된 급성 위궤양이 전처치한 PD에 의해 위궤양 보호효과를 가지는 가를 알아보기 위해 실시하였다. PD의 효능은 위점막에서의 COX-2의 발현과 더불어 위점막상피세포의 증생과 세포사멸정도에 의해서 평가하였다. 실험군은 정상대조군, ibuprofen 유발 위궤양군, 2.5 mg/kg PD 전처치군, 5 mg/kg PD 전처치군으로 분류하였다. 급성위궤양은 200 mg/kg의 ibuprofen을 하루에 3번 8시간 간격으로 경구 투여하여 유발하였다. PD는 5일간 경구로 하루에 한 번씩 전처치하였다. PD의 전처치가 ibuprofen에 의해 유발된 위궤양 병변을 유의적으로 감소시켰으며 과도한 위점액 분비로 인한 점액질의 소실을 억제하였다. 또한 PD 전처치가 위점막의 상피세포증식층에서 Ki-67 양성세포의 감소 및 세포사멸을 억제하였다. 추가적으로 PD의 전처치가 위궤양에 의해 증가된 COX-2 발현을 감소시켰다. 이상의 연구결과는 PD의 전처치가 ibuprofen에 의해 유발된 위점막손상에 있어 COX-2의 발현조절을 통하여 위점막세포의 증식과 사멸에 관여할 것으로 보여진다.

길경(桔梗)의 추출조건에 따른 사포닌함량의 변화 (제2보) (Variation of Saponin Content in the Decoctions of Platycodi Radix (II))

  • The capability of the solvents for extracting the bioactive saponins from the roots of Platycodon grandiflorum (Campanulaceae) was investigated to obtain an ideal extract which contained bioactive saponins with high quality and high quantity. The content of eight representative saponins in extracts, such as deapioplatycoside E, platycoside E, platyconic acid A and platycodin D, platycodin $D_3$, platycodin $D_2$, polygalacin $D_2$, polygalacin D were analyzed simultaneousely by the modified HPLC analytical method. The validation test of the modified qualitative and quantitative analytical method employing the ELSD equipped HPLC for eight representative saponins in the roots extract of P. grandiflorum showed a good linearity, precision and accuracy. the correlation coefficient ($r^2$) values of the calbration curves for each saponins were observed to be over 0.9990. LOD and LOQ of each saponin was calculated as $0.10{\mu}g{\sim}0.40{\mu}g$ (LOD) and $0.40{\mu}g{\sim}0.80{\mu}g$ (LOQ), respectively. Recovery rates of each saponin were also calculated as over 98%, respectively. With exception of two saponins, platyconic acid A and platycodin D, The content of eight saponins in extracts was decreased proportionally to the increment of the water ratio of solvent for extraction. Consequently, as aquous alcohol was used as a solvent for extracting the saponin components from powdered roots of P. grandiflorum, the water content in the aquous alcohol was seemed to be a critical factor for extracting efficacy. The 60-80% ratio of alcohol in the aquous alcohol were deduced to be suitable and recommendable for the preparation of roots extract of P. grandiflorum which contained saponins with high quality and high quantity.

In Vitro Inhibitory Effect of Triterpenoidal Saponins from Platycodi Radix on Pancreatic Lipase

  • In the process of investigating anti-obesity effect of Platycodi Radix, we found that aqueous extract of Platycodi Radix might inhibit intestinal absorption of dietary fat by inhibiting pancreatic lipase (PL) activity. In order to clarify the anti-obesity mechanism of Platycodi Radix, activity-guided isolation was performed to find active components. The total saponin fraction of Platycodi Radix appeared to have a potent inhibitory activity against the hydrolysis of triolein emulsified with phosphatidycholine by pancreatic lipase in vitro. Based on these results, further purification of active components yielded 10 known triterpenoidal saponins, among these compounds, platycodin A, C, D, and deapioplatycodin D exhibited significant inhibitory effects on PL at the concentration of $500\;{\mu}g/mL$ with 3.3, 5.2, 34.8, and $11.67\%$ pancreatic lipase activity vs control, respectively. Platycodin D was found to inhibit the PL activity in a dose-dependent manner. Therefore, the anti-obesity effect of Platycodi Radix might be due to the inhibition of pancreatic lipase by its saponins.

Platycodin D Induces Apoptosis, and Inhibits Adhesion, Migration and Invasion in HepG2 Hepatocellular Carcinoma Cells

  • Background: Platycodin D (PD), a triterpenoid saponin isolated from the Chinese medicinal herb Platycodonis radix, possesses anti-cancer effects in several cancer cell lines. The aim of this study was to evaluate its anticancer activities in hepatocellular carcinoma cells. Materials and Methods: MTT and colony formation assays were performed to evaluate cell proliferation, along with flow cytometry and Western blotting for apoptosis. Cell adhesion was tested by observing cellular morphology under a microscope, while the transwell assay was employed to investigate the cell migration and invasion. Results: PD concentration-dependently inhibited cell proliferation in both HepG2 and Hep3B cells, and significantly suppressed colony formation and induced apoptosis in HepG2 cells. The protein levels of cleaved poly ADP-ribose polymerase (PARP) and Bax were up-regulated while that of survivin was down-regulated after treatment with PD. Moreover, PD not only obviously suppressed the adhesion of HepG2 cells to Matrigel, but also remarkably depressed their migration and invasion induced by 12-O-tetradecanoylphorbol 13-acetate (TPA). Conclusions: PD presents anti-cancer potential in hepatocellular carcinoma cells via inducing apoptosis, and inhibiting cell adhesion, migration and invasion, indicating promising features as a lead compound for anti-cancer agent development.
