Cytological changes of cowpea root at the early stage of root nodule formation (within 5 days after inoculation) were viewed by light and electron microscopy. The root region affected by the rhizobial infection, which was composed of a redial array of cortical cells, had prominent cell divisions, mostly anticlinal in the inner cortical cells and in addition oblique and periclinal in the outer cells. An infected root hair cell (or root hair-producing epidermal cell) had numerous infection threads and degenerated cytoplasm. Module meristem was formed adjacent to the infected root hair cell, and characterized by dense cytoplasm, prominent nucleus, numerous small vacuoles, and increased plastids, containing infection threads as well. Bacterial cells were dividing inside the infection thread, the wall materials of which appeared to be dissolved ad accumulated in small vacuoles. inner cortical cells contiguous to the nodule meristem appeared to be actively dividing and dedifferentiating; however, they were not infected by the rhizobia. These structural characteristics are similar to those in the Bradyrhizobium-soybean association previously reported, and may reflect the similar cytological process in cowpea in the early nodule formation.

DNA metabarcoding is currently used for large-scale taxonomic identification to understand the community composition in various marine ecosystems. However, before being widely used in this emerging field, this experimental and analytic approach still has several technical challenges to overcome, such as polymerase chain reaction (PCR) bias, and lack of well-established metabarcoding markers, a task which is difficult but not impossible to achieve. In this study, we present an adapted PCR-free small-organelles enriched metagenomics (SoEM) method for marine biodiversity assessment. To avoid PCR bias and random artefacts, we extracted target DNA sequences without PCR amplification from marine environmental samples enriched with small organelles including mitochondria and plastids because their genome sequences provide a valuable source of molecular markers for phylogenetic analysis. To experimentally enrich small organelles, we performed subcellular fractionation using modified differential centrifugation for marine environmental DNA samples. To validate our SoEM method, two marine environmental samples from the coastal waters were tested the taxonomic capturing capacity against that of traditional DNA metabarcoding method. Results showed that, regardless of taxonomic levels, at least 3-fold greater numbers of taxa were identified in our SoEM method, compared to those identified by the conventional multi-locus DNA metabarcoding method. The SoEM method is thus effective and accurate for identifying taxonomic diversity and presents a useful alternative approach for evaluating biodiversity in the marine environment.
Transgenic rice plants expressing a Bacillus subtilis protoporphyrinogen oxidase (Protox), the last shared enzyme of the porphyrin pathway in the expressed cytoplasm or the plastids, were compared with non-trangenic rice plants in their growth characteristics such as tiller number, plant height, biomass, and yield. Transgenic rice plants of $\textrm{T}_3$ generation had 8 to 15 % and 25 to 43% increases in tiller number compared to non-transgenic rice plants at 4 and 8 weeks after transplanting(WAT); similar values were observed for $\textrm{T}_4$ generation at 4 and 8 WAT. However, the plant height in both $\textrm{T}_3$ and $\textrm{T}_4$ generations was similar between transgenic rice plants and non-transgenic rice plants at 4 and 8 WAT. Transgenic rice plants had 13 to 32% increase in above-ground biomass and 9 to 28% increase in grain yield compared to non-transgenic rice plants, demonstrating that biomass and yield correlate with each other. The increased grain yield of the transgenic rice plants was closely associated with the increased panicle number per plant. The percent of filled grain, thousand grains and spikelet number per panicle were similar between transgenic and non-transgenic rice plants. Generally, the growth and yield of transgenic generations ($\textrm{T}_2$, $\textrm{T}_3$, and $\textrm{T}_4$) and gene expressing sites (cytoplasm-expressed and plastid-targeted transgenic rice plants) were similar, although they slightly varied with generations as well as with gene expressing sites. The transgenic rice plants had promotive effects, indicating that regulation of the porphyrin pathway by expression of B. subtilis Protox in rice influences plant growth and yield.
Eukaryotic cells consist of a complex network of thousands of proteins present in different organelles where organelle-specific cellular processes occur. Identification of the subcellular localization of a protein is important for understanding its potential biochemical functions. In the post-genomic era, localization of unknown proteins is achieved using multiple tools including a fluorescent-tagged protein approach. Several fluorescent-tagged protein organelle markers have been introduced into dicot plants, but its use is still limited in monocot plants. Here, we generated a set of multicolored organelle markers (fluorescent-tagged proteins) based on well-established targeting sequences. We used a series of pGWBs binary vectors to ameliorate localization and co-localization experiments using monocot plants. We constructed different fluorescent-tagged markers to visualize rice cell organelles, i.e., nucleus, plastids, mitochondria, peroxisomes, golgi body, endoplasmic reticulum, plasma membrane, and tonoplast, with four different fluorescent proteins (FPs) (G3GFP, mRFP, YFP, and CFP). Visualization of FP-tagged markers in their respective compartments has been reported for dicot and monocot plants. The comparative localization of the nucleus marker with a nucleus localizing sequence, and the similar, characteristic morphology of mCherry-tagged Arabidopsis organelle markers and our generated organelle markers in onion cells, provide further evidence for the correct subcellular localization of the Oryza sativa (rice) organelle marker. The set of eight different rice organelle markers with four different FPs provides a valuable resource for determining the subcellular localization of newly identified proteins, conducting co-localization assays, and generating stable transgenic localization in monocot plants.
Initial subcellular responses in susceptible (PI 274420) and resistant (cv. Hartwig) soybeans infected with the soybean cyst nematode (SCN) were examined 2 and 4 days after inoculation (DAI). Subcellular features common to both soybeans at 2 DAI included hypertrophied initial syncytial cells (ISCs) and syncytium-component cells (SCs) with a dense cytoplasm containing proliferated rough and smooth endoplasmic reticulum (RER and SER), a hypertrophied nucleolus, and reduced vacuoles, suggesting that the nematode-infected cells were dedifferentiated. In the resistant soybean, a striking initial subcellular difference from the susceptible soybean was the dilation of the RER, indicating ER dysfunction and leading to cell death. This disturbed nematode feeding, as evidenced by disrupted feeding tubes. In PI 274420, the ISC cytoplasm was depleted, with the exception of ER membranes, at 4 DAI, while the SC cytoplasm was dense with proliferation of starch-containing plastids around multiple nuclei that might be derived from the congregation of nuclei in the neighboring SCs and in part by nuclear division without cytokinesis. In cv. Hartwig, syncytia were necrotized with secondary cell wall thickening outside the plasma membrane and an extremely dense cytoplasm containing a nucleus with an electron-lucent nucleolus, accompanied by the proliferation of closely stacked parallel RER and ribosomes. These results suggest that syncytia develop continuously in PI 274420 to produce and store nutritional substances in SCs, providing for the nematode through ISC until maturation, but in cv. Hartwig, syncytia degenerate early due to excessive metabolism, blocking nematode feeding and cytoplasmic connections with adjacent intact cells.
우기자혹응애 (Eriophyes kuko KISHIDA)의 기생에 의하여 구기자 나무 (Lycium Chinens MILL) 잎에 형성되는 흑 (mite gall) 의 행태적 변화를 광학현미경 및 전자현미경으로 관찰한 결과는 다음과 같다. 1. 광현적 관찰 : 구기자혹응애가 구기자나무의 어린잎의 뒷면 표피세포에 침입되면 먼지 표피세포가 이상비대를 하면서 혹이 형성되기 시작한다. 혹이 성장함에 따라 붕상조직과 해면상조직의 세포도 이상비대를 하게되고, 점차적으로 각 조직의 세포는 이상비대로 인한 불규칙한 형태적 변화를 일으켜 조직의 구별이 힘들게 된다. 이상비대한 혹세포의 핵은 건전세포의 핵보다 크며, 혹이 성장함에 따라서 핵의 크기도 커진다. 2. 전현적 관찰 : 혹이 성장함에 따라 혹세포의 mitochondria, golgi-apparatus. plastid 등은 퇴 화 및 붕괴되고, 특히 큰 흑의 이상비대한 혹세포에는 성상체 (star body) 와 환상 (ring form) 의 구조등이 나타난다.

Water temperature affects plankton survival and growth. The dinoflagellate Shimiella gracilenta survives using the plastids of ingested prey, indicating kleptoplastidy. However, studies on the effects of water temperature on kleptoplastidic dinoflagellates are lacking. We explored the growth and ingestion rates of S. gracilenta as a function of water temperature. Furthermore, using data on its spatiotemporal distribution in Korean coastal waters during 2015-2018, we predicted its distribution under elevated temperature conditions of +2, +4, and +6℃. Growth rates of S. gracilenta with and without Teleaulax amphioxeia prey as well as ingestion rates were significantly affected by water temperature. Growth rates of S. gracilenta with and without prey were positive or zero at 5-25℃ but were negative at ≥30℃. The maximum growth rate of S. gracilenta with T. amphioxeia was 0.85 d-1, achieved at 25℃, and 0.21 d-1 at 20℃ without prey. The ingestion rate of S. gracilenta on T. amphioxeia at 25℃ (0.05 ng C predator-1 d-1) was greater than that at 20℃ (0.04 ng C predator-1 d-1). Thus, feeding may shift the optimal temperature for the maximum growth rate of S. gracilenta from 20 to 25℃. In spring and winter, the distributions of S. gracilenta under elevated temperature conditions were predicted not to differ from those during 2015-2018. However, S. gracilenta was predicted not to survive at some additional stations under elevated temperature conditions of +2, +4, and +6℃ in summer or under elevated temperature conditions of +6℃ in autumn. Therefore, global warming may affect the distribution of S. gracilenta.
과피가 녹색이고 배의 길이가 $250{\mu}$인 미성숙 과정을 거쳐 발아시까지 배 및 배유세포의 미세구조 변화를 관찰한 결과를 요약하면 다음과 같다. 1. 배의 길이가 $250{\mu}$인 미성숙 종자의 배세포는 세포질이 비교적 충실하나 mitochondria의 Cristae 및 plastid는 미분화상태이고 가끔 dictyosome이 관찰되었다. 액포안에는 전자밀도가 높은 globoid 를 함유하는 것도 있었다. 소포체는 대부분 한면소포체이고 외형질에는 spherosome을 많이 함유하고 있었다. 배유세포는 spherosome과 이들로 둘러싸인 전자밀도가 높고 균질성인 protein body로 충만되었고 이들로 인하여 세포기관은 관찰되지 않았다. 2. 과피가 홍색인 종자의 배세포는 cristae가 발달한 mitochondria가 관찰되었으며, 전자밀도가 높은 globoid가 증가하였고 액포는 확장되었다. 배유세포에는 spherosome이 증가하고 있었으며 protein body는 확장되어. 그 내부에는 전자밀도가 높은 globoidal crystal이 분산되어 있었다. 3. 종피가 열개한 종자의 배축의 전형성층과 유아세포에서는 Golgi vacuole과 vesicle이 발달되었으며 mitochondria의 cristae는 매우 분화되었다. spherosome은 매우 많이 나타났고 유근세포, 배축의 주변세포 및 자엽세포의 액포는 매우 분화되었다. 배유세포는 spherosome으로 충만되어 있고 그 내질에는 전자밀도가 높고 작은 입자를 지니고 있었다. Protein body는 단일막으로 둘러싸여 있음이 확인되었다. 세포질내에는 구형체와 spherosome 주변의 세포질에 acid phosphatase의 활성을 나타냈다. 4. 파종시 자엽의 외면부 세포는 spherosome이 급격히 증가하였고 protein body도 관찰되었다. 세포기관들은 분화되어 있었고 전분을 함유하는 색소체가 출현하였다. 5. 파종시킨 후 발아할 때까지 자엽의 외측 $2{\sim}3$열세포, 배축의 주변세포, 유근부 세포에서는 spherosome이 증가하였고 전분립을 함유하는 plastid 도 증가하였다. 배를 구성하는 세포의 내형질의 액포부근에는 mitochondria와 microbody 가 혼재하고 있음이 관찰되었다. 발아시기가 가까워 질수록 배와 연접하는 배유세포에는 액포가 매우 분화되고 spherosome은 감소되는 반면 액포내에는 무정형의 전자밀도가 높은 물질이 증가하였다. 그밖의 배유세포에는 spherosome이 감소됨에 따라 mitochondria는 증가되고 전자밀도가 높은 소구체가 형성되어 증가되었다. Protein body의 외질은 감소되고 전자밀도가 낮은 부위는 확장되며 이는 서로 융합하여 외연부는 없어지고 기질속에 소입자들이 형성되었다. 6. 발아기의 배유세포안에는 spherosome이 감소되고, protein body는 서로 융합되어 한 세포내에 $2{\sim}3$개로 충만되어 있었다.
일부 수생식물은 휴면구조로 엽상체가 변형된 잠아를 형성하여 적응 생존한다. 부유부엽성 식물인 개구리밥은 가을에 형성된 잠아로 동절기 물속에 가라앉아 휴면상태로 겨울을 난다. 이들 잠아는 엽상체의 특정 부위에서 기원하나 모엽상체와 달리 통기조직의 발달이 미흡하고 조직의 밀도가 높아 수중 침강에 유리한 구조로 분화된다. 잠아조직 내에는 일반적으로 기온상승 시 생장재개에 바로 이용할 수 있는 영양분이 저장되어 있다. 이러한 특성을 지닌 개구리밥 잠아의 형성 및 발아에 대해서는 생리적, 생화학적으로 다양한 연구가 진행되고 있으나 잠아조직 세포의 미세구조 연구는 미흡한 상태이다. 이 연구에서는 엽상체와 잠아 엽육조직의 분화, 통기조직의 발달, 녹말의 분포, 세포수준에서의 미세구조 발달양상을 비교 연구하였다. 엽육조직이 비교적 분화되어 있는 모엽상체는 통기조직이 잘 발달되어 엽상체 용적의 약 $37{\sim}45%$를 차지하였다. 반면, 잠아는 엽육조직의 분화 없이 조직 내 통기조직의 발달이 미흡하여 전체 용적의 약 $9{\sim}15%$를 차지하였다. 모엽상체 엽육세포와 달리 잠아 세포내 세포질은 매우 치밀하며 엽록체 기질에는 녹말이 다량 축적되어 있다. 엽록체, 세포벽, 액포, 세포막은 모엽상체 세포내 특성과 다른 양상으로 분화하였다. 이와 같이 잠아는 통기조직의 미흡한 발달, 세포질의 높은 밀도, 녹말의 다량 축적 등으로 조직의 밀도를 높게 하여 겨울철 휴면상태에 잘 적응된 구조로 발달시켜 축소된 개구리밥 식물체 생활사에 중요한 역할을 수행하는 독특한 구조로 분화하였다. 매우 빠른 생장으로 수면에 넓게 분포하는 개구리밥은 수생환경에 필요한 양분 및 빛을 차단하여 수생의 생태에 나쁜 영향을 미치기도 한다. 그러나 잠아형성 촉진 조건을 가해 주면 식물체는 쉽게 물속에 가라앉아 휴면상태를 유지할 수 있는 것으로 알려져 있다. 다른 식물 종들은 생장 및 형성 조건을 조절하기 어렵지만 개구리밥 식물체의 잠아는 이러한 구조적 특징으로 형성이 쉽게 조절될 수 있어 수생환경 개선에 유망한 후보가 되고 있다. 향후 본 연구의 모엽상체와 잠아의 생장 및 구조 특성을 이용하여 유사 수생식물 잠아의 생장특성에 적용하면 수생환경정화 등의 영역에서 유용하게 활용될 수 있는 휴면구조가 될 것이다.
녹조류는 cellulose가 세포벽의 주요 구성성분이며 그 양이 다른 해조류에 비해 월등하고, 세포벽에 lignin이 없어 lignin 제거공정을 거치지 않고 cellulase를 사용하여 쉽게 당화시킬 수 있을 뿐만 아니라 저장산물인 전분도 당 성분으로 사용할 수 있다. 이에 바이오에너지 생산을 위한 좋은 바이오매스가 될 것으로 기대되는 녹조류인 구멍갈파래(Ulva pertusa Kjellman)를 사용하여 마이크로웨이브 장치를 통한 열수 전처리 효과가 cellulase를 사용한 가수분해 효율에 미치는 영향을 알아보았다. 열수의 온도에 따른 전처리 효과를 확인하기 위해 $100{\sim}150^{\circ}C$에서 전처리를 수행하였으며, $140{\sim}150^{\circ}C$에서 가장 높은 전처리 효과를 얻었다. 또한 전처리 최적조건인 마이크로웨이브 장치의 출력 50W와 온도$150^{\circ}C$에서 열수 전처리한 구멍갈파래에 포함되어 있는 탄수화물의 효소적 가수분해 효율을 높이기 위해 cellulase 외에 ${\alpha}$-amylase와 ${\beta}$-glucosidase를 함께 사용하여, 효소 혼합의 효과를 확인하였다. 전처리한 구멍갈파래 시료에 cellulase와 ${\alpha}$-amylase 그리고 ${\beta}$-glucosidase 활성을 가지는 Novozyme 188을 사용하여 가수분해하였을 경우 전처리하지 않은 구멍갈파래 시료와 비교하면 초기 가수분해속도가 6배 이상 월등히 높았고, 3시간 만에 구멍갈파래에 포함되어 있는 탄수화물의 96 wt%에 해당할 정도의 환원당이 생성되었으며, 이 양은 전처리하지 않은 구멍갈파래 시료를 24시간동안 효소적 가수분해해야 얻을 수 있는 환원당의 양으로 열수 전처리한 효과가 월등함을 보여주었을 뿐만 아니라 대부분의 탄수화물이 전환되는 최대 당화 효율을 얻을 수 있음을 보여준다.

④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
