Antimicrobial Drug Resistance and R Plasmids of Serratia marcescens (Serratia marcescens의 항균제 내성 및 R plasmid)

  • Forty clinical isolates of Serratia marcescens were tested for their susceptibility to 19 antimicrobial drugs and studied on the molecular characteristics of R plasmids. Cefotaxime (Ct) was the most effective drug and only 2 (5%) strains were resistant to this drug. Thirteen to 18% of strains were resistant to cefoperazone (Cz), amikacin (Ak). and trimethoprim (Tp), and 28 to 40% were resistant to piperacillin (Pi), nalidixic acid (Na), gentamicin (Gm), and cefoxitin (Cx). A majority of strains were resistant to carbenicillin (Cb), tobramycin (Tp), kanamycin (Km), and cefamandole (Cd), and all to cephalothin. One half of the isolates were resistant to 10 or more drugs. $MIC_{90}$ of Pi to Gm-resistant strains (Gm') were 8 times higher than that to Gm-susceptible strains (Gm'), but $MIC_{90}$ of Ak, Cx, Ct, and Cz were almost the same between both Gm' and Gm' strains. Nine (23.7%) strains among 38 of multiply drug-resistant S. marcescens transferred conjugally their partial patterns of resistance to E. coli or Klebsiella strains, and two S. marcescens strains producing bacteriocin transferred their resistance to Klebsiella only, but not to E. coli. The plasmid profiles of S. marcescens were studied by the methods of SDS lysis and agarose gel electrophoresis. Twenty-four (60%) strains carried one to four plasmids of 1.4. to 144 Mdal, and conjugative R plasmids of 49 to 127 Mdal were noted in transconjugants. MIC levels of drugs in transconjugants were variable by the R plasmids and recipient strains.

  • Staphylococcus aureus KH13 and Staphylococcus aureus KH28 were resistant to chloramphenicol, ampicillin, clindamycin, erythromycin, gentamicin, kanamycin, streptomycin, tobramycin, and norfloxacin. A plasmid (pKH13) and two plasmids (pKH14, pKH15) were isolated from Staphylococcus aureus KH13 and Staphylococcus aureus KH28, respectively and complete nucleotide sequences of three plasmids were determined. It was found that pKH13 and pKH15 mediated chloramphenicol resistance and pKH14 was a cryptic plasmid.

  • Promoters which are useful for constructing expression vectors for lactic acid bacteria were obtained from the chromosomal DNA of Lactococcus lactis ssp. lactis MG1363. pBV5030, a promoter-selection vector, replicates in L. lactis and Escherichia coli and carries a promoterless chloramphenicol acetyltransferase gene (cat-86). After examining E. coli transformants which grew on LB media containing chloramphenicol (Cm, 20$\mu\textrm{g}$/mL) , many MG1363 derived DNA fragments which encompass promoter sequences were identified. Some recombinant E. coli cells can grow at the Cm concentration of 1,000$\mu\textrm{g}$/mL. When plasmids from those highly resistant E. coli cells were purified and introduced into L. lactis ssp. lactis MG1614 cells by electroporation, lactococcal transformants showing Cm resistance were obtained. So far, five plasmids with different promoter inserts were introduced into L. lactis MGl614 cells. The maximum level of Cm resistance in L. lactis MG1614 transformants was quite low (20$\mu\textrm{g}$/mL) when compared with that observed in recombinant E. coli cells harboring the same plasmids.

Identification of Responsible Region for the Polymerization of Plasmid pEC-3 (Plamid pEC-3의 중합에 필요한 부위의 동정)

  • In order to find specific acting site of Rec A protein in plasmic polymerization in E. coli, we randomly deleted various part of pEC-3 (a derivative of pBR322) with SI nuclease treatment. Self-ligated plasmids were introduced into E. coli WA802(Rec $A^+$). A number of colonies were analyzed if they contained monomeric or polymeric plasmids by gel electrophoresis. The plasmid (pEC-43), which was deleted the region of tetracycline gene, revealed only monomeric form in Rec $A^+$ E. coli. When two plasmids, pEC-3 and pEC-43, were co-transformed in the same E. coli, the original pEC-3 showed polymerization but pEC-43 revealed monomeric form only. These results suggest that Rec A protein requires the specific site for polymerization.

Relationships between in-vitro virulence-associated characteristics, plasmid-bearing and production of Outer Membrane Protein(OMP) of Yersinia enterocolitica isolated from pigs (Yersinia enterocolitica의 시험관내 병원성 성상, plasmid 보유 및 외막 단백질(OMP) 생산간의 관계)

  • Two hundred and eighty nine strains of Yersinia enterocolitica isolated from healthy pigs were tested for the presence of 40~50 Megadalton virulence-associated plasmids and plasmidmediated in vitro virulence-associated properties, i.e., congo red uptake, calcium dependency, autoagglutination, CRMOX reaction, crystal violet binding and pyrazinamidase reaction. The correlationships between in vitro virulence-associated properties and the presence of 220 Kdalton outer membrane protein(OMP) were examined in strains with or without virulence-associated plasmids. The correlationships between the presence of plasmids on the production of the OMP and the expression of in vitro virulence-associated properties were studied with $CRMOX^+$ strains and acridine orangecured $CRMOX^-$ mutants. The results were as follows : 1. Of the in vitro virulence-associated tests with 289 strains of Y enterocolitica, 275 strains (95.2%) were positive for pyrazinamidase test, and followed by in order of crystal violet binding test, 226 (79.2% ) ; CRMOX test, 190 (65.7%) ; autoagglutination test, 1.85(64.0%) : calcium dependency test, 86 (29.8%) and congo red uptake test, 47(16.3%). 2. The correlationship between autoagglutination and CRMOX test(r=0.90) was highly significant (p<0.01). 3. In 190 strains(65.7%) bearing the virulence-associated plasmids(MW 40~50 Mdalton), the correlation between the presence of plasmids and their in vitro virulence-associated properties were highest with CRMOX test(r=0.93) and followed by in orders of AAG test(0.81), CV test(0.46), PYZ test(0.37) and CD test(0.18), but no correlationship between the presence of plasmids and CR test(-0.11). 4. The $CRMOX^+$ strains produced the 220 Kdalton OMP when they were cultured at $37^{\circ}C$, but not at $26^{\circ}C$. The presence of 220 Kdalton OMP was correlated significantly with in vitro virulence properties and the presence of virulence-associated plasmid, respectively. 5. In the isogenic $CRMOX^-$ mutant strains, of which plasmid were cured by treatment with acridine orange not only in vitro virulence-associated properties(CR 100%, CD 100%, AAG 82.6%, CV 58.3%) disappeared but also 220 Kdalton OMP(100%) was not produced. These results indicate that the positive CRMOX reaction is plasmid-mediated and the CRMOX test is potential as an in vitro virulence tests with Y enterocolitica.

Transfer of R plasmids of Bacterial Isolates and Their Cloned R Genes in Natural Wastewater Environments (I) -Cloning of $Km^rCm^r$Gene- (하폐수의 자연환경에서 R plasmid와 재조합 유전자의 전이특성( I ) -$Km^rCm^r$유전자의 클로닝-)

  • In order to study the transfer of antibiotics resistance genes of the genetically cloned bacteria in water environments, DK1 strain, which is resistant to kanamycin (Km), chloramphenicol (Cm), streptomycin (Sm), and sulfadiazine (Su), was selected from the Gram-negative bacterial isolates from wastewater. One of 4 plasmids harboured in the DK1 strain was found to possess Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ gene and be about 68 kb in size, and it was designated as pDK101. The plasmid of pDK101 was also found to have 16, 32, and 6 restriction sites for EcoRI. .PstI, and SalI, respectively. From the digestion fragments of pDK101 plasmid and pKT230 used as a vector by EcoRI restriction endonuclease, pDT309 and pDT529 were constructed as chimeric plasmids which possess Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ gene and are 30.9 and 52.9 kb in size, respectively. When the chimeric plasmids were trasformed into E. coli C600 or E. coli HB101, transformants of DKC601, DKC602, DKH102, and DKH103 were obtained as cloned bacterial cells. The Km$^{${\gamma}$}$Cm$^{${\gamma}$}$ genes were well expressed in those cloned cells and the chimeric plasmids were clearly detected in the cloned cells of DKC601 and DKH103.

Rearrangement of $Km^{r}$ Gene and Plasmid by Conjugal Transfer in aquatic Environments (수계에서 접합에 의하여 전이된 $Km^{r}$ 유전자 및 Plasmid 의 재배열)

  • The $Km^{r}$ gene and plasmid of natural isolate and genetically modified microorganisms (GMM) rearranged by conjugation in water environments were comparatively analyzed by agarose gel electrophoresis and Southern analysis. The transfer rates of the $Km^{r}$ gene from GMM strains were generally 100 times higher than thosc of natural iso]ate(DKI) under laboratory cnvironments, but their transfer rate was not much different in Moosimcheon River water. The conjugants obtained in LB(Luria-Bertani broth) and FW(filtered river water) water under laboratory conditions showed same number of the plasmids. but the sizes of the plasmids were changed. The $Km^{r}$ gene in the conjugants was found in the same position as the pDKJO] $Km^{r}$ plasmid. In case of the GMM strains as donor. the large plasmids of 180 kb appeared in conjugants obtained in LB and FW water. Especially, the $Km^{r}$ gene in the donor of DKC600 was found to be inserted into chromosome of the conjugant obtained in FW water. However. in the conjugants obtained from DKl and DKB 701 in Moosimcheon River water, the plasmids were rearranged by 4 and 8. respectively, and all of them showed hybridization by the $Km^{r}$ probe. But the small plasmids of the recipient disappeared in the conjugant from DKC600 as donor, and the rearranged plasm ids and chromosome in the conjugants were observed to be hybridized with the $Km^{r}$ probe. Therefore, rearrangement of $Km^{r}$ gene and plasmids by conjugation was found to be afTected diversely by cellular characteristics as well as by environmental factors.

A Profile of Naturally Occurring Plasmids from Selected Strains of Vibrios

  • The naturally occurring plasmids of Vibrio species have been isolated in part to investigate their genetic traits. Among six different Vibrio species tested, Vibrio anguillarum, Vibrio fluvialis, Vibrio vulnificus, Vibrio mimicus and Vibrio furnissi did not show any presence of plasmid. One environmental isolate of Vibrio pnrahemolyticus harboring plasmid was observed. The isolated plasmid was 8.7 kb by analysis with restriction endonuclease digestion. No common feature was shown relationships between the presence of plasmid and resistance against commonly used antibiotic compounds from the tested Vibrios. Key words . plasmid, Vibrio anguillarum, Vibrio fluvialis, Vibrio uulnincus, Vibrio mimicus, Vibrio furnissi, Vibrio parahemolyticus.

  • Pseudomonas syringae Pv. syringae is a causal agent of bacterial blossom blight of kiwifruit in Korea. Eleven strains of the pathogen were isolated from different kiwifruit orchards in Korea and the plasmid profiles were obtained by pulsed-field gel electrophoresis. They could be clustered into six groups according to the number and size of plasmids. The number of plasmids per strain and size of these plasmids ranged from 0 to 4 and from 22 to 160 kb, respectively. Among them, four strains belonging to Group III which harbored two plasmids were resistant to copper sulfate. Southern blot hybridization of the plasmid DNA indicated that the copper resistance determinant was carried on a 48 kb plasmid.
