Transfer of Insecticidal Toxin Gene in Plants: 2. Subcloning of B. thuringiensis Insecticidal Protein Gene and Rapid Plantlet Regeneration from Nicotiana tabacum Protoplast and Callus (식물세포에 살충독소유전자의 전이연구: 2. B. thuringiensis 살충독소유전자의 Subcloning과 Nicotiana tabacum의 원형질체와 칼루스로부터 신속재생연구)

  • The insecticidal protein gene in the pKL-20-1 clone derived from Bacillus thuringiensis serovar. kurstaki plasmid was subcloned in the plant shuttle vector, pGA643. The 7.3 kb fragment was cloned in the BglII and Hpal sites of pGA643 vector and expressed in E. coli S17-1, which produced insecticidal proteins killing Bombyx mori larvae. The clone was named pHL-20. The protoplast formation, calli induction and plantlet regeneration of Nicotiana tabacum was carried out. A tremendous number of mesophyll protoplasts of N. tabacum were formed, up to 7$\times$105 protoplast per ml, for 20 hours in darkness in the enzyme solution of 0.5% cellulase and 0.1% macerosin, pH 5.8. The viabilities of the protoplasts were maintained above 80% for 6 days in the media containing 2mg/1 of NAA and 1mg/1 of kinetin. Calli were induced from the protoplasts and leaves of the N. tabacum on MS medium containing 0.5mg/1 BAP. Under the culture conditions the protoplasts underwent repeated cell division into calli. Plantlets were regenerated from callus cultures derived from protoplast and leaves. Shoots were induced in a medium containing 1mg/1 of BAP.

  • The medicinal value of the genus Cleome justifies bio-technological studies of Cleome rosea, a Brazilian annual species from sandy coastal ecosystems (restinga), which have been submitted to an intense process of antropogenic degradation. In the present work, was analyzed the influence of cytokinins, 6-benzyladenine (BA) and 6-furfurylaminopurine (kinetin) added to the Murashige and Skoog medium (MS), on the proliferation capacity of explants from the stem axis (hypocotyl, node and internode) for a period of five monthly subcultures (150 days). Regardless of the explant sources, plantlet regeneration by direct and indirect organogenesis was observed. The largest number of shoots proliferated through direct organogenesis was obtained on medium with 4.4 $\mu{M}$ BA. Also, the highest proliferation capacity through indirect organogenesis was found on medium with 4.4 $\mu{M}$ BA + 4.6 $\mu{M}$ kinetin. The presence of kinetin alone was not effective for multiplication of the species. Elongation and rooting were obtained when shoots were transferred onto growth regulator-free medium, and acclimatization rates from 70% to 81% were achieved depending on explant sources used. Plants were then successfully established in soil and showed normal phenotypes.

  • An efficient plant regeneration of C. asiatica was achieved from organogenesis using petiole explants of in vitro plantlet on MS basal medium controled with different plant growth regulators (NAA,2,4-D, IAA kinetin, and BA). Best results that 50%, efficiency of regeneration per explant for regeneration were obtained with IAA $17.13\;{\mu}M$ and BA $8.9\;{\mu}M$. Formation of adventitious shoots via organogenesis from the petiole explant was verified by histological sectioning of plantlets. Regenerated plants were transplanted into soil.

  • Xanthoceras sorbifolium Bunge (yellowhorn) is a woody tree in the soapberry family, Sapindaceae, native to northern China. This species has been identified as a major woody bioenergy plant for bio-diesel production because of high oil content in seed. But the flowers do not bear fruit well while the many flowers blooming. This study was performed to regenerate in vitro plantlet using adventitious shoot formation. To establish the protocol of plant regeneration, adventitious shoots formation rate in the culture of cotyledon of immature zygotic embryos was 68.6% in 1/2 MS medium with 0.5 mg l-1 BA and 3% sucrose (w/v). In the culture of cotyledons of mature zygotic embryos, induction of adventitious shoots was needed to contain high sucrose in pre-culture medium and the frequency of shoot induction was 64.4%. Multiple shoots were induced in 0.5 mg l-1 TDZ, and rooting of shoot was induced 4.0 mg l-1 IBA. Flow cytometry analysis revealed that all the regenerated plantlets were diploid.

Effects of GA3 and Charcoal on Plant Regeneration from Somatic Embryos of Acanthopanax sessiliflorus (오가피(Acanthopanax sessiliflorus)의 체세포배로부터 식물체 재생에 미치는 GAa3와 Charcoal의 영향)

  • To establish the optimum condition for plant regeneration from somatic embryos of Acanthopanax sessiliflorus Rupr. et Maxim, a medicinal plant, somatic embryos were induced from zygotic embryo-derived embryogenic callus in hormoen-free MS medium. To induce plantlet conversion, cotyledonary somatic embryos were cultured on MS solid medium with GA$_3$at various concentrations (0~10 mg/L) for three weeks. Plantlets were transferred to 1/3 MS solid medium with 0.5% charcoal for 7 weeks. Stem length was increased proportionally to the concentration and treatment period of GA$_3$. Also, the highest leaf width (8.9 mm) and leaf number (2.84) of plantlet were obtained when plantlets were converted on 5,10 mg/L GA$_3$pretreatments, respectively. The highest plant conversion frequency (66.7%) was obtained when the somatic embryos were cultured on medium containing 5 mg/L GA$_3$ for 3 weeks and then were transferred to 1/3 MS medium with 0.5% charcoal. The highest survival rate of soil transfer was 90% when plantlets were regenerated on medium with 5 mg/L GA$_3$ for 3 weeks and then transferred to plastic pots containing vermiculite and sand mixture for 4 weeks.

  • The present study was carried out to assess the possibility of rapid multiplication of Zingiber mioga ROSC. through in vitro culture of shoot-apex. The factor investigated was effect of various growth regulators on shoot-apex culture. The shoot-apex cultured of M. S. (Murashige and Skoog) medium developed into plantlet in 12 Weeks. M. S. medium containing NAA at 05ppm and BA 5.0ppm was found to be optimal for growth of in vitro plantlet.

  • This study was carried out to determine the optimal conditions for the production of plants derived from the unpollinated ovary culture of Chinese chive (Allium tuberosum Rottl.). The Chinese chive collected from Korea showed much higher frequency of plantlet formation than those from Japan in the culture of unpollinated ovaries. Among the collections, 'Youngiljaerae' showed the highest frequency of plantlet formation. The MS basal medium was superior to B/sub 5/ in plantlet formation. The ovaries inoculated on the 2,4-D-free medium were directly induced plantlets without callus formation. Floral parts inoculated as a unit played important roles in callus formation and plant production. The frequency of callus and plantlet formation was higher in the culture of ovary with anthers than that of ovary alone.

  • Single cells obtained from suspension culture of mature embryo-derived callus in wheat(Triticum aestivum L. cv Jang Kwang) were cultured to regenrated into the plantlet. Cell clusters and embryogenic calluses were efficiently developed from when the single cells clutured on the MS medium supplemented with 10${\mu}{\textrm}{m}$ 2,4-D. Upon transfer to hormone-free MS medium containing 10 mg/I AgNO3, embryogenic calluses gave rise to shoots, probably through somatic embryogenesis.

The Protein and Isozyme Patterns During in vitro Plant Regeneration of Yooja (Citrus junos Sieb.) and Trifoliate Orange (Poncirus trifoliata Rafin.)

  • In this study, plant regeneration through in vitro culture from plantlet stems of Yooja (C. junos Sieb.) and trifoliate orange (P. trifoliata Rafin.) was attempted to make mass-production system of virus-free plants having the same genotype with mother plant. In order to investigate physiological change depending on the developmental stage of plant regeneration, the changes of total protein, peroxidase and esterase activity and their isozyme patterns as well were examined in 1/2 MS medium. The results are as follows : 1. The MS medium for the optimal callus induction and shoot formation was utilized. The medium was supplemented either with 2,4-D and Kinetin or with BA and NAA. The optimal concentrations were the combination of 1.0mg/ 2,4-D +0.3mg/ Kinetin and 1.0mg BA +0.3mg NAA in callus induction and shoot formation, respectively. 2. For the plant regeneration from somatic embryos, 1/2 MS medium was used with supplements of growth regulators (free, 1.0mg/ IBA +1.0mg/ BA ,0.5mg/ IBA +0.5mg/ BA). Shooting and rooting were the best in the treatment of 0.5mg/ IBA and 0.5mg/ BA combination. 3. The total protein content has a tendency of increase with the developmental stage of embryo, but it was decreased at the plantlet. Also it was the highest at 8 and 6 weeks stage in C. junos Sieb. and P. trioliata Rafin, respectively. In the SDS-PAGE pattern of protein, C. junos Sieb. showed bands of 29.0 and 40kDa at 10 weeks. The 45,66 and 97.4 kDa bands at 10 weeks of culture were shown in P. trifoliata Rafin. 4. The highest esterase activity was shown at the 6 and 8 weeks of culture in C.junos Sieb. and P. trifoliata Rafin.., respectively. 5. Esterase isozyme patterns were shown difference according to the developmental stage. In C. junos Sieb. a new band was observed at pl 7.7 following 4 weeks culture. On the other hand, new bands in P. trifoliata Rafin. were observed at pl 7.5~6.5 following 4 and 6 weeks culture, respectively.
