풋마름병균, Ralstonia solanacearum의 길항세균 SKU-78 균주의 분리 동정 및 특성 (Isolation, Identification and Biological Control Activity of SKU-78 Strain against Ralstonia solanacearum)

  • 종자발아실험과 뿌리에서의 집락 형성실험으로 고추와 토마토의 근권 토양에서 plant growth promoting rhizobacteria(PGPR) 균주를 선발하였다. 그 중 포트실험에서 종자처리와 토양 관주처리 하였을 때 60% 이상 풋마름병 방제효과를 나타낸 SKU-78 균주에 대해서 포장에서의 방제효과 검증에 의해 생물농약으로의 개발 가능성을 확인하였으며, 생화학적 특성조사와 16S rDNA sequence 분석에 의해 SKU-78 균주를 Bacillus sp. SKU-78로 동정하였다.

Biological Control of Bacterial Fruit Blotch of Watermelon Pathogen (Acidovorax citrulli) with Rhizosphere Associated Bacteria

  • Bacterial fruit blotch (BFB), which is caused by Acidovorax citrulli, is a serious threat to watermelon growers around the world. The present study was conducted to screen effective rhizobacterial isolates against 35 different A. citrulli isolates and determine their efficacy on BFB and growth parameters of watermelon. Two rhizobacterial isolates viz. Paenibacillus polymyxa (SN-22), Sinomonas atrocyanea (NSB-27) showed high inhibitory activity in the preliminary screening and were further evaluated for their effect on BFB and growth parameters of three different watermelon varieties under greenhouse conditions. The greenhouse experiment result revealed that SN-22 and NSB-27 significantly reduced BFB and had significant stimulatory effect on total chlorophyll content, plant height, total fresh weight and total dry weight compared to uninoculated plants across the tested three watermelon varieties. Analysis of the 16S ribosomal RNA (rRNA) sequences revealed that strains SN-22 belong to P. polymyxa and NSB-27 to S. atrocyanea with the bootstrap value of 99% and 98%, respectively. The isolates SN-22 and NSB-27 were tested for antagonistic and PGP traits. The result showed that the tested isolates produced siderophore, hydrolytic enzymes (protease and cellulose), chitinase, starch hydrolytic enzymes and they showed phosphate as well as zinc solubilizing capacity. This is the first report of P. polymyxa (SN-22) and S. atrocyanea (NSB-27) as biocontrol-plant growth promoting rhizobacteria on watermelon.

Plant growth promoting rhizobacteria influence potato tuberization through enhancing lipoxygenase activity

  • Molecular insights on the role of plant growth promoting rhizobacteria (PGPR) in potato tuberization are reported in the present study. The PGPRwere isolated from the soil collected from potato fields of Highland Agricultural Research Centre, Pyeongchang, Korea and they were identified to the genus level based on the 16S rRNA sequence analysis. These PGPR were heat-killed, filtered and the filtrates were addedindividually at a concentration of $10^7\;cfu\;mL^{-1}$ in MS (Murashige and Skoog's) medium supplemented with 7% (w/v) sucrose to study their influence on in vitro potato tuberization. Tuber initiation occurred early in untreated control, while tuber growth was pronounced in case of PGPR treatments. The control explants showed tuber formation as a result of sub-apical swelling of stolons while several sessile tubers formed directly in the axils of nodal cuttings in case of PGPR treatments, which is an indication of strong induction for tuberization. Theexplants cultured on MS medium supplemented with bacterial isolate 6 (Bacillus firmus strain 40) showed highest average tuber yield (Ca. 12.56 g per treatment) after 30 days of culture, which was 3 folds increase over the untreated control. A significant increase in lipoxygenase (LOX1) mRNA expression and activity of LOX enzyme were also detected in the tubers induced on PGPR treatments as compared to untreated control. This LOX expression level correlated with increased tuber growth and tuber yield. Further studies focused on the role of bacteria cell wall components, growth regulators and signal molecules released by PGPR are under investigation to elicit clues for PGPR-mediated signal pathway controlling potato tuberization.

해안 사구에서 서식하는 토착식물로부터 분리된 근권미생물 Bacillus aerius MH1RS1의 식물성장 촉진 능력 연구 (Plant Growth-promoting Ability by the Newly Isolated Bacterium Bacillus aerius MH1RS1 from Indigenous Plant in Sand Dune)

  • 다양한 난개발과 해안도로, 방파제 등의 인공 구조물 설치로 인한 해안 침식과 해안선 파괴 등으로 해안사구가 크게 훼손되고 있다. 이에 본 연구에서는 해안사구에서 서식하고 있는 토착식물의 근권으로부터 식물 성장 촉진 능력이 있으면서 동시에 염분에 강한 내성을 가지는 근권세균의 library를 구축하였고, 이들 균주를 대상으로 식물 성장 촉진 능력을 평가하였다. 또한, 내염성 식물을 대상으로 사구 토양에서의 성장률에 근권세균이 미치는 영향을 평가한 후, 훼손된 사구의 복원에 가장 유용한 미생물을 선별하고 분리된 근권세균이 식물 성장에 미치는 영향을 평가하였다. 실험은 선정된 균주와 갯기름나물과 줄무늬갈대를 해안사구에 식재한 후 성장에 미치는 영향을 평가하였다. 그 결과, MH1RS1는 (IAA) production, siderophores, 1-aminocyclopropane-1-carboxylate deaminase (ACC deaminase) 그리고 염분내성을 가지고 있다. 갯기름나물의 경우는 줄기의 길이와 뿌리의 무게는 크게 향상되었다. 특히, 줄기의 생체와 뿌리 무게는 control과 비교했을 때 25% 성장이 향상되었다. 줄무늬 갈대는 Bacillus aerius MH1RS1 에 의해 초장은 18%와 뿌리의 생체 중은 20%로 크게 향상 되었다.

Rapid Detection of Cadmium-Resistant Plant Growth Promotory Rhizobacteria: A Perspective of ELISA and QCM-Based Immunosensor

  • Plant growth-promoting rhizobacteria (PGPR) pseudomonads have a large number of lipopolysaccharides on the cell surface, which induces immune responses. Cd-resistant PGPR prevalent at the Cd-affected sites under biophytostabilization was monitored. Transmissiom electron microscopy was used to the study the behavior of tolerance of PGPR to cadmium level and its effect on pseudomonad strains (Z9, S2, KNP2, CRPF, and NBRI). An immunosensor was developed by immobilizing antibody (anti-Z9 or anti-S2) against selected PGPR on a piezoelectric quartz crystal microbalance (QCM). Immunosensors were found to supplement the inherent specificity of antigen-antibody reactions with the high sensitivity of a physical transducer. On comparison of the efficiency of detection with ELISA, the spectrophotometric technique, the developed immunosensor was found to be more sensitive, fast, and reliable even after regeneration for several times. Thus, the immunosensor may be used for future detection of PGPR strains after automation of the screening process.

Isolation and Characterization of Plant Growth-Promoting Bacteria for the Phytoremediation of Diesel- and Heavy Metal-Contaminated Soil

  • Plant growth-promoting (PGP) bacteria can be used as bioresources to enhance phytoremediation through their PGP traits and pollutant removal capacity. In this study, 49 rhizobacteria were primarily isolated from the rhizosphere of tall fescue grown in diesel- and heavy metal-contaminated soil. Their biosurfactant production, phosphate (P) solubilization, and siderophore production were qualitatively and quantitatively evaluated to identify superior PGP bacteria. The optimal conditions for the growth of PGP bacteria and the stability of their PGP traits were a temperature of 35℃, a pH of 7, and 2 days of cultivation time. Four superior PGP bacteria (Pseudomonas sp. NL3, Bacillus sp. NL6, Bacillus sp. LBY14, and Priestia sp. TSY6) were finally selected. Pseudomonas sp. NL3 exhibited superior biosurfactant production and P solubilization. Bacillus sp. NL6 showed the highest P solubilization and superior production of biosurfactants and siderophores. Bacillus sp. LBY14 offered the best siderophore production and impressive P solubilization. Priestia sp. TSY6 had superior capacity for all three PGP traits. Through their secretion of beneficial PGP metabolites, the four bacteria isolated in this study have the potential for use in the phytoremediation of contaminated soil.

양송이 수확 후 배지로부터 식물생장촉진세균의 분리 및 생육특성 (Isolation and Characterization of Plant Growth Promoting Rhizobacteria from Waste Mushroom bed from Agaricus bisporus)

  • An auxin-producing bacteria (3YN11-02) was isolated from waste mushroom bed from Agaricus bisporus of Chungnam Buyeo-Gun area. The strain 3YN11-02 was identified as a novel species belongs to Rahnella aquatica by a chemotaxanomic and phylogenetic nalysis. The isolate was confirmed to produce indole-3-acetic acid (IAA) which is one of auxin hormone by TLC and HPLC analysis. When the concentration of IAA was assessed by performing HPLC quantity analysis, the maximal $290mg\;L^{-1}$ of IAA detected in ether fraction extracted from the culture filtrate which was cultured in R2A broth containing 0.1% tryptophan for 24 h at $35^{\circ}C$. The molecular weight of the main peak obtained by LC-mass analysis was correspondent well to 175, that of IAA. To investigate the growth promoting effect of crop, when the culture broth of R. aquatica 3YN11-02 was infected onto water culture and seed pot of mung bean, the adventitious root induction and root growth of mung bean were 2.0 times higher than control.

Different Mechanisms of Induced Systemic Resistance and Systemic Acquired Resistance Against Colletotrichum orbiculare on the Leaves of Cucumber Plants

  • Defense mechanisms against anthracnose disease caused by Colletotrichum orbiculare on the leaf surface of cucumber plants after pre-treatment with plant growth promoting rhizobacteria(PGPR), amino salicylic acid(ASA) or C. orbiculare were compared using a fluorescence microscope. Induced systemic resistance was mediated by the pre-inoculation in the root system with PGPR strain Bacillus amylolquefaciens EXTN-1 that showed direct antifungal activity to C. gloeosporioides and C. orbiculare. Also, systemic acquired resistance was triggered by the pre-treatments on the bottom leaves with amino salicylic acid or conidial suspension of C. orbiculare. The protection values on the leaves expressing SAR were higher compared to those expressing ISR. After pre-inoculation with PGPR strains no change of the plants was found in phenotype, while necrosis or hypersensitive reaction(HR) was observed on the leaves of plants pre-treated with ASA or the pathogen. After challenge inoculation, inhibition of fungal growth was observed on the leaves expressing both ISR and SAR. HR was frequently observed at the penetration sites of both resistance-expressing leaves. Appressorium formation was dramatically reduced on the leaves of plants pre-treated with ASA, whereas EXTN-1 did not suppress the appressorium formation. ASA also more strongly inhibited the conidial germination than EXTN-1. Conversely, EXTN-1 significantly increased the frequency of callose formation at the penetration sites, but ASA did not. The defense mechanisms induced by C. orbiculare were similar to those by ASA. Based on these results it is suggested that resistance mechanisms on the leaf surface was different between on the cucumber leaves expressing ISR and SAR, resulting in the different protection values.

A Two-Strain Mixture of Rhizobacteria Elicits Induction of Systemic Resistance Against Pseudomonas syringae and Cucumber Mosaic Virus Coupled to Promotion of Plant Growth on Arabidopsis thaliana

  • We evaluated a commercial biopreparation of plant growth-promoting rhizobacteria (PGPR) strains Bacillus subtilis GB03 and B. amyloliquefaciens IN937a formulated with the carrier chitosan (Bio Yield) for its capacity to elicit growth promotion and induced systemic resistance against infection by Cucumber Mosaic Virus (CMV) and Pseudomonas syringae pv. tomato DC3000 in Arabidopsis thaliana. The biopreparation promoted plant growth of Arabidopsis hormonal mutants, which included auxin, gibberellic acid, ethylene, jasmonate, salicylic acid, and brassinosteroid insensitive lines as well as each wild-type. The biopreparation protected plants against CMV based on disease severity in wild-type plants. However, virus titre was not lower in control plants and those treated with biopreparation, suggesting that the biopreparation induced tolerance rather than resistance against CMV. Interestingly, the biopreparation induced resistance against CMV in NahG plants, as evidenced by both reduced disease severity and virus titer. The biopreparation also elicited induced resistance against P. syringae pv. tomato in the wild-type but not in NahG transgenic plants, which degrade endogenous salicylic acid, indicating the involvement of salicylic acid signaling. Our results indicate that some PGPR strains can elicit plant growth promotion by mechanisms that are different from known hormonal signaling pathways. In addition, the mechanism for elicitation of induced resistance by PGPR may be pathogen-dependent. Collectively, the two-Bacilli strain mixture can be utilized as a biological inoculant for both protection of plant against bacterial and viral pathogens and enhancement of plant growth.

Potential for Augmentation of Fruit Quality by Foliar Application of Bacilli Spores on Apple Tree

  • Previous studies have addressed the management of phyllosphere pathogens by leaf and root-associated microbes. The present study evaluated the effect of the foliar application of three strains of Bacillus spp. on plant growth and fruit quality. The application of a bacilli spore preparation significantly improved leaf growth parameters such as leaf thickness and photosynthesis capacity, indicating that bacilli treatment directly promoted leaf growth. In addition, foliar treatment resulted in an improvement in the key indicators of fruit quality including water, glucose, and sucrose contents. The present results suggest that foliar spraying of beneficial bacilli is a potential treatment of wide application for the improvement of apple quality. Foliar application of bacilli preparation as effective plant growth-promoting rhizobacteria broadens the spectrum of their availability for orchard application.
