오차드그래스의 종자유래의 캘러스로부터 식물체 재분화율에 미치는 Copper의 영향 (Effect of Copper on the Plant Regeneration from Seed Derived Callus of Orchardgrass (Dactylis glomerata L.))

  • 오차드그래스의 완숙 종자배양에서 배지 내에 첨가되는 copper의 농도가 캘러스 형성과 식물체 재분화율에 미치는 영향을 조사하였다. 캘러스의 유도는 $2\;mg/{\ell}$의 2,4-D와 $0.1-100\;{\mu}mol$의 copper sulfate를 첨가한 MS 배지를 이용하였으며, 식물체 재분화는 $1\;mg/{\ell}$의 NAA, $5\;mg/{\ell}$의 kinetin 및 $0.1-100\;{\mu}mol$의 copper sulfate가 첨가된 N6 배지를 이용하였다.그 결과, copper의 첨가는 캘러스 형성능력에는 유의성있는 차이를 나타내지 않았으나, copper가 첨가된 배지에서 생성된 캘러스에서 백색이면서 조직이 치밀한 즉, 식물체 재분화에 적합한 캘러스의 비율이 높게 나타났다. 식물체 재분화율은 copper가 첨가된 배지에서 생성된 캘러스가 첨가되지 않은 배지에서 생성된 캘러스보다 높게 나타났으며, 특히 $70\;{\mu}mol$의 copper가 첨가된 백지에서 생성된 캘러스가 대구조에 비해 2.5배 이상 증가된 가장 높은 재분화 능력을 나타내었다. 또한, 캘러스당 재분화된 식물체의 수는 $70\;{\mu}mol$의 copper가 첨가된 배지에서 생성된 캘러스에서 대조구에 비해 2.6배 이상 증가된 것으로 나타났다.

무궁화 품종 '난파'의 효율적인 재분화 체계 확립 (Establishment of an efficient regeneration system for Hibiscus syriacus 'Nanpa')

  • Hibiscus syriacus L., the national flower of Korea, is a commonly grown ornamental species found in parks, gardens and along roadsides in Korea. This study sought to establish a plant regeneration system of H. syriacus 'Nanpa' using mature leaves as an explant. Sterilized mature leaf explants were cultured on modified MS medium with combinations of NAA and BAP at various concentrations for 6 weeks. Among the various media evaluated, modified MS media containing 0.46 mg/L NAA and 0.5 mg/L BAP (CI-1) or 0.92 mg/L NAA and 1 mg/L BAP (CI-4) were the most effective for callus formation. Whitish-yellow calluses were observed on CI-1 medium, while green calluses formed on CI-4 medium. The whitish-yellow callus derived from CI-1 medium showed a higher frequency of shoot induction than the green callus derived from CI-4 on modified MS medium containing 0.5 mg/L BAP. Among the various media evaluated in this study, modified MS medium containing 0.46 mg/L NAA and 0.5 mg/L BAP was the most optimal for shoot-forming callus production. Our findings show that mature leaves of H. syriacus 'Nanpa' can be used for in vitro plant regeneration, and the regeneration system described here may be a powerful tool for molecular breeding of H. syriacus 'Nanpa' in the future.

  • 히아신스 Pink Pearl 품종의 인편배양에서 자구의 재생과 생장은 1.0 mg/l와 3.0 mg/l의 농도에서 IAA보다 IBA가 더 효과적이었으며, 절편체의 기부가 배지에 삽식되는 정상적인 치상 방향이 역방향의 치상 방법에 비해 자구의 재생과 뿌리의 생장에 더 효율적인 것으로 나타났다. 계대배양에서 재생된 자구와 뿌리의 생장은 한천배지보다 펄라이트가 첨가된 액체배지에서 증가되었다. 이와 같은 히아신스 Pink Pearl 품종의 기내배양의 경우 자구 재생을 위한 초대배양은 한천 고체배지를, 재생 된 자구의 생장은 펄라이트의 액체배지를 사용하는 2 단계의 교대배양 시스템이 더 유용함을 시사한다.

Micropropagation of Lobelia chinensis Lour.: Influence of Medium Parameters on Plant Regeneration, Antioxidant Activity, and Secondary Metabolite Accumulation

  • Chinese lobelia (Lobelia chinensis Lour.) is an important medicinal plant that is used in traditional Chinese, Korean, and Japanese medicine. The goal of the current study was to develop an in vitro propagation technique for Lobelia chinensis. We have examined the effects of different media compositions on the regeneration of shoots from nodal cultures of Lobelia chinensis, including Murashige and Skoog (MS), Gamborg (B5), Schenk and Hildebrandt (SH), Woody plant (WPM), Chu (N6), and Nitsch and Nitsch (NLN) media. Similar to this, shoot regeneration was examined using MS medium of double (2.0), full (1.0), half (0.5), and quarter (0.25) strengths. The regeneration of shoots was also examined with additions of 0, 1, 3, 5, and 7% (w/v) sucrose to MS media. For axillary shoot regeneration, full-strength MS medium supplemented with 3% (w/v) sucrose was shown to be the most effective of all the evaluated factors. On this medium, nodal explants optimally regenerated 4.5 shoots per explant and subsequently shoots involved in rooting on the same medium. The regenerated plants possessed abundant phenolics, flavonoids, and DPPH, ABTS, and FRAP antioxidant activities. High performance liquid chromatographic examination (HPLC) of the regenerated plants revealed an accumulation of myricetin and catechin in higher amounts.

Effects of Media Composition on Plant Regeneration and Callus Formation of Abeliophyllum distichum Nakai

  • This experiments were carried out to find out the effects of different explant materials, kinds and concentration of plant growth regulators, and total nitrogen and sucrose contents on the in vitro regeneration of Abeliophyllum distichum Nakai. The effects of growth regulators on regeneration from 3 explant sources (leaf, internode and node) were more or less same. Leaf explants produced only callus with 2ip (Isopentenyladenine) and NAA (Naphthaleneacetic acid) treatment and other regulators had no effects. Test with internode explants yielded about same results but callus was obtained with 2,4-D (2,4-Dichlorophenoxyacetic acid). Node explants resulted in shoot regeneration by all regulator treatment except NAA and 2,4-D, but control also showed similar results. Callus formation from internode and node explants was vigorous by 2ip, zeatin, and 2,4-D treatments and high NAA concentration resulted in higher callus formation. In this experiment, various mixed treatment of growth regulators were also employed, using node as explant material. Shoot regeneration was obtained with BA (Benzyl adenine) + NAA treatments but the results were comparable with control. Generally shoot and root regeneration was poor with all combined treatment except 2ip + NAA and 2,4-D + NAA. However, callus was formed readily with all treatments. In this experiment, combined treatments of regulators were applied on the callus derived from singular regulator treatment. The results showed no shoot and root regeneration with any combination of 2,4-D, IAA (Indoleacetic acid) and NAA, but soft milky white callus was formed in all the treatments. No shoot and root regeneration was observed with any combination of 2iP, NAA and IAA, but somewhat hard, light green callus was formed in all the treatments. Callus formation decreased with high kinetin concentration in case of kinetin + NAA treatment. The experiments with total nitrogen content of media showed that low concentrations of 15 and 30mM were effective for the shoot and root regeneration. Sucrose experiment demonstrated shoot regeneration with 1${\sim}$4% concentration, and root and callus formation with 2${\sim}$4%. No root and callus formation was observed with 0 and 1% sucrose.

오차드그래스의 종자유래 캘러스배양 및 재분화에 미치는 배지첨가물질의 영향 (Effects of Medium Supplements on Seed-Derived Callus Culture and Regeneration of Orchardgrass)

  • 오차드그래스의 최적 조직배양조건을 확립하기 위하여 'Roughrider' 품종의 성숙종자로부터 최적 배발생 캘러스 유도 및 식물체 재분화에 미치는 배지첨가물질의 영향을 조사하였다. 성숙종자로부터 배발생 캘러스 유도시 첨가되는 생장조절 물질로는 3 mg/L 2,4-D가 가장 효율적이었으며, 식물체 재분화에는 1 mg/L 2,4-D와 3 mg/L BA가 첨가된 배지에 캘러스를 배양했을 때 36.3%의 재분화율을 나타내었다. 캘러스 유도배지와 재분화배지에 1 g/L의 casein hydrolysate와 300 mg/L의 proline을 동시에 첨가해주었을 때 캘러스 유도율과 재분화율이 각각 57.3%와 60.7%로 증가되었다. 또한 항산화물질로서 10 mg/L의 $\textrm{AgNO}_3$ 와 40 mg/L의 cysteine을 첨가해준 과 켈러스 유도율 재분화율 각각 68.7%와 71.7%까지 증가되었다. 본 연구를 통하여 확립된 성숙종자로부터 효율적인 배발생 캘러스의 유도 및 식물체 재분화 체계는 유전자 형질전환을 통한 신품종 오차드그래스 개발에 유용하게 응용되어질 수 있을 것이다.

The Protein and Isozyme Patterns During in vitro Plant Regeneration of Yooja (Citrus junos Sieb.) and Trifoliate Orange (Poncirus trifoliata Rafin.)

  • In this study, plant regeneration through in vitro culture from plantlet stems of Yooja (C. junos Sieb.) and trifoliate orange (P. trifoliata Rafin.) was attempted to make mass-production system of virus-free plants having the same genotype with mother plant. In order to investigate physiological change depending on the developmental stage of plant regeneration, the changes of total protein, peroxidase and esterase activity and their isozyme patterns as well were examined in 1/2 MS medium. The results are as follows : 1. The MS medium for the optimal callus induction and shoot formation was utilized. The medium was supplemented either with 2,4-D and Kinetin or with BA and NAA. The optimal concentrations were the combination of 1.0mg/ 2,4-D +0.3mg/ Kinetin and 1.0mg BA +0.3mg NAA in callus induction and shoot formation, respectively. 2. For the plant regeneration from somatic embryos, 1/2 MS medium was used with supplements of growth regulators (free, 1.0mg/ IBA +1.0mg/ BA ,0.5mg/ IBA +0.5mg/ BA). Shooting and rooting were the best in the treatment of 0.5mg/ IBA and 0.5mg/ BA combination. 3. The total protein content has a tendency of increase with the developmental stage of embryo, but it was decreased at the plantlet. Also it was the highest at 8 and 6 weeks stage in C. junos Sieb. and P. trioliata Rafin, respectively. In the SDS-PAGE pattern of protein, C. junos Sieb. showed bands of 29.0 and 40kDa at 10 weeks. The 45,66 and 97.4 kDa bands at 10 weeks of culture were shown in P. trifoliata Rafin. 4. The highest esterase activity was shown at the 6 and 8 weeks of culture in C.junos Sieb. and P. trifoliata Rafin.., respectively. 5. Esterase isozyme patterns were shown difference according to the developmental stage. In C. junos Sieb. a new band was observed at pl 7.7 following 4 weeks culture. On the other hand, new bands in P. trifoliata Rafin. were observed at pl 7.5~6.5 following 4 and 6 weeks culture, respectively.

국화 '오블랑'의 재생 효율 증진을 위한 식물생장조절제와 AgNO3 적정 농도 선별 (Optimal concentrations of plant growth regulators and AgNO3 for the improvement of regeneration efficiency in Chrysanthemum morifolium 'Ohblang')

  • 국화 '오블랑'의 기내 배양 잎 절편체로부터 신초 기관형성을 통한 식물체 재생 시스템을 확립하는데 있어 효율적인 신초 재생 및 갈변억제를 위한 식물생장조절제와 AgNO3의 농도를 선정하고자 실험을 수행하였다. 잎 절편체를 식물생장조절제인 BA 0.5, 1.0, 2.0 mg/L와 NAA 0.2, 0.5, 1.0, 2.0 mg/L를 조합 처리한 12가지 조성의 배지에 치상하여 암조건에서 6주간, 명조건에서 6주간 배양하였을 때, BA 1.0 mg/L, NAA 1.0 mg/L를 첨가한 배지를 사용하는 것이 가장 효과적이었다. 이 결과를 기반으로 잎 절편체로부터 갈변 감소를 위해 MS 염류와 비타민, BA 1.0 mg/L, NAA 1.0 mg/L, sucrose 30 g/L, agar 6 g/L를 포함한 신초유도배지에 AgNO3 0, 1, 3, 5, 7, 10 mg/L를 각각 첨가하여 실험을 실시하였다. AgNO3 처리는 잎 절편체로부터의 갈변을 감소시키는 효과를 보였으나, 처리 농도가 증가됨에 따라 신초로의 발달은 원활하게 이루어지지 않았다. 결과적으로 AgNO3 1 mg/L를 포함한 신초유도배지(MS 염류와 비타민, BA 1.0 mg/L, NAA 1.0 mg/L)에 잎 절편체를 치상하였을 때, 갈변지수 0.7, 절편체당 신초 수가 2.6개로 '오블랑' 재생에 있어 가장 적합한 농도임을 확인하였다.

Establishment of Cell Suspension Cultures and Plant Regeneration in White Dandelion (Taraxacum coreanum NAKAI.)

  • In this study, we established a novel somatic embryogenesis and plant regeneration system through cell suspension culture of white dandelion (Taraxacum coreanum NAKAI.). Embryogenic calli could be initiated from leaf and root explants of sterile seedlings on solid Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) after 3-week cultures. To proliferate embryogenic calli rapidly, cell suspension culture was performed with transferred to liquid MS medium with various combinations of plant growth regulators (PGRs) including 2,4-D, ${\alpha}$-naphthalene acetic acid (NAA), indole-3-acetic acid (IAA), $N^6$-benzylamino purine (BAP), thidiazuron (TDZ), and kinetin. During suspension cultures, embryogenic calli not only greatly proliferated, but shoot organogenesis also simultaneously occurred from the surface of somatic embryos. Among them, TDZ at lower concentration, 0.1 mg/L produced the highest efficiency of somatic embryo formation and shoot organogenesis. Rooting of embryogenic calli with adventitious shoots was done on solid MS medium containing 0.1 mg/L NAA and 0.3% activated carbon. Nearly 80% of embryogenic calli with shoot organogenesis could be rooted normal. Well-rooted plantlets were transferred into pots under a greenhouse condition, and plants derived from this system appeared phenotypically normal.

벼 Callus의 계대배양(繼代培養)이 식물체(植物體) 재분화(再分化)에 미치는 영향(影響) (Effect of Subculture on Plant Regeneration in Rice Callus Culture)

  • 벼 완숙현미(完熟玄米)의 배(胚)로부터 유기(誘起)된 callus의 계대배양(繼代培養)에 영향(影響)을 미치는 계대배양(繼代培養) 회수(回數), 기간(期間) 및 배지내(培地內)에 첨가(添加)되는 생장조정제(生長調整劑)의 조성(組成) 등(等)과 같은 몇가지 요인(要因)에 대한 실험(實驗)을 수행(遂行)하여 얻어진 결과(結果)는 다음과 같다. 벼 callus의 계대배양(繼代培養)은 4주간격(週間隔)보다는 2주간격(週間隔)으로 배양(培養)하는 것이 효과적(效果的)이었고 식물체(植物體) 재분화율(再分化率)은 품종(品種)과 계대배양(繼代培養) 기간(期間)에 따라 크게 상이(相異)한 것으로 나타났다. "영덕(盈德)벼"의 경우는 2주간격(週間隔)으로 4회(回) 계대배양(繼代培養)된 callus에서 72.5%의 가장 높은 재분화율(再分化率)을 보였으며, 4주간격(週間隔) 2회(回) 계대배양(繼代培養)된 callus에서도 58.3%의 재분화율(再分化率)을 보였다. Callus유기(誘起) 배지(培地)에 CH(2g/1), MI(200mg/1), TH(2mg/1)등(等)을 첨가(添加)하면 callus생장량(生長量)도 많았고 식물체(植物體) 재분화율(再分化率)도 향상(向上)되었으며, 식물체(植物體) 재분화(再分化)에는 kinetin단용(單用)보다는 kinetin과 NAA를 혼용(混用)하는 것이 효과적(效果的)인 것으로 나타났다. Callus 계대배양(繼代培養)에 CH, MI, TH 등(等)을 첨가(添加)한 배지(培地)를 이용(利用)하면 embryogenic callus의 발생량(發生量) 증가(增加)하는 경향이었고 품종(品種)에 따라서는 식물체(植物體) 재분화율(再分化率)도 높게 유지(維持)되는 것으로 나타났다.
