Bacteriophage MS2를 이용한 소독제 효력시험 확립에 관한 연구 (Method development for efficacy testing of veterinary disinfectants using bacteriophage MS2)

  • In virucidal efficacy testing, the chemical inactivation cannot be determined for all viruses due to the difficulties or the inability to culture sufficiently or the risk of exposure to the viruses. Therefore, disinfectants against these viruses could be evaluated by different methods and surrogate viruses are used as alternative. In this study we developed a method for efficacy testing of veterinary disinfectants using one of the candidate surrogate viruses, bacteriophage MS2, as part of the research on the selection of surrogate viruses for efficiency of efficacy testing of veterinary disinfectants. This method is based on the Animal and Plant Quarantine Agency (APQA) guidelines for efficacy testing of veterinary disinfectants. Bacteriophage and disinfectant are reacted in suspension in accordance with the APQA guidelines and then a newly established double agar layer method is applied for the efficacy test. The double agar layer method is summarized as follows: 1) The bottom agar with 1.5% agar is boiled and cooled before poured into petri dishes at volume of 20 mL, and dried under biological safety cabinet. 2) The top agar with 0.7% agar is boiled and kept at 50℃ before E. coli culture was seeded. 3) The serially diluted bacteriophage MS2-disinfectant mixtures 0.05 mL and E. coli host 0.01 mL (OD600 0.2~0.3) are mixed with 5 mL of top agar and incubate them at 50℃ for 5 min for reaction. 4) The resulting mixture is poured over top of a bottom agar plate and rocked sufficiently to ensure that the top agar covers the entire surface of the bottom agar. 5) The double agar layer is then placed under biological safety cabinet to allow the agar layer to solidify and subsequently incubated at 37℃ for 24 hr. 6) Following incubation, the plates may be inspected for plaques and record results.

국내 감귤류에 발생한Citrus vein enation virus 분포조사 (Incidence of Citrus vein enation virus in Citrus spp. and Poncirus trifoliata in Korea)

  • 2017년, 감귤류에 Citrus vein enation virus (CVEV)의 발생보고가 있었다. 식물방역법상 관리급 병원체인 CVEV에 대한 검역조치를 위해 감귤의 주요 재배지인 제주도를 포함한 4개도 29개 시군에서 온주밀감, 유자, 만숙성 감귤류(한라봉, 천혜향, 레드향, 황금향) 재배과원 등 203개소에서 시료를 수집하고 reverse transcription polymerase chain reaction (RT-PCR) 방법과 시퀀싱을 통해 진단하였다. 진단결과, 양성반응을 보인 시료를 대상으로 외피단백질 유전자 부위를 분석하기 위해 GenBank에 등록된 분리주들과의 염기서열과 아미노산 서열을 비교한 결과, 98% 이상의 매우 높은 상동성을 확인하였다. 전체 시료에 대하여 RT-PCR 진단 결과, 136개 과원(67%)에서 CVEV가 검출되었으며, 유자 과원의 85.4%, 온주밀감 과원의 77.8%가 CVEV에 감염된 것으로 파악됐다. 또한, 감귤의 주요 재배지인 제주도에서 90.6%의 과원이 CVEV에 감염된 것으로 확인하였다. 기주별·지역별 발생실태 조사를 토대로 국내 감귤류에 CVEV가 만연되어 있음을 확인하였다. 본 연구를 바탕으로, CVEV의 발생실태 조사와 검역병원체 여부를 검토하여 2018년 5월 CVEV를 비검역 병원체로 변경하였다.

제주 및 경북지역에서 cuelure 제형별 호박꽃과실파리(Zeugodacus scutellata)의 유인효과 (The Attraction Effect of Different Types of Cuelure on Striped Fruit Flies, Zeugodacus scutellata, in Jeju and Gyeongbuk Regions)

  • 호박꽃과실파리(Z. scutellata)는 박과작물에 피해를 주는 중요한 해충이다. 현재 큐루어(Cuelure)는 국내에서 검역 대상 해충으로 규정된 오이과실파리, 타우과실파리, 및 퀸슬랜드과실파리와 더불어 호박꽃과실파리 수컷 성충의 예찰에 다양한 제형으로 활용되고 있다. 본 연구는 액체, 고체, 및 왁스제형의 큐루어에 대한 호박꽃과실파리 수컷 성충의 야외 유인효과를 검증하기 위한 것으로 2021년 4월부터 10월까지 제주 2개 지역과 경북 1개 조사에서 2주 간격으로 모니터링하였다. 호박꽃과실파리의 발생밀도가 높은 제주지역에서는 7월부터 8월에 액체와 왁스제형의 큐루어에 호박꽃과실파리가 더 많이 유인되어 통계적인 유의차가 있었으나, 호박꽃과실파리의 발생밀도가 낮은 경북지역에서는 제형별 유인력에 차이가 없었다. 야외조사 결과를 바탕으로 액체와 왁스제형의 큐루어가 호박꽃과실파리 수컷 성충 예찰에 권장되며, 이 두 제형의 큐루어에 유인되는 검역 대상 과실파리류의 국내 예찰에 효과적일 수 있다.

원유 세균수 검사장비의 정도관리를 위한 표준시료의 개발 및 평가 (Development and evaluation of standard samples for quality control of automated total bacterial counter in raw milk)

  • Standard samples were prepared for this study, and applied for BactoScanTM and BactoCountTM in order for quality control evaluation for total bacterial count in raw milk. Accordingly, the preparation of two lots of standard samples for quality control were lyophilized, which contain Lactobacillus lactis. The standard samples were prepared into three different levels of bacterial counts, those were Low 30,000~40,000, Medium 70,000~90,000, High 150,000~220,000 CFU/mL, respectively. Then, the proficiency tests were performed in total 19 laboratories for measuring total bacterial counts. The total bacterial counts in the standard samples showed 37~42, 82~105, 214~240 CFU/mL in the first lot, and it showed 30~36, 67~75, 131~163 CFU/mL in the second lot in low, medium and high levels, respectively. Based on these results, the absolute values of z-scores of six standard samples in 18 laboratories were ≤2, which means the samples are satisfactory. However, z-score in one laboratory was ≤3, which means the sample is questionable. Using two standard samples, the correlation between BactoScanTM and BactoCountTM was 0.9982, which means the results of total bacterial count measurement of both equipment were equivalent. Therefore, the standard samples manufactured in this study for quality control of total bacterial count using BactoScanTM and BactoCountTM in the raw milk could be applied to proficiency tests.

한국호흡기형 닭전염성기관지염 생독백신주의 작성 (Development of an attenuated vaccine strain from a korean respiratory type infectious bronchitis virus)

  • An attenuated vaccine strain AVR1/08 of Korean respiratory type of infectious bronchitis virus (IBV) was developed by 89th passages of IBV D85/06 strain in chicken eggs. The AVR1/08 strain had higher virus titer at least 20 times ($10^{1.3}$) than the parent virus D85/06 by egg inoculation method. The AVR1/08 strain had a single point mutation (S to Y) at position 56 of spike protein of IBV compared to parent virus IBV D85/06 strain. The mutation was observed consistently at viruses after 47th passage in chicken eggs. The AVR1/08 strain showed no virulence even after 6 passages in chickens and all chickens inoculated induced anti-IBV antibody 14 days after vaccination. The AVR1/08 strain had broad protective efficacy against QX type Korean nephropathogenic virus (Q43/06 strain), KM91 type Korean nephropathogenic virus (KM91 strain) and Korean respiratory virus (D85/06 strain). In contrast, Massachusetts (Mass) type attenuated vaccine strain H120 showed protection of 37.5 to 50% against these three viruses. Our results indicate that the AVR1/08 strain has potential as an attenuated vaccine effective in controlling IBVs circulating in Korea.

Zonate Leaf Spot of Sorghum Caused by Gloeocercospora sorghi in Korea

  • Gloeocercospora sorghi caused the zonate leaf spot on sorghum (Sorghum bicolor Moench) in fields in Korea. The zonate spots were conspicuous on sorghum leaves as circular, reddish purple bands alternating with tan or straw-colored areas, which formed a coarsely zonate pattern. The lesions often occurred in semicircular patterns along the margin of leaves. The fungus produced sporodochia on the surface of infected leaves in a moist chamber. The conidia, born in a pinkish to salmon-colored slimy matrix, were hyaline, elongate to filiform, straight or slightly curved, 3- to 17-septate, and of variable lenght (28-197 x 1.6-$3.4\mu\textrm{m}$). G. sorghi isolated from diseased leaves of sorghum caused characteristic reddish-brown water-soaked leaf spots when inoculated to seedlings.

식물검역 종자전염 Wheat Streak Mosaic Virus의 PCR 검사시스템 개발 (Development of PCR Diagnosis System for Plant Quarantine Seed-borne Wheat Streak Mosaic Virus)

  • Wheat streak mosaic virus (WSMV)는 Potyviridae 과 Tritimovirus 속으로 분류되는 식물병원성 바이러스로 밀과 옥수수 종자 등에 많은 피해를 주고 있다. 본 연구에서는 관리급 식물검역 종자전염바이러스인 WSMV를 신속하고 정확하게 진단하기 위한 2단계 PCR 시스템을 개발하였으며, 실험실 오염의 여부를 확인 할 수 있는 새로운 양성 대조구를 제작하였다. 본 연구에서 개발된 PCR 시스템으로 일본에서 수입한 스위트콘 종자와 미국에서 수입한 밀 종자의 검역과정에서 WSMV-JSweet-corn2868, WSMVUwheat1944-1 및 WSMV-Uwheat1944-2를 검출하였다. 검출된 증폭 산물은 다른 WSMV의 isolates와 80.60-100.00%의 유사성을 보였다. WSMV는 계통수에서 4가지 genotype이 확인되며, 이번 연구에서 검출한 isolate WSMV-JSweet-corn2868 (JX845574)은 Clade B로 분류되었고, isolate WSMV-Uwheat1944-1 (KC754959)과 WSMV-Uwheat1944-2 (KC754960)는 Clade D로 분류되었다.

검역잡초종자 발아력 제거를 위한 수입농산물 열처리 효과 (Heat-treatment Effects of Agricultural Imports for Devitalization of Quarantine Weed Seeds)

  • 우리나라는 열처리방법을 포함한 식물검역규정에 의하여 외래잡초의 유입방지를 위한 잡초검역을 실시하고 있다. 현행 열처리기준의 적절성을 확인하고 검역잡초의 발아력을 완전히 제거하기 위한 열처리조건을 구명하여 식물검역에 활용하기 위하여 검역잡초 9종과 검역잡초와 유사한 잡초 1종의 종자를 재료로 하여 건열처리와 상대습도 40%의 습열처리에 의한 사멸조건을 연구하였다. 다양한 온도 및 시간별 건열처리 결과 $95^{\circ}C$에서 48시간, $100^{\circ}C$에서 36시간, $110^{\circ}C$에서 24시간, $121^{\circ}C$에서 4시간, $130^{\circ}C$에서 30분 처리 후 모든 종자가 사멸되었고, 상대습도 40%에서의 습열처리 결과 $85^{\circ}C$에서 36시간, $90^{\circ}C$에서 16시간 습열처리 후 모든 종자가 사멸하였다. 상기의 결과는 현행 열처리기준이 검역잡초 몇 종에는 적용하기에 적합하지 않음을 보여주며, 검역잡초종자의 발아력을 제거할 수 있는 이 열처리조건은 수입농산물을 통한 검역잡초의 유입을 방지하기 위한 검역의 한 방법으로 적용할 수 있을 것이다. Picris echioides와 같이 강한 내열성이 있는 종과 새삼속(Cuscuta spp.)과 같이 열에 민감한 종을 위한 별도의 열처리조건이 필요하며 이는 다른 내열성의 검역잡초 종자를 사멸시키는 검역업무의 효율성을 향상시킬 수 있을 것으로 사료된다.

Epidemiological characteristics of bovine brucellosis in Korea, 2000~2004

  • This paper describes the epidemiological characteristics of bovine brucellosis in Korea during January 2000~September 2004, which encompasses the period when the incidence of bovine brucellosis increased abruptly. Data from the National Animal Infectious Disease Data Management System were used for this study. A range of epidemiological measures was calculated including annual herd and animal incidence. During the study period, there were 1,183 outbreaks on 638 farms. In beef cattle, annual herd incidence increased from 0.2 (2000) to 11.5 (2004, to September) outbreaks per 10,000 and annual animal incidence varied between 3.4 (2000) and 105.8 (2004, to September) per 100,000, respectively. On 401 (62.9%) infected farms during this period, infection was eradicated without recurrence. Recurrence of infection was significantly higher on farms where abortion was reported (53.3%), compared to farms where it was not (30.0%). On beef cattle farms, infection was introduced most frequently through purchased cattle (46.2%). Based on the results of this study, the establishment and spread of brucellosis in the Korean beef cattle population were mainly due to incomplete or inappropriate treatment of aborted materials and the movement of infected cattle.

Isolation of novel bovine parainfluenza virus type 5 (bPIV5) and its incidence in Korean cattle

  • Four viruses showing cytopathic effects in MDBK cells were isolated from brains of cattle showing downer cattle syndrome in 2012. The isolates were confirmed to belong to the genus Rubulavirus of the subfamily Paramyxovirinae. Isolate QIA-B1201 had the ability to hemagglutinate red blood cells from several species of animals and was capable of adsorbing guinea pig erythrocytes on the surface of infected Vero cells. Nucleotide sequence analysis showed that two isolates (QIA-B1201 and QIA-B1204) had high similarity with other human and animal PIV5 isolates ranging from 98.1 to 99.8%. The highest sequence similarity of the two isolates corresponded to strain KNU-11 (99.8% at the nucleotide and amino acid level) isolated from suckling piglets in Korea in 2012. To evaluate the virulence of strain QIA-B1201, we inoculated bPIV5 into 5 week-old mice via both the intraperitoneal and intracranial route. Body weight was not significantly altered in mice inoculated with QIA-B1201. In this study, we isolated and characterized novel bPIV5s from brain samples showing downer cattle syndrome, but were not able to elucidate the pathogenicity of the bPIV5s in mice.
