Description and Application of a Marine Microalga Auxenochlorella protothecoides Isolated from Ulleung-do (울릉도 거북바위 조수웅덩이에서 분리된 해양 미세조류 옥세노클로렐라 프로토테코이드 균주의 기술 및 응용)

  • A unicellular green alga was axenically isolated from a tidal pool on Ulleung-do, Korea. Morphological, molecular, and biochemical analyses revealed that the isolate belonged to Auxenochlorella protothecoides. The current study is the first record of this species in Korea. The microalgal strain was named as A. protothecoides MM0011 and its growth, lipid and pigment compositions, and biomass properties were investigated. The strain is able to thrive in a wide range of temperatures ($5{\sim}35^{\circ}C$) and to withstand up to 1.5 M NaCl. The results of GC/MS analysis showed that the isolate was rich in nutritionally important polyunsaturated fatty acids (PUFAs). Its major fatty acids were linoleic acid (27.6%) and ${\alpha}-linolenic$ acid (39.6%). Thus, this indigenous microalga has potential as an alternative source of ${\omega}3$ and ${\omega}6$ PUFAs, which currently come from fish and plant oils. Also, the HPLC analysis revealed that the value-added antioxidant, lutein, was biosynthesized as the accessory pigments by the microalga. A proximate analysis showed that the volatile matter content was 85.6% and an ultimate analysis indicated that the gross calorific value was $20.3MJ\;kg^{-1}$. Since 40.5% of total nitrogen and 27.9% of total phosphorus were removed from the medium, respectively, it also has potential as a feedstock for biofuel applications which could be coupled to wastewater treatment. In addition, the biomass may also serve as an excellent animal feed because of its high protein content (51.4%). Therefore, A. protothecoides MM0011 shows promise for application in production of microalgae-based biochemicals and as a biomass feedstock.

A Local Governments' Preferences in Selecting Modern Eight Scenic Landscapes (지자체가 선정한 현대팔경에 나타난 경관 선호 양상)

  • The followings are the landscape preference aspects from the 816 landscapes(景, Kyung), which comprise the 78 modern Palkyungs, presented by the 78 local governments in Korea. First, the natural environment elements selected as Kyung(景), which are topographical landscapes, mostly consist of mountain elements such as mountains, terrace(臺), rocks and stones and water elements classified as rivers, oceans, and lakes. Natural elements also include old-growth and giant trees such as pines, ginkgos, Japanese cornels and fringe trees, tree-lined streets and forests, and plant elements such as azaleas, rhododendrons, lotuses, reeds, and silver grasses which provide seasonal landscapes. Second, more than half of Kyung, selected as human environment elements, are historical and cultural heritages such as graveyards, mountain fortresses, town fortresses, traditional villages, pavilion in villas, and temples. And it is followed by leisure tourism facilities such as traditional markets, exhibition halls, theme parks, beaches, and food streets, green-based structures such as trails, plazas, parks, and botanical gardens, and industrial heritages such as ranches, abandoned coal mines, stations, ports and bridges. Third, modern Palkyungs include objects not related to the views such as local representative facilities, regional products, and festivals. Fourth, although most of the modern Palkyungs consist of eight, some include 20, 38, or 100 in order to increase the number of objects of public relations. Fifth, a certain local government makes two modern Palkyungs with different subjects by introducing traditional Palkyung and modern Palkyung altogether. In this case, it presents several modern Palkyungs like by selecting Palkyungs in a limited area. Furthermore, one Palkyung includes numerous place names at a time in some cases. Sixth, Sosangjeonhyeong(瀟湘典型)-style modern Palkyung uses 'NakAn(落雁)' as the name of Kyung. Sosangyusahyeong(瀟湘類似型)-style modern Palkyung expresses 'Hyojong(曉鐘)' and landscape of glow of the setting sun, sunset, night view, dawn, sunrise and depicts cloud, sunset, moon, and snow. There are many Myeongsocheheomhyeong(名所體驗型)-style Palkyungs exhibiting the behavior of tourism and Myeongseunghyeong(名勝型)-style Palkyungs raising the awareness only by the names of the places. Seventh, modern Palkyung's naming styles are diverse, such as using only four letters instead of specifying Kyungmul(景物) or Kyungsaek(景色) in combination with Chinese characters or adding modifiers specializing in places.

Managing Mixtures of Tall Fescue (Festuca arundinacea Schreb) and Zoysiagrass (Zoysia japonica Steud.) for Athletic Turf (톨 페스큐와 한국들잔디로 혼합 조성된 운동장 잔디관리)

  • Managing a mixture of zoysiagrass with tall fescue has been proposed in transitional zone as a practical practice to combine the advantages of the two species and compensate the limitations. To manage the mixture is a challenge because two species are involved. The objective of this study was to determine if zoysiagrass/tall fescue mixture can be maintained with proper mowing and fertilization under simulated sport traffic at an acceptable quality level. Zoysiagrass was seeded in June and tall fescue was overseeded in August 1996. In November 1996, zoysiagrass coverage was 62.36, 29.88, and 30.02% for 0, 50, and $100\;Kg\;ha^{-l}\;N$ rates, respectively. At the same time, zoysiagrass coverage was 23.53, 41.95, and 57.40% for the mowing heights of 6.5, 5.0, and 3.5 cm, respectively. Zoysiagrass and tall fescue coverage in July 1997 was showing the same trend as in the late season of 1996 although the differences were not as big. There were significant interactions between N fertilization rates and mowing heights. In November 1998, the zoysiagrass coverage was different among the two tall fescue variety mixtures, 21.68, and 32.25% in 'Arid' and 'Grasslands Garland', respectively. Zoysiagrass coverage was favored in lower mowing height, lower N rates, and lower traffic. Interaction effects on zoysiagrass were found between tall fescue variety and nitrogen rate, tall fescue variety and mowing height, and traffic and nitrogen rate. Zoysiagrass shoot density was 7.42, 25.47, and 58.95% for mowing heights of 6.5, 5, and 3.5 cm, respectively; and it was 47.27, 20.27, and 26.26% for N rates of 0, 50, and $100\;Kg\;ha^{-l}\;$, respectively in 1998. The effects on zoysiagrass shoot density from the interaction of N rate and tall fescue variety was significant in 1998. Shoot density responded to the N rate, mowing height, and traffic differently from the ground coverage, indicating that shoot and leaf growth have different adaptation strategies.

Influence of Dichlorprop and MCPB on the Reduced Effect of Fruit Drop and Fruit Quality before and after Storage in Apples (생장조정제(生長調整劑) Dichlorprop 및 MCPB 처리(處理)가 사과의 후기낙과(後期落果) 경감효과(輕減效果) 및 저장후기(貯藏後期) 과실(果實)의 품질(品質)에 미치는 영향(影響))

  • This experiment was conducted to evaluate the effect of plant growth regulators, dichlorprop and MCPB on the reduced effect of fruit drop and fruit quality before and after storage in apples. Dichlorprop was tested with dilution of 1000 at 30, 40, 50 days before harvesting, and MCPB with dilution of 4000 at 15, 25, 35 days before harvesting. The results are summarized as follows : Percentage of fruit drop was appeared to the notable reduction as compared with the untreated control when regulators was applied with dilution of 1000 at 30 days before harvesting by dichlorprop and with dilution of 4000 at 35 days before harvesting by MCPB. Degree of fruit colour showed to the remarkable promotion at all the treatment of 30, 40, 50 days before harvesting by dichlorprop as compared with the untreated control. Sugar contents in flesh was increased a little at the treatment of 30 days before harvesting by dichlorprop, but acid contents in flesh was reduced at all the treatment of 30, 40, 50 days before harvesting by dichloroprop and at 15, 25, 35 days before harvesting by MCPB. Passed firmness of fruit after storage was maintained at the treatment with dilution of 4000 at 35 days before harvesting. Therefore, it was repressed a softening of fruit, but by dichlorprop treatment at 30, 40, 50 days before harvesting, fruit firmmess was appeared to reduce according to the passage of storage period. Amount of ethylene evolution after storage was showed to reduce at all the treatment by early treated time of dichoroprop and MCPB, but carbon dioxide increased at treatment conditions such as the front. Accordingly, these relationship showed to be contrary each other.

Evaluation for the Antimicrobial, Antioxidant and Anti-thrombosis Activity of Korean Traditional Liquors (시판 전통주의 항균, 항산화 및 항혈전 활성 평가)

  • To identify the useful biological activities and the superiority in quality of Korean traditional liquors, we prepared the alcohol-free concentrates from different types of commercial traditional liquors, Takju ($T1{\sim}T3$), Yakju/Cheongju ($Y-1{\sim}Y-5$), Fruit wine (F-1) and Soju (S-1, S-2), respectively. We investigate their compositions as well as antimicrobial, antioxidant, and anti-thrombosis activity. Among the 11 traditional liquors, Y-3, Y-4, Y-5 and F-1 showed higher total-polyphenol, total-flavonoids and reducing sugars than the others. The strong antibacterial and anti-thrombosis activities were identified in Y-3, Y-4, Y-5 and F-1, and a minor antioxidant activity was found in F-l. The antibacterial activity of the Y-3, Y-4, Y-5 and F-1 alcohol-free concentrates showed a broad-spectrum, and growth inhibition was found in gram-positive, gram-negative, and ampicillin-resistant bacteria. The sequential solvent fractionation of Y-3, Y-4, Y-5 and F-1, and following analysis showed that ethyl acetate fractions of Y-3, Y-4, Y-5 and F-1 possess strong antibacterial and anti-thrombosis activity. Especially, the ethyl acetate fractions of Y-3, Y-4 and F-1 showed superior anti-thrombosis activity compared than that of aspirin. Our results suggest that the useful substances are produced from substrates and edible plant added during the fermentation, and the Korean traditional liquors could be developed as strong antibacterial and anti-thrombosis agents.

Study on Effect of Saengbal-eum-II(Shēngfà-yĭn-II)on Hair Regrowth Promotion in C57BL/6 Mice (생발음(生髮飮)-II 피부도포가 C57BL/6 마우스의 육모촉진에 미치는 효과)

  • Objectives : Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) is a hair care product which is composed of ten plant extracts used in oriental medicine. This study was carried out to investigate the effects of Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) on hair regrowth and cytokine changes in a shaving model of C57BL/6 mice. Materials and Methods : Five-week-old mice were acclimated for 1 week at a temperature between $21-23^{\circ}C$, 40-60% relative humidity, and 12h of a light/dark cycle before beginning of the experiment. There were three experimental groups including 50% ethanol (EtOH, control), a positive control of 3% Minoxidil, and 30% Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) in 50% ethanol in 18 female mice. The test compounds were topically treated once a day over 12 days. The hair regrowth was photographically and histologically determined during the experimental period of 12 days. Revelation of EGF, $TGF-{\beta}1$ and IL-6 in hair follicle were also determined using immunohistochemistry. In addition to that, IL-6, $TNF-{\alpha}$, and $IL-1{\beta}$ in skin tissue were determined using enzyme-linked immunosorbent assay(ELISA). Results : Hair regrowth in 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups was promoted earlier and faster than the control group. Concentrations of hairs and thick-hair ratio in 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups were promoted than the control group. EGF was moderately positive in hair follicle of 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups, but negative in the control group. $TGF-{\beta}1$ was not significantly difference between the groups. IL-6 in hair follicle of Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) group was negative, but weakly positive in 3% Minoxidil and control group. IL-6 and $IL-1{\beta}$ in skin tissue were significantly decreased in Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) group, but there was not significantly decreased in 3% Minoxidil and control group. $TNF-{\alpha}$ in skin tissue was significantly decreased in 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups. Conclusions : These results suggest that Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) has hair growth promoting activity and it can be used for treatment of alopecia. And these effects relate to EGF revelation of hair follicle and a decrease IL-6, $TNF-{\alpha}$, and $IL-1{\beta}$ in skin tissue.

Deposition of Atmospheric Pollutants in Forest Ecosystems and Changes in Soil Chemical Properties (대기오염물질(大氣汚染物質)의 산림생태계내(山林生態系內) 유입(流入)과 토양(土壤)의 화학적(化學的) 특성(特性) 변화(變化))

  • Environmental pollution has recently been progressed in the metropolitan and industrial areas of Korea and concerns have been evolved against the chronic effects of the pollution on natural ecosystem. This study was carried out to investigate the environmental pollution impacts on ion input into forest ecosystems and soil environmental changes. Study plots were established at Seoul, Ulsan, Yeochon, and Seosan for pollution sites and at Pyungchang for a non-pollution site. Atmospheric deposition was measured with rain, throughfall, and stem flow samples collected in the forest areas. Soil chemical properties were investigated to compare the pollution impacts on the sites. Precipitation acidity in the metropolitan and industrial areas ranged from pH 4.5 to 5.5, showing the levels lower than pH 5.8 of mountain area. Ion concentrations in the precipitation had increased significantly while passing the crown layer in the metropolitan and industrial areas, showing the increase by 4 times at the maximum. Total ion input in the metropolitan and industrial areas was greater than that in mountain area by approximately 2-3 times. Soil acidification caused by acidic ion input seemed to be greatest at Seoul, showing pH 1 decrease compared to that of Pyungchang. Soil canon contents were relatively high in the metropolitan and industrial areas. Although the canon leaching loss was not apparent, soil acidification process seemed to be continued by acidic ion input. Environmental pollution in the metropolitan and industrial areas exerted changes in ion input into the forest ecosystems and soil conditions. The chronic effects of environmental pollution should be monitored and investigated further to explain the processes of ecosystem change and the impacts on plant growth.

Effect of Concentration and Time of GA3 Treatment on Flowering and Cut Flower Yield of Limonium spp. 'Ocean Blue' ($GA_{3}$처리 농도와 시기가 스타티스 '오션 블루'의 개화와 절화수량에 미치는 영향)

  • This study was carried out to investigate effect of $GA_{3}$ foliar application on the growth and flowering of Limonium spp. 'Ocean Blue' to develop an effective $GA_{3}$ treatment method for a year round commercial production. Plants were treated with different concentrations of $GA_{3}$ 0, 100, 200, 400, and 800 mg.$L^{-1}$ in summer, and 0, 100,200, and 400 mg.$L^{-1}$ in winter at seven weeks after transplanting. The effects of $GA_{3}$ treatment time (0, 10, 20, 30, 40, and 50 days after planting) and plant age (raising during 3, 5, 7, and 9 weeks) was investigated. The $GA_{3}$ treatment advanced bolting and flowering with little difference between the two seasons. In a summer culture the effective concentration of $GA_{3}$ ranged from 200 to 400 mg. $L^{-1}$, while in winter it was found to be 400 mg. $L^{-1}$. The $GA_{3}$ treatment at 10 days after planting significantly reduced days to flowering. However, the greatest cut flower yield was observed in plants treated at 20 days after planting. The $GA_{3}$ treatment to different age plants did not show a significant effect, except in the oldest (nine weeks old plants). In conclusion, to induce early flowering in Limonium spp. 'Ocean Blue', the proper concentration and time of application of $GA_{3}$ was 400 mg.$L^{-1}$ $GA_{3}$ treated to nine weeks old plants at 20 days after transplanting.

Management Planning and Change for Nineteen Years(1993~2011) of Plant Community of the Pinus densiflora S. et Z. Forest in Namhan Mountain Fortress, Korea (남한산성 소나무림의 19년간(1993~2011년) 식생구조 변화와 관리방안)

  • This study, targeting Namhan Mountain Fortress which was designated as a No. 57 national historic site and placed on the World Heritage Tentative List in 2010, was intended to identify the change of vegetation structures by reviewing past references, pictures, research data and additionally conducting a site survey. Also, it was designed to draw up measures for restoring vegetation suitable for historically and culturally valuable Namhan Mountain Fortress. According to the biotope mapping of study site, Quercus spp. forest distributed a greatest part of area with 40.8% of $2,611,823m^2$. Pinus densiflora forest, highly likely to go through ecological succession, was dispersed in the whole region of Cheongryangsan, the area from West Gate to North Gate and the ranges between South Gate to Cheongryangsan with taking 16.5%. Pinus densiflora forest with a low probability of succession amounted to 4.7% and was dispersed mainly in the forest behind Namhansan elementary school. Pinus densiflora going on the ecological succession is distributed a portion of 2.9%. And the currently dying out Pinus densiflora forest amounted to 2.1%. As a result of analysis of the vegetation structure for 19 years, the succession from Pinus densiflora forest to Pinus densiflora and succession from Quercus spp. mixed forest to Quercus spp. forest to Carpinus laxiflora forest were predicted. Additionally, Quercus spp. expanded its dominance over time. According to the characteristics of each classified zone, the site was categorized into $553,508m^2$ area of Pinus densiflora forest area for the landscape maintenance, $114,293m^2$ area of Pinus densiflora forest area for the landscape restoration, $205,306m^2$ area of Pinus densiflora forest area for the disclimax, and $1,169,973m^2$ area of Pinus densiflora forest area for inducing ecological succession.

A Study on Ecological Characteristics and Changes of Vegetation in Hamyangsangrim (함양상림 식생의 생태적 특성 변화 연구)

  • The purpose of this study was to provide data for the basic research to found the effective conservation and management plan for the Hamyangsangrim designated as Natural Monument No. 154 and surround areas by analyzing the ecological characteristics such as landuse and actual vegetation, plant community structure, soil chemical property and also to identify the tendency of the ecosystem changes through comparison with past studies. According to analysis the results, the landuse and actual vegetation of survey site was classified as 8 types which were forest, artificial greenspace, plaza and open-space, bare ground and the pavement, water bodies, facilities, agricultural land and etc.. It was also categorized as 38 types according to detailed characteristic. 15 kinds of forest vegetation community types comprising silva were classified among them. The changes of actual vegetation showed little variation that the Quercus serrata-Carpinus tschonoskii forest was the most widely distributed in 2003(forest area ratio is 48.3%) and 2010(forest area ratio is 48.1%), whereas, the Quercus serrata forest declined a more dramatic from 11.6% to 23.2%. The Quercus serrata-Carpinus tschonoskii forest increased sharply(11.6% ${\rightarrow}$ 23.2%) and also the Quercus serrata-Zelkova serrata forest increased steadily(2.2% ${\rightarrow}$ 7.9%). The agricultural land around forest was transformed into artificial green zone. Moreover, water bodies, bare ground and the pavement of areas have increased. In addition, the urbanized area has decreased because the damaged areas inside forest have been restored to the woodlands. According to the result of typical 6 types vegetation communities change, the middle layer trees grew up to the canopy layer trees and was formed the canopy layer structure. The middle layer trees expanded their forces widely. Also new species appeared. And the shrub individuals also more dramatically increased due to the growth of shrubs. The force of Quercus serrata declined as well. However the force of Carpinus tschonoskii and Zelkova serrata expanded and increased in the forest. It must be the result of the ecological vegetation succession and environmentally soundly health recovery by influence of the projects of forest surrounding environment improvement and limitation of access to forest have managed and maintained since 2003. Those sorts of changes seems to be going to develop continuously. In the future Carpinus tschonoskii and Zelkova serrata will not be only codominant in the canopy layer but also Carpinus Tschonoskii, Zelkova serrata, Meliosma myriantha, Sapium japonicum. Styrax obassia and Acer pseudo-sieboldianum will be codominant in the middle layer. As a result, the forest's codominance species are going to be changed such like that.
