한약재 주정 추출물이 종자발아와 유묘생장에 미치는 영향 (Effect of Ethanol Extract of Herbal Medicine on Crop Growth Inhibitor)

  • BACKGROUND: Herbal extracts have been screened for their inhibitory effect of seed germination and root development on weeds, but there is a scarcity of reports for crop growth regulation. The objective of this research was to develop a growth inhibitor on Brassica campestris, and its effective extraction method from herbal medicine extract. METHODS AND RESULTS: Eighty four herbal medicine extracts were tested for their plant growth inhibition activity on B. campestris. The alcohol extracts of Artemisia annua, Cinnamomum cassia, and Mentha arvensis inhibited over 30% of germination and the extract of A. annua, and C. cassia inhibited over 70% of radicle growth at 0.1 % w/w treatment. The partially purified extracts of A. annua, and C. cassia with dichloromethane and hexane showed stronger radicle growth inhibition than the crude extracts on B. campestris. The diethyl ether extract of A. annua showed a similar 50% radicle growth inhibition ($RI_{50}$ = 45 mg/L) to its partially purified extract with dichloromethane or hexane, but the diethyl ether extract of C. cassia showed a worse $RI_{50}$ than the purified extract. CONCLUSION: The alcohol extracts of A. annua, and C. cassia showed potent radicle growth inhibition properties on B. campestris. Diethyl ether proved to be a good solvent for simple extraction from A. annua.

Antioxidant and antibacterial activities of Trema orientalis Linn: an indigenous medicinal plant of indian subcontinent

  • Methanol and aqueous extract of leaves of Trema orientalis Linn. were subjected to the potential antioxidant and antibacterial activities. The pharmacological interest of this plant coupled with traditional use (antidiarrhoeal, antiseptic, analgesic etc) prompted to test for antioxidant and antibacterial activities. The antioxidant potential of the methanolic extract was determined on the basis of their scavenging activity of the stable 1,1-diphenyl-2-picryl hydrazyl free radical. $IC_{50}$ of the methanol extract of T. orientalis was $110.25\;{\mu}g/ml$ which indicated the strong antioxidant activity of the plant. However the aqueous extract showed mild antioxidant activity. In case of antibacterial activities test, the extract was subjected for its effectiveness against both Gram-positive and Gram-negative bacteria in agar diffusion method. The zones of inhibition produced by the crude methanol and aqueous extract against few sensitive strains were measured and compared with those of standard antibiotic Gentamycin. It is evident that both extracts are active against the bacteria at low concentrations. The obtained results provide a support for the use of this plant in traditional medicine and suggest its further advance investigation.

Antifungal Activity of Nor-securinine Against Some Phytopathogenic Fungi

  • Crude extracts and active principles from medicinal plants have shown potential role in controlling plant diseases in glasshouses as well as in fields as one of the safest and ecofriendly methods. The effect of nor-securinine (an alkaloid) isolated from Phyllanthus amarus has been seen against spore germination of some fungi (Alternaria brassicae, A. solani, Curvularia pennisetti, Curvularia sp., Erysiphe pisi, Helminthosporium frumentacei) as well as pea powdery mildew (Erysiphe pisi) under glasshouse conditions. The sensitivity of fungi to nor-securinine varied considerably. Nor-securinine was effective against most of the fungi. H. frumentacei was more sensitive even at the lowest concentration ($1,000\;{\mu}g/ml$). Likewise conidia of E. pisi were also inhibited in partially or completely appressorium formation. Pre-inoculation treatment showed greater efficacy than post-inoculation in inhibiting powdery mildew development on pea plants in a glasshouse. Maximum inhibition occurred at $2000\;{\mu}g/ml$.

양치식물류의 메탄올 추출물에 항균활성 분석 (Antimicrobial Activities of Methanol Extracts Obtained from Several Ferns)

  • 10종 양치식물의 지상부와 지하부 메탄올 추출물을 대상으로 Bacillus subtilis, Escherichia coli, Listeria monocytogenes, Propionibacterium acnes, Streptococcus mutans, Streptococcus sobrinus 등 병원성 미생물에 대한 항균활성을 broth microdilution법과 paper diffusion법을 이용하여 분석하였다. 억제환이 15 mm 이상으로 나타난 추출물을 대상으로 0.125, 0.25, 0.5 1, $2\;mg{\cdot}mL^{-1}$의 농도에서 3일 동안 각 미생물의 생육 억제에 미치는 영향을 분석하였다. 그 결과, B. subtilis, E. coli, L. monocytogenes에 대해서는 꿩고비의 지하부 추출물에서 억제활성이 가장 우수하였으며, 살균처리하지 않은 조추출물 상태에서 $2\;mg{\cdot}mL^{-1}$의 농도에서 72시간 이후에도 각 39, 33, 58%의 억제활성을 보였다. P. acnes에 대해서는 꿩고비 지상부 추출물이 가장 우수한 항균활성을 보였으나, 15 mm 미만의 억제환을 보여 항균력이 약한 것으로 나타났다. S. mutans와 S. sobrinus에 대해서는 다수의 양치식물이 비교적 우수한 항균활성을 보여 양치식물의 추출물은 전반적으로 Streptococcus에 대한 감수성이 높은 것으로 생각되었다. 그러나 고비의 지하부 추출물이 $2\;mg{\cdot}mL^{-1}$의 농도에서 72시간 후 각 28, 39%, 그 외의 추출물은 10% 내외의 낮은 항균활성을 보여 10종의 양치식물 추출물 중에는 Streptococcus에 대하여 뛰어난 항균력을 보이는 추출물은 없는 것으로 생각되었다.

  • The plant species reported here are used by traditional healers in Gabon for different ailments such as wounds, malaria, fever, gonorrhea or diarrhea. The aim of this study was to evaluate the antimicrobial activities of 7 plants (Strombosiopsis tetrandra, Tetraberlinia bifoliolata, Dichapetalum barbatum, Guibourtia demeusii, Dacryodes normandii, Manniophytum fulvum, Paropsia grewoides) against different strains of both Methicillin-Resistant Staphylococcus aureus (MRSA) and Salmonella. Disc diffusion was first used to determine the antimicrobial effectiveness of the plants' ethanolic extracts. Then the minimum inhibitory concentrations of the crude extracts of either leaves or stem barks of the 7 plants were determined using broth micro-dilution. The ethanolic plant extracts showed very good activity against both MRSA and Salmonella strains where the MICs ranged from $250\;{\mu}g/ml$ to $1000\;{\mu}g/ml$. The study shows that many of the tested plants used by Gabonese traditional healers have antimicrobial activities and give support to their traditional use.

Antibacterial and Antifungal Activities of Stereum ostrea, an Inedible Wild Mushroom

  • Antibacterial and antifungal activities of liquid culture filtrate, water and ethanol extract (solid culture) of Stereum ostrea were evaluated against 5 bacteria and 3 plant pathogenic fungi. To determine the minimal inhibitory concentration (MIC), we studied $5{\sim}300\;mg/ml$ concentrations against bacteria and fungi separately. The MIC was 10 mg/ml for Bacillus subtilis and 40 mg/ml for Colletotrichum gloeosporioides and Colletotrichum miyabeanus. Liquid culture filtrate was more effective against Gram positive than Gram negative bacteria, and Staphylococcus aureus was the most inhibited (20.3 mm) bacterium. Water and ethanol extracts were effective against both Gram positive and Gram negative bacteria, and water extract was better than ethanol extract. In water and ethanol extract, inhibition zones were 23.6 and 21.0 mm (S. aureus) and 26.3 and 22.3 mm (Pseudomonas aeruginosa), respectively. For plant pathogenic fungi, the highest and lowest percent inhibition of mycelial growth (PIMG) was found 82.8 and 14.4 against C. miyabeanus and Botrytis cinerea in liquid culture filtrate, respectively. In water extract, the PIMG was found to be the highest 85.2 and lowest 41.7 for C. miyabeanus and C. gloeosporioides, respectively. The inhibitory effect of ethanol extract was better against C. miyabeanus than C. gloeosporioides and B. cinerea. Among 3 samples, water extract was the best against tested pathogenic fungi. This study offers that the extracts isolated from S. ostrea contain potential compounds which inhibit the growth of both bacteria and fungi.

Evaluation of Biological Activities of Invasive Alien Plants for Development of Functional Biomaterials

  • This study aims to confirm the possibility of using the invasive alien plants in Jeju as a functional biomaterial. To achieve this purpose, 70% ethanol extract and solvent fractions were prepared for five invasive alien plants (Hypochaeris radicata, Rumex acetosella, Humulus japonicus Siebold & Zucc., Solanum viarum, Lactuca scariolar) and their antioxidant, antibacterial anti-inflammatory and anti-obesity effects were investigated. The DPPH radical scavenging activity of ethanol extract from invasive alien plants was shown in the order of Rumex acetosella > Hypochaeris radicata > Humulus japonicus. Antimicrobial activity of ethanol extract against food poisoning bacteria (4 species) and oral cavity-induced microorganisms (6 species) was measured. As a result, the extract of Humulus japonicus showed high antibacterial effects against food poisoning bacteria (E. coli, V. parahaemolyticus) and oral microbes (L. casei, S. epidermidis, E. faecalis). In LPS-induced RAW 264.7 cells, the anti-inflammatory effect of ethanol extract from invasive alien plants was investigated. As a result, the NO production inhibition activity was highest in the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract, and the NO production inhibition activity was concentration-dependent. In addition, the Rumex acetosella and the Humulus japonicus Siebold & Zucc. ethanol extract showed a concentration-dependent inhibitory effect on cytokine (IL-6) production. These extracts also showed inhibitory activity of COX-2, an inflammatory protein. This suggests that NO production inhibition activity by the extract of invasive alien plants is the result of inhibition of iNOS and COX-2 expression. Currently, organic solvent fractions of crude extract are manufactured and the investigation of active ingredients is continuing along with evaluation of biological activity such as anti-inflammatory. These results are expected to be a major data for the study on the separation and utilization of active ingredients with antioxidant, antibacterial and anti-inflammatory effects using foreign plant crude extract and solvent fractions, and are highly likely to be applied to the development of functional food and cosmetics materials.

차광재배가 눈개승마(Aruncus dioicus var. kamtschaticus)의 성분 및 항산화 특성에 미치는 영향 (Effect of Shading Practices on the Chemical Compounds and Antioxidant in Aruncus dioicus)

  • 눈개승마의 총 엽록소, 총 카로테노이드 함량은 무차광에 비하여 차광율이 높을 수록 높았다. 지방산함량은 무차광 재배에 비하여 차광 재배 및 식용 가능시기간의 생육 후기에 감소되었다. 식용가능시기에서의 지방산 종류간의 함량은 oleic acid, palmitc acid, linoleic acid, stearic acid 순으로 높았으며, 무차광에 비하여 차광율이 높을 수록 낮았다. 특히 linoleic산의 함량은 타 지방산에 비하여 차광정도에 따른 함량의 감소율이 매우 높았다. 차광율 및 수확시기간의 휘발성 정유성분의 함량은 차광율이 높을 수록 증가하였고, 초기수확이 높았다. DPPH 소거활성은 $6.64-14.49(IC_{50}\;:\;{\mu}g/m{\ell})$로 항산화 활성이 매우 높게 나타났고 차광정도가 높을 수록 낮았고, 조기 수확 할 수록 높았다. POD 및 SOD 활성은 식물체에 비하여 종자가 매우 낮았고, 무차광에 비하여 차광 재배 시에 증가하였다. POD활성은 식용가능시기의 생육 초기에 비하여 후기에 높았으나, SOD활성은 감소되었다. POD Isozyme band 수는 이식묘의 종자, 잎 및 줄기에서 각 각 6개의 밴드를 나타내었으나, 뿌리에는 없었다. SOD Isozyme band는 송자와 잎에 비하여 뿌리에서 뚜렷하였다.

한약재 추출액의 S. aureus KCCM12256과 V. parahaemolyticus KCCM11965에 대한 항균 활성 (Antibacterial Activity of Medicinal Plant Extracts to S. aureus KCCM12256 and V. parahaemolyticus KCCM11965)

  • 천연식품 보존제의 개발과 이용에 대한 기초 자료를 확보하고자 31종의 한약재 추출물 중 S. aureus와 V. parahaemolyticus 등 식중독 유발균에 대해 우수한 항균 활성을 가지는 한약재를 선발하고, 선발된 한약재 추출물의 농도나 분획물의 항균 활성 등을 검토한 결과는 다음과 같다. 물과 메틸알콜을 용매로 추출하여 S. aureus와 V. parahaemolyticus에 대한 항균 활성을 조사한 결과, 지유, 황련 및 마치현 등이 강한 항균 활성을 가지며, 물보다는 메틸알콜 추출액의 항균 활성이 높은 것으로 나타났다. 특히 지유의 물 및 메틸알콜 추출물 모두 S. aureus 및 V. parahaemolyticus에 대하여 강한 항균 활성이 있었다. 지유 추출물은 S. aureus에 대해서는 5% 희석 농도까지 항균 활성이 인정되었으나, V. parahaemolyticus에 대해서는 10% 희석 농도까지 항균 활성이 인정되었다. 지유 추출물의 EtOAc, BuOH 및 $H_2O$ 분획물은 S. aureus와 V. parahaemolyticus에 대하여 항균 활성이 있었으며, S. aureus에 대해서는 EtOAc 분획물은 5%, BuOH 분획물은 2% 그리고 $H_2O$ 분획물은 1% 희석 농도까지 항균 활성이 인정되었으며, V. parahaemolyticus에 대해서는 EtOAc 분획물은 1% 희석 농도, $H_2O$ 분획물은 2% 그리고 BuOH 분획물은 5% 희석 농도까지 항균 활성이 있는 것으로 나타났다.

100종 네팔 식물 추출물로부터 $\alpha$-Amylase 및 $\alpha$-Glucosidase저해제의 선별 (Screening of $\alpha$-Amylase and $\alpha$-Glucosidase Inhibitor from Nepalese Plant Extracts)

  • 반추동물의 급성 산독증의 예방 및 치료제 개발 연구의 일환으로 100종의 네팔 식물추출물을 이용하여 미생물 유래의 $\alpha$-amylase 및 $\alpha$-glucosidase 저해 활성을 평가하였다. 그결과 Cedrus deodara(Roxb.) G. Don와 Myrica nagi Thunb.추출물에서 acarbose보다 강력한 $\alpha$-amylase 저해활성을 확인하였으며, 각각의 $IC_{50}$는 44.5, 47.5 및 $50.5\;{\mu}g/mL$로 확인되었다. 또한 고혈당 치료 효과가 보고된 Cleistocalyx operculatus(Roxb.) 추출물에서는 강력한 $\alpha$-amylase 저해활성 및 양호한 $\alpha$-glucosidase 저해 활성을 확인하였다. 최종 선정된 3종의 추출물은 1 mg/mL 농도까지 인간적혈구에 대한 용혈활성이 없었으며, $80^{\circ}C$, 또는 0.01N HCl 조건에서 60분간 처리시에 활성의 변화가 나타나지 않았으며, 특히 C. operculatus(Roxb.)와 M. nagi Thunb의 경우 산 처리시에 오히려 20% 정도의 저해활성의 증가가 나타났다. 이러한 결과는, 최종 선정된 3종의 추출물이 미생물 유래의 전분분해 효소의 저해를 통해 반추동물의 급성 산독증 예방 및 치료에 이용 가능함을 제시하고 있다.
