Yielding Ability and Characteristics of Collections of Local Sesame (Sesamum indicum L.) (참깨 지방수집종의 주요형질과 수량성에 관한 연구)

  • Fourteen hundred and one landraces of sesame cultivated in Korea were collected and evaluated for their major agronomic characters and yield potential under conditions of May 15 sowing, 50\timesl5cm planting density and 4-3-2 kg/l0a of N-P_2 $O_{5}$-K_2 O application in Suweon in 1977. Seventy nine percent of the collection had white seed coat color, and 29% was brown. One capsule type with two rooms-four cells was 75%, one capsule type with four rooms-eight cells was 20%, and three capsule type with two rooms-four cel1s were 5% of the varieties collected, But, two one-capsule type varieties with three rooms-six cells were also found. Days to flowering from emergence were long in the varieties collected in the central northern area, but short in the varieties collected in the southern area. Number of capsule per plant was more in the central northern varieties than in the southern varieties. Number of branch per plant was more in the one capsule type varieties with two rooms-four cells than in the three capsule type varieties with two rooms-four cells and in the one capsule type varieties with four rooms-eight cells. There were significant corelations between number of capsules per plant and grain yields in the one capsule type varieties with four cells. Grain yields had significant correlation with number of capsules per plant and number of branches per plant.

Effect of mixed plant-extract powder on the regulation of differentiation and oxidative stress-induced apoptosis in C2C12 cells (식물 추출물 혼합 분말이 C2C12 세포 내 분화 및 산화적 스트레스 유발 세포사멸 조절에 미치는 효과)

  • This study evaluated the differentiation and protective effects of mixed plant-extract powder in C2C12 muscle cells. Cells were differentiated into myotubes in 2% horse serum (HS)-containing medium with mixed plant-extract powder (MPEP) for 6 days. Treatment with MPEP increased the expression of myogenin and myosin heavy chain (MHC) protein in cells compared with non-treated cells. Differentiated cells were pretreated with MPEP, and hydrogen peroxide (H2O2). Our results revealed that treatment with MPEP before H2O2 treatment increased cell viability and decreased H2O2-induced lactate dehydrogenase (LDH) and creatine kinase (CK). In addition, MPEP attenuated H2O2-induced upregulation of Bax, downregulation of Bcl-2, and activation of caspase-9 and -3. These results suggest the MPEP can stimulate C2C12 muscle cell differentiation into myotubes and observe the protective effect of mixed plant-extract powder against muscle oxidative stress. In conclusion, MPEP may be useful as a prevention and treatment material for skeletal muscle disease caused by age-related diseases.

Inhibitory Effect of the Branch Extracts from Taxillus yadoriki Parasitic to Neolitsea sericea against the Cell Proliferation in Human Lung Cancer Cells, A549 (참식나무(Neolitsea sericea) 기주 참나무겨우살이(Taxillus yadoriki) 가지 추출물의 폐암세포 A549에 대한 세포생육 억제활성)

  • In this study, we evaluated the anti-cancer activity and potential molecular mechanism of 70% ethanol extracts of branches from Taxillus yadoriki parasitic to Neolitsea sericea (TN-NS-B) against human lung cancer cells, A549. TY-NS-B dose-dependently suppressed the growth of A549 cells. TY-NS-B decreased ${\beta}$-catenin protein level, but not mRNA level in A549 cells. The downregulation of ${\beta}$-catenin protein level by TY-NS-B was attenuated in the presence of MG132. Although TY-NS-B phosphorylated ${\beta}$-catenin protein, the inhibition of $GSK3{\beta}$ by LiCl did not blocked the reduction of ${\beta}$-catenin by TY-NS-B. In addition, TY-NS-B decreased ${\beta}$-catenin protein in A549 cells transfected with Flag-tagged wild type ${\beta}$-catenin or Flag-tagged S33/S37/T41 mutant ${\beta}$-catenin construct. Our results suggested that TN-NS-B may downregulate ${\beta}$-catenin protein level independent on $GSK3{\beta}$-induced ${\beta}$-catenin phosphorylation. Based on these findings, TY-NS-B may be a potential candidate for the development of chemopreventive or therapeutic agents for human lung cancer.

  • Chemically functionalized plant oils such as acrylated epoxidized soybean oil (AESO) and maleinized acrylated epoxidized soybean oil (MAESO) were used as new bio-based binders for $TiO_2$ electrodes of dye-sensitized solar cells (DSSC). More porous networks and larger porosities were fabricated on the $TiO_2$ films using plant oil binders due to the larger number of functionalities, in comparison with the film using polyethylene glycol (PEG). The charge-transfer resistance in the $TiO_2$ films was considerably shrunk due to the reduced impurity states. The short circuit photocurrent (Isc) and the open circuit photovoltage (Voc) of the cell using plant oil binders increased and the conversion efficiency improved significantly.

A low-pressure gene gun for genetic transformation of maize (Zea mays L.)

  • We have successfully used the low-pressure BioWare gene gun, developed for gene transfer in animal cells, for plant tissues. The BioWare device is easy to manipulate. Just 50 psi helium pressure was sufficient to transfer foreign genes into the aleurone layer and embryo of maize without causing tissue damage in the impact area. As shown by expression signals from invasive histochemical ${\beta}-glucuronidase$ (GUS) activity, the foreign reporter gene expressed well in bombarded tissues. This successful GUS-transient expression extends the application of this low-pressure gene gun from animal cells to plant tissues.

Partial Mitotic Synchronization and Giemsa G-banding in Allium wakegi

  • Hydroxyurea (HU), a DNA synthesis inhibitor, was tested as synchronizing agent in root-tip meristem of Allium wakegi. Roots were treated with 2.5mM HU for 14 h to accumulate meristem root-tip cells at the G1/S interface. After release from HU block, the cells re-entered the cell cycle with a high degree of synchrony. Synchronized mitotic frequency of A. wakegi was 22.7%, which was about 3.9 times as high as that of the control. The highest metaphase index(23.0%) was obtained when, 6 h after release from the HU block, the roots were treated with 0.05% colchicine for 2 h. Modifying various Giemsa staining protocols defined for animals and a few plant species, G-bands were visualized at prometaphase and metaphase chromosomes of A. wakegi. The higher degree of chromosome condensation, the less differential bands could be resolved. This is the first demonstration introduced partial mitotic synchronization into G-banding in plant.

  • Haploid cell obta-ined from microspores of Solanum nigrum were cultured on two kinds of medium, "Callus-inducing medium" and "Differentiation medium", in order to conduct histological studies of callus and examine differentiation of plantlets. On the callus-inducing medium the calli grew rapidly. The bulk of callus mass was light brown colored "Wet callus" covered on the surface with thin layers of rough and gleaming "White callus". The wet callus was consisted of parenchyma and meristematic tissues, while the white callus had no meristematic tissues. Large parenchyma cells, by successive divisions, became multicellular or poly nucleate cells which developed later to be meristematic tissues. The calli embedded on the differentiation medium quickly turned to dark brown color. Plantlets, however, came out later from these blackened callus mass. In the callus sectioned about ten weeks after imbedding on the differentiation medium, radially elongated tissue, concentric tissue, epidermis, tracheid-like structure, and plant jprimordia were observed.ure, and plant jprimordia were observed.

  • Chlorella cells were cultured with M4N media treated with glucose (5 mM) sucrose (10 mM) and raffinose (30 mM). Phospholipids and their fatty acid compositions were analyzed in the chloroplast isolated from cultured Chlorella cells. Growth rate was prominently raised in the treatment with raffinose. Glucose was the most excellent carbon source in the biosynthesis of total lipid, phosphatidylcholine(PC), phosphatidylethanolamine(PE), phosphatidylinositol(PI) of the chloroplast. Also, the major fatty acids were palmitic, linoleic and linolenic acid during the biosynthesis of phospholipid in the control and in the treatment with carbon sources.
