Promises and Risks of Unsaturated Volatile Organic Compounds: Limonene, Pinene, and Isoprene

  • Limonene, pinene, and isoprene are abundant and ubiquitous volatile organic compounds (VOCs) which are found in various natural products and also produced from various manufacture processes. Limonene and pinene are major components of food additives and household products for enrichment of good flavors and elimination of malodors, and isoprene is a basal motif of monoterpenes such as limonene and pinene. They have shown many beneficial effects such as chemopreventive, chemotherapeutic, and antioxidant activities. Upon certain conditions, however, adverse effects of these compounds on human health have also been reported. Although they do not seem to have acute and severe toxicity to human, they can easily generate secondary organic aerosols (SOAs) when they react with oxygen and/or ozone, which have shown certain toxic effects on experimental animal models as well as on humans. Numerous household and scented products containing limonene, pinene, and isoprene are widely used in these days. However, biological consequences upon exposure to these products are largely unknown. The aim of this review is to summarize and analyze the current understanding on the biological effects of VOCs, in particular limonene, pinene, and isoprene, as well as their SOAs.

순수 로즈마리 정유제품의 생산국별 향기성분과 항산화 활력 (Aroma Components and Antioxidant Activities of Pure Rosemary Essential Oil Goods Produced in Different Countries)

  • 국내외에서 유통되고 있는 로즈마리 정유 31점을 수집하여 GC통한 유효성분분석과 항산화 활력시험을 실시하였다. 주요 성분은 ${\alpha}$-pinene, camphene, ${\beta}$-pinene, 1.8-cineol, verbenone 및 borneol 등이 동정되었다. 주요성분에 따라 생산국별로 함량의 차이가 나타났으며, 프랑스 원산 정유의 품질이 우수하였고, 국내에서 재배하여 추출한 정유가 ${\alpha}$-pinene과 camphene의 함량이 높았다. ISO가 제시한 9가지 성분의 함량을 충족하는 제품은 총 31점 중 5점(16%)으로 낮았다. 전자공여능(EDA)은 4.8-96.0%로 현저한 차이를 보였으며, 60%이상의 항산화능 활력을 갖는 정유는 13%였다. 특정 성분 함량에 따른 항산화 활력정도의 차이는 일치하지 않았으며, 스위스 원산의 정유에서 가장 높은 항산화 활력을 보였다.

서양등골나물 휘발성 추출물의 알레로파시 효과 (Allelopathic Effect of Volatile Extracts from Eupatorium rugosum)

  • 서양등골나물의 휘발성 추출물에 포함된 terpenoid 화합물을 분리, 동정하였고, 주요 물질 4종에 대한 종자발아와 유식물 생장을 조사하였다. 서양등골나물 정유에 포함된 화합물의 수는 $\beta$-caryophyllene, $\alpha$-terpinenol, chamazulene, bornyl acetate, $\alpha$-pinene등 총 49종이었으며, 그 중 3종은 미확인되었다. 서양등골나물의 정유로부터 분리 확인한 휘발성 화학물질 중 다량 함유되어 있거나 중요한 성분으로 판명된 $\alpha$-pinene, bornyl acetate, linalool 및 terpinen-4-ol 등 4종의 물질에 대한 녹두의 발아 억제 효과를 조사하였다. linalool과 terpinen-4-ol에 의해서는 녹두의 종자가 전혀 발아하지 못하여 발아 억제효과가 매우 큰 것으로 보인다. 정유 내 화합물에 의한 녹두 유식물의 초기 생장 결과, $\alpha$-pinene과 bornyl acetate에 대한 묘조와 유근의 생장은 농도에 따라 점점 억제되었고, linalool과 terpinen-4-ol에 대해서는 실험구에서 생장 반응이 나타나지 않을 만큼 강하게 억제반응이 나타났다. 생체량에 있어서 $\alpha$-pinene의 경우 58 ${\mu}l$ 이상에서 유의한 영향을 미치고 있었으며, bornyl acetate의 경우에는 19 ${\mu}l$ 이상에서 감소하는 것으로 나타났으나 그 이후에는 통계적으로 유의한 차이를 나타내지는 않았다. 위의 결과를 종합해 볼 때, 정유 내에 포함된 서양등골나물의 알레로 화합물 중 생물검정에 사용된 $\alpha$-pinene, bornyl acetate, linalool과 terpinen-4-ol은 식물의 발아 억제 효과를 나타내는 주요한 물질 중 하나로 생각되며, 특히 linalool과 terpinen-4-ol은 다른 식물의 생장을 감소시키는 데 탁월한 효과가 있음을 알 수 있었다.

전남지역 숲길의 모노테르펜류 농도와 공기질 특성 (The Characteristics of Monoterpene and Air Quality in Ambient Air at Forested Road in Jeollanam-do)

  • 본 연구는 숲속 환경대기에서 모노테르펜류(${\alpha}$-Pinene, camphene, sabinene, ${\beta}$-pinene, myrcene, terpinolene, limonene, 1,8-cineol, linalool, camphor)의 농도분포, 숲속 대기의 청정도 및 공기음이온 분포를 파악하기 위해 전남도 내 5개 유명 숲을 대상으로 3 계절(봄, 여름, 가을) 동안 수행하였다. 4 시간 평균농도 기준으로 대나무가 우점하고 있는 죽녹원지점에서 ${\beta}$-pinene+myrcene이 958 pptv, 우드랜드와 순천만에서 ${\alpha}$-pinene(524 pptv)과, limonene(445 pptv)이 각각 높은 농도로 나타났다. 계절별로는 우드랜드와 선암사는 봄, 죽녹원과 순천만은 여름에 가장 최고치를 보였다. 대기환경기준물질 농도는 환경정책기본법의 환경기준보다 매우 낮은 농도로 나타났다. 공기음이온은 지점별로 공기 1 mL당 최고 400~3,000개로 조사되었다. 모노테르펜과 공기음이온사이에 유의한 상관은 나타나지 않았다.

소나무의 정상(正常)잎, 피해(被害)잎 및 솔방울의 테르페노이드성분(成分) 분석(分析) (Terpenoid Analysis of the Normal, Damaged Needle and Pinecone in Pinus densiflora)

  • To analyze terpene components, the essentail oil were extracted with steam distillation method from normal needle, damaged needle and pinecone of Pinus densiflora. The extracted essential oil was analyzed by gas chromatography and gas chromatography-mass spectroscopy. The results were summarized as follows; 1. Normal needles involve 43 kinds of terpene components, but damaged needles contained only 29 kinds. The most abundant components of normal and damaged needles were respectable ${\alpha}$-pinene and caryophyllene oxide. ${\alpha}$-pinene content in normal meedles amounted to 15.99 percents and caryophyllene oxide in damaged was 8.15 percents. 2. Pinecone showed 23 kinds of terpene components and among them the most abundant component was ${\beta}$-phellandrene, of which content showed 19.31 percents. 3. In normal needles, excluding ${\alpha}$-pinene, the contents of 8 kinds of other monoterpenes, reached to 48 percents of the total terpenes, 4. In damaged needles, excluding ${\alpha}$-pinene, the contents of 4 kinds of other monoterpenes, reached to 11 percents of the total terpenes. 5. In pinecone, excluding camphene, the contents of 6 kinds of other monoterpenes, reached to 58 percents of the total terpenes.

Study of Antimicrobial Activity of New Zealand's Tea Tree Essential Oil, Grapefruit Seed Extract and its major Component.

  • Manuka oil sometime named New Zealand's tea tree oil is soluble in oil and come from nature. The $\alpha$-pinene extracted from Manuka oil and R-limonene which is one of the component of extracted Citrex from Grapefruit were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion and broth dilution methods were used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus is similar when the concentration of Manuka oil and $\alpha$-pinene is $10{\mu}l$. However, Antimicrobial activity of Manuka oil for Escherichia coli, Staphylococcus aureus is better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene is low. Antimicrobial activity of Citrex is superior to that of R-limonene. The proper ratio of Maunka oil and Citrex can improve the antimicrobial activity. The proper ratio obtained from studies was 75% of Maunka oil and 25% Citrex for Escherichia coli, 25% of Maunka oil and 75% Citrex for Staphylococcus aureus.

Comparison of Aromatic and Natural VOC Concentrations in the Ambient Air

  • The ambient concentrations of benzene, toluene, and $\alpha$-/$\beta$-pinene were measured from July, 2002 to August, 2002 at the Gumsung mountain site in Jeonnam province. The mean concentration of benzene for this study was $62.6{\pm}43.9$ pptv (Min. 20.4 ~ Max. 151.2 pptv) , and that of toluene was 619.8 ${\pm}$ 330.2pptv (264.8~1, 386pptv). It was observed that the concentrations of $\alpha$ -pinene and $\beta$ -pinene were $169.0{\pm}97.7$ pptv (72.9~396.1pptv) and $11.2{\pm}27.9$pptv (7.5 ~95.9 pptv) , respectively. The ambient concentrations of aromatic and natural VOCs were found at the pptv level. In this study, the toluenelbenzene ratios were $13.9{\pm}11.2$ , but they were a little higher than those found in the domestic study and approximately 5 times greater than the values in the foreign study. The $\beta$ -/ $\alpha$ -pinene ratios ranged between 0.05 and 0.55, and this range was found to be similar in the foreign study.

Biotransformation of (-)-α-Pinene by Whole Cells of White Rot Fungi, Ceriporia sp. ZLY-2010 and Stereum hirsutum

  • Two white rot fungi, Ceriporia sp. ZLY-2010 (CER) and Stereum hirsutum (STH) were used as biocatalysts for the biotransformation of (-)-${\alpha}$-pinene. After 96 hr, CER converted the bicyclic monoterpene hydrocarbon (-)-${\alpha}$-pinene into ${\alpha}$-terpineol (yield, 0.05 g/L), a monocyclic monoterpene alcohol, in addition to, other minor products. Using STH, verbenone was identified as the major biotransformed product, and minor products were myrtenol, camphor, and isopinocarveol. We did not observe any inhibitory effects of substrate or transformed products on mycelial growth of the fungi. The activities of fungal manganese-dependent peroxidase and laccase were monitored for 15 days to determine the enzymatic pathways related to the biotransformation of (-)-${\alpha}$-pinene. We concluded that a complex of enzymes, including intra- and extracellular enzymes, were involved in terpenoid biotransformation by white rot fungi.

Antifungal Activities of Essential Oils from Glehnia littoralis Alone and in Combination with Ketoconazole

  • The antifungal activities of essential oils from the leaves of Glehnia littoralis, which is cultivated in Korea, were evaluated against pathogenic Trichophyton species by the broth dilution method and the disk diffusion test. Additionally, the effects of the oils together with ketoconazole were tested by the checkerboard titer test. The essential oil fraction and its main components showed significant inhibition of the tested Trichophyton fungi, with minimal inhibitor concentrations (MICs) in the range of 16-32 mg/ml. The results suggest that activities of this oil are based mainly on the contents of ${\alpha}-pinene$ (22.17%), the next prominent component of the oil fraction, while the first main components ${\beta}-pinene$ (57.83%) have relatively mild activity. The MICs of ${\alpha}-pinene$ and ${\beta}-pinene$ were 1-4 mg/ml and 4-32 mg/ml, respectively. Additionally the Glehnia oil fraction and its main components as well, exhibited significant synergism with ketoconazole against Trichophyton rubrum.
