Anthocyanin and proanthocyanidin contents, antioxidant activity, and in situ degradability of black and red rice grains

  • Objective: An experiment was conducted to assess the antioxidant contents and activities of colored rice grains and to evaluate their nutritive characteristics in terms of chemical composition and in situ ruminal degradation. Methods: Ten cultivars of colored rice grains (Oryza sativa L.) collected from several areas of Japan were studied, and control rice without pigment, maize, barley, and wheat grains were used as control grains. Their chemical compositions, pigment, polyphenol contents, total antioxidant capacity (TAC), and degradation characteristics were determined. Results: The starch contents of the colored rice grains were in the range of 73.5% to 79.6%, similar to that of the control rice grain. The black and red rice grains contained anthocyanin (maximum: $5,045.6{\mu}g/g$) and proanthocyanidin (maximum: $3,060.6{\mu}g/g$) at high concentrations as their principal pigments, respectively. There were significantly (p<0.05) positive relationships among the pigment contents, polyphenol content, and TAC values in the colored and control rice grains, indicating that the increase in pigment contents also contributed to the increased polyphenol content and TAC values in the colored rice grains. The dry matter and starch degradation characteristics, as represented by c (fractional degradation rate of slowly degradable fraction) and by the effective degradability, of the colored rice grains and the control rice grain were ranked as follows among commonly used grains: wheat>barley${\geq}rice$>maize. The colored rice grains also included the most-digestible starch, since their potential degradable fraction and actual degradability at 48 h incubation were almost 100%. Conclusion: Colored rice grains have high potential to be used as antioxidant sources in addition to starch sources in ruminants.

Relationship between Environmental Characteristics and Pigment Composition and Concentrations of Porphyra yezoensis Ueda in the Southwestern Coast of the Korean Peninsula (남서해역에서 양식되는 방사무늬김(Porphyra yezoensis Ueda)의 색소조성과 농도에 영향을 미치는 해양환경 특성)

  • The effect of water temperature, salinity and water column nutrient contents on pigment composition and concentration of purple lavers were studied at the main purple lavers production areas in Southwestern coast of Korea, during January to March, 2011. Water temperature was between 3.0 and $11.3^{\circ}C$. Salinity range was between 32.7 and 34.7, those were lower St. 1 and St. 6, which were at close to the seashore. Water column dissolved inorganic nitrogen (DIN), dissolved inorganic phosphorus (DIP) and silicate concentrations were $1.73{\sim}12.84{\mu}M$, $0.07{\sim}0.67{\mu}M$ and $4.93{\sim}18.29{\mu}M$, respectively. Chl a concentration was between 0.41 and $9.14{\mu}g\;L^{-1}$, and it was the highest at St. 1 during January. Photosynthetic pigment of fucoxanthin was dominant at all sites, which showed its highest concentration ($0.06{\sim}3.41{\mu}g\;L^{-1}$) at St. 1 on January. Water column DIN concentration was higher at January during low salinity period at all sites, but it was low at St. 1. Photosynthetic pigment of Chl a, PE and PC concentration of porphyra blades was between $1,173{\sim}8,124{\mu}g\;DW\;g^{-1}$, $3,281{\sim}10,076{\mu}g\;DW\;g^{-1}$, $388{\sim}1,346{\mu}g\;DW\;g^{-1}$, respectively. The concentration was relatively high at the St. 2 and St. 3. The pigment concentration of porphyra blades was higher at only Porphyra yezoensis was cultured than at Porphyra yezoensis and Porphyra seriata were cultured. The pigment concentration of porphyra blades was higher at St. 2 and St. 3 in only Porphyra yezoensis was cultured. This study shows that pigment concentration of porphyra blades may depend on habitat environment and culture methods.

Changes in Fatty Acid Composition of Dried Shellfish during Storage (패류 건제품의 저장 중 지방산 조성의 변화)

  • To evaluate lipid oxidation in dried shellfish during storage, brown pigment formation and composition of fatty acid were determined. Levels of brown pigment formation occurred at $25^{\circ}C$ were higher than those occurred at $4^{\circ}C$ throughout the storage period. Especially, the formation content of lipophilic brown pigment was 9$\~$10 times higher than that of hydrophilic brown pigment in all samples. Fatty acid composition of total lipid (TL) in fresh sea mussel and baby clam were $34.21\%$ and $32.83\%$ in saturated fatty acid, $19.50\%$ and $20,83\%$ in monoenoic acid, and $46.29\%$ and $46.79\%$ in polyenoic acid, respectively. The prominent fatty acids were 16:0, 16:1 and 20:5 in TL of the both samples. During storage, the contents of saturated fatty acid and monoenoic acid were slightly increased but that of polyenoic acid was decreased. In sea mussel, the content of polyenoic acid was higher in phospholipid (PL) than in neutral lipid (NL), while saturated fatty acid and monoenoic acid content were higher in the latter. The ratios of polyenoic acid/saturated fatty acid (P/S) in NL was similar in all samples. But the ratio of P/S in PL was higher in baby clam ($1.81\%$) than that in sea mussel($1.42\%$).

A Study of Variation Characteristics of the Phytoplankton Community by UPLC Located in the Jinju Bay, Korea (UPLC를 이용한 남해 진주만 식물플랑크톤 군집 변동특성 연구)

  • In order to provide important information for the efficient management of the identified farm ecosystem in Jinju Bay, we investigated the spatial and temporal distribution of the phytoplankton community using a UPLC pigment analysis and a CHEMTAX program from the timeframe of February 2013 to January 2014. In addition, we measured the available physical and chemical parameters controlling the distribution of the phytoplankton communities. As a result of this comprehensive pigment analysis, it was noted that the Diatoms were the predominant species with an average of 77.1% as noted located in Jinju Bay. It was discovered that during the summer season, the phytoplankton community composition was changed by a reduction of diatoms and noted increases of the Cryptophytes, Prasinophytes, and Dinoflagellates. Especially, it was noted that the Cryptophytes and Prasinophytes were shown with an average of 18.8% and 17.8% in June, respectively. However, it was revealed that the Cryptophytes and Prasinophytes were not shown by a microscopic observation. The phytoplankton community composition was correlated with the temperature and salinity variations as noticed in the Jinju Bay. Therefore, the water temperature and freshwater inputs in the Jinju Bay were important environmental factors for controlling the phytoplankton community composition and the varying Cryptophytes and the noted amounts of Prasinophytes as well.

  • Digital ink-jet printing system has many advantages such as fast and fine printing of various images, high efficiency and low cost process. Generally digital ink-jet printing requires ceramic pigments of cyan, magenta, yellow and black with thermal and glaze stability above $1000^{\circ}C$ for the application of porcelain product design. In this study, pink-red colored $CaO-SnO_2-Cr_2O_3-SiO_2$ pigment was synthesized using solid state reaction. The synthesis conditions of $Ca(Cr,Sn)SiO_5$ pigment such as annealing temperature, amount of mineralizer and non-stoichiometric composition were optimized. Crystal structure and morphology of the obtained $Ca(Cr,Sn)SiO_5$ pigment were analyzed using XRD, SEM, PSA, FT-IR and effect of Cr substitution on the pigment color was analyzed using Uv-vis. spectrophotometer and CIE $L^*a^*b^*$ measurement.

Antioxidant and Antimutagenic Activity of Solvent-Fractionated Layers of Colored Rice Bran (유색미 품종별 유기용매 추출물의 항산화성 및 항변이원성 검정)

  • Twenty-four cultivars of colored rice seeds were collected inside and outside Korea, and the antioxidant and antimutagenic activity was determined for the solvent-fractionated layers of their bran parts lipid soluble fraction, pigment containing fraction, and pigment component per se. As the serial organic solvent extraction proceeded, the overall tendency of antioxidant activities declined with increased chemical homogeneity of each fraction. This markedly showed the low antioxidativities of the pigment components from LK 1-3-6-12-1-1 and Gillimhukmi. Even all the colored rice cultivars, with considerable antimutagenic activity in 70% ethanolic extract, exhibited mutagenicity when measured with its pigment containing fraction (wx 124-163-45-7-1-1-1 and LKlB-2-1-1 being the strongest). The pigment content in each colored rice seeds decreased in the order of IR 17491-5-4-3-3>LK 1-3-6-12-1-1>LK 1D-2-12-1>RGS No.336, Elwee. In addition, a substantial difference in both chemical composition of the constituents and its amount could be found between the colored rice and cooking rice cultivars. This revealed that, compared to cooking rice, major components of organic solvent fractions from colored rice probably have long hydrocarbon chain moieties.

Changes in Nutrient and Pigment Contents of Laver Pyropia yezoensis Based on Heating Process and Storage (방사무늬김(Pyropia yezoensis)의 가열조건 및 저장기간에 따른 영양소와 색소함량의 변화)

  • Various types of dried laver Pyropia yezoensis have been produced in response to increasing demand and other laver goods manufactured using different processing methods are continuously being developed. The dried laver used in this experiment was initially heated at $165^{\circ}C$ for 3 seconds, followed by second heating at a high temperature ($340-350^{\circ}C$) to increase the storage period and enhance taste and flavor. Nutrient analysis of each sample heated under three conditions revealed that the protein and lipid contents were highest in samples from D company, while the carbohydrate contents remained relatively stable. After storage for 10 weeks at room temperature, changes in the composition were evaluated. The results showed decrease in protein (30%-49%) and essential amino acid contents. During storage, the major unsaturated fatty acids contained in dried laver slightly changed to 53.4%-56.0% in the form of EPA, while saturated fatty acids slightly increased to 18.4%-22.6% in the form of palmitic acid. The variables derived from fatty acid composition, such as atherogenic and thrombogenic, and hypocholesterolemic/hypercholesterolemic dietary indices, and polyunsaturated fatty acids/saturated fatty acids ratio, also indicated reasonable levels of stability. However, the laver should be consumed within 2 months.

Composition Analysis of Painted Pigments for the Jeoguibon (Patterns of the Queen's Ceremonial Robe) in Changdeok Palace (창덕궁 대조전 적의본의 채색안료 성분분석)

  • Damaged the Jeoguibon (patterns of the Queen's ceremonial robe) was found during repair work of Daejojeon, Changdeok Palace in 2011. Analytical study on seven colors painted in the patterns has been conducted by P-XRF and Micro-XRD analysis. The result displays that red lead, ultramarine blue, conichalcite or mixed pigment of emerald green and barium white, orpiment and white lead had been used on the patterns. Comparison of the pigments used for the Pyeseulbon (patterns of the Queen's belt ornament covering knees in the front) housed in National Museum of Korea has revealed that not only red lead but also cinnabar and organic pigment had been used in the Jeoguibon and the Pyeseulbon housed in National Museum of Korea. In addition the same pigments has been examined in green and yellow parts of both the Jeoguibon in Daejojeon and the Pyeseulbon housed in National Museum of Korea. The result has shown that western's synthetic green pigment such as emerald green was introduced and used in 1920's. The scientific result from this study expects to be used as useful referencing data for the comparative understanding on pigments used in palace during the 20th century.

The Result of the Pigment Analysis of the Mural in Ssangyeongchong (Tomb of Two Pillars) from Goguryeo (고구려(高句麗) 쌍영총(雙楹塚) 벽화(壁畫)의 안료분석(顔料分析))

  • The elements of the pigments used on the wall painting in Ssangyeongchong (Tomb of Two Pillars) from Goguryeo in the Nampo area of Pyeongyang were analyzed to confirm their mineral compositions and features of the painting. Specifically, the non-destructive X-ray fluorescence spectrometer (XRF) was used. On the other hand, the mineral composition of the background and pigment layers were analyzed using an X-ray diffractometer (XRD). The results of these analyses suggested that the lips of the characters in the painting were painted with HgS, and their faces, painted with HgS(Cinnabar/ vermilion) mixed with CaCO3. Note that lead white pigment [2PbCO3·Pb(OH)2] was found only on the bottom layer of the painting, indicating that the wall painting was likely to have been created using the Secco method.
