Effects of Sex Steroid Hormones on Differentiation of Pig Preadipocytes (스테로이드 성호르몬이 돼지 지방전구세포의 분화에 미치는 영향)

  • The current study was undertaken to determine the effects of sex steroid hormones(estrogen, testosterone and 19-nortestosterone) on differentiation and proliferation of pig preadipocytes. The preadipocytes were isolated from the backfat of new-born female pigs by collagenase digestion. 10-8M and 10-7M sex steroid hormones were treated to the cultured preadipocytes. Sex steroid hormones treated during the early stage of cell growth did not affect differentiation and proliferation of preadipocytes. However, testosterone and 19- nortestosterone treated during the late stage of cell growth stimulated differentiation of pig preadipocytes.

Ramie Leaf Extracts Suppresses Adipogenic Differentiation in 3T3-L1 Cells and Pig Preadipocytes

  • The present study was carried out to evaluate the anti-obesity effect of different concentrations of extracts of hot air-dried ramie leaf (HR) and freeze-dried ramie leaf (FR) in 3T3-L1 cells and pig preadipocytes. To analyze the effect on cell proliferation, cells were treated with $25{\mu}g/mL$ or $100{\mu}g/mL$ HR or FR extract for 2 days. Cell differentiation was evaluated by measuring glycerol-3-phosphate dehydrogenase and lipoprotein lipase (LPL) activities and intracellular triglyceride content. Treatment with either HR or FR extracts inhibited the proliferation of 3T3-L1 cells and pig preadipocytes in a dose-dependent manner. HR extract treatment inhibited the differentiation of both cell types more effectively than FR treatment. The extent of triglyceride accumulation decreased significantly in both cells following either HR or FR treatment. Furthermore, LPL activity significantly decreased after treatment with HR or FR extract. These results indicated that HR and FR extracts may inhibit proliferation and differentiation of 3T3-L1 cells and pig preadipocytes. Further studies are needed to explore the anti-obesity effect of HR and FR extracts.

Effects of Kohlrabi (Brassica oleracea var. Gongylodes) on Proliferation and Differentiation of Pig Preadipocytes and 3T3-L1 Cells (콜라비가 돼지 지방전구세포와 3T3-L1 cell의 증식과 분화에 미치는 영향)

  • The current study was carried out to determine the effects of Kohlrabi (Brassica oleracea var. gongylodes) on proliferation and differentiation of pig preadipocytes and $_3T_3-L_1$ cells. Pig preadipocytes were isolated from the backfat of the new-born pigs. Twenty-four hours after seeding, the cells were washed with DMEM/F-12 (designated day 0). To measure the cell proliferation, the cells were treated with 25 ng/ml and 100 ng/ml ethanol extracts of Kohlrabi (peel and flesh) for two days (day 0 ~ 2). To measure differentiation, the cells were treated with Kohlrabi for two days (day 0 ~ 2) and cell differentiation was measured on day 6. Twenty-five ng/ml and 100 ng/ml of Kohlrabi peel decreased proliferation of pig preadipocytes by 4.59% and 17.7%, respectively, compared with the control and Kohlrabi flesh by 11.4% and 19.2%, respectively. However, Kohlrabi did not inhibit cell differentiation. To measure the effects of Kohlrabi on proliferation and differentiation of $_3T_3-L_1$ cells, the cells were treated with Kohlrabi for two days in culture, like pig preadipocytes. Kohlrabi (both peel and flesh) did not show any effects on cell proliferation and differentiation. In summary, the results of the current study showed that Kohlrabi decreased proliferation of pig preadipocytes, but no inhibitory effects on differentiation of the cells. Kohlrabi had no effects on proliferation and differentiation of $_3T_3-L_1$ cells.

  • The current study was undertaken to determine the effect of conjugated linoleic acid(CLA) isomers, cis-9, cis-11(c9c11), cis-9, trans-11(c9t11), trans-9, trans-11(t9t11), trans-10, cis-12(t10c12) on differentiation of pig preadipocytes and myogenic satellite cells during culture. Cells were isolated from new born pigs. The t10c12 isomer decreased differentiation of pig preadipocytes(92%), but not that of myogenic cells. The t9t11 isomer decreased differentiation of preadipocytes(14%) and increased that of myogenic cells (26%). No other CLA isomers affected differentiation of preadipocytes or myogenic cells. The effects of CLA on proliferation of preadipocytes and myogenic cells were small, compared to the effects on differentiation. These results suggest that CLA isomers have different effects on differentiaton of pig preadipocytes and myogenic cells.

Effect of Retinoic Acid on Proliferation and Differentiation of Preadipocytes from Male and Female Pigs

  • The current study was undertaken to determine the effect of retinoic acid (RA) on proliferation and differentiation of preadipocytes from male and female pigs. The preadipocytes were isolated from new-born male and female pigs by collagenase digestion and washed three times one day after seeding (designated as day 0 of culture). RA was included in the media at various concentratives from day 0 to 2. The cell number was measured on day 2 with hematocytometer after trypsin digestion. Cell differentiation was determined on day 6 by measuring glycerol-3-phosphate dehydrogenase activity. RA (0.1, 1 and 10 uM) showed no effect on proliferation of preadipocytes from both male and female pigs. However, RA significantly decreased differentiation of pig preadipocytes. Degree of differentiation with 0.1 uM, 1 uM and 10 uM of RA treatment was 80%, 41% and 29% respectively, compared with control. Similar inhibitory effect was found in the female pigs; 77%, 28% and 16% respectively. It is interesting that RA treated on cell proliferation stage had no effect on proliferation but had a strong inhibitory effect on differentiation which is happening in the late stage of cell culture.

  • The current study was undertaken to determine the effects of sex steroid hormones (estrogen, testosterone and 19-nortestosterone) on proliferation and differentiation of preadipocytes of female and male pigs. The preadipocytes were isolated from the backfat of new-born female and male pigs by collagenase digestion and cultured in the $CO_2$ incubator. The concentration of $10^{-7}M$ and 10-6M sex steroid hormones were treated to the cultured preadipocytes. Regarding the effects on preadipocytes proliferation, high concentration ($10^{-6}M$) of all the three hormones increased proliferation of female preadipocytes,and only estrogen and testosterone increased proliferation of male preadipocytes. Regarding the effects on preadipocyte differentiation, all the three hormones increased differentiation of pig preadipocytes, regardless of hormone concentrations and sex of preadipocytes. The degree of stimulation of cell differentiation by sex steroid hormones was greater than that of cell proliferation.

Insulin-like Growth Factors-Ι and II Promote Proliferation and Differentiation of Cultured Pig Preadipocytes by Different Receptor-mediated Mechanisms (Insulin-like Growth Factors-Ι 과 II 는 서로 다른 수용체-매개 작용기전을 통해 돼지 지방전구세포의 증식과 분화를 촉진한다)

  • The current study was undertaken to investigate the mechanism of action of insulin-like growth factors (IGFs) on proliferation and differentiation of pig preadipocytes. The preadipocytes were isolated from the backfat of new-born female pigs and cultured in serum-deprived medium in the presence and absence of recombinant native IGFs or recombinant mutant IGFs that have reduced affinity for binding to both type-1 IGF receptors and insulin receptors. Fifty ng/ml of either IGF-I, [Leu60]IGF-I, IGF-Ⅱ or [Leu27]IGF-Ⅱ were included in the media in which preadipocytes were cultured for 4 days. IGF-I, [Leu60]IGF-I, IGF-Ⅱ and [Leu27]IGF-Ⅱ stimulated proliferation of pig preadipocytes by 39%, 8%, 25% and 2% respectively, as measured by increased numbers of cells. This indicates that both IGF-I and -II promote replication of pig preadipocytes by actions mediated either by type-1 IGF receptor or insulin receptor. IGF-I, [Leu60]IGF-I, IGF-Ⅱ and [Leu27]IGF-Ⅱ stimulated differentiation of pig preadipocytes by 50%, 17%, 37% and 30%, respectively, measured as glycerolphosphate dehydrogenase activity. Reducing the affinity of IGF-I for type-1 IGF receptors or insulin receptors significantly reduced the differentiation response. However, the differentiation response to [Leu27]IGF-II was not significantly different from the response to IGF-II. This shows that IGF-I and IGF-Ⅱ promote cell differentiation by different receptor-mediated mechanisms. IGF-II promotes differentiation of pig preadipocytes by actions that do not involve either type-1 IGF receptors or insulin receptors. These actions therefore appear to be mediated by binding of IGF-II to type-2 IGF receptors(also known as cation-independendent mannose-6-phosphate receptor[CIM6P/IGF2 receptor]). This is the first study to find evidence that IGF-II promotes differentiation of preadipocytes from any animal species by actions mediated by CIM6P/IGF2 receptors. In summary, this study shows that IGF-I and IGF-Ⅱ promote differentiation of pig preadipocytes by mechanisms that involve different cellular receptors.

Effects of Loquat(Eriobotrya japonica Lindl.) Extracts in Different Aerial Components on Differentiation of 3T3-L1 Cells and Pig Preadipocytes (비파 부위별 추출물이 3T3-L1 세포와 돼지 지방전구세포의 분화에 미치는 효과)

  • The current study was undertaken to determine the effects of the ethanol extracts of loquat (Eriobotrya japonica Lindl.) seeds, flesh or leaves on the differentiation of 3T3-L1 cells and male pig preadipocytes. The cell number was measured with the MTT assay after trypsin digestion. The cell differentiation was determined by measuring the glycerol-3-phosphate dehydrogenase (GPDH) activity and triglyceride(TG) content. No cytotoxicity was observed from the loquat flesh and leaf ethanol extracts at concentrations of 5, 10, 25, 50, 100 or $200{\mu}g/mL$ in 3T3-L1 cells and pig preadipocytes. However, the cell viability of neither cell line were affected by up $50{\mu}g/mL$ of loquat seed ethanol extract. Treatment with the loquat seed and leaf ethanol extracts significantly suppressed the terminal differentiation of both cell lines in a dose-dependent manner, as confirmed by the decrease in the glycerol-3-phosphate dehydrogenase(GPDH) activity and TG content. Treatment with the loquat seed and leaf ethanol extracts inhibited the GPDH activity and reduced the TG content of both cell types more effectively than that with the loquat flesh ethanol extract. The most potent anti-adipogenic effect was obtained in the case of the ethanol extract of loquat seeds.

Effects of Retinoic Acid on Differentiation and Gene Expression of Pig Preadipocytes (Retinoic Acid가 돼지 지방전구세포의 분화와 유전자 발현에 미치는 영향)

  • The current study was undertaken to determine the effect of retinoic acid(RA) on differentiation and gene expression of pig preadipocytes. The preadipocytes were isolated from the backfat of the new-born pigs. RA was treated to the cultured cells for 4 days and RNA was extracted from the cells. Isolated RNA went through in situ hybridization using the 14,688-gene cDNA microarray chip. Degree of cell differentiation was determined by measuring glycerol 3-phosphate dehydrogenase activity. RA decreased differentiation of pig preadipocytes by 78%. Fourteen genes were significantly up-regulated by RA, including genes known to be involved in lipid metabolism, particulary sphingomyelin phosphodiesterase, apolipoprotein R precursor, growth factor receptor-bound protein 14, retinoic acid receptor RXR gamma. However, the expression of vascular endothelial growth factor D precursor and growth hormone receptor precursor genes playing a central role in cell growth, was greatly decreased. These results suggest that RA inhibits differentiation of pig preadiocytes by regulation of gene expression of the growth factor or growth hormone receptor.
