Effect of Abelmoschus esculentus extract on anti-melanogenesis and skin barrier function (오크라 추출물의 멜라닌 생성저해 및 피부장벽개선 효과)

  • We investigated the application of functional materials by examining a variety of physiological activities of Abelmoschus esculentus extract. A. esculentus extract had a low cytotoxicity against murine melanoma B16F10 cells. At concentrations showing little or no cytotoxicity, A. esculentus extract showed high a DPPH radical scavenging activity ($ID_{50}$, $5.24{\mu}g/mL$), inhibited tyrosinase activity ($ID_{50}$, $102.12{\mu}g/mL$), and decreased melanin content ($ID_{50}$, $17.85{\mu}g/mL$). The treatment of B16F10 cells with A. esculentus extract suppressed the protein expression of tyrosinase in a dose-dependent manner. These findings suggest that A. esculentus extract inhibits melanin synthesis in murine melanoma B16F10 cells by suppressing intracellular tyrosinase expression, as well as directly inhibits tyrosinase activity simultaneously. Additionally, A. esculentus extract promotes the expression of involucrin, which is related to skin barrier protection. These results indicate that A. esculentus extract is an appropriate material for improving skin barrier function.

Physiological Function in vitro of Biopolymer from Bacillus coagulans CE-74 (Bacillus coagulans CE-74에 의해 생산된 Biopolymer의 생리적 기능성)

  • In order to prove physiological function of biopolymer from Bacillus coagulans CE-74, in vitro experiments simulating the passive membrane transport of gastrointestinal tract were carried out using dialysis membrane. And inhibition effect of isolated biopolymer on tyrosinase and angiotensin converting enzyme (ACE) were observed. The glucose retardation index after 30 min dialysis was 43.5% in the presence of 2% biopolymer. As the dialysis period became longer, the retarding effect toward glucose absorption decreased and the effect was close to zero after 5 hr dialysis. The bile acid retardation index after 30 min dialysis was 34% and 44.2% in the presence of 1% and 2% biopolymer, respectively. The effect decreased as the dialysis time elapsed. It was measured by arosinase inhibition activity of biopolymer that inhibition effect was 48.5% in $20\;{\mu}g/{\mu}l$. In a ACE inhibition activity, biopolymer showed inhibition activity as 97% in $10\;{\mu}g/{\mu}l$.

  • The effect of extracellular products (ECPs) prepared from highly virulent Edwardsiella tarda PoKF-000623 on the physiological and the immunological function in the olive flounder, Paralichthys olivaceus was evaluated. The virulence of ECPs on the fish, 5.6 g of average body weight was detected $0.714{\mu}{g}$ g $fish^{-1}$ as $96hr-LD_{50}$ by intraperitonial injection. After intramuscularly injected with 0, 4 and $40{\mu}{g}$ ECPs TEX>$fish^{-1}$ to fish average sized 59.1 g of body weight, the fish were measured the glucose and total protein concentration in sera, and immune activity of kidney macrophage. The fish injected with $40{\mu}{g}$ protein $fish^{-1}$ showed a significant increase in the glucose and total protein concentration in sera. But, the fish were changed in cellular immune response as lowed chemiluminescence response and bactericidal activity of head kidney macrophage. These results were suggested that ECPs of E. tarda could be effect on the physiological and the immunological factors, especially in the function of kidney macrophage concerning to edwardsiellosis infection.

  • To enjoy a healthy life, it is important to have a well-balanced diet. However, in today's society, there is an increase in the consumption of preprocessed foods and frequency of eating out. Also the western diet, which is becoming move popular worldwide, contains relatively high levels of protein and fat, and a low amount of fiber, Furthermore, the increased availability of favorite foods has created a condition were the individual diet is less variable. With these conditions, it is difficult to maintain a diet that is nutritionally balanced. With these unbalanced diets, which are difficult to change, there has been an increase in adult disease and health problems, such as colon and breast cancer, It is speculated that metabolites for carcinogens are produced from diet components and that intestinal bacteria contribute to the production of these metabolites. Therefore, it is necessary to evaluate the relationships between health, diet, and intestinal microflora. Soybean oligosaccharide is composed of water-soluble saccharides that have been extracted from soybean whey, a by-product from the production of soy protein. This is mainly a mixture of mono-, di-, tri-, and tetrasac-charides, with the principle components being the oligosaccharide raffinose and stachyose. When consumed by humans, the oligosaccharides cannot be digested in the human duodenal and small intestinal mucosa, and these are selectively utilized by beneficial bifidobacteria in intestines. The results of acute and subacute toxicity tests, soy-bean oligosaccharides were nonpoisonous. Soybean oligosaccharides promote the growth of indigenous bifido-bacteria in the colon which by their antagonistic effects, suppress the activity of putrefactive bacteria. Also, they reduce toxic metabolites, detrimental enzymes and plasma lipid, and increase in the frequency of bowel evacuation and fecal quantities. Consequently, soybean oligosaccharides as functional foods components have potential roles in the prevention and medical treatment of chronic adult diseases. The study of processing property and physiological function of soybean oligosacchavides and development of high oligosaccharide variety allow the creation of new and exciting foodstuffs that aye functional healthy.

Variation of Psychophysiological Characteristics Related with Human Errors during a Simple Pointing Task (단순 지적과업 중 인간과오 관련 심리생리학적 특성의 변화)

  • During a learning process, a human being is assumed to experience knowledge-based behaviors, rule-based behaviors, and skill-based behaviors sequentially if Rasmussen was right. If any psycho-physiological symptom to those different levels can be obtained, it can be useful as a measure whether a human being is fully trained and has gotten a skill in his work. Therefore, this study aimed to draw relationships between human performance measures and psycho-physiological measures while committing a computer-simulated pointing task by utilizing the power spectrum technique of EEG data, especially with the ratio of relative beta-to-alpha band power. The result showed that, during correct responses, the ratio came to stabilize as all the performance data went stable. However, response time was not a simple linear function of task difficulty level only, but a joint function of task characteristics as well as behavior levels. Comparing relative band power ratios from errors and correct responses, activated states of one's brain could be explained, and characteristics of the task could understood. To tell that of pointing task, correlations around C3, C4, P3, P4 and 01, 02 area were significant and high in correct response cases whereas most correlation coefficients went down in error cases standing for imbalance of psycho-motor functions. Though task difficulty was the only one factor that could influence on relative band power ratio with statistical significance, it should be comprehended to mean a different way of expression indicating task characteristics since at least error-some situation could be explained with the help of relative band power ratio that absolute band power failed.

Regioselective Oxidation of Lauric Acid by CYP119, an Orphan Cytochrome P450 from Sulfolobus acidocaldarius

  • Archaebacteria Sulfolobus acidocaldarius contains the highly thermophilic cytochrome P450 enzyme (CYP119). CYP119 possesses stable enzymatic activity at up to $85^{\circ}C$. However, this enzyme is still considered as an orphan P450 without known physiological function with endogenous or xenobiotic substrates. We characterized the regioselectivity of lauric acid by CYP119 using the auxiliary redox partner proteins putidaredoxin (Pd) and putidaredoxin reductase (PdR). Purified CYP119 protein showed a tight binding affinity to lauric acid ($K_d=1.1{\pm}0.1{\mu}M$) and dominantly hydroxylated (${\omega}-1$) position of lauric acid. We determined the steady-state kinetic parameters; $k_{cat}$ was 10.8 $min^{-1}$ and $K_m$, was 12 ${\mu}M$. The increased ratio to $\omega$-hydroxylated production of lauric acid catalyzed by CYP119 was observed with increase in the reaction temperature. These studies suggested that the regioselectivity of CYP119 provide the critical clue for the physiological enzyme function in this thermophilic archaebacteria. In addition, regioselectivity control of CYP119 without altering its thermostability can lead to the development of novel CYP119-based catalysts through protein engineering.

Use of Fraxnius rhynchophylla H.ance bark as antioxidant (물푸레나무 수피의 항산화 효과)

  • This study reviewed antioxidants and various enzymic physiological functions of Korean ash tree extractives that contains coumarine and ester component in order to examine the practical possibility of those materials as pharmaceutical and cosmetic additive applications. first, in the experiment of enzymic physiological activity, the electron donating ability showed 62.00% at 1ppm and over 90% at 10ppm or higher concentration, and the pseudo SOD ability showed 39% at the concentration of 2000ppm and 84.73% at the concentration of 5000ppm. Second, the inhibition activity of xanthine oxidase showed a relatively low inhibition effect of 45.57% at 200ppm, and the inhibition effect of tyrosinase showed 57.51% at 2000ppm. However, it showed a very low inhibition effect at the concentration of 500ppm or less. Third, the lipid oxidation, when Cu$\^$2+/ion was used as an oxidation accelerator, showed the inhibition effect of 82% at the concentration of 10ppm, and when Fe$\^$2+/ion was used also as the oxidation accelerator, it showed a very outstanding effect of 60% and more at 100ppm. Conclusively it is clear that the extracts of Korean ash tree bark can be used as cosmetic and pharmaceutical additive agents due to their excellent activities as antioxidant and enzymic physiological functions of it.

Effect of Restrict Feeding, Roxarsone or Its Analogues in Inducing Fatty Livers in Mule Ducks

  • This study is aimed at understanding the role of arsenic in Roxarsone in causing fatty livers in mule ducks. One hundred 10-week-old mule ducks were randomly divided into 5 groups. Ducks received 2 weeks of various treatments followed by 2 weeks of withdrawal. The treatments were non-treatment (control), 300 mg/kg Roxarsone inclusion for 2 weeks ($1^{st}$ and $2^{nd}$ week), Roxarsone inclusion for one week ($2^{nd}$ week only), restrict feeding, or Roxarsone analogue (3-nitro-4-hydroxyphenyl acid) inclusion. Results showed that feed intake and body weight in the Roxarsone groups and the restrict feeding group decreased significantly during the treatment period. However only the liver and heart weights were significantly decreased (p<0.05) in the restrict feeding group. Fatty acid synthetase (FAS) activity showed a significant decrease (p<0.05) in the Roxarsone groups and the restrict feeding group, two-week-Roxarsone treatment significantly increased NADP-malic dehydrogenase (MDH) activity compared to the restrict (p<0.05). After 2 weeks drug withdrawal, the 1-week-Roxarsone or restrict feeding group showed significantly increased (p<0.05) glucose-6-phosphate dehydrogenase (G-6-PDH) activity (p<0.05). Two-week-Roxarsone treatment significantly decreased (p<0.05) the high density lipoprotein (HDL) and increased (p<0.05) the low density lipoprotein (LDL) and very low density lipoprotein (VLDL) ratio. After drug withdrawal, the 1-week-Roxarsone or restrict feeding group showed significantly increased (p<0.05) creatine kinase (CK) activity. The 2-week-Roxarsone treatment group showed significantly increased (p<0.05) aspartate aminotransferase (AST) activity. The restrict feeding treatment group showed significantly decreased (p<0.05) total protein (TP) concentration. After drug withdrawal, the related enzyme activities in the blood that reflected the liver function were restored to the normal physiological range, except for the total bilirubin concentration and CK activity in the 1-week-Roxarsone group. This group showed a significant increase (p<0.05). Thus, the reasons for liver enlargement in the Roxarsone and restrict feeding groups were different.

Regulation of m-Calpain Activity by α-Synuclein and Its C-terminal Fragment (α-syn61-140)

  • The m-calpain activity hydrolyzing a fluorogenic substrate of N-Succinyl-Leu-Leu-Val-Tyr-7-amino-4-methylcourmarin (LLVY-AMC) was significantly stimulated by more than two-fold in the presence of 5$\mu$M $\alpha$synuclein at $15{^{\circ}C}$. The stimulation was also confirmed with azocasein. The stimulation of the peptide hydrolyzing activity required structural intactness of $\alpha$-synuclein since the C-terminally or N-terminally modified proteins such as $\beta$-synuclein, $\alpha$-syn1-97, and $\alpha$-syn61-140 did not increase the proteolytic activity. Instead, however, the N-terminally truncated $\alpha$-syn61-140 was shown to drastically suppress the calpain activity. Since the N-terminal truncation was known to be the primary cleaving event of calpain-mediated proteolysis of $\alpha$-synuclein and the $\alpha$-syn61-140 has been demonstrated to be resistant against the calpain digestion, it has been proposed that the intracellular calpain activity could be regulated in a reciprocal manner by $\alpha$-synuclein and its proteolyzed C-terminal fragment. Based on the results, a possible physiological function of $\alpha$-synuclein has been suggested as a calpain regulator which contains both stimulatory and inhibitory activities.

Study on Antitumor Activity and Immunomodulatory effects of Seoleosojong-tang (활어소종탕이 항종역반응에 미치는 영향)

  • In order to evaluate the antitumor activity and immunomodulatory effects of Seoleosojong-tang(SST), studies were done. We measured the cytotoxic activity for various kinds of cancer cells, inhibitory effect on activity of DNA topoisomerase I, cell adhesion to complex extracellular matrix, survival time in ICR bearing S-180, pulmonary colonization and histological changes of lung in C57BL/6 injected i.v. with B16-F10, CAM assay, expression of CD4/sup +/, CD8/sup +/, B220/sup +/, cytokine gene in spleen cell. The results were obtained as follows: 1. In cytotoxicity against A549, HT1080, 816-F10, NCL-H661 was showed cytotoxicity as compared with control. 2. The inhibitory effect on adhesion of A549, 816-F10 to complex extracellular matrix was over 40% at 100 ㎍/㎖ of SST. 3. In DNA topoisomerase I assay, SST has inhibitory effect. 4. The T/C% was 120.8 in SST treated group in S-180 bearing ICR mice. 5. In pulmonary colonization assay, a number of colonies were decreased significantly and histological changes were showed that infiltration area of cancer cells were inhibited effectively in SST treated group. 6. In CAM Assay, SST has antiangiogenic effect. 7. On the expression of positive cell to CD4/sup +/, CD8/sup +/ and 8220/sup +/ in spleen cells, CD4/sup +/ cells were increased significantly in SST treated group. 8. Effect of SST on IL-1β gene expression in splenic cell was significantly increased as function of whole concentration. 9. The gene expression of IL-4, IL-6, IL-10, IL-12, IFN-γ, TNF-α were increased in SST treated group. From above results SST could be usefully applied for antitumor activity and immunomodulatory effects, but further research of SST should be required.
