수소를 생산하는 미생물은 크게 광합성 세균(photosynthetic bacteria), 혐기성세균(non-photosynthetic anaerobic bacteria), 조류(algae) 등으로 구분되고, 이들의 수소 생성 기작, 사용가능기질 및 수소 발생량은 상당한 차이가 있다. 광합성세균은 Rhodospirillaceae, Chromatiaceae 및 Chlorobiaceae로 구분되며, 이는 각각 홍색비유황세균(purple non-sulfur bacteria), 홍색유황세균(purple sulfur bacteria), 녹색유황세균(green sulfur bacteria)으로 통칭된다. 혐기성 세균은 절대 또는 통성혐기세균중 일부가 수소생산에 관여하며, 조류는 녹조류(green algae)와 남조류(blue-green algae, cyanobacteria)가 알려져 있다. 생물학적 수소생산 기술은 (1) 녹조류(green algae)가 광합성 메카니즘에 의해 수소를 생산하는 직접 물 분해 수소생산(direct bio-photolysis) (2) 광합성 작용에 의해 물을 분해하여 산소를 발생하고, 동시에 공기 중 이산화탄소를 고정하여 고분자 저장물질로 균체 내에 저장한 후 혐기 발효 또는 광합성 발효에 의해 수소를 발생하는 간접 물 분해 수소생산(indirect bio-photolysis or two stage photolysis) (3) 빛이 존재하는 혐기상태 배양 조건에서 홍색 세균에 의한 광합성 발효(photo-fermentation) 또는 (4) 광이 존재하지 않는 조건에서 혐기 미생물에 의해 수소와 유기산을 내는 혐기 발효(dark anaerobic fermentation) (5) 균체 외(in virro) 수소 발생 (6) 일산화탄소 가스 전환 반응(microbial gas shift reaction)에 의한 수소 생산 기술로 구분할 수 있다. 물로부터 생물학적 기술에 의한 수소생산은 공기 중의 이산화탄소를 고정하고, 수소와 산소를 발생하는 원천기술로써 오래 전부터 미국, 유럽에서 태양에너지를 이용하는 광합성 미생물의 분리, 개선 및 반응기에 관한 연구가 축적되어 왔으며, 유기물 즉 바이오매스로부터 혐기 및 광합성 발효를 연속적으로 적용하는 기술은 비교적 최근에 일본을 비롯한 유기성 폐기물이 많은 국가에서 수소에너지 생산과 유기성 폐기물 처리라는 두 가지 목적에 부합하는 연구로써 활발히 진행되고 있다. 유기성 폐기물이나 폐수와 같은 수분함량이 높은 바이오매스는 대부분이 매립처리 되는 실정이지만 높은 수분 함량 때문에 매립 시 발생하는 침출수는 환경오염의 주범으로 가까운 장래에는 매립도 금지될 전망이다. 이와 같은 수소에너지 생산기술과 이용시스템 개발은 화석연료 사용을 최소화 할 수 있으며, 국내에서 다량 발생하는 유기성 폐기물을 이용한 에너지 생산으로 자원 강대국 입지에 설 수 있다. 미생물에 의한 수소생산 기술은 청정에너지 생산과 아울러, 동시에 산소 발생, 공기 중 이산화탄소 고정, 식품공장 폐수 및 음식쓰레기와 같은 유기성 폐기물 처리 등 환경에 이로운 방향으로 진행될 뿐만 아니라, 미생물 자체가 갖는 생물 산업성도 높아서 비타민류, 천연색소, 피부암 치료제등의 고부가가치 의약품 생산도 활성화할 수 있다.
광합성세균을 이용한 농축산가의 소규모 가축 폐수처리공정 개발을 위한 연구의 일환으로 고농도에서 유기산의 자화율이 높은 Rhodopseudomonas palustris KK14를 사용하여 돈분폐수처리공정의 최적화를 flask-scale 및 laboratory-scale에서 실시하였다. 반연속식으로 실시한 flask-scale에서 광합성세균 반응조는 HRT 6일, 광합성세균오니량 5%(v/v), 반송오니량 10%(v/v)로 했을 때 안정된 83%의 COD 제거율 및 적절한 $4{\sim}5\;g/l$의 MLSS가 유지되었다. 산생성 반응조와 광합성세균 반응조의 working volume을 각각 5.2 l와 15 l로 scale-up한 실험실적 실험에서는 원폐수 대비 COD와 BOD의 제거율이 각각 95%와 96%를 나타냈으며 광합성세균 반응조의 COD 용적부하율 및 오니부하율은 각각 $3kg\;COD/m^3/day$와 1.1kg COD/kg MLSS/day를 유지함으로써 적당한 MLSS농도가 유지되었고 부가적으로 폐수처리가 진행되면서 악취가 제거되는 효과를 나타내었다.
본 연구에서는 R rubrum을 이용한 석탄합성 가스로부터 수소 생산공정에 있어서의 세포성장 및 일산화탄소 전환을 최적화하 는 여러 조건들을 조사하였다. 그 중 pH의 영향을 살펴보면 R. rubrum 세포성장에는 pH 6~7이 최적이었고 수소생산에는 pH 7 7-7.5이 최적이었으며 pH가 5.5에서는 세포성장이거의 이루어 지지 않았다. 또한 온도가 34 'C 이상 증가되었을 때 세포성장이 둔화되어 멈추고 안정적인 co 전환속도를 얻을 수 없으므로 $30^{\circ}C$가 R. rubrum 균주 성장과 co 전환에 최적온도라 생각된다. 또한 R. rubrum은 photosynthetic bacteria인데 이 세포의 성장에 는 벚의 세기가 1,700-2,400 Lux가 최적임을 알 수 있었고 co 전환에는 계속적인 빛의 공급이 꼭 필요하지는 않고 간헐적인 빛의 노출만으로도 충분하다고 생각된다. 또한 연속반응기를 이 용하여 600 rpm, $30^{\circ}C$, pH 7에서 합성가스 체류시간 110분시 co 전환율 약 53%정도를 얻을 수 있었다. 이 연구가 계속 진 행되어져서 photobioreactor의 개발, high pressure bioreactor의 이 용, 균주의 mutatIOn 및 전환능력 우수 균주 등의 selection을 수 행한다면 매우 높은 합성가스 전환율을 갖는 생물반응기 공정개 발도 가능하리라 생각된다.

Vertebrata lanosa is an abundant and obligate red algal epiphyte of Ascophyllum nodosum that forms part of a complex and highly integrated symbiotic system that includes the ascomycete, Mycophycias ascophylli. As part of ongoing studies to resolve interactions among species in the symbiosis, we used pulse amplitude modulation fluorimetry of chlorophyll a fluorescence, from photosystem II (PSII), to measure the maximum quantum yield ($F_v/F_m$) of PSII [$QY(II)_{max}$] and relative photosynthetic electron transport rates (rETR), as a function of light intensity, in order to evaluate the photosynthetic capacity of the two algal symbionts in the field and in the laboratory under different treatments. Our primary question was 'Is the ecological integration of these species reflected in a corresponding physiological integration involving photosynthetic process?' In the laboratory we measured changes in $QY(II)_{max}$ in thalli of V. lanosa and A. nodosum over one week periods when maintained together in either attached or detached treatments or when maintained separated from each other. While the $QY(II)_{max}$ of PSII of A. nodosum remained high and showed no significant variation among treatments, V. lanosa showed decreasing performance in the following conditions: V. lanosa attached to A. nodosum, V. lanosa in the same culture, but not attached to A. nodosum, and V. lanosa alone. These results are consistent with observations in which rETR was reduced in V. lanosa maintained alone versus attached to A. nodosum. Values for $QY(II)_{max}$ in V. lanosa measured in the field in fully submerged thalli were similar to those measured in the laboratory when V. lanosa was attached to it obligate host A. nodosum. Our results provide evidence of a physiological association of the epiphyte and its host that reflects the known ecology.
Photosynthesis and respiration rate of microalgae are important factors during advanced wastewater treatment research using microalgae, There are several equipments and measurement methods for measuring photosynthesis and respiration, with different challenges that occur during pretreatment and stabilization of the analysis process. Therefore, in this study, for accurate Photosynthesis and Respiration (P&R) analyzer measurement, the analysis process was divided into pre-processing, DO stabilization, and analysis stages and each was optimized to enable accurate evaluation. For this purpose, the effect of DO saturation of the sample on P&R analysis, DO stabilization according to the degassing flow rate, and photoinhibition of the OD level on photosynthesis was investigated. Based on our study results, when DO was supersaturated, photosynthetic efficiency decreased due to photorespiration, making it inappropriate as a P&R sample. In addition, 0.5 L-N2/min level was the optimal nitrogen degassing flow rate for DO desaturation. The inhibition of photosynthetic efficiency by self-shading caused by the increase in OD was observed from OD 2.0, and it was found that P& R analysis is preferably performed on samples with OD less than 2.0. In addition, based on the above three optimization results, an optimized P&R Analyzer instruction for accurate P&R analysis was also presented.
The synthesis and excited-state dynamics are described for a tetrad (ZC - ZP - ZP - I) consisting of zinc chlorin (ZC), zinc porphyrin (ZP), zinc porphyrin (ZP), and pyromellitimide (I), which upon photoexcitation provides a fully charge-separated state (ZC$^+$- P - ZP - l$^- $) with lifetimes of 230 $\mu$s in THF and > 50$ \mu$s in DMF at room temperature via a stepwise electron-transfer relay that has been detected by the ps-time resolved transient absorption spectroscopy.
Photoinhibition and its recovery of spectrally adapted Anacystis nidulans were studied. Phycocyanin and Chl content and phycocyanin/Chl ratio were increased in cells grown under blue-green light compared with those grown in white light. Photosynthetic activities of white light and blue-green light grown cells were reduced by 50% after 15 min and 10 min of photoinhibitory light treatment (1.2 mmol·m-2s-1), respectively, largely due to the decline of PSII activities. However, their activities were recovered fully after 30 min incubation under weak light. Treatment of rifampicin and chloramphenicol magnified the photoinhibitory effects and suppressed the recovery with disappearance of susceptibility to photoinhibition and delayed the recovery process, indicating no significant differences in phosphorylation, dephosphorylation and protease activity between two cells. Therefore, it is suggested that the increased sensitivity of blue-green adapted cells might be attributed to the decline of protein synthesis, and phosphorylation-dephosphorylation of protein and protease activity might be involved in the recovery process.
Plants dissipate excess excitation energy from their photosynthetic apparatus by a process called non-photochemical quenching (NPQ). The major part of NPQ is energy dependent quenching (qE) which is dependent on the thylakoid pH and regulated by xanthophyll cycle carotenoids associated with photosystem (PS) II of higher plants. The acidification of the lumen leads to protonation and thus conformational change of light harvesting complex (LHC) proteins as well as PsbS protein of PSII, which results in the induction of qE. Although physiological importance of qE has been well established, the mechanistic understanding is rather insufficient. However, recent finding of crystal structure of LHCII trimer and identification of qE mutants in higher plants and algae enrich and sharpen our understanding of this process. This review summarizes our current knowledge on the qE mechanism. The nature of quenching sites and components involved in this process, and their contribution and interaction for the generation of qE appeared in the proposed models for the qE mechanism are discussed.
Recently, systems biology has been increasingly applied to gain insights into the complexity of living organisms. Many inaccessible biological information and hidden evidences fur example flux distribution of the metabolites are simply revealed by investigation of artificial cell behaviors. Most bio-models are models of single cell organisms that cannot handle the multi-cellular organisms like plants. Herein, a structured and multi-cellular model of potato was developed to comprehend the root starch biosynthesis. On the basis of simplest plant cell biology, a potato structured model on the platform of Berkley Madonna was divided into three parts: photosynthetic (leaf), non-photosynthetic (tuber) and transportation (phloem) cells. The model of starch biosynthesis begins with the fixation of CO$_2$ from atmosphere to the Calvin cycle. Passing through a series of reactions, triose phosphate from Calvin cycle is converted to sucrose which is transported to sink cells and is eventually formed the amylose and amylopectin (starch constituents). After validating the model with data from a number of literatures, the results show that the structured model is a good representative of the studied system. The result of triose phosphate (DHAP and GAP) elevation due to lessening the aldolase activity is an illustration of the validation. Furthermore, the representative model was used to gain more understanding of starch production process such as the effect of CO$_2$ uptake on qualitative and quantitative aspects of starch biosynthesis.
사방오리나무(Alnus firma)가 공해에 대해서 내성을 가지는 이유를 구명하기 위해서 광합성 등 생리적인 특성과 몇 가지 antioxidant 관여 효소를 분석하였다. 이를 위해서 공단이 많은 오염지역(여천산업단지)에서도 잘 자라는 사방오리나무 개체목과 피해를 받은 개체목을 각각 선발하였다. 그리고 비교를 위해서 오염원이 거의 없는 청정지역(수원)에서도 비슷한 생장을 하고 있는 개체목을 각각 1997년도에 선발하였다. 광합성능력, 기공전도도, 증산량, 엽록소함량, Rubisco, Antioxidant 효소(Superoxide dismutase(SOD) 그리고 Glutathione reductase(GR))의 활성 모두, 오염지역에서 잘 자라며 가시적인 피해를 보이지 않는 개체목의 경우 가시적인 피해가 있는 개체의 경우보다 높은 것으로 조사되었다. 이는 오염물질로부터 자신을 보호하여 정상적인 생장을 유지하기 위한 생물학적인 보상(Biological compensation)을 하는 것이라고 추정된다.
