Production of Lysophospholipid Using Extracellular Phospholipase $A_1$ from Serratia sp. MK1

  • For the efficient production of lysophospholipid the hydrolysis of phospholipid using phospholipase $A_1$ from Serratia sp. MK1 was studied in an aqueous-solvent, a two-phase and an emulsion system. Judged on the basis of productivity and the degree of hydrolysis, the yield of lysophospholipid in a two-phase system was found to be better than that obtained in an emulsion system. Among the 13 organic solvents tested phospholipase $A_1$ showed the most efficient catalytic activity and stability in butyl acetate. When 20% phospholipid was used it was completely hydrolyzed in this two-phase system.

Effect of Dietary Grapeseed Oil on Serum Lipids in Spontaneously Hypertensive Rats (포도종자유 급여가 본태성고혈압쥐의 혈청지질 성분에 미치는 영향)

  • This study was performed to investigate the effects of different fat diets on the serum lipid composition and on the morphology of liver tissue of the spontaneously hypertensive rats(SHR), Rats were fed for 17 weeks one of four fat diets, control(corn oil : beef tallow=1:1), beef tallow, grapeseed oil and soybean oil. Blood was withdrawn by heart puncture from each SHR, and used to determine the serum levels of total cholesterol, HDL-cholesterol, trigly ceride and phospholipid. No significant difference in body weight was observed in all diet groups. the total serum cholesterol was significantly lower in SHR fed grapeseed oil than that of other groups. The cholesterol lowering effect of grapeseed oil seemed to results from its high contents of phytosterol and linoleic acid. Also, serum HDL-cholesterol, triglyceride and phospholipid contents were low in all the groups. photochemical microscopic observations of the liver tissue revealed the congestion of sinosoid which is regarded as the characteristics of SHR, but no significant difference was observed among the groups.

  • The hypolipidemic effect of the exe-biopolymer (EXBP) and endo-biopolymer (ENBP) produced from a submerged mycelial culture of Ganoderma lucidum was investigated in dietary-induced hyperlipidemic rats. Hypolipidemic effects were achieved in both the EXBP- and ENBP-treated groups, however, the former proved to be more potent than the latter. The administration of the EXBP (100 mg/kg body weight) substantially reduced the plasma total cholesterol, low-density lipoprotein (LDL) cholesterol, triglyceride, phospholipid levels, and atherogenic index by 31.0, 39.0, 35.4, 28.1, and 53.5%, respectively, when compared to the control group. The EXBP also lowered the liver total cholesterol, triglyceride, and phospholipid levels by 22.4, 23.1, and 12.9%, respectively. Furthermore, the high-density lipoprotein (HDL) cholesterol and ratio of HDL cholesterol to total cholesterol were significantly increased by as much as 24.2% and 47.6%, respectively.

  • The identification of eicosapentaenoic acid (EPA) by Pseudomonas sp. CH-414 were investigated under the optimal culture conditions. The maximum dry cell weight and phospholipid production showed about 10 mg/ml and 0.6 mg/ml, respectively, at the 48 hr cultivation. The phospholipid form produced by Pseudomonas sp. CH-414 was elucidated as a phosphatidyt ethanolamine by thin layer chromatography. EPA was contained about 15% in the. extractable lipid, and the amount of EPA was about 83 $\mu $g/ml from the culture broth. Also myristic, palmitic, palmitoleic, stearic and oleic acids were identified with gas chromatography.

  • A bacterial consortium capable of utilizing a variety of polycyclic aromatic hydrocarbons has been isolated from a former manufactured gas plant site. The consortium consisted of four members including Arthrobacter sp., Burkholderia sp., Ochrobacterium sp., and Alcaligenes sp., which were identified and characterized by the patterns of fatty acid methyl esters (FAME analysis) and carbon source utilization (BIOLOG system). With the individual members, the biodegradation characteristics of aromatic hydrocarbons depending on different growth substrates were determined. FAME analyses demonstrated that microbial fatty acid profiles changed to significant extents in response to different carbon sources, and hence, such shift profiles may be informative to characterize the biodegradation potential of a bacterium or microbial community.

  • [ $^{31}P$ ] powder pattern spectra were measured to investigate the aspects of the interaction between the MLV (Multilamellar vesicle) and PMAP-23, a membrane of cathelicidin family and then CSAs(chemical shift anisotropy) were calculated to indentify the extent of perturbation of phospholipid mobility by the peptides. We found that acidic phospholipid interacts strongly with PMAP-23, and the analogues which modified to increase the amphipathic property showed that larger change of CSA. The analogue which introduced positive charge showed the same effects with amphipathic property.

Effect of Maternal Selenium Nutrition on pulmonary Selenium, Glutathione Peroxidase, and Phospholipid Levels in Neonatal Rats

  • The present study was designed to determine if prenatal and postnatal Se nutriture affects Se concentration, glutathione peroxidase(GSHPx) activity and phospholipid distribution of the neonatal rat lung. Female SD rats were bred and fed a semipurified Se-deficient(0.04ppm, Se-) or a Se-adequate(0.5ppm, Se+) diet through pregnancy and lactation. On d 2 of lactation, maternal dietary Se had no significant effect on pulmonary Se concentration of pups. On d 16 of lactation, mean milk Se concentration in Se- dams was significantly lower than that in Se+ dams. Milk Se concentration was reflected on lung Se concentration and GSHPx activity of d 16 pups, which were dramatically decreased in Se- pups. In addition, pulmonary disaturated phosphatidyl choline/total phosphatidyl choline ratio was also significantly decreased in Se- pups, implying impaired function of pulmonary surfactant. These data indicate that adequate Se nutrition is important in the maturation of neonatal rat lungs.

  • The effects of sodium molybdate(Mo) on phospholipid metabolism in sciatic nerve isolated from lead-intoxicated rat were investigated. Four weeks aged albino rats were divided into four groups : normal control group, lead(10 ppm)-treated group, Mo(1 mg/kg)-treated group, lead and Mo-treated group. As a result of the study, Mo significantly increased the rate of incorporation of $2-[^3H]$myo-inositol into polyphosphinositides in lead-intoxicated rat. Mo also increased the rate of phosphatidylcholine metabolism in normal and lead-intoxicated rats. However, Mo did not affect the metabolism of phosphatidylserine and phosphatidylethanola-mine in normal and lead-intoxicated status. These results suggest that Mo might improve lead-intoxicated status by principally enhancing the metabolism of myo-inositol-related phospholipids and by partly phosphatidylcholine in sciatic nerve.

  • Liposomes have been widely employed as biomembrane-mimetic system and drug-delivery system. In these applications, the low stability of liposomes has been the most serious problem. They have relatively short half-lives and easily lysed through interactions with biological components. This study was performed to investigate the effects of godulbaegi extracts on the fludity of phospholipid liposomes. We used dipalmitoyl phosphatidylcholine(DPPC) liposomes which make most stable liposomes among the other phosphatidylcholines. The thermograms of the DPPC liposomal bilayers incorporated with the hexane extract of godulbaegi(Ixeris sonchifolia H.) were obtained, and the enthalpy changes and the sizes of cooperative unit of the transition were calculated. The incorporation of the Ixeris sonchifolia H. into the liposomal bilayers effectively reduced the transition temperature at which the transition from gel state to liquid-crystalline state occurs, broadened the thermogram peaks, and reduced the ratio of van't Hoff to calorimetric enthalpies. These results indicate indicate that the godulbaegi extracts (Ixeris sonchifolia H.) have significant effects on the fluidity of biological membrance.

  • The purpose of this study was to determine pharmacokinetic parameters of BR-28702-2, a new antineoplastic agent which is the conjugate of nucleotide and phospholipid, and to compare them with those of ara-C. Male rats were cannulated in the left femoral vein and received a single i.v. bolus dose of either BR-28702-2 or ara-C. BR-28702-2 was also administered i.p. and plasma samples were analyzed by reversedphase HPLC. The t$_{1}$2($\beta$)/ of ara-C(1.22 hr.) was significantly smaller than that of BR-28702-2(4.420 hr.). The absolute bioavailability of BR-28702-2 after i.p. injection was 1.125%. This lower bioavailability, together with previous reports that marked antineoplastic activity was observed when given i.p., indicates that BR-28702-2 would act as a depot system to release active moieties. Further works, therefore, need to be done to characterize active metabolites.
