Preparation and Quality Characteristics of Soymilk added with Buckwheat Sprout (메밀 싹 첨가 두유의 제조와 품질 특성)

  • This study investigated the physicochemical properties and antioxidative activity of soymilk added with buckwheat sprout (SBS), and sensory properties of SBS were analyzed with control soymilk (CS). Moisture content decreased while protein, ash, and lipid contents increased according to content of buckwheat sprout. Solids content and viscosity tended to increase with addition of buckwheat sprout from 0% (CS) to 2.5% (2.5SBS), 3.5% (3.5SBS), and 4.5% (4.5SBS). SBS was found the significant pH drop from 7.08 to 6.43, 6.34, and 6.21. Suspension stability of soymilk slightly decreased with addition of buckwheat sprout. Hunter's color value L of SBS decreased while a and b values increased in comparison with CS. Rutin content was measured 23.78 mg/100 g (2.5SBS), 39.68 mg/100 g (3.5SBS), and 44.80mg/100 g (4.5SBS). Vitamin C content in SBS was higher than CS. Daidzin content increased as buckwheat sprout was added. Total phenolic content increased from 100.95 mg/100 g (CS) up to 315.71 mg/100 g (4.5SBS). For free radical scavenging activity, SBS was significantly higher than CS. In conclusion, SBS can be used as a functional food with higher amounts of rutin, vitamin C, total phenolics, and daidzin. 2.5SBS and 3.5SBS also showed better overall quality characteristics.

Sulforaphane and Total Phenolics Contents and Antioxidant Activity of Radish according to Genotype and Cultivation Location with Different Altitudes (재배지 고도에 따른 무 품종별 설포라판, 총페놀함량 및 항산화 특성)

  • Sulforaphane (SFN) and total phenolics (TPC) contents and antioxidant activity (AA) were analyzed from 13 radish genotypes (Rhaphanus sativus L.), cultivated at 3 locations with different altitudes (Gangneung: asl 5 m, Jinbu: asl 550 m, and Daegwallyeong: asl 750 m). SFN varied greatly from 0.1 to $120.5{\mu}g{\cdot}g^{-1}$ in dry weight test and was significantly affected by location ($P{\leq}0.001$), genotype ($P{\leq}0.001$) and $location{\times}genotype$ interaction ($P{\leq}0.01$). Radishes, cultivated at Daegwallyeong site, showed higher SFN than those of other locations. Among different genotypes, the root of 'Black radish' and leaves of 'Purunmu' of Daegwallyeong had the highest SFN (107.8 and $120.5{\mu}g{\cdot}g^{-1}$, respectively). TPC in root was affected by genotype ($P{\leq}0.001$), and $location{\times}genotype$ interaction ($P{\leq}0.01$), but not by location. In leaves, TPC was affected by location ($P{\leq}0.01$), genotype ($P{\leq}0.001$), and $location{\times}genotype$ interaction ($P{\leq}0.001$). AA expressed as electron donating ability was significantly influenced by location, genotype and $location{\times}genotype$ interaction and correlated positively with TPC ($Pearson's$ $r$=0.897) in root. These results suggest that radish could be a good source of functional food and high altitude location such as Daegwallyeong has potential for the production of radish with high content of health promoting factors.

Protective Effect of Ethyl Acetate Fraction from Hibiscus Sabdariffa L. Extract against High Glucose-induced Oxidative Stress (고포도당으로 유도된 산화 스트레스에 대한 로젤 아세트산에틸 분획물의 신경세포 보호효과)

  • To investigate the physiological effect of Hibiscus sabdariffa, in vitro antioxidant activities and neuroprotective effects against high glucose-induced oxidative stress were examined. The ethyl acetate fraction (EtOAc-Fr) from H. sabdariffa contained high total phenolic contents compared with other fractions but total anthocyanin contents were lower than 80% Ethanol extract showed the highest 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging activity and malondialdehyde inhibitory effect. Furthermore, the EtOAc-Fr decreased the intracellular reactive oxygen species level, and protected the neuron-like PC12 cells from high glucose-induced cytotoxicity. The EtOAc-Fr also presented inhibitory effects against acetylcholinesterase as an acetylcholine hydrolase enzyme. Finally, chlorogenic acids as main phenolics by high performance liquid chromatography analysis.

Nutritive Evaluation of Some Browse Tree Legume Foliages Native to Semi-arid Areas in Western Tanzania

  • Browse tree legume leaves from Acacia spp (A. nilotica, A. tortilis, A. polyacantha), Dichrostachys sp, Flagea villosa, Piliostigma thonningii, Harrisonia sp were evaluated for nutritive potential (chemical compositions and degradability characteristics) compared to Gliricidia sepium. Effect of tannins anti-nutritive activity on digestibility was also assessed by polyethylene glycol (PEG) tannin bioassay. Crude protein (CP), ash, neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) differed (p<0.05) between legume foliages. Mean CP, ash, NDF, ADF and ADL for fodder species tested were 158, 92, 385, 145, and 100 g/kg DM, respectively. CP ranged from 115 (P. thonningii) to 205 g/kg DM (G. sepium). Acacia spp had moderate CP values (g/kg DM) of 144 (A. nilotica), to high CP in A. tortilis (188) and A. polyacantha (194) comparable to G. sepium. The forages had relatively lower fiber compositions. A. nilotica had (p<0.05) lowest NDF, ADF and ADL (182, 68 and 44) compared to P. thonningii (619, 196 and 130) g/kg DM, respectively. Except G. sepium, all fodder species had detectable high phenolic and tannin contents greater than 5% DM, an upper beneficial level in animal feeding and nutrition. Mean total phenolics (TP), total tannins (TT) and condensed tannins (CT) (or proanthocyanidins) for fodder species tested were 139, 113 and 43 mg/g DM, respectively. F. villosa had (p<0.05) lowest TP and TT of 65 and 56 mg/g DM, respectively, compared to A. nilotica (237 and 236 mg/g DM, respectively). The CT varied (p<0.05) from 6 (F. villosa) to 74 mg/g DM (Dichrostachys sp). In vitro organic matter (OM) degradability (OMD) differed (p<0.05) between fodder species. G. sepium had (p<0.05) high degradability potential compared to A. polyacantha that had (p<0.05) the lowest OMD values. Forage degradability ranked: G. sepium>A. nilotica>P. thonningi>F. villosa>Dichrostachys sp>A. tortilis>A. polyacantha. Addition of PEG resulted to (p<0.05) improvement in in vitro OM digestibility (IVD). Increase in IVD was mainly due to binding action of PEG on tannins; and represents potential nutritive values previously depressed by tannins anti-nutritive activity. Browse fodder has potential as sources of ruminal nitrogen especially for ruminants consuming low quality roughages due to high protein, lower fiber compositions and high potential digestibility. However, utilization of browse supplements in ruminants is hampered by high phenolic and tannin contents. Deactivation of tannin anti-nutritive activity, possibly by feeding tanniniferous browse with other readily available nitrogen sources to dilute tannin anti-nutritive activity could improve utilization of browse fodder supplements. Further studies are needed to assess browse fodder palatability and intake, and their effect on growth performance in ruminants.

A Study on the Functional Characteristics of Cuscuta australis R.Brown (실새삼(Cuscuta australis R.Brown)의 기능적 특성에 관한 연구)

  • To investigate the usefulness of natural plants, physicochemical characteristics, functional components and physiological activities of dodder(Cuscuta australis R.Brown) were experimented. The extraction of dodder was carried out at 60℃ in shaking bath with 10 times of distilled water for three hours. Harmful metals such as lead, cadmium and arsenic were not detected, but the content of mercury from dodder was 0.0004ppm in acceptable standards. The extraction yield was 23.45%, total content of phenolics equivalent to gallic acid was 699.8 mg/100g and total content of flavonoid equivalent to naringin was 373.8 mg/100g. 28 kinds of free amino acids were detected with the contents of 252mg/100g in the dodder extract, and the contents of anserine and sarcosine were analysed highest in all amino acids. From the comparison of physiological activities of dodder extract with those of ascorbic acid, total reducing power, electron donating ability based on DPPH radical scavenging and ferric reducing antioxidant power was equivalent to 88%, 56% and 40%, respectively, of ascorbic acid. The nitrite scavenging activity of dodder extract was 78% of ascorbic acid on pH 1.5, but the antioxidant index of dodder on perilla oil was about 4 times greater than that of ascorbic acid.

Characterization of flaxseed and flaxseed oil as edible oil resources (아마인 및 아마인유의 유지자원으로서의 특성)

  • The objective of this research was to find out the potential value of flaxseed as a dietary supplement as well as an edible oil resource. The characteristics of yellow flaxseed oil and brown flaxseed oil were compared to check which oil is better in the aspects of cooking purpose and of nutritional value. The quality of flaxseed oil was evaluated based on the composition of fatty acid, the content of phenolic compounds and the anti-oxidant activity. The total phenolics of yellow flaxseed oil and brown flaxseed oil were $10.78{\pm}0.46$ and $29.88{\pm}3.25mg/100g$, respectively. Their ${\gamma}$-tocopherol contents were 20.59 and 17.94 mg/100 g, respectively. Contents of linolenic acid were 56.60 and 31.38% and oleic acid were 18.24 and 39.16 %, respectively. Yellow flaxseed oil showed higher ratio of unsaturated fatty acid than brown flaxseed oil. However, brown flaxseed oil showed higher electron-donating abilities than brown flaxseed oil, which might be due to its higher content of phenolic compounds. In conclusion, flaxseed has a great potential as a good edible oil resource due to its high content of unsaturated fatty acid and anti-oxidant activity.

Antioxidative and Anti-Diabetes Activity, and Free Amino Acid and Mineral Contents of Beverage with Gugija (Lycii fructus) Extracts (구기자 추출물 함유 음료의 산화방지활성, 항당뇨효과, 유리아미노산과 무기질 함량)

  • Free amino acid and mineral analysis of beverages with Gugija (Lycii fructus) extracts performed to determine the antioxidative and anti-diabetes activities of the beverages. The four major free amino acids in the water- or ethanol-extracts of the Gugija beverages were asparagine (115.23, 51.95 mg%), methionine (20.02, 22.07 mg%), aspartic acid (19.65, 13.72 mg%) and taurine (18.64, 22.44 mg%). The mineral contents of the Gugija beverages with water- and ethanol-extracts were in the order K>Mg>Na>Ca>Zn>Fe. Antioxidant activity determined by DPPH and ABTS radical scavenging activity. There was no significant difference in DPPH and ABTS radical scavenging activities between the water-extract beverages and the ethanol-extract beverages, although more phenolics and flavonoids were found in the ethanol-extract beverage. Higher anti-${\alpha}$-glucosidase activity was observed in the ethanol-extract beverage compared to acabose, which was used as the control and is an inhibitor of ${\alpha}$-glucosidase, suggesting that the Gugija beverage with ethanol extract could be a potential hypoglycemic agent.

Biological Activities and Total Phenolic Content of Ethanol Extracts of White and Flesh-colored Solanum tuberosum L. Potatoes (일반감자와 유색감자 에탄올 추출물의 생리활성 및 폴리페놀 함량)

  • This study was aimed to evaluate and compare the biological activities and total phenolic content of ethanol extracts of white (Superior) and colored potatoes (Red, Rose, Haryoung, Blue, Jaseo, and Jasim). Various ethanol extracts (250, 500, 1,000, and $2,000{\mu}g/mL$) were assessed for antioxidant activities of DPPH free radicals and hydroxyl radical scavenging. The cytotoxic activity of the extracts (0, 30, 60, $100{\mu}g/mL$) was also carried out by a human cancer cell lethality bioassay method. Results showed that the DPPH and hydroxyl radical scavenging activities, of all extracts increase in a concentration-dependant manner. All color-fleshed potato extracts, except for Red and Haryoung, showed higher radical scavenging activities than the general white potato extract. Cytotoxicity of the extracts as determined in THP-1 cells showed that Blue and Jasim was highly toxic at most concentrations tested. In addition, the Blue extract had the highest cytotoxic activity on $HepG_2$ cells at the concentration $100{\mu}g/mL$. The contents of total phenolic (TPC) in the extracts significantly varied from 120.0 mg/100 g powder (Superior) to 151.1 mg/100 g powder (Blue), and TPC of all color-fleshed potato extracts was higher than that of white. The greatest positive correlation was found between phenolic contents and hydroxyl radical scavenging activity (r=0.776, p<0.05), and a strong negative correlation was found between hydroxyl radical scavenging activity and cytotoxic activities on $HepG_2$ cells (r=-0.795, p<0.05). The present study demonstrates that color-fleshed potatoes have higher antioxidant and stronger anticancer activities than general potato. These results provide useful and important information for potato breeders and researchers, in order to increase the antioxidant capacity and functional value of colored potatoes for use in the food and pharmaceutical industries.

Comparison of Physicochemical Properties between Extracts and Soaked Liquor of Lespedeza cuneata G.Don (비수리(Lespedeza cuneata G.Don) 추출물과 담금주의 이화학적 특성비교)

  • Lespedeza cuneata (LC) is a herbaceous plant that grows throughout Korea. To investigate the physicochemical properties and the antioxidant activities of LC extracts and LC-soaked liquor (s. liquor), extraction with distilled water (DW) and ethanol was conducted and the extracts were compared with s. liquor. A total of 22 free amino acids from LC extract were detected, with the main ones being phosphoethanolamine 20.36>L-proline 18.02>ammonia 14.48>L-aspartic acid 12.96>${\gamma}$-amino-n-butyric acid 10.67%. The total flavonoid contents (TFC) and total phenolics contents (TPC) were in the order of s. liquor>DW>ethanol extract. The electron donating ability based on DPPH radical scavenging ability was highest for s.liquor, which equivalents 81.4% ascorbic acid in the order of s. liquor>ethanol>DW extract. The orders of ferric reducing antioxidant power and ABTS radical scavenging ability were proportional to the TFC and TPC of extracts, and in the order of s. liquor>ethanol>DW extract. The nitrite scavenging ability of s. liquor was highest among the three extracts (96.6% at pH 1.2), which was 2.6 times greater than that of DW extract.

Antioxidative Activity of Cheonggukjang Prepared with Purple Sweet Potato (자색고구마 첨가 청국장의 항산화능 평가)

  • The characteristics and antioxidative activities of yellow and black soybean cheonggukjang prepared with the addition of purple sweet potato (Ipomoea batatas cv. Jami) were evaluated. In sensory evaluation experiments involving the addition of purple sweet potatoes to cheonggukjang at 0, 5, 10, 20, and 30% ratios (w/w), yellow (YCAP) and black (BCAP) soybean cheonggukjang with 20% purple sweet potato were most preferred. The total phenolics and flavonoid contents of YCAP and BCAP were higher than those of yellow (YC) and black (BC) cheonggukjang prepared without added purple sweet potato. YCAP and BCAP also showed higher DPPH radical-scavenging activities than other samples. Moreover, rat blood plasma samples taken 1 h after oral administration of YCAP and BCAP showed higher inhibitory effects against lipid peroxidation than did rats fed YC or BC. These results suggest that the long-term intake of cheonggukjang prepared with purple sweet potato may increase the antioxidative activity in the blood.
