Evaluation of antioxidant and cancer cell growth inhibition activities of red rice and black rice (적미와 흑미의 항산화 활성 및 암세포성장 억제 효과)

  • The extraction yield, total phenolics content, antioxidant activity, cancer cell growth inhibition (A549 and MCF 7), and lung cancer cell (A549) viability of red rice and black rice were investigated, to evaluate the functional properties of colored rice. The extraction yields and the total phenolics contents of the rice cultivars were Heugseol > Heugkwang > Hongjinju > Jeogjinju > Ilpum. Also, the DPPH radical scavenging activity of the black rice did not differ from that of the red rice, whereas its activity among its cultivars was Jeogjinju > Heugseol > Heugkwang > Hongjinju > Ilpum. The ABTS antioxidant activity of the black rice (Heugkwang, Heugseol) was greater than that of the red rice. The total phenolics was partly attributed to its high antioxidant. On the other hand, the effect of the red rice on the lung cancer cell (A549) viability was higher than that of the black rice. The breast cancer cell (MCF 7) growth inhibition activity of the black rice did not differ from that of the red rice. Our results indicate that the ABTS antioxidant activity of black rice is better than that of red rice, and the lung cancer cell (A549) viability of red rice is better than that of black rice.

Effect of Sulfur Fertilization Method on Quality of Safflower Seed (황 시용방법에 따른 잇꽃 종실의 품질)

  • This study was carried out to clarify the effect of sulfur fertilization method on the qualities of safflower (Carthamus tinctorious L.) seed. Sulfur application increased the nitrogen and sulfur content of seed. In nitrogen content, there was no significant difference among sulfur fertilizers, whereas sulfur content was increased by raising ammonium sulfate (AS) amounts by soil application. In application method of AS, there was no significantly difference between foliar application and soil application with 8 kg S/10a. N/S ratio in seed was decreased by sulfur application, and also decreased with the increase of AS amounts by soil application. The contents of lipid and protein were increased by sulfur application. Lipid content was highest in foliar application, and protein content was lowest in AS 4 kg S/10a. By sulfur application, the content of total phenolics and electron donating ability (EDA) were increased by $3.1{\sim}4.7%$ and $3.6{\sim}8.5%$, respectively compared with control. In content of total phenolics, there was no significant difference among sulfur fertilizers, but EDA was higher in sulfur powder 20 kg/10a and AS 8 kg S/10a than that in other fertilizers. Sulfur application showed positive effects on the qualities of safflower seed. In application effects, AS and foliar application were more efficient than sulfur powder and soil application, respectively.

Antioxidant Activities of Various Solvent Extracts from Canola Meal (추출용매별 canola meal의 항산화 활성)

  • This study was performed to investigate antioxidant activity of various solvent extracts (methanol, ethanol, acetone and water) from canola meal. As the concentration of all extracts increased, antioxidant activities increased gradually. Among solvent extracts from canola meal, methanol extract showed the highest antioxidant activities with $EC_{50}$ values of 0.9, 3.5, 1.3, and 8.5 mg/mL for DPPH radical scavenging ability, ABTS radical scavenging ability, reducing power, and chelating ability, respectively, and it had the highest total phenolics and total flavonoids contents. Furthermore, methanol extract was fractionated into three fractions (MF 1, MF 2, and MF 3) using Sephadex LH-20 column. The highest antioxidant activity was found on MF 2 for DPPH radical scavenging ability, on MF 3 for ABTS radical scavenging ability and reducing power, and on MF 1 for chelating ability at the same concentration of 1.5 mg/mL. The results indicated that each fraction containing various antioxidant materials seemed to have different influence at each antioxidant activity.

Optimization of Extraction of Marker Compounds from Red Ginsengs by Accelerated Solvent Extraction Using Response Surface Methodology (반응표면분석법을 사용한 가속 용매 추출에 의한 홍삼 지표성분의 추출 최적화)

  • A new method based on accelerated solvent extraction (ASE) combined with response surface methodology (RSM) has been developed for optimization of the extraction of ginsenoside [Rb1, Rg1, and Rg3(20S)], total phenolics, and benzopyrene in red ginseng. The RSM method, based on a five level and two variable central composite design, was employed to obtain the optimal combination of extraction conditions. In brief, ginsenosides Rb1, Rg1, and Rg3(20S) and total phenolics with undetectable benzopyrene were optimally extracted with 50% ethanol as an extraction solvent, extraction temperature of $158^{\circ}C$, extraction time of 20 min, extraction pressure of 2,500 psi, flush volume of 60%, and one extraction cycle. The contents of ginsenosides and total phenolics in red ginseng extracted by ASE under optimum conditions were significantly higher than those extracted by sonication and reflux extraction.

Anti-oxidative activity of the herb mixture prescribed to induce blood glucose level and effect on the differentiation of 3T3-L1 fibroblast (혈당저하를 위해 처방된 천연물 복합제의 항산화 활성 및 3T3-L1 세포의 분화에 미치는 영향)

  • We prepared five different kinds of herb mixtures prescribed for hypoglycemic effect. And the physicochemical properties of their water extracts were assessed to identify functional materials. Yields were in the range $19.52{\sim}29.79%$. Total phenolics and flavonoid contents were $349.24{\sim}1,752.21\;mg%$ and $163.06{\sim}1,118.47\;mg%$, respectively, and herb mixtures No. 2, 3 and 5 showed particularly high levels greater than 1,000 mg%. Electron-donating ability was best in herb mixtures showing high levels of total phenolics and flavonoids. Nitrite-scavenging abilities were more than 70% in herb mixtures No. 2 and 5, and decreased as pH increased. Herb mixture extracts strongly inhibited differentiation of 3T3-L1 fibroblasts, with potencies ranked in the herb mixture order 5, 1, 4, 3, and 2. The five different kinds of herb mixtures prescribed for their hypoglycemic effects may be useful as functional food materials.

Optimization of β-Glucan Extraction Process from Rice Bran and Rice Germ Using Response Surface Methodology (미강과 배아로부터 β-glucan의 추출조건 최적화 및 기능성 생리활성)

  • This study was investigated on optimal conditions of the functional activities of ${\beta}$-glucan which was extracted from rice bran (RB) and rice germ (RG) using response surface methodology. The extraction temperature was varied in the $80-100^{\circ}C$, the extraction time between 2-10 min, and the ethanol concentration was in the interval of 30-70%. A central composite design was applied to investigate the effects of independent variables of extraction temperature ($X_1$), extraction time ($X_2$) and ethanol concentration ($X_3$) on dependent variables such as electron donating ability of RB ($Y_1$), electron donating ability of RG ($Y_2$), total phenolics of RB ($Y_3$), total phenolics of RG ($Y_4$), ${\beta}$-glucan contents of RB ($Y_5$) and ${\beta}$-glucan contents of RG ($Y_6$). As a result, the highest $Y_1$ level was 84.02% at $92.60^{\circ}C$, 2.75 min and 60.41% in saddle point. This value was affected by extraction temperature (P<0.05). The value of $Y_2$ was found to be the highest at $87.52^{\circ}C$, 2.23 min and 54.40% in saddle point. The highest $Y_3$ level was $98.56^{\circ}C$, 6.69 min and 40.26% in saddle point, and this extraction was greatly influenced by extraction temperature (P<0.01) and ethanol concentration (P<0.05). The value of $Y_4$ was found to be highest at $95.73^{\circ}C$, 9.19 min and 53.67% in minimum point. The value of $Y_5$ was found to be the highest at $96.23^{\circ}C$, 7.70 min and 63.69% in saddle point. The value of $Y_6$ was found to be highest at $87.82^{\circ}C$, 2.10 min and 50.03% in minimum point, and this extraction was greatly influenced by extraction time (P<0.01).

Antioxidant and Cytotoxic Potentials of Methanol Extracts from Taraxacum officinale F. H. Wigg. at Different Plant Parts (서양민들레 부위별 추출물의 항산화 및 세포독성 연구)

  • A laboratory experiment was conducted to determine the content of phenolics and flavonoids, antioxidant activity and cytotoxicity from methanol extracts of different plant parts of $T.$ $officinale$ F. H. Wigg. Total phenolics [mg chlorogenic acid equivalents (FAE) $kg^{-1}$ DW] was highest in flower extracts (72.0 mg $kg^{-1}$), followed by leaf, root, and stalk extracts of $T.$ $officinale$ ($p$ < 0.05). The result of total flavonoid level [mg naringin equivalents $kg^{-1}$ DW] had same tendency to differential total phenolics contents among plant parts, but showed lower ranges of amount. The antioxidant activity of the methanol extracts from all the plant parts dose-dependently increased. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was highest in flower extracts ($IC_{50}$ value = 624.3 mg $kg^{-1}$ ), and followed by leaf, root, and stalk extracts of $T.$ $officinale$ ($p$ < 0.05). By means of MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, cell viability of Calu-6 for human pulmonary carcinoma and SNU-601 for human gastric carcinoma showed the lowest $IC_{50}$ value in the flower extracts ($IC_{50}$ value = 85.7 and 311.4 mg $kg^{-1}$, respectively), indicating the highest cytotoxicity. The results suggested that total phenolics content and total flavonoids level in different plant parts of $T.$ $officinale$ were highly correlated with antioxidative ($r^2$=0.7280 to 0.9971) or with cytotoxic activities ($r^2$=0.5795 to 0.9515).

Effects on antioxidant, whitening, and anti-wrinkle improvement of ethanol extracts from fermented Trigonotis radicans var. sericea by Lactobacillus brevis (Lactobacillus brevis 발효가 참꽃마리(Trigonotis radicans var. sericea) 에탄올 추출물의 항산화, 미백 및 주름 개선 활성에 미치는 영향)

  • This study was designed to compare the effects on antioxidant, whitening, and anti-wrinkle improvement of 60% ethanol extracts from Trigonotis radicans var. sericea (TR) and Lactobacillus brevis-fermented T. radicans var. sericea (FTR). The total phenolics contents of TR was 18.6 mg/g, and FTR was 18.2 mg/g. The HPLC analysis confirmed more phenolic compounds were produced in FTR by bioconversion than in TR. The FTR showed higher 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity than TR. The results of the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation decolorization assay, FTR and TR showed higher activity than BHT (positive control). The FTR (1.21-1.29 PF) showed higher antioxidant protection factor (PF) activity than the TR (1.15-1.22 PF) and showed higher tyrosinase, collagenase and elastase inhibitory activities. This results showed L. brevis -fermentation of Trigonotis radicans var. sericea changed the composition profile and has effects on antioxidant, whitening and anti-wrinkle improvement.

General Characteristics of Korean Propolis

  • The propolis was collected from the whole part of Korea Peninsula, such as central, southern, and Jeju island, to analyze the general composition including total flavonoid and phenolic contents, heavy metals, colors, amino acids, and crude lipid. The total flavonoid and phenolic contents of middle region in Korea Peninsula (4.26% and 13.06 g/100 g gallic acid) were higher than southern region (2.53%, 10.9 g/100 g gallic acid) and Jeju island (0.03%, 7.22g/100 g gallic acid), respectively. The heavy metals contents showed that Zn contents were found in $0.44{\pm}0.2ppm$ and a harmful heavy metal such as Cr, Cd, Pb, Ni, Cu, As, and Hg were not detected at all in our experiment. The colors of extracted propolis showed a various spectrum from yellow to red purple. Various amino acids were also detected as proline, valine, methionine and the average crude lipid contents of propolis were 42.4%. All data collected in South Korea were very first executed in our institution for the purposes of utilizing and developing the industrial insects, honeybees as well as silkworm.

Quality Characteristics of Gastrodia elata Extract Glutinous Rice Porridge Jochung and Principal Component Analysis of Antioxidant Activity (천마추출액을 이용한 찹쌀죽 조청의 다변량 분석기법을 통한 항산화 활성 및 품질 특성)

  • This study was performed to investigate the effects of Gastrodia elata extract on the quality characteristics of glutinous rice porridge Jochung saccharide by barley malt. Glutinous rice porridge Jochung contatining 0%, 5%, 10%, 15%, 20% of Gastrodia elata extract were produced. Soluble solid contents and pH increased as content of Gastrodia elata extract increased. Free acidity contents were 24.87 meq/kg (control) and 25.67~42.03 meq/kg (GR2~GR5) (p<0.001 among samples). There was significant total phenolics and TEAC obtained for antioxidant activity of Gastrodia elata extract Jochung samples. Increased the ratio of Gastrodia elata extract in Jochung tended to decrease the color value. Results show Jochung containg less than 15% Gastrodia elata extract resulted in the highest scores for quality characteristics and sensory evaluation. Correlations between antioxidant activity and parameters were found to be statistically significant.
