의약품 원료 DIET 합성 중 H2O2를 이용한 붕소제거 반응공정에서의 폭주반응 위험성 평가 (Hazard Evaluation of Runaway Reaction in Deboronation Process Using H2O2 in DIET Synthesis of Pharmaceutical Raw Material)

  • 원료의약품 제조회사에서는 화학반응에 의해 제품이 생산되기 때문에 화학반응 전 단계인 원료 분말을 투입하는 과정에서 화재 폭발사고가 자주 발생하고 있다. 이에 대한 실질적인 화학반응 단계에서 사고원인 분석을 통한 안전대책 연구는 많지 않다. 본 연구에서는 화학반응 단계에서의 위험성을 알아보고자 붕소제거 반응공정에서 발열에 대한 실험을 진행했다. 연구대상 반응공정은 실제 원료의약품 공장에서 합성하고 있는 제품을 대상으로 반응열량계를 이용하여 열적 거동을 조사하였다. 실제 제조현장의 반응공정에서 냉각실패 등의 이유로 발열할 수 있는 합성반응의 최대온도와 기술적 근거에 의한 최대온도를 비교해서 위험도를 예측하였다. 이러한 결과를 가지고 실제 제조현장에 적용하여 발열에 따른 폭주반응 위험성을 제어하는 안전대책을 제시하였다.

화장품소재로서 곽향 추출물 및 발효물의 생리활성 연구 (A Study on the Physiological Activity of Agastachis herba Extract and Fermented Products as Cosmetic Materials)

  • 곽향은 서양에서 Korea mint라고 불릴 정도로 허브와 쓰임새가 매우 유사하다. 서양의 허브는 화장품의 소재로 많이 사용하지만 곽향은 우리나라에서 약재로 더 많이 사용되어 화장품 소재로서의 기능성을 비교해보았다. 곽향 추출물과 추출물을 이용한 유산균과 효모 발효물, 에센셜오일, 플로럴워터에 대한 항산화, 미백, 폴리페놀, 플라보노이드, 항균력 실험을 진행하였다. 항산화 활성은 모든 샘플에서 높은 효과가 보였고, 미백과 폴리페놀, 항균력에서 에센셜오일이 가장 높은 효과가 보였다. 반면에 플라보노이드는 70% 에탄올 추출물에서 가장 높은 효과가 보였다. 이런 결과로 에센셜오일이 기능적으로 가장 큰 효과가 있었다. 하지만 추출물을 가지고 효모 발효를 진행한 시료들이 에센셜오일 만큼의 효과가 있다는 것을 확인해 에센셜오일의 적은 추출량에 비해 효모 발효를 진행한 시료가 화장품소재로서 충분한 가치가 있다고 본다.

  • Heat is generated during the synthesis and mixing process of chemical compounds due to a change in activation energy during the reaction. A runaway reaction occurs when sufficient heat is not removed during the heat control process within a reactor, rapidly increasing the temperature, reaction speed, and rate of heat generation inside the reactor. A risk assessment was executed using an RC-1 (Reaction Calorimeter) during Friedel-Crafts acylation. Friedel-Crafts acylation runs the risk of rapid heat generation during Active Pharmaceutical Ingredient (API) manufacturing; it was used to confirm the risk of a runaway reaction at each synthesis stage and during the mixing process. This study used experimental data to develop a safety efficiency improvement plan to control the risks of runaway and other exothermic reactions, which was implemented at the production site of a chemical plant.

  • Aegle marmelos (Rutaceae) is a deciduous shrub or tree and typically known as bael throughout Southeast Asia and has been used as a medicinal plant and a food ingredient. In this study eight compounds were determined to be O-(3,3-dimethylallyl) halfordinol (1), (R)-aegeline (2), (R)-marmeline (3), imperatorin (4), xanthotoxol (5), valencic acid (6), vanillic acid (7) and rutin (8). The chemical structures of the isolates were elucidated through spectroscopic evidence including 1D, 2D NMR, ESI-Q-TOF-MS and optical rotation.

Growth and Ingredient Contents of Platycodon grandiflorum Roots under Sensor-based Soil Moisture Contents of Farmland Conditions

  • Growth characters and ingredient contents of two-year-old bellflower (Platycodon grandiflorum) roots were investigated under both control and soil moisture treatment condition using soil moisture control system including soil sensing and automatic water supply chain in this study. Root diameter, fine root number, root length, fresh weight and dry weight of the plant were significantly influenced by the automatic water treatment, 20%, 30%, 40% and 50%, respectively. Ingredient contents of the two-year-old roots in bellflower plants were detected in the 20% and 50% of controlled soil moisture content. Contents of amino acids were decreased by the soil moisture treatment, meanwhile, contents of minerals were not showed significant decrease except for phosphorus content. Showing no difference in proline and tyrosine, fourteen of the amino acid contents were gradually decreased by the increased soil moisture contents, with significant decrease in serine, glycine, alanine, leucine, lysine and histidine at 20% treatment.

Hyaluronidase Inhibitory and Antioxidant Activities of Enzymatic Hydrolysate from Jeju Island Red Sea Cucumber (Stichopus japonicus) for Novel Anti-aging Cosmeceuticals

  • An active ingredient with hyaluronidase (HAse) inhibitory effect is one of the anti-aging approaches in cosmeceuticals. Here, red sea cucumbers (RSCs), Stichopus japonicus, from Jeju Island were evaluated to examine their HAse inhibitory and antioxidant activity effect. In this study, RSCs were extracted by six enzymatic hydrolysis (Alcalase; Al, Trypsin; Try, Neutrase; Neu, Pepsin; Pep, Alpha-chymotrypsin; Chy and Protamex; Pro). Alcalase hydrolysate (AlH) showed the highest antioxidant capacities for both of oxygen radical absorbance capacity (ORAC) and trolox equivalent antioxidant capacity (TEAC) methods, compared to those of other hydrolysates, at $66.59{\pm}0.78{\mu}M\;TE/mg$ and $135.78{\pm}3.24{\mu}M\;TE/mg$, respectively. Furthermore, AlH performed the highest capacity of HAse inhibitory with $IC_{50}$ value of 3.21 mg/ml. Thus, RSCs hydrolyzed by Al were chosen to determine the cellular antioxidant activity and hyaluronic acid (HA) production effect on Human immortalized keratinocyte cell line (HaCaT). The results showed that AlH improved the cell viabilities and intracellular reactive oxygen species (ROS) induced by 2,2'-Azobis(2-amidinopropane) dihydrochloride (AAPH) were significantly decreased. In addition, AlH increased HA amount by regulating HYAL2 and HAS2 expressions in the HaCaT cells. Taken together, AlH of RSCs collected from Jeju Island showed HAse inhibitory and antioxidant activities against skin-aging which shows its potentials can be an optional natural bioactive ingredient for novel cosmeceuticals.

Effects of Avocado Yoghurt, Three Beans Cookie, and Tiger Nut Galsu Drink on Fibroblasts (Hs68) and Keratinocytes (HaCaT)

  • The present study aimed to investigate the effects of skin care foods on the synthesis of pro-collagen type I C peptide and suppression of matrix metalloproteinase (MMP)-1 secretion through an in vitro study using fibroblasts (Hs68 cells) and keratinocytes (HaCaT cells). Among the three skin care foods developed in this study, three beans cookie and avocado yoghurt influenced the production of pro-collagen type I C peptide and suppressed MMP-1 secretion; however, tiger nut Galsu drink did not exhibit these effects. All skin care foods, including three beans cookie and plain yoghurt ($50{\mu}g/mL$, p<0.001) influenced the suppression of MMP-1 in addition to other commercially available breast milk production support foods examined, such as Heath Heather ($50{\mu}g/mL$, p<0.001), Happy Mama ($50{\mu}g/mL$, p<0.01), BioLys ($50{\mu}g/mL$, p<0.001), Enfamama ($25{\mu}g/mL$, p<0.0001), and Pregnagen ($25{\mu}g/mL$, p<0.001). Avocado fruit yoghurt ($25{\mu}g/mL$, p<0.05), avocado fruit jam yoghurt ($50{\mu}g/mL$, p<0.01), Enfamama ($100{\mu}g/mL$, p<0.05), and Pregnagen ($100{\mu}g/mL$, p<0.05) influenced the production of pro-collagen type I C peptide and suppressed MMP-1 secretion. This result indicates that only avocado jam yoghurt significantly influenced both the prevention of skin keratinization and acceleration of recovery of skin fibrous structure. Therefore, avocado is a favorable ingredient for nutrition-balanced dietary foods or an essential ingredient in products for revitalization of human skin.

6-Shogaol, an Active Ingredient of Ginger, Improves Intestinal and Brain Abnormalities in Proteus Mirabilis-Induced Parkinson's Disease Mouse Model

  • Parkinson's disease (PD) which has various pathological mechanisms, recently, it is attracting attention to the mechanism via microbiome-gut-brain axis. 6-Shogaol, a representative compound of ginger, have been known for improving PD phenotypes by reducing neuroinflammatory responses. In the present study, we investigated whether 6-shogaol and ginger attenuate degeneration induced by Proteus mirabilis (P. mirabilis) on the intestine and brain, simultaneously. C57BL/6J mice received P. mirabilis for 5 days. Ginger (300 mg/kg) and 6-shogaol (10 mg/kg) were treated by gavage feeding for 22 days including the period of P. mirabilis treatment. Results showed that 6-shogaol and ginger improved motor dysfunction and dopaminergic neuronal death induced by P. mirabilis treatment. In addition, they suppressed P. mirabilis-induced intestinal barrier disruption, pro-inflammatory signals such as toll-like receptor and TNF-α, and intestinal α-synuclein aggregation. Moreover, ginger and 6-shogaol significantly inhibited neuroinflammation and α-synuclein in the brain. Taken together, 6-shogaol and ginger have the potential to ameliorate PD-like motor behavior and degeneration of dopaminergic neurons induced by P. mirabilis in mice. Here, these findings are meaningful in that they provide the first experimental evidence that 6-shogaol might attenuate PD via regulating gut-brain axis.

Multi-Layered Matrix Tablets with Various Tablet Designs and Release Profiles

  • Tablet dosage forms have been preferred over other formulations for the oral drug administration due to their low manufacturing costs and ease of administrations, especially controlled-release applications. Controlled-release tablets are oral dosage forms from which the active pharmaceutical ingredient (API) is released over an intended or extended period of time upon ingestion. This may allow a decrease in the dosing frequency and a reduction in peak plasma concentrations and hence improves patient compliance while reducing the risk of undesirable side effects. Conventional singlelayered matrix tablets have been extensively utilized to deliver APIs into the body. However, these conventional single-layered matrix tablets present suboptimal delivery properties, such as non-linear drug delivery profiles which may cause higher side effects. Recently, a multi-layered technology has been developed to overcome or eliminate the limitations of the singlelayered tablet with more flexibility. This technology can give a good opportunity in formulating new products and help pharmaceutical companies enhancing their life cycle management. In this review, a brief overview on the multi-layered tablets is given focusing on the various tablet designs, manufacturing issues and drug release profiles.

Apoptosis Induction of Persicae Semen Extract in Human Promyelocytic Leukemia (HL-60) Cells

  • The major ingredient of Persicae Semen is a cynogenic compound, amygdalin (D-mandelonitrile-$\beta$-gentiobioside). Controversial results on the anticancer activity of amygdalin were reported due to its conversion to its inactive isomer, neoamygdalin. In order to inhibit the epimerization of amygdalin, we used newly developed simple acid boiling method in preparation of Persicae Semen extract. HPLC analysis revealed most of amygdalin in Persicae Semen extract was active D-form. Persicae Semen extract was used to analyze its effect on cell proliferation and induction of apoptosis in human promyelocytic leukemia (HL-60) cells. Persicae Semen extract was cytotoxic to HL-60 cells with $IC_{50}$ of 6.4 mg/mL in the presence of 250 nM of $\beta$-glucosidase. The antiproliferative effects of Persicae Semen extract appear to be attributable to its induction of apoptotic cell death, as Persicae Semen extract induced nuclear morphology changes and internucleosomal DNA fragmentation.
