Sibutramine is an anorectic that has been banned since 2010 due to cardiovascular safety issues. However, counterfeit drugs or slimming products that include sibutramine are still available in the market. It has been reported that illegal sibutramine-contained pharmaceutical products induce cardiovascular crisis. However, the mechanism underlying sibutramine-induced cardiovascular adverse effect has not been fully evaluated yet. In this study, we performed cardiovascular safety pharmacology studies of sibutramine systemically using by hERG channel inhibition, action potential duration, and telemetry assays. Sibutramine inhibited hERG channel current of HEK293 cells with an $IC_{50}$ of $3.92{\mu}M$ in patch clamp assay and increased the heart rate and blood pressure ($76{\Delta}bpm$ in heart rate and $51{\Delta}mmHg$ in blood pressure) in beagle dogs at a dose of 30 mg/kg (per oral), while it shortened action potential duration (at $10{\mu}M$ and $30{\mu}M$, resulted in 15% and 29% decreases in $APD_{50}$, and 9% and 17% decreases in $APD_{90}$, respectively) in the Purkinje fibers of rabbits and had no effects on the QTc interval in beagle dogs. These results suggest that sibutramine has a considerable adverse effect on the cardiovascular system and may contribute to accurate drug safety regulation.
멜리틴은 봉독의 주요 성분 중 하나로 항염증과 항암활성 효과를 가지고 있다. 우리는 폐암세포에서 멜리틴이 EMT 억제를 통해 암세포 이동과 침투를 억제하는 사실을 확인하였다. 멜리틴은 EGF로 유도된 폐암 세포 이동과 침투를 억제하였을 뿐만 아니라 EMT와 관련된 단백질인 이카드헤린의 발현을 증가시켰으며, 바이멘틴과 피브로넥틴 발현은 감소시켰다. 또한 멜리틴에 의한 EMT조절 전사인자인 ZEB2, Slug, Snail의 발현을 확인한 결과 멜리틴 처리에 의해 농도의존적으로 발현이 감소하였다. 또한 작용 메커니즘을 확인하기 위해 mTOR와 FAK 메커니즘을 확인한 실험에서 EGF 처리에 의해 증가한 AKT, mTOR, p70S6K, 4EBP1의 인산화가 멜리틴 농도의존적으로 감소하였다. 그러나 FAK는 EGF에 의해 변화가 없었으며, EKR, JNK 메커니즘은 EGF 처리에 의해 인산화가 증가하였으나 멜리틴 처리에 의해 아무런 영향을 받지 않았다. 그러므로, 폐암세포의 세포 이동과 침투에 대한 멜리틴의 억제효과는 AKT/mTOR/P70S6K/4EBP1 기전 억제를 통해 EMT를 억제하여 세포 이동과 침투를 억제하는 것으로 보인다.

Background: Glucocorticoids (GCs) are commonly used in many chemotherapeutic protocols and play an important role in the normal regulation of bone remodeling. However, the prolonged use of GCs results in osteoporosis, which is partially due to apoptosis of osteoblasts and osteocytes. In this study, effects of Korean Red Ginseng (KRG) on GC-treated murine osteoblastic MC3T3-E1 cells and a GC-induced osteoporosis mouse model were investigated. Methods: MC3T3-E1 cells were exposed to dexamethasone (Dex) with or without KRG and cell viability was measured by the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Realtime polymerase chain reaction was performed to evaluate the apoptotic gene expression; osteogenic gene expression and alkaline phosphatase (ALP) activity were also measured. Western blotting was performed to evaluate the mitogen-activated protein kinase (MAPK) proteins. A GC-induced osteoporosis animal model was used for in vivo study. Results and conclusion: The MTT assay revealed that Korean Red Ginseng (KRG) prevents loss of cell viability caused by Dex-induced apoptosis in MC3T3E1 cells. Real-time polymerase chain reaction data showed that groups treated with both Dex and KRG exhibited lower mRNA levels of caspase-3 and -9, whereas the mRNA levels of Bcl2, IAPs, and XIAP increased. Moreover, groups treated with both Dex and KRG demonstrated increased mRNA levels of ALP, RUNX2, and bone morphogenic proteins as well as increased ALP activity in MC3T3-E1 cells, compared to cells treated with Dex only. In addition, KRG increased protein kinase B (AKT) phosphorylation and decreased c-Jun N-terminal kinase (JNK) phosphorylation. Moreover, microcomputed tomography analysis of the femurs showed that GC implantation caused trabecular bone loss. However, a significant reduction of bone loss was observed in the KRG-treated group. These results suggest that the molecular mechanism of KRG in the GC-induced apoptosis may lead to the development of therapeutic strategies to prevent and/or delay osteoporosis.
The objective of this study is investigation of contamination levels and transfer rate of heavy metals and assessment of health risk effects in animal medicines. A total of sixteen items with 148 samples were obtained for this investigation from major herbal medicine producing areas and herbal markets from 2011 to 2012. Inductively coupled plasma mass spectrometer method was conducted for the quantitative analysis of Pb, Cd and As. In addition, the mercury analyzer system was conducted for that of Hg without sample digestion. The average contents of heavy metals in animal medicines were as follows : 1.55 mg/kg for Pb, 0.28 mg/kg for Cd, 0.70 mg/kg for As and 0.047 mg/kg for Hg respectively. The average contents of heavy metals in decoctions were as follows : 0.03 mg/kg for Pb, 0.01 mg/kg for Cd, 0.17 mg/kg for As and 0.003 mg/kg for Hg respectively. The average transfer rates of heavy metals from animal medicines to decoctions were 1.9% for Pb, 3.6% for Cd, 24.2% for As and 6.0% for Hg respectively. Cancer risks of animal medicines and decoctions through slope factor by ministry of environment republic korea and environmental protection agency was $1.36{\times}10^{-6}$ and $2.50{\times}10^{-8}$ for Pb and $7.46{\times}10^{-5}$ and $1.98{\times}10^{-5}$ for As (assuming that the total arsenic content was equal to the inorganic arsenic) respectively. The hazard index of heavy metals in animal medicines and decoctions was below recommended hazard index as 0.52 and 0.05 respectively. Taken together, cancer risks about Pb and hazard indexes of animal medicines and decoctions indicate that the majority of samples except for some samples with relative high contents of heavy metals were safe. But cancer risks about As of animal medicines indicate that the majority of samples were not relatively safe.
올벚나무 잎 추출물을 제조하고 LPS로 자극시킨 RAW 264.7 대식세포에 대한 항염 효과를 조사하였다. 추출물 및 분획물(n-hexane, ethyl acetate, n-butanol 및 water)에 대하여 활성 실험을 진행한 결과, $100{\mu}g/mL$ 농도에서 ethyl acetate (EtOAc) 분획물이 세포독성 없이 76.3%의 NO 생성 억제 효과를 나타내었다. 또한 EtOAc 분획물이 농도 의존적으로 NO 생성을 효과적으로 억제하고 있음을 확인하였다. 이러한 항염 효과에 대한 추가적인 기전 연구를 진행한 결과, EtOAc 분획물이 $PGE_2$, $IL-1{\beta}$ 및 IL-6의 생성을 감소시켰으며, iNOS와 COX-2 단백질의 발현도 억제시키는 것을 확인하였다. EtOAc분획물에서 활성 성분을 규명하기 위하여 column chromatography를 진행하였으며 5개의 화합물을 분리 동정하였다. ursolic acid (1), prunasin (2), methyl p-coumarate (3), kaempferol (4), astragalin (5). 분리된 compound 1 - 5는 모두 올벚나무 잎에서 처음 분리된 물질이다. 분리된 화합물을 대상으로 활성 실험을 진행한 결과, 플라보노이드 compound 4와 5가 NO 생성을 억제하고 있음을 확인하였다. 이상의 결과를 바탕으로, 플라보노이드를 유효성분으로 함유하는 올벚나무 잎은 항염 효과를 갖는 천연 소재로서 활용이 가능할 것이라 사료된다.
콩[Glycine max (L.) Merr. 2n=40]은 국내에서 벼 다음으로 많이 재배되고 있는 식량작물이며 성숙 콩 종실에는 3대 영양소외 페놀성 화합물, 이소플라본, 사포닌, 루테인 토코페롤, 피틴산, 루테인, 안토시아닌등 생리활성 물질이 다양하게 존재하지만 기능성과 가공 적성 및 품질을 저해시키는 성분들도 다수 존재한다. 성숙 콩 종실에서 품질, 기능성 및 가공 적성을 저하시키는 KTI 단백질이 없으면서 난소화성 올리고당인 stachyose 함량이 낮은 속 푸른 검정콩 유전자형을 육성하기 위하여 진행되었다. 3개의 모본으로 육종집단을 창성하여 검정종피, 녹색자엽 및 KTI 단백질이 없는 11개의 $F_2$ 종자를 얻었고 stachyose 함량이 낮은 2개의 계통을 선발하여 농업형질 평가를 통하여 검정 종피, 녹색자엽, stachyose 함량이 낮은 titirs2rs2 유전자형을 가진 계통을 선발하였다. 선발된 계통의 초장은 66 cm 정도였으며 백립중은 28.4 g으로 대립이었으며 종피색과 제색은 모두 검정색이었고 성숙 자엽색은 녹색이었다. 선발된 계통의 stachyose 함량은 일반품종(13.01-16.81 g/kg)보다 매우 낮은 2.59(g/kg)이었다. 본 연구를 통하여 선발된 계통은 Kunitz Trypsine Inhibitor (KTI) 단백질이 없으며 난소화성 올리고당인 stachyose 성분의 함량이 낮고 검정종피와 녹색자엽을 가진 기능성 고품질 유색콩 육성을 위한 중간모본으로 이용될 수 있을 것으로 사료되었다.
본 연구는 화장품 소재로서 우절 에탄올추출물의 가능성을 확인하기 위한 것이다. 이를 위해 우리는 연근의 마디부분을 70%에탄올로 추출하여 사용하였으며 미백효능 및 주름개선 효과에 대한 생물학적 활성 평가를 수행하였다. 시료의 미백활성을 알아보기 위해 멜라닌세포(B16F10 cells)의 MTT assay를 이용한 샘플의 독성평가와 멜라닌 생성에 영향을 주는 관련 단백질의 발현량을 확인하였다. 시료의 주름활성억제 평가를 위해 collagenase inhibition assay를 수행하였다. 또한 UVB로 유도된 섬유아세포(CCD-986sk cells)의 MTT assay를 이용한 샘플의 독성평가와 matrix metalloprotease (MMP-1) inhibition 및 procollagen synthesis를 평가하였다. 미백활성 평가를 알아보기 위한 western bolt 결과, 멜라닌 생성과 관련된 두 단백질 Tyrosinase related protein-1 (TRP-1, Tyrosinase related protein-2 TRP-2)의 합성이 농도의존적으로 감소됨을 나타내었다. 게다가 우절 에탄올추출물의 collagenase inhibiion 실험의 결과, $500{\mu}g/ml$의 농도에서 대조군인 EGCG와 효능이 비슷한 80% 이상의 효과를 나타내었다. Procollagen synthesis 결과 UVB 자극군에 의해 콜라겐 합성은 68.8%로 감소되었으며 시료의 $25{\mu}g/ml$의 농도에서 80.2%의 콜라겐 합성에 회복능을 보였다. MMP-1 inhibition 실험결과는 $25{\mu}g/ml$ 농도에서 20.2%의 저해능을 나타내었다. 실험 결과는 NRN의 미백 및 주름 억제 효과를 검증하고, 향후 안전한 천연 화장품 재료로 사용될 수 있음을 확인했다.
사람의 장내 미생물은 숙주의 대사 조절에서 중요한 역할을 한다. 장내 미생물 균총의 불균형은 비만, 대사 증후군과 밀접하게 병리학적, 생리학적 상호작용을 한다. A. muciniphila는 최근 인간의 대변에서 분리되었으며, 장내 미생물의 약 1-4%를 차지하는 우점균이다. A. muciniphila 유래 외막(external membran) 단백질 Amuc_1100과 세포외 소포(EVs)의 이용은 비만치료를 위한 새로운 전략이 될 수 있다. A. muciniphila는 비만과 같은 대사 장애치료를 위한 차세대 probiotics로 고려되고 있다. F. prausnitzii는 next-generation probiotic로서 건강한 성인의 경우 장내미생물 균총의 약 5%를 차지하며 성인 장 건강의 지표이다. F. prausnitzii는 butyrate-producing bacterium로서 항염증 효과를 나타내며, 면역 질환과 당뇨병 치료를 위한 next-generation probiotics로 기대된다. Postbiotics는 probiotics에 의해 분비되는 세포 상층액에 포함된 대사산물의 복잡한 혼합물이다. 반면, parabiotics는 probiotics를 불활성화 시킨 미생물 세포이다. Paraprobiotics와 postbiotics는 probiotics에 비해 명확한 화학구조(clear chemical structures)와 안전한 투여 용량(safety dose parameters), 장기간의 유통 기간(longer shelf life) 등 많은 장점을 가지고 있어서 probiotics를 대체할 수 있는 잠재력을 가지고 있다. 장 생태계의 불균형을 정상적으로 복원하기 위해서 장내 공생 박테리아(commensal bacteria) 중 next-generation probiotics (NGPs)를 사용하는 것이 가장 자연스런 방법이다. 따라서 next-generation probiotics 를 대상으로 parabiotics와 postbiotics와 같은 새로운 식품이나 약품으로 개발하는 것이 필요하다.
부작용을 줄인 사물탕을 개발하고자 오미자를 사용한 변형 사물탕을 난소를 절제한 랫드에 경구투여하고 여성갱년기 기능성 효능의 평가지표 변화를 측정하였다. 체중 증가, 자궁과 질의 상대 장기중량, 혈중 지질관련 지표, 혈중 estradiol 의 변화에서 대조군과 비교하여 변형 사물탕이 통계적으로 유의한 개선효과는 나타나지 않았다. 그러나 자궁 조직내 단백질 변화에서 estrogen receptor alpha 와 beta 의 발현이 증가하는 경향성이 관찰되었고 estrogen receptor 신호전달에 관여하는 것으로 알려진 ERK와 AKT에서 활성화에 기여하는 인산화 ERK의 발현량이 유의한 증가를 나타내었고 인산화 AKT의 발현량은 유의하진 않지만 증가하는 경향성을 나타내었다. 난소절제로 유도한 여성갱년기 모델에서 시험물질 변형 사물탕을 비교적 적은 용량 투여했음에도 이러한 결과를 나타내는 것은 시험물질인 변형 사물탕에 estrogen receptor를 활성화시키는 능력이 있는 것으로 판단된다. 추후 위장장애가 적으면서 사물탕의 효능을 나타내고자 할 때 유용한 천연물 배합으로 이용이 가능할 것으로 본다.

BACKGROUD/OBJECTIVES: Allomyrina dichotoma larva (ADL), one of the many edible insects recognized as future food resources, has a range of pharmacological activities. In a previous study, an ADL extract (ADLE) reduced the hepatic insulin resistance of high-fat diet (HFD)-induced diabetic mice. On the other hand, the associated molecular mechanisms underlying pancreatic beta-cell dysfunction remain unclear. This study examined the effects of ADLE on palmitate-induced lipotoxicity in a beta cell line of a rat origin, INS-1 cells. MATERIALS/METHODS: ADLE was administered to high-fat diet treated mice. The expression of apoptosis-related molecules was measured by Western blotting, and reactive oxidative stress generation and nitric oxide production were measured by DCH-DA fluorescence and a Griess assay, respectively. RESULTS: The administration of ADLE to HFD-induced diabetic mice reduced the hyperplasia, 4-hydroxynonenal levels, and the number of apoptotic cells while improving the insulin levels compared to the HFD group. Treatment of INS-1 cells with palmitate reduced insulin secretion, which was attenuated by the ADLE treatment. Furthermore, the ADLE treatment prevented palmitate-induced cell death in INS-1 cells and isolated islets by reducing the apoptotic signaling molecules, including cleaved caspase-3 and PARP, and the Bax/Bcl2 ratio. ADLE also reduced the levels of reactive oxygen species generation, lipid accumulation, and nitrite production in palmitate-treated INS-1 cells while increasing the ATP levels. This effect corresponded to the decreased expression of inducible nitric oxide synthase (iNOS) mRNA and protein. CONCLUSIONS: ADLE helps prevent lipotoxic beta-cell death in INS-1 cells and HFD-diabetic mice, suggesting that ADLE can be used to prevent or treat beta-cell damage in glucose intolerance during the development of diabetes.

일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
