Immunostimulatory Effect of Heat-Killed Probiotics on RAW264.7 Macrophages

  • Probiotics modulate the gut microbiota, which in turn regulate immune responses to maintain balanced immune homeostasis in the host. However, it is unclear how probiotic bacteria regulate immune responses. In this study we investigated the immunomodulatory effects of heat-killed probiotics, including Lactiplantibacillus plantarum KC3 (LP3), Lactiplantibacillus plantarum CKDB008 (LP8), and Limosilactobacillus fermentum SRK414 (LF4), via phagocytosis, nitric oxide (NO), and pro-inflammatory cytokine production in macrophages. We thus found that heat-killed LP8 could promote the clearance of foreign pathogens by enhancing the phagocytosis of macrophages. Treatment with heat-killed LP8 induced the production of NO and pro-inflammatory cytokines, including TNF-α, IL-6, and IL-1β. In addition, heat-killed LP8 suppressed the production of NO and cytokines in LPS-induced RAW264.7 cells, suggesting that heat-killed LP8 exerts immunomodulatory effects depending on the host condition. In sum, these results indicate that heat-killed LP8 possesses the potential for immune modulation while providing a molecular basis for the development of functional probiotics prepared from inactivated bacterial cells.

Investigation of Immunostimulatory Effects of Heat-Treated Lactiplantibacillus plantarum LM1004 and Its Underlying Molecular Mechanism

  • Postbiotics are defined as probiotics inactivated by heat, ultraviolet radiation, sonication, and other physical or chemical stresses. Postbiotics are more stable than probiotics, and these properties are advantageous for food additives and pharmacological agents. This study investigated the immunostimulatory effects of heat-treated Lactiplantibacillus plantarum LM1004 (HT-LM1004). Cellular fatty acid composition of L. plantarum LM1004 isolated form kimchi was analyzed by gas chromatography-mass spectrometry detection system. The nitric oxide (NO) content was estimated using Griess reagent. Immunostimulatory cytokines were evaluated using enzyme-linked immunosorbent assay. Relative protein expressions were evaluated by western blotting. Phagocytosis was measured using enzyme-labelled Escherichia coli particles. L. plantarum LM1004 showed 7 kinds of cellular fatty acids including palmitic acid (C16:0). The HT-LM1004 induced release of NO and upregulated the inducible NO synthase in RAW 264.7 macrophage cells. Tumor necrosis factor-α and interleukin-6 levels were also increased compared to control (non-treated macrophages). Furthermore, HT-LM1004 modulated mitogen-activated protein kinase (MAPK) subfamilies including p38 MAPK, extracellular signal-regulated kinase 1/2, and c-Jun N-terminal kinase. Therefore, these immunostimulatory effects were attributed to the production of transcriptional factors, such as nuclear factor kappa B (NF-κB) and the activator protein 1 family (AP-1). However, HT-LM1004 did not showed significant phagocytosis of RAW 264.7 macrophage cells. Overall, HT-LM1004 stimulated MAPK/AP-1 and NF-κB expression, resulting in the release of NO and cytokines. These results will contribute to the development of diverse types of food and pharmacological products for immunostimulatory agents with postbiotics.

Korean Ginseng Berry Polysaccharide Enhances Immunomodulation Activities of Peritoneal Macrophages in Mice with Cyclophosphamide-Induced Immunosuppression

  • Korean ginseng (Panax ginseng C. A. Meyer), a member of the Araliaceae family, is known as a traditional medicinal plant to have a wide range of health properties. Polysaccharides constitute a major component of Korean ginseng, and its berries exhibit immune-modulating properties. The purpose of this study was to investigate the immune effects of crude polysaccharide (GBPC) extracted from Korean ginseng berry on peritoneal macrophages in mice with cyclophosphamide (CY)- induced immunosuppression. BALB/c mice were divided into eight groups: normal control, normal control + CY, levamisole + CY, ginseng + CY, and four concentrations of 50, 100, 250, and 500 mg/kg BW/day of GBPC + CY. Mice were orally administered with samples for 10 days. Immunosuppression was established by treating mice with CY (80 mg/kg BW/day) through intraperitoneal injection on days 4 to 6. The immune function of peritoneal macrophages was then evaluated. Oral administration of 500 mg/kg BW/day GBPC resulted in proliferation, NO production, and phagocytosis at 100%, 88%, and 91%, respectively, close to the levels of the normal group (100%) of peritoneal macrophages. In CY-treated mice, GBPC of 50-500 mg/kg BW/day also dose-dependently stimulated the proliferation, NO production, and phagocytosis at 56-100%, 47-88%, and 53-91%, respectively, with expression levels of immune-associated genes, such as iNOS, COX-2, IL-1β, IL-6, and TNF-α, of about 0.32 to 2.87-fold, compared to those in the CY group. GBPC could be a potential immunomodulatory material to control peritoneal macrophages under an immunosuppressive condition.

Cytotoxicity of two self-adhesive resin cements and their interference in the phagocytic activity of murine macrophages

  • Objectives: This study aimed to evaluate in vitro the effects of the self-adhesive resin cements RelyX U200 (3M ESPE) and seT PP (SDI Limited) on murine macrophages and the interference of the photoactivation. Materials and Methods: Cell viability assays, cell adherence, yeast phagocytosis of Saccharomyces boulardii and production of reactive oxygen species (ROS) were performed in the presence of capillaries containing the respective self-adhesive cement when photoactivated or not. Results: After long periods of contact, both types of cements, when not photoactivated, are more cytotoxic for macrophages. The seT PP cement when only chemically activated seems to interfere more negatively in the process of phagocytosis of yeasts S. boulardii. Both types of cements interfere in the cell adhesion process, independent of photoactivation. None of the types of cements tested was able to induce the production of ROS. Conclusions: Our results highlight the great importance of the photoactivation of self-adhesive resin cements in the dental clinic, since RelyX U200, when photoactivated, presented the best results within the evaluated parameters.

산양삼의 TRL2/4 의존성 MAPK, NF-κB 및 PI3K/AKT 신호전달 활성화를 통한 면역증진활성 (Immune-Enhancing Activity of Wild Simulated Ginseng through TRL2/4-Dependent Activation of MAPK, NF-κB and PI3K/AKT Pathways)

  • Ginseng (Panax ginseng Meyer) is a very well-known traditional herbal medicine that has long been used to enhance the body's immunity. Because it is a type of ginseng, it is believed that wild simulated ginseng (WSG) also has immune-enhancing activity. However, study on the immune-enhancing activity of WSG is quite insufficient compared to ginseng. In this study, we evaluated immune-enhancing activity of WSG through macrophage activation to provide a scientific basis for the immune enhancing activity of WSG. WSG increased the production of immunomodulators such as NO, iNOS, COX-2, IL-1β, IL-6 and TNF-α and activated phagocytosis in mouse macrophages RAW264.7 cells. Inhibition of TLR2 and TLR4 reduced the production of immunomodulators induced by WSG. WSG activated MAPK, NF-κB and PI3K/AKT signaling pathways, and inhibition of such signaling activation blocked WSG-mediated production of immunomodulators. In addition, activation of MAPK, NF-κB and PI3K/AKT signaling pathways by WSG was reversed by TLR2 or TLR4 inhibition. Based on the results of this study, WSG is thought to activate macrophages through the production of immunomodulators and phagocytosis activation through TLR2/4-dependent MAPK, NF-κB and PI3K/AKT signaling pathways. Therefore, it is thought that WSG have the potential to be used as an agent for enhancing immunity.

단핵구세포주 THP-1의 대식세포로의 분화 및 활성화에서 CO의 억제 효과 (Carbon Monoxide Inhibits PMA-induced Differentiation in Human Monocytic THP-1 Cells)

  • Carbon monoxide (CO)는 세포 보호의 기능을 가지는 항산화 효소인 heme oxygenase-1 (HO-1)의 대사산물로 세포성장, 아폽토시스, 염증에 대한 억제 효과를 보이는 것으로 보고가 이어지고 있고, 이에 관련된 연구가 활발히 진행되고 있는 실정이다. 본 연구에서는 CO가 단핵구의 대식세포로의 분화 및 그 활성화 과정에 미치는 영향을 인간 단핵구세포주 THP-1을 이용하여 조사하였다. CO-releasing compound인 CORM-2는 phorbol 12-myristate 13-acetate (PMA)로 자극한 THP-1 세포에서 viability와 증식에는 큰 영향을 주지 않았으나 부착능의 뚜렷한 감소를 보였다. 그리고, CORM-2는 대식세포의 막표면 분화 인자인 CD14, CD11b 및 CD18의 발현과 latex beads를 이용한 포식 기능을 현저히 억제하였다. 다음으로, 배양중인 THP-1 세포를 PMA로 6일 동안 대식세포로 분화시킨 후 inflammatory cytokines의 분비와 포식 기능을 조사하였다. CORM-2의 처리는 lipopolysaccaride (LPS)로 자극한 대식세포로부터 분비되는 IL-6와 $TNF-{\alpha}$의 분비를 감소시켰다. 또한, 분화된 대식세포에 E. coli (K-12 strain) bioparticles를 이용하여 포식 기능을 측정한 결과 CORM-2를 처리한 세포에서는 현저히 감소되는 경향을 보였다. 이를 종합해 볼 때, CO는 항원 인식과 포식 기능에 관여하는 막단백질의 발현을 저해함으로써 단핵구의 분화과정을 억제하였고, 분화된 대식세포의 inflammatory cytokines의 분비 및 포식 기능을 저해함으로써 활성화 과정도 억제하는 것으로 보인다.

결핵균의 약제내성과 말초혈액단핵구의 결핵균 탐식능에 관한 연구 (Phagocytosis of Drug-Resistant Mycobacterium Tuberculosis by Peripheral Blood Monocytes)

  • 연구배경 : 세포내 병원균(intracellular pathogen)으로 감염초기에 단핵식세포(mononuclear phagocytes)에 탐식되는데, 독성(virulent) 결핵균주(Erdman, H37Rv)가 약독화(attenuated) 균주(H37Ra)보다 탐식이 더 잘 되며, 이러한 현상으로부터 결핵균의 입장에서 보면 단핵식세포에 탐식이 잘되는 것이 생존 및 증식에 유리할 것이라는 의견이 제시되어 왔다. 그리고 난치성 결핵환자에서 분리한 결핵균은 임상적으로 독성이 강하며 대부분에서 다제내성을 보이므로, 약제내성 결핵균여 감수성균에 비해 독성이 강하다는 가정하에 약제내성 결핵균이 감수성균에 비해 단핵식세포에 탐식이 더 잘 되는지, 그리고 난치성 결핵환자에서 분리한 단핵식세포가 정상인에 비해 결핵균을 더 잘 탐식하는지 알아보고자 하였다. 방 법 : 약제내성의 정도에 따라 모든약제에 감수성인 결핵균, Isoniazid와 Rifampicin을 포함한 일부 약제에 내성인 결핵균, 모든 약제에 내성인 결핵균을 각각 $70^{\circ}C$에서 1시간 가열하여 사멸시킨 뒤 부유액을 만들어 10명의 난치성 결핵환자와 12명의 정상인에서 분리한 말초혈액탄핵구 배양액에 첨가하여 탐식시킨 뒤 단핵구의 결핵균을 탐식율을 관찰하였다. 결 과 : 약제내성 결핵균이 감수성균에 비하여 말초혈액단핵구에 탐식이 더 잘되었으며(정상 대조군에서 분리한 말초혈액단핵구의 결핵균 탐식율 : 모든약제 감수성균 : $32.3{\pm}2.9%$, 모든약제 내성균 : $49.6{\pm}3.4%$, p = 0.0022, 난치성 결핵환자군에서 분리한 말초혈액단핵구의 결핵균 탐식율 : 모든약제 감수성균 : $34.9{\pm}3.6%$, 모든약제 내성균 : $50.7{\pm}4.5%$, p = 0.0069), 난치성 결핵환자와 정상인에서 분리한 말초혈액단핵구 사이의 결핵균의 탐식능에는 차이가 없었다. 결 론 : 약제 내성균이 감수성균보다 단핵식세포에 탐식이 더 잘 되며, 숙주 요인은 결핵균의 탐식의 차이에 영향을 미치지 않는 것으로 보인다.

Pharmacological Activities of a New Glycosaminoglycan, Acharan Sulfate Isolated from the Giant African Snail Achatina fulica

  • Acharan sulfate (AS) is a glycosaminoglycan (GAG) prepared from the giant African snail, Achatina fulica. In this study, some biological activities of AS were evaluated on the basis of structural similarities to heparin/heparan sulfate and the biological functions of GAGs. We demonstrated that it exhibited strong immunostimulating activities as measured by carbon clearance test in mice and in vivo phagocytosis. It also exhibited a significant hypoglycemic activity in epinephrine (EP)-induced hyperglycemia as well as antifatigue effects by weight-loaded forced swimming test. And it showed hypolipidemic activities in cholesterol-rich mixture induced hyperlipidemia in rats. The above results indicate that AS has diverse biological activities and suggest therapeutically important target molecules.

Regulation of Melanosome Transfer to and Distribution in Keratinocytes

  • Skin pigmentation, also known as complexion coloration, results from the biosynthesis of melanin within the melanocytes of the Stratum basalum and the subsequent transfer, translocation, and degradation of this pigment to, in, and by the neighboring keratinocytes respectively, Melanins are produced and retained in melanosomes synthesized in the cell body that are translocated along the dendrites using microtubules via motor proteins. Melanosomes are eventually captured and retained at the tips of dendrites by attachment to the peripherally localized actin. Melanosomes reaching the dendritic tips are transferred to keratinocytes, primarily via phagocytosis of released melanosomes by keratinocytes. Molecules responsible for cell/cell recognition and interaction that regulate transfer are being identified. Some of these putative mediators appear to be affected by ultraviolet radiation. After the keratinocytes receive melanosomes, the granules are distributed individually or as clusters in dark versus light skin respectively. These melanosomes are then aggregated over the nucleus for photoprotection ofkeratinocyte DNA and eventually degraded.

가미대보탕이 생쥐의 복강 Macrophages에 미치는 영향 (Effect of Kamidaebo-tang on Murine Peritoneal Macrophages)

  • The purpose of this research was to investigate the effects of Kamidaebo-tang water extract (KDT) on murine peritoneal macrophages. KDT (50 or 250 mg/kg) was administerd p.o. once a day for 7 days to mice. KDT increased the production of tumor necrosis factor-α and nitric oxide from murine peritoneal macrophages, but decreased the production of interleukin-1β. Also, KDT enhanced the production of lucigenin chemiluminescence from peritoneal macrophages. These results suggest that KDT enhances the non-specific immune response via increase of tumor necrosis factor-α and nitric oxide and phagocytic activity from peritoneal macrophages.
