Bacterial components and their derivatives have been reported to mediate various immunomodulating activities and to activate immune cells including macrophage. In this study, the secretory and cellular macrophage response to teichoicated fragments (TFs) from pneumococcal cell wall subcomponent were examined. Tumoricidal activity was measured by MTT assay and secretory molecules were assessed by biological assay. After stimulation of macrophages with various doses of TFs for 18hrs, secretion of TNF-$\alpha$, nitrite and $H_2O$$_2$ were significantly increased as compared to medium-treated control. In addition, tumorcidal activity of TFs-treated macrophages was enhanced, whereas production of IL-1 and IL-6, and phagocytic activity were not induced. These data suggest that TFs is a potent inducer of macrophage secretory and cellular activities.
Effects of squalene on cellular and non-specific immune responses and antitumor activity in mice were investigated. Cellular and non-specific immunological assay parameters adopted in the present study were delayed-type hypersensitivity reaction and resette forming cells (RFC) for cellular immunity, activities of natural killer (NK) cells and phagocyte for non-specific immunity. Squalene resulted in marked increases of cellular and non-specific immune functions and enhancement of host resistance to tumor challenge in dose-dependent manner.

Cellular components of Lactococcus lactis ssp. lactis (heat-killed whole cells, cytoplasm, and cell walls) were tested for their in vivo immunopotentiating activity. Peritoneal macrophages from mice orally administered with heat-killed whole cells exhibited significantly greater phagocytic activity than the groups administered with cell-wall fraction or cytoplasm fraction. The cytotoxicity of natural-killer cells was the highest in the group administered with whole cells, and the production of cytokines ($IFN-\gamma$, IL-2, and IL-12) in spleen cells was significantly higher, when cellular components were injected, and it tended to be higher in the cell-wall and cytoplasm groups than in the whole-cell group. Interestingly, the cytokine production of Peyer's patch cells was high, when cytoplasm fractions were administered. These results demonstrate that whole cells and cytoplasm and cell-wall fractions of L. lactis ssp. lactis have immunopotentiating activities, which are related to the stimulation of Peyer's patches.

This study were done to know the effects of Dangkiyeumja on the in vivo and in vitro immune responses of mice. The recipes of Dangkiyeumja used in this study enhanced such, cellular functions of immunocytes as phagocytic capacity of macrophages, rossett-eforming abilities of splenocytes and metabolic activities of lymphocytes, However, the same recipes decreased the formation of such reactive oxygen intermediates(ROI) as superoxide and hydrogenperoxide from the macrophages. The effects of the same recipes on the in vim immune responses was suppressive on the cellular immune response(CIR)measured by delayed-type hypersensitivity against dinitrofluorobenzene and mildly enhancing for the humoral immune response measured by antibody production against sheep red blood cells. The results of this study could be summarized as follow: 1. Administration of Dangkiyeumja enhanced the phagocytic activity of the murine macrophage. 2. Administration of Dangkiyeumja decreased the formation of ROI in the murine macrophage 3. Administration of Dangkiyeumja increased the number of the splenic rotte forming cells in the mouse. 4. Administration of DangKiyeumja did not effect the antibody production against sheep red blood cells. 5. Administration of Dangkiyeumja depressed the delayed-type hypersenitivity against dinitrofluoro benzene in the mouse. The result of this study suggest that Dangkiyeumja could ameliorate the hypersensitivity reactions by reducing the formation of ROI and decreasing the CIR without affecting the other functions of immunocytes.
Objectives : Anticancer and immune-modulating effects of several Korean medical prescriptions including Yukgunja-tang, Bohwa-tang, Sogam-Won, and Kamiboa-tang were investigated. Methods : In vitro anti-cancer effects were measured by cytotoxicity MTT assay using SNU-1 gastric cancer cell lines, In vivo anti-cancer effects were measured by increased life span of S-180 sarcoma-injected ICR mouse. Immune-modulating effects were analyzed by measuring hemagglutinin titer, appearance of rosette forming cells, lymphocyte proliferation, and phagocytic index in methotrexate-pretreated mice. Results : In vitro assay showed that only Sogam-won showed cytotoxic effect with $IC_{50}$ of 87.9 ${\mu}g/ml$. All other prescriptions showed no cytotoxic effects against SNU-1 gastric cancer cell line. However, in vivo assay showed that Sogam-won showed lowest anti-cancer effects in contrast to its highest cytotoxic effects, Kamiboa-tang, which showed no cytotoxic effect, showed the highest in vivo anticancer effects, with increased life span of 140%. Kamiboa-tang showed significant immune-enhancing activities by significantly increasing rosette forming cells, lymphocyte proliferation, and phagocytic index in methotrexate-pretreated mice (P<0.05). Conclusion : The anticancer effect of Kamiboa-tang is not mediated by direct inhibition of cancer cells but is mediated by improving immune reactions against cancer cells.

Immunological activities of the combined-administration of Acanthopanacis Cortex and Lycii Cortex Radicis were examined in BALB/C mice. The 40% ethyl alcohol extract of Acanthopanacis Corter (AE) or the 40% ethyl alcohol extract of Acanthopanacis Coriex and Lycii Cortex Radicis (ALE) were administered p.o. once a day for 7 days, respectively. AE did not affect the viability of thymocytes, but ALE decreased the viability of thymocytes. ALE enhanced the viability of splenocytes increased by AE. Also, AE enhanced the population of cytotoxic T cell in thymocytes, and ALE enhanced the population of helper T cell compared with AE. Furthermore, AE increased the population of $Thy1^+$ cells in splenocytes, and increased the population of splenic $CD4^+$ cells. In addition, ALE enhanced the phagocytic activity which was decreased by AE ALE decreased the production of nitric oxide increased by AE in peritoneal macrophages. These results suggest that ALE enhance an immune-regulative action of AE.
The effects of temperatures on pearl oyster, Pincdata fucata were studied by evaluating some functional immune responses of the haemocytes. Water temperature is one of the most important factors in bivalve immune defense. Haemocytes comprise a primary line by inflammation, encapsulation and phagocytosis. These phagocytic abilities of haemocytes were observed in different temperatures. The number of the circulating haemocytes by migratory assay, phagocytic activities by MTT assay and reactive oxygen species production of haemocytes by CL assay were measured at different temperatures. Results showed that pearl oyster maintained at 20℃ and 25℃ displayed significantly higher values for all the measured immune parameters in comparison to maintained at 10, 15, and 30℃.

Inhibitory effects of cockle extracts on carcinogen-induced cytotoxicity in C3H/10T1/2 cells were studied. Soup (62$\mu\textrm{g}$/mL), solubility (28$\mu\textrm{g}$/mL) and liposolubility (9 $\mu\textrm{g}$/mL) of the cockle inhibited 3-methyl-cholanthrene(MCA)-induced cytotoxicity in C3H/10T1/2 cells by 53 and 94%, respectively. These results suggest that the extracts cockle might have anticarcinogen-induced cytotoxicity of C3H/10T1/2 cells. The effects of cockle extracts on the immune response related to its antitumor activity in vitro and in vivo were investigated. The cockle extracts showed a direct cytotoxic effect on sarcoma-180 cells, tumor cells in vitro. Soup (0.49 mg/mL), solubility (0.11 mg/mL) and liposolubiliy (0.05 mg/mL) of the cockle markedly decreased the total numbers of sarcoma-180 cells, but not their viability. The phagocytic acitivity of peritoneal macrophage of mice was significantly augmented by these extracts of the cockle compared with that of control in vivo. These extracts also raised the phagocuytic index, indicating that the number of phagocytize dmicrobes per macrophage increased. Thus, cockle extracts might show a antitumor activity by enhancing the phagocytic cell activities.
Two dietary experiments were performed to evaluate the impact of the herb Mentha piperita as a dietary supplement on Catla catla. In Experiment 1, fingerlings (0.45-2.60 g) were fed on diets supplemented with M. piperita at different levels (P1, P2, P3, and P4 at 2, 4, 6, and $8g\;Kg^{-1}$ of feed) and C1 served as the control with no peppermint supplementation. In Experiment 2, fingerlings (0.40-3.15 g) were fed with diets containing autochthonous probiotic bacteria Bacillus coagulans (3000 colony forming unit $g^{-1}$) along with M. piperita (PP1-PP4 at 2, 4, 6, and $8g\;Kg^{-1}$ of feed) and feed C2 served as control containing probiotics but no peppermint. Significantly (P < 0.05) high values of growth parameters, digestive enzyme activities, low reed conservation ratio, low excretion of ammonia, and orthophosphates were observed in fingerlings fed with P3 for Experiment 1 and PP3 for Experiment 2. Hematological counts, phagocytic activity, and respiratory burst activity were also enhanced in fingerlings fed with M. piperita supplementation revealing that dietary administration of peppermint at optimum level can enhance the growth, digestibility, and immunity of C. catla fingerlings; however, incorporation of B. coagulans showed better growth and immunity revealing that synergistic effects of M. piperita and autochthonous probiotic in C. catla diets can lead to its sustainable aquaculture.
Ovotransferrin (OTF), an egg protein known as transferrin family protein, possess strong antimicrobial and antioxidant activity. This is because OTF has two iron binding sites, so it has a strong metal chelating ability. The present study aimed to evaluate the improved immune-enhancing activities of OTF hydrolysates produced using bromelain, pancreatin, and papain. The effects of OTF hydrolysates on the production and secretion of pro-inflammatory mediators in RAW 264.7 macrophages were confirmed. The production of nitric oxide (NO) was evaluated using Griess reagent and the expression of inducible nitric oxide synthase (iNOS) were evaluated using quantitative real-time polymerase chain reaction (PCR). And the production of pro-inflammatory cytokines (tumor necrosis factor [TNF]-α and interleukin [IL]-6) and the phagocytic activity of macrophages were evaluated using an ELISA assay and neutral red uptake assay, respectively. All OTF hydrolysates enhanced NO production by increasing iNOS mRNA expression. Treating RAW 264.7 macrophages with OTF hydrolysates increased the production of pro-inflammatory cytokines and the phagocytic activity. The production of NO and pro-inflammatory cytokines induced by OTF hydrolysates was inhibited by the addition of specific mitogen-activated protein kinase (MAPK) inhibitors. In conclusion, results indicated that all OTF hydrolysates activated RAW 264.7 macrophages by activating MAPK signaling pathway.
