Occurrence of Leaf Blight of Sweet Persimmon Tree Caused by Pestalotiopsis theae in Korea (단감나무 둥근갈색무늬병의 발생 상태)

  • Leaf blight outbreak was investigated in sweet persimmon tree orchards in Korea during a three-year period from 1995 to 1997. The man percentage of sweet perismmon leaves blighted by Pestalotiopsis theae was 15.9%. The highest disease incidence was surveyed to 20% in Milryang. The disease began from early June to late October, and peaked in September and October. The lesion size on leaf ranged mainly 1-3 cm. The typical symptoms were large grayish concentric lesions of oval patterns of either mesophyll or margin of the leaf, and olde trees were more susceptible than younger ones.

Environmental Factors Affecting Conidial Germination of Persimmon Leaf Blight on Sweet Persimmon Tree (단감나무 둥근갈색무늬병균(Pestalotiopsis theae)의 분생포자 발아에 미치는 환경요인)

  • The conidial germination of Pestaotiopsis theae was occurred in cells attched with pedicels, the inferior cell of conidium. The cells were swollen like a ball, and then germinated in 4 hours under favorable conditions. Generally, it was considered that fifty percent of whole conidia was germinated in the range 6~12 hours. The optimum temperature and pH for conidial germination of P. theae (SP-3) causing leaf blight on sweet persimmon was $25^{\circ}C$ and pH 5, respectively. Conidial germination rate was higher than 90% at 100% relative humidity, but never germinated at a relative humidity lower than 88.5%. Conidial germination was highest at the concentration of 1~4$\times$104 conidia/ml. In case of cultural media, the conidial germination was higher than 90% on PDA, PSA, OME and Leonian agar. There was no relationship between light and conidial germination. It was concluded that the key environmental factors affecting conidial germination of the fungus (SP-3) were temperature and moisture.

Effects of Persimmon leaf on the Photoaging Skin Improvement(2) (감잎의 광노화 피부 개선에 미치는 효과(2))

  • This study was performed to investigate the anti-photoaging effects of Persimmon leaf tea(PLT) in hairless mice(SKH-1) exposed to UVB radiation. The animals were divided into non-treated group (normal, N) and UV-radiated groups. UV-radiated groups were divided into only UV-radiated group(control, C) and UV-radiated and PLT treated experimental groups[first extraction treated group(PLT-I), second extraction treated groupe(PLT-II), and third extraction treated group(PLT-III)]. Three PLT treated experimental groups of mice were treated with both oral administration(300mg/Kg B.W./day) and topical application (100 ul of 2% conc./mouse/day) for 4 weeks. Anti-photoaging effects of Persimmon leaf were evaluated by MTT assay, anti oxidative reaction, MMP immunohistochemistry, gelatin zymography assay and RT-PCR observations. Treatment with Persimmon leaf tea(PLT)-I, and -III groups decreased immunohistochemical density of matrix metalloproteinases(MMP)-3 and -9 related to degradation of extracellular matrix in skin. Especially, immunohistochemical density of MMP-2 decreased in PLT-I, -II and -III groups in skin. On the effects of antioxidant function on the treatment with Persimmon leaf tea(PLT), treatment of HaCaT cells with extracts of PLT-I and PLT-II had also significantly reduced intracellular ROS produced by UVB irradiation in a dose dependent manner(PLT-I, p<0.05, p<0.01, p<0.001; PLT-II, p<0.01, p<0.001). Gelatin zymography assay revealed that PLT-II and PLT-III (200 ug/ml) had inhibitory effect on MMP-9 expression in UVB-radiated HaCaT cells. Western blot analysis revealed that PLT-1, -II and -III groups down-regulates the expression of inflammatory associated genes(IL-$1{\beta}$) and PLT-1 and -II groups down-regulates the expression of TNF-${\alpha}$ in a dose dependent manner. Our study suggests that Persimmon leaf tea(PLT) extracts participates in inhibitory effects on the morphological and molecular experiments related to photoaging skin on UVB irradiated hairless mice.

Effects of persimmon leaf extracts on proteasome activity in HepG2 human liver cancer cells (감잎 추출물이 HepG2 인간 간암 세포의 proteasome 활성에 미치는 영향)

  • Proteasome inhibitors can promote apoptosis and cell cycle arrest in cancer cells by inhibition of nuclear factorkappaB ($NF-{\kappa}B$) activation. The purpose of this study was to investigate the effects of persimmon leaf extract (PSE) on proteasome activity in HepG2 human liver cancer cells. PSE treatment inhibited the proteasome activity and $NF-{\kappa}B$ activation in a dose-dependent manner in HepG2 human liver cancer cells (p<0.05). PSE treatment increased the population of cells in G2/M and sub-G1 phases. The results suggested that PSE is one of the candidate substances that may be developed into a proteasome inhibitor.

The Aroma Components of Duchung Tea and Persimmon Leaf Tea (두충차와 감잎차의 향기성분)

  • The aroma components of duchung tea and persimmon leaf tea were collected simultaneous distillation and extraction method. Those were analyzed and identified by GC-MS using a fused sillica capillary column. Seventy seven components, including 17 alcohols, 3 hydrocarbons, 15 ketones, 16 aldehydes, 8 esters,8 acids, 8 heterocyclic compounds and 2 phenols were confirmed in duchung tea. Seventy one components, including 17 alcohols, 10 hydrocabrons, 11 ketones, 13 aldehydes, 8 esters, 6 acids, 5 heterocyclic compounds and 1 phenol were confirmed in persimmon leaf tea. The most abundant components of duchung tea were aldehydes(23.31%) including 2-pentenal and heterocyclic compounds(16.71%) including $epoxy-{\beta}-ionone$. The most abundant components of persimmon leaf tea were alcohols(25.57%) including linalool and aldehydes(19.45%) including hexanal and hydrocarbons(10.40%) including ${\alpha}-copaene$.

Extraction of Flavonoid Components from Persimmon Leaf, Thistle and New Green (감잎, 엉겅퀴, 뉴그린으로부터 플라보노이드성분의 추출)

  • In this study, we extracted active components from thistle, persimmon leaf, and new green which are known to have a high content of antioxidants and also analyzed the 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavening activity and flavonoid content. Both ultrapure water and alcohol were used as extraction solvents and the ratio of both solvents, sample, amunts extraction time, and extraction temperature were varied. The optimal extraction condition of each natural compounds were 2.5~3.5 h of the extraction time and 50 g/L of the sample amount. The optimal ratio of ultrapure water and alcohol and extraction temperature were as follows; persimmon leaf (55~65 vol%, $50{\sim}60^{\circ}C$), thistle (40~50 vol%, $55{\sim}65^{\circ}C$) and new green (55~65 vol%, $50{\sim}60^{\circ}C$). In addition, the antioxidant capacity and flavonoid content of the extract increased in the order of persimmon leaf, thistle, and new green.

  • The aim of this study was to examine the efficiency of Rhodosporidium toruloides as a new tool to evaluate the triglyceride (TG) reduction effects of anti-obesity candidate materials. Unfermented and fermented persimmon leaf hot water extracts (UFPLE and FPLE) were used as anti-obesity agents. The content of TG in R. toruloides treated with FPLE was less than those with UFPLE by about 11% (p < 0.05) relative to the control (R. toruloides incubated in YPD medium without the agents). Fat reduction in 3T3-L1 cells achieved by FPLE was about 13% higher than that achieved by UFPLE.

  • The antimutagenic effects of persimmon leaf tea extract (PLTE) at concentration levels consumed by human were examined in mice using micronuleus induction with MMC(mitomycin C) or 4-NQO (4-nitroquinoline-1-oxide). When mice received oral gavage of 10 equivalent to PLTE 24 hr and 6 hr before, and 5 equivalent to PLTE 6 hr before and 3 hr after intraperitoneal injection of MMC, a significant decrease in the frequency of micronuclei were observed. The induction of micronuclei by 4-NQO was suppressed by oral dosage of PLTE at 5 equivalent to PLTE 6 hr before and 3 hr after, 10 equivalent to PLTE 3 hr before and 3 hr after intraperitoneal injection of MMC. Though the components of PLTE have not been analyzed so far, our present results suggest the existence of several bio-antimutagens and/or desmutagens in PLTE, beside catechin, well-known antimutagen.

Qualities of Bread Added with Korean Persimmon(Diospyros kaki L.folium)Leaf Powder (한국산 감잎가루를 첨가한 빵의 품질)

  • The purpose of this study was to investigated quality properties of breads prepared from wheat flour with persimon leaf power. Results showed that the volume of bread was lessened, the weight of bread went up, and baking loss rate diminished as persimmon leaf powder content incremented. The experiment showed that bread was maintained in the low level of springiness and cohesiveness was relatively reduced as time lapsed, whereas 3.0 and 5.0% test group remained significantly high level. There was a rapid increment in chewiness and gumminess, which tended to show significantly low as compared to control group, Results of sensory evaluation showed that the preference scores decreased as the persimmon leaf powder contents increased (p<0.05), flavor made no significant differences up to 3.0% test group, however, 5.0% test group containing elutionsuch as catechin or tannin produced sour and bitter taste so strongly that made if feel uncomfortable with. The augment of content indicated a low prederence in terms of texture, in an overall preference had no significant differences as compared to the control group, and 0.5% test group. Through the results of these of these experiments, we can conclude that the highest quality of persimmon leaf powder content is no more than 0.5% in making bread added with persimmon leaf powder.

Studies on Antioxidative Effect and Lactic Acid Bacteria Growth of Persimmon Leaf Extracts (감잎 추출물의 유산균 생육 및 항산화 효과에 관한 연구)

  • This study was investigated the growth effect of persimmon leaf extracts on various Lactic acid bacteria, the effect as freeze-dry cryoprotectan, by the changes of vitamin C content, electron donating ability, total cell count and SOD-like activity. The total cell count of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 in the absence of persimmon leaf extracts(10%) at 37℃ after 48hr were 3.2×10/sup 9/cfu/mL, 2.9×10/sup 9/cfu/mL, 1.1×10/sup 9/cfu/mL, 1.6×10/sup 9/cfu/mL and 3×10/sup 9/cfu/mL, respectively, and pH and total acidity of those were 3.82∼3.88, and 1.697∼1.842. On the other hand, the total cell count of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 in the presence of persimmon leaf extracts at 37℃ after 48hr were 4.4×10/sup 9/cfu/mL, 4.3×10/sup 9/cfu/mL, 2×10/sup 9/cfu/mL, 3.3×10/sup 9/cfu/mL, and 3.4×10/sup 9/cfu/mL, respectively, and pH and total acidity of those were 3.74∼3.82 and 1.528∼1.805. The total cell counter of lactic acid bacteria in the presence of persimmon leaf extracts(10%) at 37℃ after 48hr higher than those in the absence of persimmon leaf extracts. In freeze-dry chryoprotectan, the survival rate of L. acidphilus MG501 decreased about 30%. In case of L. brevis MG19 and L. bulgaricus MG515, the survival rate decreased about 10%. However, the survival rate of L. lactis MG530 increased about 10% and L. casei MG311 showed the almost same effect. The changes of vitamin C content were the highest in L. lactis MG530(190.26mg/mL) and the lowest in L. acidophilus MG501(56.05 mg/mL). The electron donating ability indicated to L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 added by 10% persimmon leaf extracts, respectively. when 10% persimmon leaf extracts were added L. brevis MG19, the electron donating ability is the highest of all lactic acid bacteria. The SOD-like activity of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 showed lower than that of control.
