Anti-wrinkle Activity of a Curdrania tricuspidata Extract on Ultraviolet-induced Photoaging (Hairless Mice를 이용한 광노화 모델에서 꾸지뽕나무 추출물의 피부주름 형성 억제효과)

  • We investigated the anti-wrinkle activity of an 80% ethanol extract of Curdrania tricuspidata leaves (CTL80) on ultraviolet-induced photoaging in hairless mice. Skin wrinkles were induced by 10 weeks of UVB-irradiation on the back of Skh-1 hairless mice three times a week. Mice were divided into ten groups; normal control (-UVB), UVB irradiated control group (+UVB), dietary groups (UVB+ascorbic acid 0.1%, UVB+CTL80 0.1%, UVB+CTL80 0.25%) and topical application groups (-UVB+base lotion (BL), UVB+BL, UVB+ascorbic acid 1%+BL, UVB+CTL80 1%+BL, UVB+CTL80 2%+BL). Wrinkle formation, histological changes, superoxide dismutase (SOD) activities, glutathione peroxidase (GSH-Px), and the expression of matrix metalloproteinases (MMP-1, MMP-3 and MMP-9) were analyzed. Wrinkles for the +UVB groups formed as a pattern of deep furrows and thick crests. Wrinkles with CTL80 treatment formed as a pattern of shallow furrows and thin crests, with wrinkle areas were lower than the +UVB group. In an antioxidant analysis of mouse blood, SOD and GSH-Px activities were significantly higher in the CTL80 topical application group compared to the +UVB group. The mRNA expression of MMPs in the +UVB group was significantly higher than the normal control group, and significantly lower in the CTL80-treated group. In conclusion, CTL80 exerted anti-wrinkle activity on ultraviolet-induced photoaging by regulating antioxidative defense systems and MMPs expression.

Effect of buchu (Allium tuberosum) on lipid peroxidation and antioxidative defense system in streptozotocin-induced diabetic rats (부추가 Streptozotocin 유발 당뇨쥐의 지질과산화와 항산화방어체계에 미치는 영향)

  • The pathogenic effort of high glucose, possibly in concert with fatty acids, is mediated to vascular complications of diabetes via increased production of reactive oxygen species(ROS), reactive nitrogen species(RNS), and subsequent oxidative stress. This study was carried out to investigate the suppressive effect of buchu(Allium tuberosum) on oxidative stress in streptozotocin(STZ)-induced diabetes in Sprague Dawley male rats. The effect of buchu supplementation (10%) on lipid peroxidation, and antioxidative defense system in blood and liver was compared among normal rats fed basal diet(normal) and diabetic rats fed basal diet(DM-control) or 10% buchu-supplemented diet(DM-buchu). Diabetes was experimentally induced by the femoral muscle injection of 50 mg STZ per kg of body weight. Animals were sacrificed after 4 wks of experimental diets feeding. The induction of diabetes by STZ elevated the level of lipid peroxidation represented by thiobarbituric acid-reactive substances(TBARS) and conjugated dienes in plasma, LDL, liver, and erythrocytes. 10% buchu-supplemented diet significantly reduced the levels of conjugated dienes in erythrocytes(p<0.05) and lowered TBARS in liver and LDL to the levels of control. Induction of diabetes by STZ elevated Mn-superoxide dismutase(Mn-SOD) activity and lowered activities of glutathionine reductase(GSH-red) and glutathionine peroxidase(GSH-px). Catalase activity was not affected by the induction of diabetes by STZ. However, buchu supplementation to diabetic rats significantly elevated catalase activity(p<0.05) and slightly elevated GSH-px and GSH-red activities in liver. GSH levels of blood and liver were lowered or not changed by induction of diabetes by STZ, respectively, while buchu supplementation to diabetic rats significantly elevated hepatic GSH level (p<0.05). In conclusion, it can be concluded that buchu might be a food source to attenuate oxidative stress in diabetic patients by inhibiting lipid peroxidation, by increasing hepatic GSH level, and by inducing anti-oxidative enzyme systems.

In vivo Study of the Renal Protective Effects of Capsosiphon fulvescens against Streptozotocin-induced Oxidative Stress (스트렙토조토신 유발 당뇨 쥐의 산화스트레스에 대한 매생이 추출물의 신장 보호 효과)

  • In this study, we evaluated the effect of Capsosiphon fulvescens extract (CFE) and its active compound, pheophorbide A (PhA), on diabetic kidney failure. Diabetes mellitus (DM) was induced by a single intraperitoneal injection of streptozotocin (STZ; 40 mg/kg body weight (BW)). After a week, the rats were orally administered CFE (4 and 20 mg/kg BW) or PhA (0.2 mg/kg BW) once a day for 9 weeks. After scarification, renal tissue samples were collected for biochemical and histochemical analyses. Our study showed that the treatment with CFE and PhA significantly decreased lipid peroxidation level and the activities of glutathione peroxidase and glutathione-S-transferase (p<0.05), but it increased glutathione level and the activities of glutathione reductase, superoxide dismutase, and catalase in the renal tissues (p<0.05). The CFE- and PhA-treated rats with DM showed improved histochemical appearance and decreased abnormal glycogen accumulation. Therefore, we suggest that PhA-containing CFE could exert renal protective effects against STZ-induced oxidative stress.

The Effects of Solanum nigum Linne extract on the Hepatotoxicity of Rats Induced by Lipopolysaccharide (Lipopolysaccharide로 유발된 간독성에 대한 까마중의 효과)

  • This study aimed to investigate the protective effect of Solanum nigum Linne total extract (SNT), Solanum nigum Linne leaf extract (SNL), Solanum nigum Linne root extract (SNR) on liver injury induced by Lipopolysaccharide(LPS) in Sprague-Dawley rats. SNT, SNL, SNR of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. on the day 1.5 ml/kg of LPS was injected. Four hours later, they were anesthetization with ether and dissected. Glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) were measured in serum and superoxide dismutase (SOD), catalase and glutathione peroxidase (GPX) were measured in liver homogenate. SNT, SNL, SNR extract inhibited GOT and GPT activities in LPS-induced rats, whereas increased SOD, Catalase and GPX activity in liver tissue of LPS-induced rats. These suggested that SNT, SNL, SNR could be used for functional beverage.

Decontamination of Waste Water Polluted with Phenolic and Anilinic Compounds Using Plant Materials (식물체를 이용한 Phenol 및 Aniline성 폐수의 정화)

  • This study was carried out to estimate the possibility on the removal of phenols and anilines, which were contained in pulp or dye waste water, and the reusability of plant materials, shepherd's purse and turnip. Most of phenols catalyzed with shepherd's purse were removed more than 90% in the presence of $H_2O_2$, and the removal was ranged from 53.1% for 2,6-DMP to more than 99% for 2,4,6-TCP when turnip was used as catalysts. The removal of anilines catalyzed with shepherd's purse was ranged from 42.2% for 2-CA to 78.7% for 3,4-DCA in the presence of $H_2O_2$, and in case of turnip, from 31.5% for 2-CA to 90.0 % for 2,4-DCA. The reuse of plant materials was proved to be possible for not only the batch method but also the continuous method. No decreasing removal was observed during 30 cycles in waster water contaminated with 100ppm of 2,4-DCP. However, it was observed that the removal was decreased with increasing the number of cycles in higher concentration of 2,4-DCP(800ppm). Therefore, it could be suggested that the number of reusable cycles depends on the initial concentration of substrates.

Protective Effects of Extract of Puerariae radix on Hepatic injury Induced by Carbon Tetrachloride In Rat. (갈근 추출물이 사염화탄소로 유발된 흰쥐의 간 손상에 대한 보호효과)

  • This study was done to investigate the protective effects of ethanol extract puerariae radix(Pr) on carbon tetrachloride($CCl_4$) intoxicated rats. Male sprague Dawley rats(200~210g)was used. experimental groups were divided into normal group, $CCl_4$-control group, and ethanol extract $CCl_4$-treated group. $CCl_4$-treated groups were injected with $CCl_4$ 0.6mg/kg.b.w(i.p). The activities of Alanine aminotransferase(ALT), Aspartate aminotransferase(AST), Alkaline phosphatase(ALP), Glutamyltranspeptidase(${gamma}$-GT), Lactate dehydrogenase(LDH) in extract pretrated group was significantly decreased(p<0.05) compared to the $CCl_4$-control group. The contents of triglyceride, cholesterol and lipid peroxide were significantly decreased(p<0.05). whereas content of HDL-choresterol was significantly increased.(p<0.05). In addition, activities of hepatic catalase(CAT), glutathione peroxidase(GSH-Px) in the extract pretreated rats were significantly decreased(p<0.05) compared to the $CCl_4$-control group. but the content of glutathione(GSH) was significantly increased(p<0.05). These results suggest that extract of puerariae radix(Pr) has hepatoprotective effect in the $CCl_4$-intoxicated rats.

Effects of Dietary Methionine Level on Lipid Peroxidation and Hepatic Morphology in Rat (식이중의 Methionine첨가수준이 흰쥐의 체내 지질 과산화와 간조직 형태에 미치는 영향)

  • The effect of dietary methionine level on lipid peroxidation of rats was studied. Rats were fed vitamin E- selenium- deficient diet or diet supplemented with various levels (0.3, 0.6, 0.9%) of methionine. In rat fed MF diet, body weight gain and feed efficiency ratio were decreased compared with those of control rats, but reversed by supplementation with 0.3 and 0.6% methionine. Lipid peroxide levels in plasma and hepatic mitochondrial fraction of MF group rats were significantly higher than those of control rats. However, supplementation with 0.6% methionine modified this increment. GSH-Px activity was decrased to varying degrees in erythrocyte and hepatic mitochondrial fraction from rats fed MF diet. Methionine supplementation did not affect induction of this enzyme activity. Examination of hepatocytes by electronmicroscopy showed that Influence of vitamin E, selenium, and methionine deficiency was mainly characterized by lipid droplets, swollen mitochondria and microvilli destruction. Supplementation with various levels of dietary methionine modified these changes to some extent. The results of this experiment indicated that MF diet causes significant change in lipid peroxide level, GSH-Px activity and morphology of rats which these changes may lessen by supplementation with 0.6% methionine.

Degradation of human immunoglobulins and cytotoxicity on HeLa cells by live Trichomonas vaginalis (질편모충 단백질분해효소의 세포독성 및 인체면역글로불린 분해능)

  • The present study was undertaken to determine whether live T. uaginnlis degrades human secretory IgA, serum If and IgG molecules. Human immunoglobulins were exposed to live trophozoites, parasite Iysate, and excretory-secretory product (ESP) of T ucginnlis. To determine the fragmentation of immunoglobulins, the reaction sample was subjected to SDS-PAGE and EITB, and peroxidase conjugated antihuman IgA and IgG were used as probes. Live trophozoites degraded secretory IgA, serum IgA and IgG, and degradation were pressed forward by the prolongation of the incubation time and by increasing the number of trichomonads respectively. Also the Iysates and ESP of trichomonads degraded IgA and IgG. The cysteine and serine proteinase inhibitors such as I-64, antipain, iodoacetic acid, iodoacetamide, TLCK reduced the ability of cleaving immunoglobulins. The proteinase activity and cytotoxicity of T. uaginnlis to HeLa cells were decreased when live T. vusinalis was treated with metallo-proteinase inhibitor as well as cysteine and serine proteinase inhibitors. These results suggest that proteinase secreted from live T ucginclis may play a part role in host pathogenesis by T. uosinnlis, and the cleaving ability of host immunoglobulins by the proteinase may contribute as a one of immune evasion mechanism for parasite survival in the host.

Radioprotective Effect of Post-treatment with Rutin on γ-Irradiation-induced Cellular Damage in Mice (감마선 조사로 유도된 세포 손상 마우스에서 루틴 투여 후의 방사선 방호 효과)

  • This study aimed to investigate the therapeutic effect of rutin against whole-body ${\gamma}$-irradiation in BALB/c mice. BALB/c mice were randomly divided into four groups and exposed to 6 Gy ${\gamma}$-irradiation. One hour later, mice were orally administered rutin (50 and 100 mg/kg) for seven consecutive days. ${\gamma}$-Irradiation (6 Gy) resulted in cellular damage as manifested by elevated levels of plasma hepatic marker enzymes and lipid peroxidation in liver tissue, accompanied with decreased spleen and thymus indices, and white blood cell count. In addition, ${\gamma}$-irradiation significantly decreased the levels of antioxidant enzymes such as superoxide dismutase, glutathione peroxidase and catalase. Rutin treatment significantly protected against ${\gamma}$-irradiation-induced cellular damage, which was evident by the improvement in the status of most of the investigated parameters. Therefore, rutin has beneficial effects against radiation-induced damage.

Expression of Cu/Zn SOD according to H2O2 in Hepatoma cell line (Hepatoma 세포주에서 H2O2 처리에 의한 Cu/Zn SOD의 발현)

  • Oxygen is required for many important aerobic cellular reactions, it may undergo electrontransfer reactions, which generate highly reactive membrane-toxic intermediates (reactive oxygen species, ROS), such as hydrogen peroxide, singlet oxygen, superoxide radical, hydroxyl radical, hydroperoxyl radical, hydroxy ion. Various mechanisms are available to protect cells against damage caused by oxidative free radicals, including scavenging enzyme systems such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx). This antioxidant defense system is a very complex and finely tuned system consisting of enzymes capable of detoxifying oxygen radicals as well as low molecular weight antioxidants. In addition, repair and turnover processes help to minimize subcellular damage resulting from free radical attack. $H_2O_2$,one of the major ROS, is produced at a high rate as a product of normal aerobic metabolism. The primary cellular enzymatic defense systems against $H_2O_2$ are the glutathione redox cycle and catalase. From Northern blot analysis of total RNAs from cultured cell with $H_2O_2$ treatment, various results were obtained. Expression of Cu/Zn SOD decreased when cell passage increased, but the level of the Cu/Zn SOD was scarcely expressed in 35 passage.
