1,2-benzopyrone이 개 말초혈액 백혈구의 유주활성에 미치는 영향 (Effect of 1,3-Benzopyrone on Chemotactic Activity of Peripheral Blood Leukocytes in the Dog)

  • 개 말초혈액 백혈구의 유주성에 있어서 1,2-benzopyrone의 효과를 검토하였다. 백혈구의 유주성은 Boyden chamber 변법으로 측정하였다. 1.2-benzopyrone 그 자체는 PMN과 PBMC의 유주성에 직접적인 효과를 보이지 않았다. PMN과 PBMC는 1,2-benzopyrone으로 배양한 PMN의 배양상층액에 대해서도 유주활성을 보이지 않았다. 그러나 1,2-benzopyrone으로 배앙한 PBMC의 배양상층액은 PBMC에 대해서는 유주활성을 보이지 않았으나 PMN에 대해서는 현저한 유주활성을 나타내었다. 또한 유주인자인 IL-8에 의한 PMN의 유주활성 측정 결과도 1,2-benzopyrone으로 배양한 PBMC의 배양상충액의 그것과 유사하였다. 이러한 유주활성은 anti-IL-8 pAb를 처리했을 때 PMN의 유주활성이 억제되어, 본 유주활성은 PBMC에서 분비되는 IL-8일 것으로 강하게 시사되었다. 이상의 결과로부터, 1,2-benzopyrone은 개 말초혈액 PMN의 유주성에 대하여 면역자극 작용을 가지고 있으며, 이것은 1,2-benzopyrone의 자극에 의해 PBMC에서 분비되는 IL-8양 가용성 물질에 의해 나타나는 것으로 사료되었다

가미삼령백출산(加味蔘苓白朮散)의 B16 흑색종 암모델에 대한 항종양효과(抗腫瘍效果)와 면역증강효과(免疫增强效果)에 관한 연구(硏究) (The Effect of Gamisamryungbaekchul-san(加味蔘苓白朮散) on the Tumor and Immune Response in Mouse B16 Melanoma Tumor Model)

  • Background : Gamisamryungbaekchul-san(加味蔘?白朮散) is a herbal medicine which has been used for the traditional therapeutic agent of augmentation of the spleen and reinforcement of the Qi. Objective : This Study was performed to investigate the effect of Gamisamryungbaekchul-san on the tumor and immune response in the moose B16 melanoma tumor model. Materials and Methods : The tumor was induced by subcutaneous inoculation of B16BL6 melanoma cells in the shaved dorsal region of mice. Mice were orally administered with Gamisamryungbaekchul-san extract(26.3mg/mouse) for 14days after inoculation. For making examination of antitumor effect, the Increase of life span, Tumor growth inhibition rate, change of body weight were measured and evaluated. For the immune response increasing effect, the percentage of T lymphocyte and B Lymphocyte in the peripheral blood, the percentage of CD4+ T-cell, CD8+ T-cell and CD4+/CD8+ ratio in the peripheral blood and spleen, interleukin-2 productivity were measured and evaluated. Results : Gamisamryungbaekchul-san showed 16.59% increase of life span, 31.64% tumor growth inhibition rate and increase of body weight. Gamisamryungbaekchul-san increased the percentage of T lymphocyte in the peripheral blood, CD4+ T cell percentage of peripheral blood and spleen, and Interleukin-2 productivity as compared with the Control group. Whereas Gamisamryungbaekchul-san had no effect on the percentage of B lymphocyte in the peripheral blood, the percentage of CD8+ T cell, CD4+/CD8+ T-cell ratio in both of peripheral blood and spleen as compared with the Control group. Conclusion : This study shows that Gamisamryungbaekchul-san has anti-tumor effects and immunoregulatory effects on the B16 melanoma tumor model. It is suggested that Gamisarmyungbaekchul-san could be a useful immunomodulator and anti-tumor agent.

폐결핵환자의 말초혈액에서의 ${\gamma}{\delta}$ T 림프구에 관한 연구 (${\gamma}{\delta}$ T Cells in the Peripheral Blood of Pulmonary Tuberculosis)

  • 연구배경: ${\gamma}{\delta}$ T 림프구가 새로 발견된 이후 아직 명확한 기능이 밝혀지지 않았지만 M.tuberculosis의 HSP65에 반응함이 보고되었다. 아직도 결핵의 유병률이 높은 현실을 감안하여 폐결핵환자를 대상으로 하여 본 연구를 시행하였다. 방법: 16예의 건강대조군과 22예의 폐결핵군에서 말초혈액을 채취하여 PPD, Con-A, 그리고 H37Ra lysate로 자극하였으며, 자극전후의 ${\gamma}{\delta}$ T 림프구의 비율과 활성도를 유세포계측을 이용하여 측정하였다. 결과: 1) 건강대조군의 말초혈액의 총 T 림프구 중에서 ${\gamma}{\delta}$ T 림프구의 비율은 $10.0{\pm}4.8%$ 이었고, 폐결핵군에서는 $7.5{\pm}5.2%$로 통계적으로 유의한 차이는 없었다. 2) PPD, Con-A, 그리고 H37Ra lysate로 자극한 후에도 ${\gamma}{\delta}$ T 림프구의 비율은 유의한 변화가 없었다. 3) ${\gamma}{\delta}$ T 림프구 중에서 IL-2R(+)인 비율은 건강대조군에서 $1.8{\pm}2.8%$, 그리고 폐결핵군에서는 4.8%로 폐결핵군에서 유의하게 높았다. 4) PPD 또는 Con-A로 자극후 양군 모두에서 IL-2R(+)의 비율은 통계적으로 유의하게 증가되었다. 5) PPD 피부반응검사와 말초혈액의 ${\gamma}{\delta}$ T 림프구의 비율간에는 유의한 상관관계가 없었다. 결론: 본 연구에서는 폐결핵환자의 말초혈액에서 ${\gamma}{\delta}$ T 림프구의 역할을 규명할 수 없었다. 국소병변에서의 ${\gamma}{\delta}$ T 림프구에 대한 연구가 더 필요하리라 생각된다.

갑상선 종양에 있어서 말초형 림프구의 Purine Nucleoside Phosphorylase (PNP) 활성과 T 세포 아형에 관한 연구 (Purine Nucleoside Phosphorylase (PNP) Activity of Lymphocytes and T Cell Subsets in Peripheral Blood in Thyroid Tumors)

  • 저자는 1991년 1월부터 동년 8월 사이에 부산대학교 병원 내과 외래에서 임상증상, 이학적 소견 및 각종 검사소견과 병리조직학적으로 진단된 단순 증식성 갑상선종 환자 20예, 갑상선선종 환자 9예 및 갑상선암환자 20예와 건강대조군 11례에서 말초혈 림프구의 PNP활성을 측정하고 $CD4^+$$CD8^+$ 세포를 동시에 검색하여 분석할 성적을 다음과 같이 요약한다. 1) 말초혈 림프구의 PNP 활성은 건강 대조군 및 단순 증식성 갑상선종 보다 갑상선선종 및 암환자에서 의의있게 증가하거나 증가하는 경향이 있었다. 2) 말초혈의 $CD8^+$ 세포 비율은 갑상선암환자에서 건강 대조군, 단순 증식성 갑상선종 및 갑상선선종환자 보다 각각 의의 있게 감소하거나 감소하는 경 향이 있었다. 3) 말초혈의 CD4/CD8비는 갑상선암환자에서 건강 대조군, 단순 증식성 갑상선종 및 갑상선선종환자보다 각각 의의 있게 증가하거나 증가하는 경향이 있었다. 이상의 결과에 의하면 갑상선암환자에서는 말초혈의 억제/세포상해 T 세포의 감소에 의한 세포성 면역능의 이상이 있고, 말초혈 림프구의 PNP활성의 측정은 갑상선종양환자의 면역 상태를 파악하는데 도움이 되는 검사라고 생각된다.

The Role of Dendritic Cells in Central Tolerance

  • Dendritic cells (DCs) play a significant role in establishing self-tolerance through their ability to present self-antigens to developing T cells in the thymus. DCs are predominantly localized in the medullary region of thymus and present a broad range of self-antigens, which include tissue-restricted antigens expressed and transferred from medullary thymic epithelial cells, circulating antigens directly captured by thymic DCs through coticomedullary junction blood vessels, and peripheral tissue antigens captured and transported by peripheral tissue DCs homing to the thymus. When antigen-presenting DCs make a high affinity interaction with antigen-specific thymocytes, this interaction drives the interacting thymocytes to death, a process often referred to as negative selection, which fundamentally blocks the self-reactive thymocytes from differentiating into mature T cells. Alternatively, the interacting thymocytes differentiate into the regulatory T (Treg) cells, a distinct T cell subset with potent immune suppressive activities. The specific mechanisms by which thymic DCs differentiate Treg cells have been proposed by several laboratories. Here, we review the literatures that elucidate the contribution of thymic DCs to negative selection and Treg cell differentiation, and discusses its potential mechanisms and future directions.

관절7호방이 Type II collagen으로 유발된 흰쥐의 관절염에 미치는 영향 (Suppressive Effect of GJB-7 on Collagen-Induced Arthritis in Mice)

  • Objectives: This study was carried out to know the effects of Gwan-Jul-Bang-7 (hereafter referred to GJB-7) on the inhibition of arthritis induced by collagen on the mouse. Methods: To assess the effects of GJB-7 on mouse with arthritis induced by collagen II, we conducted several experiments such as analysis of cytotoxicity, hepatotoxicity, arthritis index, total cell number of draining lymph nodes and paw joints, value of immunocyte in PBMC (peripheral blood mononuclear cell), DLN (draining lymph node) and paw joint, measurement of cytokine and anti-collagen II, observation of the histological changes of joint. Results: 1. Cytotoxicity against HFC (human fibroblast cells) was not observed in any concentration and hepatotoxicity was not observed in the GJB-7 treated group. 2. The incidence of arthritis significantly decreased. 3. Total cell number of draining lymph nodes significantly increased and total cell number of paw joints significantly decreased. 4. The percentage of $CD8^+$ cells in PBMC (peripheral blood mononuclear cell) significantly increased. The percentage of $CD3^+/CD69^+$, and $CD3^+/CD49b^+$ cells in PBMC significantly decreased. 5. The percentage of $CD19^+,\;CD3^+$, and $CD4^+/CD25^+$ cells in DLN (draining lymph nodes) significantly increased. The percentage of $B220^+/CD23^+$ cells in DLN significantly decreased. 6. The percentage of $CD3^+,\;CD4^+$, and $CD11b^+/Gr-1^+$ cells in paw joints significantly decreased. 7. The production of TNF-$\alpha$, IL-6, and MCP-1 significantly decreased. 8. Anti-collagen II in serum significantly decreased. 9. With the hematoxylin and eosin stain, inflammation of joint decreased. Under Masson's trichrome stain, the cartilage destruction and synovial cell proliferation and the expression of collagen fibers decreased. Conclusions: Comparison of the results for this study showed that GJB-7 had immunomodulatory effects. So we expect that GJB-7 could be used as an effective drug for not only rheumatoid arthritis but also another auto-immune diseases.

CD103+ Cells and Chemokine Receptor Expression in Breast Cancer

  • Mucosal environments harbour lymphocytes, which express several adhesion molecules, including intestinal homing receptors and integrin αE/β7 (CD103). CD103 binds E-cadherin, an integrin receptor expressed in intestinal endothelial cells. Its expression not only enables homing or retention of T lymphocytes at these sites but is also associated with increased T lymphocyte activation. However, it is not yet clear how CD103 expression is related to the clinical staging of breast cancer, which is determined by factors such as the size of the tumor (T), the involvement of nearby lymph nodes (N), and presence of metastasis (M). We examined the prognostic significance of CD103 by FACS in 53 breast cancer patients and 46 healthy controls enrolled, and investigated its expression, which contributes to lymphocyte recruitment in tumor tissue. Patients with breast cancer showed increased frequencies of CD103+, CD4+CD103+, and CD8+CD103+ cells compared to controls. CD103 was expressed at a high level on the surfaces of tumor-infiltrating lymphocytes in patients with breast cancer. Its expression in peripheral blood was not correlated with clinical TNM stage. To determine the localisation of CD103+ cells in breast tissue, tissue sections of breast tumors were stained for CD103. In tissue sections of breast tumors stained for CD103, its expression in T lymphocytes was higher compared to normal breast tissue. In addition, CD103+ cells expressed higher levels of receptors for inflammatory chemokines, compared to CD103- cells. CD103+ cells in peripheral blood and tumor tissue might be an important source of tumor-infiltrating lymphocyte trafficking, homing, and retention in cancer patients.

Di(2-ethylhexyl) phthalate에 의해 유도된 DNA손상과 소핵 형성 (DNA Damage and Micronuclei Induced by Di (2-ethylhexyl) phthalate in Human Breast Carcinoma MCF-7 cells)

  • Di-2-ethylhexyl phthalate (DEHP) is the most commonly used phthalate ester in polyvinyl chloride formulations including food packing and storage of human blood. DEHP is a well known as non-genotoxic carcinogen and endocrine disrupting chemical (EDC). DEHP have shown all negative results in ICH-guildeline recommended standard genotoxicity test battery. In this study, to assess the clastogenic and DNA damaging effect in human-derived tissue specific cells, DEHP was treated in human derived MCE-7 cells, HepG2 cells, LNCap cells, BeWo cells, MCE-10A cells, and female peripheral blood cells using micronucleus assay and in human breast carcinoma MCF-7 cells up to $1.28$\times$10^{-2}$ M using Comet assay. The in vitro micronucleus assay is a mutagenicity test system for the detection of chemicals which induce the formation of small membrane bound DNA fragment i.e. micronuclei in the cytoplasm of interphase cells, originated from clastogenic and/or aneugenic mechanism. The single cell gel electrophoresis assay (Comet assay) is used to detect DNA strand-breaks and alkaline labile site. In our results, DEHP increased significantly and/or dose-depentently and time-dependently micronucleus frequency at the 6 and 24 hr without metabolic activation system only in MCE-7 cells. DEHP treated with 2 hrs in MCF-7 cells using Comet assay induced DNA damage dose-depentantly.
