Isolation and Culture Characteristics of Strains for Color Removal of Reactive Dyes (반응성 염료의 색도제거를 위한 균주의 분리 및 성장 특성)

  • Strains degrading and decolorizing reactive dyes, Procion blue HEGN and Procion red HE7B were isolated from water system, are named as RBK1 and RRK, the growth characteristics of which were investigated. Decolorization efficiencies after 42 hrs in batch culture were 95% and 77%, respectively. and the optimal culture condition of temperature and pH were $30^{\circ}C$, 7.0. Decolorization efficiencies in condition of aerobic shaking culture by strains RBK1 and RRK conspicuously increased, and culture by strain RBK1 was found as 95% after 42 hrs, while standing culture was 64%, Optimum nitrogen source was peptone, and It was found that decolorization efficiencies by strains RBK1 and RRK increased up to 4,000mg/l of peptone concentration as nitrogen source, but peptone concentration did' nt influence the decolorization efficiency in above 4,000mg/l. When the concentration of dyes were more than 800 mg/l and 400 mg/l respectively, the strains RBK1 and RRK, which degrade Procion bule HEGN and Procion red HE7B, showed a sharply decreased decolorization efficiencies; then the specific growth rate were $0.25hr^{-1}$ and $0.09hr^{-1}$.

Optimizing the composition of the medium for the viable cells of Bifidobacterium animalis subsp. lactis JNU306 using response surface methodology

  • This research improved the growth potential of Bifidobacterium animalis subsp lactis strain JNU306, a commercial medium that is appropriate for large-scale production, in yeast extract, soy peptone, glucose, L-cysteine, and ferrous sulfate. Response surface methodology (RSM) was used to optimize the components of this medium, using a central composite design and subsequent analyses. A second-order polynomial regression model, which was fitted to the data at first, significantly lacked fitness. Thus, through further analyses, the model with linear and quadratic terms plus two-way, three-way, and four-way interactions was selected as the final model. Through this model, the optimized medium composition was found as 2.8791% yeast extract, 2.8030% peptone soy, 0.6196% glucose, 0.2823% L-cysteine, and 0.0055% ferrous sulfate, w/v. This optimized medium ensured that the maximum biomass was no lower than the biomass from the commonly used blood-liver (BL) medium. The application of RSM improved the biomass production of this strain in a more cost-effective way by creating an optimum medium. This result shows that B. animalis subsp lactis JNU306 may be used as a commercial starter culture in manufacturing probiotics, including dairy products.

Studies on the Production of Mycelium by Lyophyllum decastes in Submerged Culture (Lyophyllum decastes의 심부배양(深府培養)에 의한 균체(菌體) 생산(生産)에 관한 연구(硏究))

  • Cultural and nutritional conditions for Lyophyllum decastes and its chemical composition in a synthetic medium were investigated. The optimal temperature and pH for the production of mycelium were $25^{\circ}C$ and pH 7.5. The mycelium yield was the highest on 15th day. Among the carbon sources, glucose and CMC were the best for the production of mycelium and their optimal concentrations were 3 and 6%, respectively. As an organic nitrogen, proteose peptone was the best and $(NH_4)_2HPO_4$ as an inorganic nitrogen was good. The optimal concentration of proteose peptone and $(NH_4)_2HPO_4$ were 2 and 0.2%, respectively. The optimal ratio of glucose to proteose peptone for production of mycelium was 10 : 1. Also, the optimal concentrations of $K_2HPO_4$ and $MgSO_4$ were 0.2 and 0.06%, respectively and that of $CaCl_2$ was 0.1%. Among the bioextracts, yeast extract was the most effective and its optimal concentration was 1.5%. In chemical components of the mycelium of Lyophyllum decastes, total sugar, crude protein and crude fat were 34.80, 28.35 and 2.50%, respectively. Its ash was 7.57% and crude fiber 11.99%.

Application of Response Surface Methodology in Medium Optimization to Improve Lactic Acid Production by Lactobacillus paracasei SRCM201474 (반응표면분석법을 이용한 Lactobacillus paracasei SRCM201474의 생산배지 최적화)

  • The aim of this study was to establish the optimal medium composition for enhancing L(+)-lactic acid (LLA) production using response surface methodology (RSM). Lactobacillus paracasei SRCM201474 was selected as the LLA producer by productivity analysis from nine candidates isolated from kimchi and identified by 16S rRNA gene sequencing. Plackett-Burman design was used to assess the effect of eleven media components on LLA production, including carbon (glucose, sucrose, molasses), nitrogen (yeast extract, peptone, tryptone, beef extract), and mineral (NaCl, K2HPO4, MgSO4, MnSO4) materials. Glucose, sucrose, molasses, and peptone were subsequently chosen as promising media for further optimization studies, and a hybrid design experiment was used to establish their optimal concentrations as glucose 15.48 g/l, sucrose 16.73 g/l, molasses 39.09 g/l, and peptone 34.91 g/l. The coefficient of determination of the equation derived from RSM regression for LLA production was mathematically reliable at 0.9969. At optimum parameters, 33.38 g/l of maximum LLA increased by 193% when compared with MRS broth as unoptimized medium (17.66 g/l). Our statistical model was confirmed by subsequent validation experiments. Increasing the performance of LLA-producing microorganisms and establishing an effective LLA fermentation process can be of particular benefit for bioplastic technologies and industrial applications.

  • Effects of culture conditions and Mn(II) addition were investigated for production of extracellular manganese peroxidase by lignin-degrading fungus LSK-27, Nitrogen source was shown to more influence the production of extracellular manganese peroxidase by LSK-27 than carbon source. When peptone or yeast extract as nitrogen source was added, high MnP activity was obtained. Especially, nitrogen-sufficient culture condition was effective in MnP activity, showing significantly increase up to 1.0% peptone concentration, but carbon-sufficient was not. Mn(II) was shown to strongly induce the MnP production in culture fluids of LSK-27. Increase of MnP actiyity was obeserved up to addition of 100ppm Mn(II), and over this Mn(II) concentration appeared to be inhibitory. The highest level of MnP activity was attained when Mn(II) was added after 2 day incubation.

Pigment Production and Color Diference of Liquid Beni-koji under Submerged Cultural Conditions (액체 홍국의 배양조건에 따른 색소생산과 색조의 변화)

  • Mycelial growth, color difference and productivity of red pigment of beni-koji by Monascus anka KCCM 11832 were examined with respect to it's pigment in submerged culture with various medium and culture conditions. Shaking incubation was more promoted mycelial growth and the production of pigments than that for non-shaking incubation, and red color became ten times deeper. The production of red pigment was the highest when incubated at 25$\circ$C for 7 days in pH 6.0, but mycelial growth was showed the highest at 32.5$\circ$C. The levels of carbon and nitrogen source for maximum red pigment production were 2% rice powder and 0.05% peptone, respectively and the level of peptone for maximum pigment production was lower than that for maximum mycelial growth. Among pigmentation promoting agents tested, MgSO$_{4}$, was found to be suitable for the production of red pigment, and the optimum level was 0.1%.

  • From 182 non-pathogenic wild yeast isolates from flowers, Pichia silvicola UL6-1 and Sporobolomyces carnicolor 402-JB-1 were selected for potent ${\gamma}$-aminobutyric acid production and microbiological characteristics were investigated. Pichia silvicola UL6-1 formed ascospores and pseudomycelia. The strain was also halotolerant, growing well in 5% NaCl-containing yeast extract-peptone-dextrose (YPD) medium. Sporobolomyces carnicolor 402-JB-1 did not form ascospores or pseudomycelia and grew well on 10% glucose-yeast extract-peptone medium.

  • Lactobacillus spp. PAP1 과 Lactobacillus spp. MG-G2를 혼합구조로 사용하여 밤 발효제품을 제조하기 위한 제반 조건을 조사하였다. 밤 발효액은 phytone peptone, yeast exract, glucose를 각각 0.4, 0.2. 0.5%이상으로 첨가할 때 산도와 생균수가 가장 컸다. 첨가물 중 phytone peptone은 대두박이나 순물로 대체해도 같은 효과가 관찰되었다. 버섯추출물의 첨가는 산도와 생육을 약간 촉진하였으나 안정제는 약간의 감소를 가져왔다. 기본 발효액은 관능성적이 나빴으나 산도를 0.54%로 낮추고, 당을 10%로 보강하고, so-dium alginate를 0.2% 첨가하였을 때, 산미, 감미, 음용감이 크게 개선되었다

Production Condition and Utilities of Extracellular Biopolymer from Bacillus licheniformis (Bacillus licheniformis가 생산하는 Extracellular Biopolymer의 생산조건 및 이용특성)

  • A bacterium that produce biopolymer was isolated from Gochujang, one of Korean traditional fermented foods, and identified as Bacillus licheniformis. The production of biopolymer was highest and 34mg/250ml, when the baterium was cultivated in condition of sucrose 6.0%, Yeast extract 0.1%, peptone 0.l%, NaCl 3.0%, and pH 6.0. The 1% solution of this biopolymer was able to form a translucent and glossy film. And the solution of biopolymer was found to precipitate Kaolin solution and also stabilize the suspension of lactic acid bacteria within the test range of 0.25-1.00%.   • Arthrobacter simplex (ATCC 6946) was cultured, induced and immobilized in acrylamide polymer. The characteristics of the immobilized whole-cell enayme were studied using hydrocortisone as the substrate. The enzyme activity was increased during the incubation of the gel particle in 0.5% peptone media. The ennzyme reaction kinetics of the Δ'-dehydrogenase (3-oxosteroid Δ'-oxydo reductase, E. C. 1.3.99.4) foliowed the Michaelis-Menten type. Km and Vm values were different significantly after immobilization of the cell. The optimum pH and temperature were changed, too. Nitrogen sources such as casitone, peptone or tryptone were good media for the enzyme reaction. And there was no need to add cofactors of the enzyme in the pre-sence of energy sources used in the test. The effect of metal ions on the enzyme activity was insignificant. Organic solvents were used increase the substrate concentration and there was no optimum solvent concentration depending on the substrate concentration.
