Epigallocatechin Gallate (EGCG)에 노출된 용혈성 Bacillus cereus MH-2의 세포 반응 및 프로테옴 분석 (Cellular responses and proteomic analysis of hemolytic Bacillus cereus MH-2 exposed to epigallocatechin gallate (EGCG))

  • 본 연구의 목적은 시중에 판매되고 있는 쌈장에서 용혈성을 가지는 Bacillus cereus MH-2를 분리하여, EGCG 노출에 따른 MH-2 균주의 세포 반응과 프로테옴 분석을 위해 수행되었다. 다양한 농도의 EGCG에 노출된 MH-2 균주는 노출시간이 증가함에 따라 생존률은 점차 감소함을 보였다. MH-2 균주의 alginate 생성량은 EGCG의 농도가 증가함에 따라 감소하였으며, 특정 EGCG 농도에서 노출시간이 진행됨에 따라 그 생성량은 증가하는 것으로 나타났다. SDS-PAGE 및 anti-DnaK와 anti-GroEL의 단일항체를 이용한 Western blot 통한 분석으로, 두 가지 스트레스 충격단백질인 70 kDa의 DnaK와 60 kDa의 GroEL의 발현은 대수생장기의 배양에서 EGCG의 농도에 비례하여 감소하는 것을 확인하였다. EGCG에 노출된 세균의 세포 외부형태 변화를 주사전자현미경을 이용하여 관찰한 결과, 세포 표면의 돌출부 생성과 함께 세포의 뭉그러짐이 관찰되었다. EGCG에 노출된 Bacillus cereus MH-2 배양의 수용성 단백질 부분에 대한 2-DE에서 20개의 단백질 스팟이 EGCG 노출에 의해 크게 변화하는 것이 확인되었다. 장독소(hemolysin BL lytic component L1, hemolysin BL-binding protein), chaperon (DnaK, GroEL), 세포방어요소(peptidase M4 family proteins), 에너지 및 물질대사 등에 수반되는 이들 단백질은 MALDI-TOF를 사용한 peptide mass fingerprinting에 의해 동정되었다. 이들 결과는 B. cereus MH-2에 대한 EGCG-유도 스트레스와 세포독성의 기작을 이해하는데 중요한 단서를 제공할 것이다.

Proteomic Analysis of Protein Expression Patterns Associated with Astaxanthin Accumulation by Green Alga Haematococcus pluvialis (Chlorophyceae) Under High Light Stress

  • Two kinds of Haematococcus pluvialis cells (green vegetative cells cultivated under optimal cell culture conditions and red cyst cells maintained under high light stress conditions to induce astaxanthin production) were used to investigate the protein expression profiles by two-dimensional electrophoresis, image analysis, and peptide mass fingerprinting. The cellular accumulation of astaxanthin was evident after exposure to high light intensity and reached the maximum cellular level after 78 h of high light stress. In a 2-D electrophoresis analysis, 22 proteins were upregulated over 2-fold in the red cyst cells when compared with the green vegetative cells and selected for further analysis by chemically assisted fragmentation (CAF)-MALDI-TOF sequencing to identify the protein functions. Among 22 different spots, several key enzymes specific to the carotenoid pathway, including isopentenyl pyrophosphate isomerase (IPP) and lycopene $\beta$-cyclase, appeared in H. pluvialis after exposure to high light intensity. Therefore, IPP and lycopene $\beta$-cyclase would appear to be involved with carotenoid accumulation in the cytoplasm, as these peptides were preferentially upregulated by high light intensity preceding an increase in carotenoid, and only these forms were detected in the red cyst cells.

  • Adipose tissue is an important endocrine organ involved in the control of whole body energy homeostasis and insulin sensitivity. Considering the increased incidence of obesity and obesity-related disorders, including diabetes, it is important to understand thoroughly the process of adipocyte differentiation and its control. Therefore, we performed a differential proteome mapping strategy using two-dimensional gel electrophoresis combined with peptide mass fingerprinting to identify intracellular proteins that are differentially expressed during adipose conversion of 3T3-L1 pre-adipocytes in response to an adipogenic cocktail. In the current study, we identified 46 differentially expressed proteins, 6 of which have not been addressed previously in 3T3-L1 cell differentiation. Notably, we found that phosphoribosyl pyrophosphate synthetase (PRPS), a regulator of cell proliferation, was preferentially expressed in pre-adipocytes than in fully differentiated adipocytes. In conclusion, our results provide valuable information for further understanding of the adipogenic process.

Proteomic Analysis of the Hydrophobic Fraction of Mesenchymal Stem Cells Derived from Human Umbilical Cord Blood

  • Mesenchymal stem cells (MSCs) are promising candidates for cell therapy and tissue engineering, but their application has been impeded by lack of knowledge of their core biological properties. In order to identify MSC-specific proteins, the hydrophobic protein fraction was individually prepared from two different umbilical cord blood (UCB)-derived MSC populations; these were then subjected to two-dimensional (2D) gel electrophoresis and peptide mass fingerprinting matrix-assisted laser desorption/ionization (MALDI)-time of flight (TOF)-mass spectrometry (MS). Although the 2D gel patterns differed somewhat between the two samples, computer-assisted image analysis identified shared protein spots. 35 spots were reliably identified corresponding to 32 different proteins, many of which were chaperones. Based on their primary sub-cellular locations the proteins could be grouped into 6 categories: extracellular, cell surface, endoplasmic reticular, mitochondrial, cytoplasmic and cytoskeletal proteins. This map of the water-insoluble proteome may provide valuable insights into the biology of the cell surface and other compartments of human MSCs.

  • The maize pathogen Gibberella moniliformis produces fumonisins, a group of mycotoxins associated with several disorders in animals and humans, including cancer. The current focus of our research is to understand the regulatory mechanisms involved in fumonisin biosynthesis. In this study, we employed a proteomics approach to identify novel genes involved in the fumonisin biosynthesis under nitrogen stress. The combination of genome sequence, mutant strains, EST database, microarrays, and proteomics offers an opportunity to advance our understanding of this process. We investigated the response of the G. moniliformis proteome in limited nitrogen (N0, fumonisin-inducing) and excess nitrogen (N+, fumonisin-repressing) conditions by one- and two-dimensional electrophoresis. We selected 11 differentially expressed proteins, six from limited nitrogen conditions and five from excess nitrogen conditions, and determined the sequences by peptide mass fingerprinting and MS/MS spectrophotometry. Subsequently, we identified the EST sequences corresponding to the proteins and studied their expression profiles in different culture conditions. Through the comparative analysis of gene and protein expression data, we identified three candidate genes for functional analysis and our results provided valuable clues regarding the regulatory mechanisms of fumonisin biosynthesis.

Two-Dimensional Reference Map of Schizosaccharomyces pombe Proteins (Update)

  • Based on the first 2D reference map of the fission yeast Schizosaccharomyces pombe protein reported previously, we expanded and updated the map using narrower pI ranges. In this paper, 240 protein spots were identified on our reference map. In the pI 4-7 range, 144 spots corresponding to 86 different proteins were identified. In the pI 6-9 range, 43 spots corresponding to 35 different proteins were identified. Fifty-three new spots corresponding to 39 different proteins were further identified in the pI 5-6 range.

  • In this review, we described the catalogues of the rice proteome which were constructed in our program, and functional characterization of some of these proteins was discussed. Mass-spectrometry is the most prevalent technique to rapidly identify a large number of proteome analysis. However, the conventional Western blotting/sequencing technique has been used in many laboratories. As a first step to efficiently construct protein cata-file in proteome analysis of major cereals, we have analyzed the N-terminal sequences of 100 rice embryo proteins and 70 wheat spike proteins separated by two-dimensional electrophoresis. Edman degradation revealed the N-terminal peptide sequences of only 31 rice proteins and 47 wheat proteins, suggesting that the rest of separated protein sports are N-terminally blocked. To efficiently determine the internal sequence of blocked proteins, we have developed a modified Cleveland peptide mapping method. Using this above method, the internal sequences of all blocked rice proteins(i, e., 69 proteins) were determined. Among these 100 rice proteins, thirty were proteins for which homologous sequence in the rice genome database could be identified. However, the rest of the proteins lacked homologous proteins. This appears to be consistent with the fact that about 45% of total rice cDNA have been deposited in the EMBL database. Also, the major proteins involved in the growth and development of rice can be identified using the proteome approach. Some of these proteins, including a calcium-binding protein that tuned out to be calreticulin, gibberellin-binding protein, which is ribulose-1.5-bisphosphate carboxylase/oxygense active in rice, and leginsulin-binding protein in soybean have functions in the signal transduction pathway. Proteomics is well suited not only to determine interaction between pairs of proteins, but also to identify multisubunit complexes. Currently, a protein-protein interaction database for plant proteins(http://genome.c.kanazawa-u.ac.jp/Y2H)could be a very useful tool for the plant research community. Also, the information thus obtained from the plant proteome would be helpful in predicting the function of the unknown proteins and would be useful be in the plant molecular breeding.

  • 인삼 모상근의 프로테옴 분석에 의해 얻은 질량분석 스펙트럼 데이터는 MALDI/TOF/MS에서 얻는 질량 스펙트럼과 ESI/Q-TOF/MS에서 얻는 탄뎀 질량 스펙트럼으로 구분된다. 질량 스펙트럼은 단백질이 효소에 의해 분해된 펩타이드들의 분자량 정보를 제공하며, 탄뎀 질량 스펙트럼에서는 아미노산 단위로 분해된 절편 단백질의 분자량으로부터 아미노산 서열을 결과로 얻는다. 펩타이드의 아미노산 서열을 BLAST로 검색하면 유사한 단백질을 GenBank에서 검색할 수 있다. 이러한 단백질 동정 방법은 완전한 유전체 서열이 알려진 생물체의 경우 높은 정확도로 단백질을 동정할 수 있으나, 그렇지 않은 경우는 유사한 단백질이 데이터베이스에 존재하지 않아 분석이 용이하지 않다. 본 연구에서는 질량 스펙트럼 및 절편 단백질의 아미노산 서열을 EST (expressed sequence tag) 서열과 비교하여 프로테옴 데이터와 일치하는 EST 서열을 찾아내고 이를 BLAST검색에 의해 단백질 동정에 활용하였다. ESI/Q-TOF/MS 에서 얻은 아미노산 서열은 길이는 짧지만 데이터의 신뢰도가 높으므로 EST 서열과의 연관 관계를 밝힘으로써 단백질에 대한 정보를 보완할 수 있었다. ESI/Q-TOF/MS에서 얻은 펩타이드의 아미노산 서열을 EST 서열과 비교한 결과 90%의 아미노산 서열이 EST DB에서 발견되었다. NCBI의 nr 데이터베이스에서 아미노산 서열을 검색하여 찾은 단백질이 68%임에 비하여, 인삼 EST 서열에 의한 검색이 22% 더 많은 결과를 얻었다. MALDI/TOF/MS의 질량 스펙트럼에서 nr 데이터베이스로 검색한 결과와 인삼 EST 데이터베이스를 검색한 결과가 일치하는 경우는 47개 중 9개인 19%에 불과하여, 탄뎀 질량 분석으로 아미노산 서열을 얻지 않고, 단지 질량 스펙트럼으로부터 단백질을 동정하는 방법으로는 단백질 동정의 정확한 결과를 기대하기 어려움을 확인하였다.

Differential Expression of Kidney Proteins in Streptozotocin-induced Diabetic Rats in Response to Hypoglycemic Fungal Polysaccharides

  • Diabetic nephropathy remains a major cause of morbidity and mortality in the diabetic population and is the leading cause of end-stage renal failure. Despite current therapeutics including intensified glycemic control and blood pressure lowering agents, renal disease continues to progress relentlessly in diabetic patients, albeit at a lower rate. Since synthetic drugs for diabetes are known to have side effects, fungal mushrooms as a natural product come into preventing the development of diabetes. Our previous report showed the hypoglycemic effect of extracellular fungal polysaccharides (EPS) in streptozotocin (STZ)-induced diabetic rats. In this study, we analyzed the differential expression patterns of rat kidney proteins from normal, STZ-induced diabetic, and EPS-treated diabetic rats, to discover diabetes-associated proteins in rat kidney. The results of proteomic analysis revealed that up to 500 protein spots were visualized, of which 291 spots were differentially expressed in the three experimental groups. Eventually, 51 spots were statistically significant and were identified by peptide mass fingerprinting. Among the differentially expressed renal proteins, 10 were increased and 16 were decreased significantly in diabetic rat kidney. The levels of different proteins, altered after diabetes induction, were returned to approximately those of the healthy rats by EPS treatment. A histopathological examination showed that EPS administration restored the impaired kidney to almost normal architecture. The study of protein expression in the normal and diabetic kidney tissues enabled us to find several diabetic nephropathy-specific proteins, such as phospholipids scramblase 3 and tropomyosin 3, which have not been mentioned yet in connection with diabetes.

Proteomic Response of Alfalfa Subjected to Aluminum (Al) Stress at Low pH Soil

  • In order to reveal the aluminum (Al) stress tolerance mechanisms in alfalfa plant at low pH soil, a proteomic approach has been conducted. Alfalfa plants were exposed to Al stress for 5 days. The plant growth and total chlorophyll content are greatly affected by Al stress. The malondialdehyde (MDA) and $H_2O_2$ contents were increased in a low amount but free proline and soluble sugar contents, and the DPPH-radical scavenging activity were highly increased. These results indicate that antioxidant activity (DPPH activity) and osmoprotectants (proline and sugar) may involve in ROS ($H_2O_2$) homeostasis under Al stress. In proteomic analysis, over 500 protein spots were detected by 2-dimentional gel electrophoresis analysis. Total 17 Al stress-induced proteins were identified, of which 8 protein spots were up-regulated and 9 were down-regulated. The differential expression patterns of protein spots were selected and analyzed by the peptide mass fingerprinting (PMF) using MALDI-TOF MS analysis. Three protein spots corresponding to Rubisco were significantly down-regulated whereas peroxiredoxin and glutamine synthetase were up-regulated in response to Al stress. The different regulation patterns of identified proteins were involved in energy metabolism and antioxidant / ROS detoxification during Al stress in alfalfa. Taken together, these results provide new insight to understand the molecular mechanisms of alfalfa plant in terms of Al stress tolerance.
