辛夷散에 의한 아나필락시 反應의 抑制에 관한 硏究 (Studies on the Inhibitory effect of anaphylactic reaction by Shinisan)

  • Shinisan has been used for treatment of allergic disease in Oriental Medicine. However, its effect in experimental models remains unknown. The mast cell plays a pivotal role in initiating allergic response by secreting intracytoplasmic granular mediators such as histamine. The present report describes an inhibitory effect of Shinisan on mast cell-mediated immediate type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal ($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Shinisan inhibited concentration-dependently mast cell-dependent ear swelling response induced by compound 48/80. Shinisan inhibited concentration-dependently passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) IgE in rats by topical application. Shinisan also inhibited concentration-dependently the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Shinisan had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Shinisan inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

Magnoliae flos has been used for treatment of allergic disease in Korea. However, its effect in experimental models remains unknown. The present report describes an inhibitory effect of Magnoliae flos on mast cell-mediated immediate-type allergic reactions.   • In this report, we investigated the effect of aqueous extract of vinegar treated small black soybean (Glycine max Merr.) (Leguminosae) (VSBS) on mast cell-mediated allergic reaction and pro-inflammatory cytokine secretion. VSBS inhibited compound 48/80-induced systemic reactions. VSBS attenuated immunoglobulin (Ig) E-mediated passive cutaneous anaphylaxis. In addition, VSBS decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated secretion of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6 and interleukin (IL)-8 in human mast cells. Our findings provide evidence that VSBS inhibits mast cell-derived allergic reactions.

Ginsenoside Rg3 suppresses mast cell-mediated allergic inflammation via mitogen-activated protein kinase signaling pathway

  • Background: Ginsenoside Rg3 (G-Rg3) is the major bioactive ingredient of Panax ginseng and has many pharmacological effects, including antiadipogenic, antiviral, and anticancer effects. However, the effect of G-Rg3 on mast cell-mediated allergic inflammation has not been investigated. Method: The antiallergic effects of G-Rg3 on allergic inflammation were evaluated using the human and rat mast cell lines HMC-1 and RBL-2H3. Antiallergic effects of G-Rg3 were detected by measuring cyclic adenosine monophosphate (cAMP), detecting calcium influx, and using real-time reverse transcription polymerase chain reaction, enzyme-linked immunosorbent assay, Western blotting, and in vivo experiments. Results: G-Rg3 decreased histamine release from activated mast cells by enhancing cAMP levels and calcium influx. Proinflammatory cytokine production was suppressed by G-Rg3 treatment via regulation of the mitogen-activated protein kinases/nuclear factor-kappa B and receptor-interacting protein kinase 2 (RIP2)/caspase-1 signaling pathway in mast cells. Moreover, G-Rg3 protected mice against the IgE-mediated passive cutaneous anaphylaxis reaction and compound 48/80-induced anaphylactic shock. Conclusion: G-Rg3 may serve as an alternative therapeutic agent for improving allergic inflammatory disorders.

RBL-2H3 비만세포와 동물병태모댈에서 소청룡탕(小靑龍湯)의 항알레르기 효과 (Anti-allergic Effects of Socheongyoug-tang on RBL-2H3 Mast Cell and Mice-mediated Allergy Model)

  • The discovery of drugs on the treatment of mast cell-mediated allergic disease is a very important subject in human heath. The Socheongyoug-tang(SCYT) has been used for centruries as a traditional medicine in Korea and is known to have an anti-inflammatory effect. However, its specific mechanism of action is still unknown. In this report, we investigated the effect of hot water extract from SCYT on RBL-2H3 mast cell-mediated allergic reaction and studied its possible mechanism of action. SCYT inhibited compound 48/80-induced systemic anaphylaxis and serum histamine release in mice. SCYT decreased the passive cutaneous anaphylaxis reaction activated by Anti-lgE antibody-HSA. SCYT dose-dependently reduced histamine release from mice peritoneal mast cells activated by Anti-lgE antibody-HSA. SCYT increased cAMP and decreased compound 48/80-induced intracellular $Ca^{2+}$ levels. Our findings provide evidence that SCYT inhibits mast-derived allergic reactions, and also demonstrate the involvement of cAMP and intracellular $Ca^{2+}$ in these effects.

사물탕(四物湯)이 알러지 염증반응에 미치는 영향 (Effect of Samul-Tang on the Allergic Inflammatory Response)

  • Samul-Tang (SMT) has been used for nourishing of the blood, hematopoiesis as a herbal medicine history. The purpose of this study is to find out anti-allergic inflammatory reaction of SMT. To clarify the mechanism, the effect of SMT on vascular permeability of rat cutaneous tissue and histamine and cytokines (IL-6, IL-8, TNF-${\alpha}$) release from mast cells were observed. The results are the pretreatment with SMT significantly decreased the compound 48/80-induced degranulation and histamine release from RPMC, SMT also inhibited the anti-DNP lgE-induced increment of vascular permeability of rat cutaneous tissue. SMT significantly reduced the PMA plus A23187-induced increment of expression of IL-6, IL-8, and TNF-${\alpha}$ in HMC-1 Cell. The Present study provide evidence that SMT inhibits mast cell-derived inflammatory allergic reactions by blocking histamine release and pro-inflammatory cytokine expression, and suggest the mechanisms of action. Furthermore, in vivo and in vitro anti-allergic effect of SMT suggests a possible therapeutic application of this agent in inflammatory allergic diseases.

Platycodin D를 포함하는 도라지 추출물이 IgE/Ag 유도 제 1형 과민반응에 미치는 영향 (Effects of Platycodon Grandiflorum Including Platycodin D in IgE/Ag-Induced Type I Hypersensitivity)

  • 최근 알러지성 비염, 알러지성 피부염, 알러지성 천식, 알러지성 과민반응 등과 같은 질병이 점차 늘어나고 있으며 그에 따른 효과적인 치료제나 효율적인 치료법이 시급한 실정이다. 본 연구에서 동물모델과 비만세포 탈과립작용에서 Platycodin D가 포함된 도라지 추출물(PG-Platycodin D)의 항알러지 효과를 연구하였다. $In$ $vivo$ 상에서 PG-Platycodin D가 항원-항체 반응에 의한 알러지 반응을 효과적으로 억제하는지 살펴보기 위한 아나필락틱 쇼크 평가에서 Platycodin D의 함량이 1%에서 5%까지 증가한 도라지 추출물일수록 $in$ $vivo$ 수준에서의 알러지반응이 억제되는 것을 확인할 수 있었다. 또한 항원-항체 반응에 의해 매개된 RBL-2H3 비만세포의 탈 과립현상에 대한 PG-Platycodin D의 효과를 알아보기 위한 ${\beta}$-hexosaminidase의 방출량 측정에서 Platycodin D의 함량이 1%에서 5%까지 포함된 도라지 추출물에서 농도가 증가함에 따라 ${\beta}$-hexosaminidase의 방출량이 농도 의존적으로 감소하는 것을 알 수 있었다. 또한, PG-Platycodin D의 처리가 항원-항체 반응에 의해 매개된 RBL-2H3 비만세포 내의 IL-3의 발현이 감소하는 양상을 확인할 수 있었다. 이들의 결과로부터 Platycodin D가 포함된 도라지 추출물이 IL-3의 유전자 발현을 억제함으로써 항원-항체 반응에 의한 탈 과립현상을 억제하여 알러지 작용을 제어하는 가능성을 확인하였다.

  • Mast cells are key effector cells in the early phase allergic inflammation and in diverse immunological and pathological processes. In order to understand the effect on reduction of the anti-dinitrophenyl (DNP) IgE antibody-induced β-hexosaminidase release in RBL-2H3 rat mast cells, a novel series of 4-senecioyloxymethyl- 6,7-dimethoxycoumarins (SMDC) was prepared by reacting 4-chloromethyl-6,7-dimethoxycoumarin with various carboxylic acids. Compounds 8-11 with cyclic moiety such as phenyl, thiophenyl, pyridinyl, and furanyl group were found to inhibit-hexosaminidase release more potently (5.98-9.62 μM) than compounds 3- 7 and 12 with acyclic moiety (19.32-76.78 μM). Furthermore, compounds 8 and 9 inhibited IgE-induced ear swelling and significantly reduced systemic passive cutaneous anaphylaxis reaction in mice.

Antiallergic effect of ginseng fermented with Ganoderma lucidum

  • Ginseng (the root of Panax ginseng CA Meyer, family Araliacease), which is used in Korea, China and Japan as a herbal medicine, was fermented with Ganoderma lucidum (GL) and their antiallergic effects were investigated. Of GLs used for fermentation, KCTC 6283 potently produced ginsenoside Rh2, followed by KFRI M101. KCTC 6532, and ginsenoside Rd, followed by KFRI M101. Oral administration of these fermented ginseng extracts inhibited allergic reactions, passive cutaneous anaphylaxis reaction induced by IgE and scratching behaviors induced by compound 48/80. Of them, the ginseng extract fermented by KCTC 6532 and KFRI M101 potently inhibited allergic reactions compared to that fermented by KCTC 6283. These findings suggest that the fermentation of ginseng with GL can increase its antiallergic activity and the increment of its antiallergic effect may be due to the biotransformation of ginseng saponins to ginsenosides Rd and Rh2.

Syk kinase 억제를 통한 관중의 항앨러지 효과 (Crassirhizomae rhizoma Exhibits Anti-Allergic Activity through Inhibition of Syk Kinase in Mast Cells)

  • This study aimed to investigate the anti-allergic activity and the mechanism of action of Crassirhizomae rhizoma (CR). The extract of CR exhibited potent inhibitory activity in mast cells; its $IC_{50}$ values were $31.2{\pm}1.5{\mu}g/m{\ell}$ for rat basophile leukemia (RBL)-2H3 mast cells and $51.5{\pm}2.1{\mu}g/m{\ell}$ for bone marrow-derived mast cells by antigen stimulation. It also suppressed the expression of TNF-${\alpha}$ and IL-4 mRNAs in RBL-2H3 cells. In an in-vivo animal allergy model, it inhibited a local allergic reaction, passive cutaneous anaphylaxis (PCA), in a dose-dependent manner. With regard to the mechanism of action, CR inhibited the activating phosphorylation of Syk kinase, a key signaling protein for the activation of mast cells. Taken together, these results strongly suggested that the anti-allergic activity of CR is mediated through the inhibition of histamine release and allergic cytokine production by the inhibition of Syk in mast cells.
