Docosahexaenoic acid가 전기충격성 뇌장애 마우스의 기억력 및 Acetylcholine량 변화에 미치는 영향 (The Effect of Docosahexaenoic Acid on Brain Function and Acetylcholine Level in Cerebral Cortex of Electroconvulsive Shock Induced Mice)

  • Electroconvulsive shock (ECS) increases the activity of acetylchohnesterase and decreases in brain acetylcholine levels. A large amount of free fatty acids accumulated in the brain tissue affects cerebral blood flow, brain edema and inflammation and results in brain injury. The present study examined the effect of docosahexaenoic acid (DHA) and D,L-pyroglutamic acid (D,L-PCA) on the learning and memory deficit using the passive avoidance failure technique and on the change of acetylcholine and choline level in the cerebral cortex of ECS-induced mice. The application of ECS (25mA, 0.5sec) induced a significant decrease in memory function for 30 min. ECS-induced a significant decrease in cortical acetylcholine and choline levels 1 min following the ECS application, which were almost recovered to ECS control level after 30 min. DHA (20 mg/kg, i.p.). administered 24 hr before shock. prevented the ECS-induced passive avoidance failure and the decrease of acetylcholine level 1 min following the ECS application. DHA failed to elicit a change in cortical choline level. DHA did not affect memory function and the cortical Ach and choline level of normal mice. The administration of D,L-PCA (500 mg/kg, i.p.) increased the effect of DHA on memory function and the change of cortical acetylcholine level of ECS induced mice. These results suggest that DHA treatment may be contributed to the prevention against memory deficit, and to the activation of cholinergic system in the ECS induced mice.

기억력 감퇴모델에서 산사의 기억력 개선 효과에 관한 연구 (The Fruits of Crataegus pinnatifida Bunge ameliorates Learning and Memory Impairments Induced by Scopolamine)

  • Objectives : In the present study, we assessed the effects of the ethanolic extract of Crataegus pinnatifida Bunge on the learning and memory impairments induced by scopolamine using the passive avoidance and the Morris water maze tasks in mice. Methods : The cognition-enhancing effect of C. pinnatifida was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The ethanolic extract of C. pinnatifida (100, and 200 mg/kg) significantly reversed the scopolamine-induced cognitive impairments in the passive avoidance test (p < 0.05). Moreover, C. pinnatifida (200 mg/kg) also improved escape latencies in training trials and increased swimming times and distances within the target zone of the Morris water maze (p < 0.05). On the Y-maze test, C. pinnatifida (100, and 200 mg/kg) also significantly reversed scopolamine- induced cognitive impairments in mice (p < 0.05). Conclusions : The ethanolic extract of Crataegus pinnatifida dramatically possesses the anti-amnestic and cognitive-enhancing activities related to the memory processes, and these activities were parallel to treatment duration and dependent on the learning models.

Effects of Chongmyung-tang on Learning and Memory Performances in Mice

  • Chongmyung-tang(CMT, 聰明湯), oriental herbal medicine which consists of Polygaglae Radix(遠志), Acori Graminei Rhizoma(石菖蒲) and Hoelen(白茯神) has effect on amnesia, dementia. In order to evaluate effect of CMT on memory and learning in mice, CMT extract was used for studies. This paper describes the effects of CMT extract on memory and learning processes by using the passive and active avoidance performance tests, novel object recognition task and water maze task. The CMT extract ameliorated the memory retrieval deficit induced by ethanol in the passive avoidance responses but did not affect ambulatory activity of normal mice. These results suggest that CMT has an ameliorating effect on memory retrieval impairment. CMT extract decreased spontaneous motor activity(SMA) in the latter sessions of memory registration in active avoidance responses. These results suggest that CMT has partly transquilizing or antianxiety effects. In novel object recognition task to measure visual recognition memory, CMT-administered mice enhanced in long term memory for 1-3 days. In water maze task to measure spatial learning, which requires the activation of NMDA receptors in the hippocampus, spatial learning in CMT-administered mice was faster than in wild-type mice. These results suggest that CMT enhances memory and activates NMDA receptors.

지골피(地骨皮)가 스트레스로 인한 기억저하와 우울행동에 미치는 영향 (Antistress effects of Lycii Radicis Cortex in the passive avoidance test and the forced swimming test)

  • Objectives : Investigation of the anitstress effects of Lycii Radicis Cortex. Methods : Behavior was measured in PAT and FST after applying stress to rats. Also, TH expression-degree in the regions of VTA and LC was measured, respectively. Results : The memory was improved in the LH 50mg/kg treated group in the passive avoidance test(P<0.01). The duration of immobility in the forced swimming test was significantly decreased in the LR 50mg/kg and 400mg/kg treated groups(P<0.0l). Stress-induced TH increases were meaningfully suppressed in the LR 100mg/kg and 400mg/kg treated group(P<0.001). Conclusion : Lycii Radicis Cortes has the effect of memory improving and antidepressant.

  • Anti-amnestic activities of Korean red ginseng (Ginseng Radix Rubra) and Crude drug-combined preparations (RGCDP-1, RGCDP-2, and RGCDP-3) were evaluated by the animal experiment. RGCDP-1 and RGCDP-2 were prepared based on Korean folk prescriptions, 'Chongmyongtang' and 'Guibitang', respectively, while RGCDP-3, by a combination of both. Among the three preparations, RGCDP-3 was found to show the most potent anti-amnestic activity as evaluated by the passive avoidance test with mice, indicating synergistic action by combined effect of RGCDP-1 and RGCDP-2.

  • The present study investigated the passive avoidance and spatial learning in the ${\mu}$-opioid receptor gene knockout mice and wild type mice. In the step-through passive avoidance task. the ${\mu}$-opioid receptor knockout mice did not differ from the wild type mice. In Morris water maze. however. the ${\mu}$-opioid receptor knockout mice showed significant memory deficit compared to wild type mice. (omitted)

Cherry Tomatoes Ameliorate Scopolamine-induced Amnesia in Mice

  • Cherry tomatoes are rich in antioxidants, which may protect against neurodegeneration and consequent memory loss. This study was conducted to investigate the effect of cherry tomatoes on scopolamine-induced amnesia in mice. Male ICR mice (4 weeks old) were maintained for 4 weeks on a diet containing 10 or 20% tomato powder (TP), and then administered scopolamine (1 mg/kg body weight, i.p.) 45 min before memory testing. Passive avoidance and Morris water maze testing revealed that scopolamine-induced amnesia was significantly reduced in the TP groups compared to the non TP-received (control) group. Accordingly, acetylcholinesterase activities in the serum and brain of TP groups were lower than those in the control group. These findings suggest that cherry tomatoes may be useful for the prevention of neurodegenerative diseases such as amnesia and Alzheimer's disease.

Cognitive Enhancing Activity of Betula platyphylla Sap in Scopolamine Induced Amnesic Mice

  • Cognitive enhancing activity of Betula plantyphylla sap was determined in scopolamine induced amnesic mice using passive avoidance test. Oral acute administration of the sap effectively reversed memory deficit in a dose dependent manner. Then the sap was standardized on the basis of sugar contents using HPLC combined with refractive index detector. Glucose and fructose were the main sugars present in the sap.

  • Objective : Investigation of the anti-stress effects of Sihosogansan Methods : Passive avoidance test(PAT) was performed after applying immobilization stress in water to rats. Also, forced swimming test(FST) was performed to another rats and after FST, the degree of Tyrosine Hydroxylase(TH) expression was measured with immunohistochemical method in the regions of locus coeruleus(LC) and ventral tegmental area (VTA). Results : In the PAT after immobilization stress in water, response latency was significantly increased in the Sihosogansan(400mg/kg) group in comparison with the control group. In the FST, immobility was significantly decreased in the Sihosogansan groups (100mg/kg, 400mg/kg), comparing with the control group. Stress-induced TH increases were suppressed in the Sihosogansan groups (100mg/kg, 400mg/kg) at the LC and the VTA region respectively. Conclusion : Sihosogansan can improve memory ability of rats, reduce behavior of depression in rats, decrease TH-immunoreactive cells at the LC and VTA region in rat, and it may be concluded that Sihosogansan has significant effect in reducing stress.

숙지황이 기억과 망각 속도에 미치는 영향에 대한 실험적 연구 (Effect of Rehmanniae Radix Preparata on Memory in Rats)

  • Background: Rehmanniae radix preparata is a medicinal herb widely used in Asian countries, to replenish Yin and Vital Essence through strengthen the kidney function. In Oriental Medicine the brain has physiological relationship with the Vital Essence. And central nerve system (CNS) is also refered to as 'Sea of the Vital Essence'. So if the Vital Essence is strong it will nourish the brain and memory will all be keen. The purpose of this study was to investigate the effect of 7 to 14 days treatment with Rehmanniae radix preparata (3.4 g /100 g, per os) on retention performance of rats in a passive avoidance situation. Methods: Male Sprague-Dawley rats were trained on a one-trial passive avoidance task using a two-way shuttle box by giving a foot shock. They were tested for retention of 6h, 24h, 72h, 168h, and 336h after training. Results: Rehmanniae radix preparata treated rats showed a increased performance in retention test as compared to saline at 6hr (P<0.05) and 24hr (P<0.05). Conclusion: These data suggest that Rehmanniae radix preparata treatment can improve memory in the rat.
