Enzyme-linked immunosorbent assay(ELISA) for determination of progesterone concentration in bovine serum (ELISA에 의한 소의 혈청 progesterone 농도 측정)

  • This study was carried out the determine the progesterone concentration for serum by enzyme-linked immunosorbent assay(ELISA) in bovine adult at estrous, pregnant, after patuation and male, female calves of 1 month old, respectively. The results obtained were summarized as follows ; 1. The assay has a sesitivity of $0.1ng/m{\ell}$. 2. Intra and inter-assay coefficient of variation were 4.5%, 6.1~9.4% when used for the determination of progesterone in samples of bovine serum. 3. The percentages of recovery for progesterone added were between 88.0 to 88.9%. 4. Progesterone concentration of adult bovine serum at estrus, pregnant and after 1 day of parturition were $0.37{\pm}0.16$, $7.1{\pm}1.0$, $0.13{\pm}0.02ng/m{\ell}$, respectively. 5. There was no differences in serum progesterone concentration of calves both male($0.16{\pm}0.03ng/m{\ell}$) and female($0.15{\pm}0.04ng/m{\ell}$) on 1 month old. From these results, progesterone determination by ELISA is very applicable to detect of early pregnancy diagnosis, factorial analysis of reproductive disorder, and also reproductive physiological functions such as veterinary therapeutic measures and monitoring of cyclicity.

Studies on the Farrowing Induction of Sow with Prostaglandin $F_2\alpha$ I. Effect of Administration Method on the Farrowing Induclion (Prostaglandin $F_2\alpha$의 투여에 의한 돼지의 분만유기에 관한 연구 I. 투여방법이 분만유기에 미치는 영향)

  • This experiment was carried out to clarify the practical use of farrowing induction with PGF2$\alpha$ in swine production. Total 320 heads of pregnant sow were used and the effects of administration time, dosage, administration method of PGF2$\alpha$ and parity of sow on the farrowing induction were investigated. The results obtained in this experiment were summarized as following: 1. Total 228 heads out of 240 head which were treated with PGF2$\alpha$ in order to farrow artificially were farrowed normally. the rate of farrowing induction was 95%. 2. Several farrowing symptoms such as restlessness, bloody eye, udder swelling, milk discharge and nest building for piglets were observed in a few minutes after administration of PGF2$\alpha$. 3. Artifical parturition of sow treated with PGF2 at the 113rd, 112nd, 111st, 110th and 109th day of pregnancy were induced 17.0$\pm$7.3, 24.2$\pm$7.0, 31.3$\pm$14.7, 43.0$\pm$12.2 and 62.2$\pm$15.7 hours after administration, and the duration of pregnancy was significantly (P<0.01) shortened by PGF2$\alpha$ anministration. 4. Minimum amount of PGF2$\alpha$ per head for effective farrowing inductin was considered to be more than 5mg, and the more the amount, the shorter time needed for farrowing induction was resulted. 5. Effective farrowing induction was obtained from the sow treated with PGF2$\alpha$ via vulvomucous rather than from the sow treated via intromuscular, and intervals from PGF2$\alpha$ administration to farrowing in 4th-6th farrowed sows were shorter than those of 1st-3rd farrowed sows but no significant difference was observed.

  • Bovine colostrum contains various bio-functional Proteins. Especially, transforming: growth factor-${\beta}$1 (TGF-${\beta}$1) has a function in concerns with immune response. The purpose of this study was to establish the purification Processing of transforming growth factor-${\beta}$1(TGF-${\beta}$1). The highest concentration of TGF-${\beta}$1 was measured within 48 h after parturition in bovine colostrum using ELISA kit. Purification of TGF-${\beta}$1 from whey protein was carried out by the gel filtration, AF-heparin chromatography and AF-heparin rechromatography. After final purification step, TGF-${\beta}$1 with a molecular weight of 25 kD was obtained, and confirmed by silver staining and western blotting. Finally, TGF-${\beta}$1 was identified native form of 25 kD and reducing form of 12.5 kD by reducing agent.

Reproductive fecundity of Iraqi Awassi ewes immunized against synthetic inhibin-α subunit or steroid-free bovine follicular fluid

  • Objective: The present study was conducted to investigate the impacts of active and passive immunization against synthetic inhibin and steroid-free bovine follicular fluid, respectively, on reproductive fecundity out of breeding season in Iraqi Awassi ewes. Methods: Follicular fluid was aspired from mature bovine follicles, treated with activated charcoal, and used for immunization of male rabbits for obtaining steroid free bovine follicular fluid (SFBFF) antiserum. Forty non-pregnant Awassi ewes were allocated into 4 groups (n = 10 each). At day 38 of experiment, ewes were treated with intra-vaginal MPA sponge (60 mg for 12 days). At days 0, 28, and 50, ewes were treated with 4, 2, and 2 mL of normal saline (control; C-ve), 400, 200, and $200{\mu}g$ of ovalbumin (C+ve), 400, 200 and $200{\mu}g$ of inhibin (SI group), respectively, and 4 mL of normal saline at day 0, and 4 and 2 mL of SFBFF antiserum at days 28 and 50, respectively, (AI group). After mating with Awassi rams, pregnancy and embryo number were diagnosed, at day 38 of pregnancy, using ultrasonography. Blood samples were collected at days 30, 60, 90, and 120 of pregnancy, for assessment of estradiol-$17{\beta}$ (E2) and progesterone (P4) levels. After parturition, numbers of delivered lambs were recorded. Results: The results revealed significant increase of P4 and significant decrease of E2 levels in SI and AI pregnant ewes than controls at days 30, 60, 90, and 120. Newborn number increased significantly in SI and AI treated than control ewes. Conclusion: Active or passive immunization against endogenous inhibin could augment reproductive fecundity out of breeding season in Iraqi Awassi ewes.

A Field Trial as Dry-cow Therapy Using Nophazin (norfloxacin mixed cephalexin) in Mastitis Control (건유기 유방염 치료제 Nophazin리 야외응용시험)

  • A field test using Nophazin (drying off period therapeutics) was done from May, 1989 to October, 1989. 299 quarters of 75 cows from 28 herds were tested. The results were summarized as follows. 1. At the day 7 after parturition, CMT-was 81.60%, clots were removed perfectly, removal rate of hard, hot, pain was 99.6%, only one hard 0.35% was maintained. Recovery rate of general symptoms was 100%. 2. Recovery rate in Nophazin treatment was 100% in Micro-coccus SPP, Corynebacterium SPP, and unisolated, 97.06% in Enterobacterid SPP, 95.58% in Staphylococcus SPP, 91.67% in other Bacilli, 89.19% in Streptococcus SPP, Overall recovery rate was 95.32%, And fungi did not react. 3. The ratio of isolated microorganisms was Staphylococcus SPP 49.8%, Streptococcus SPP 16.3%, Enterobacteria SPP 15.0%, Micrococcus SPP 7.9% Corynebacterium SPP 4.4%, Pseudomonas SPP 2.6%, Bacilli 2.6% and Fungi 1.3%. 4. The range of sensitivity of each microorganism to therapeutics is as follow : Staphylococcus SPP 61.1∼94.7%, Streptococcus SPP 51.4∼91.9%, Micrococcus SPP 44.4∼94.4%, Corynebacterium SPP 70∼80%, Enterobacteria SPP 11.8∼88.2%, and other Gram negative bacilli 8.3∼83.3%. Overall sensitivity was in the range of 50.4∼92.0%, Microorganisms showed the highest sensitivity (80∼94.7%) to Nophazin. 5. New intramammary infection rate was 3.96% (9 strains) at paturition, 8.37% (19 strains) at the day 7 after paturition. Persistent organism wits 6.17% .(14 strains). 6. There was no side effect of Nophazin treatment.

Biochemical Characterization of 20α-Hydroxysteroid Dehydrogenase

  • In this review, we have tried to summarize the evidence and molecular characterization indicating that $20{\alpha}$-hydroxysteroid dehydrogenase ($20{\alpha}$-HSD) is a group of the aldo-keto reductase (AKR) family, and it plays roles in the modulation and regulation of steroid hormones. This enzyme plays a critical role in the regulation of luteal function in female mammals. We have studied the molecular expression and regulation of $20{\alpha}$-HSD in cows, pigs, deer, and monkeys. The specific antibody against bovine $20{\alpha}$-HSD was generated in a rabbit immunized with the purified recombinant protein. The mRNA expression levels increased gradually throughout the estrous cycle, the highest being in the corpus luteum (CL) 1 stage. The mRNA was also specifically detected in the placental and ovarian tissues during pregnancy. The $20{\alpha}$-HSD protein was intensively localized in the large luteal cells and placental cytotrophoblast villus, glandular epithelial cells of the endometrium, syncytiotrophoblast of the placenta, the isthmus cells of the oviduct, and the basal part of the primary chorionic villi and chorionic stem villus of the placenta and large luteal cells of the CL in many mammalian species. Further studies are needed to determine the functional significance of the $20{\alpha}$-HSD molecule during ovulation, pregnancy, and parturition. This article will review how fundamental information of these enzymes can be exploited for a better understanding of the reproductive organs during ovulation and pregnancy.

  • Dairy cow undergoes major adaptations in glucose and lipid metabolism to meet fetal and mammary nutrient requirements during the periparturient period. These adaptations are characterized by major changes in response to the homeostatic signal of insulin. In response to insulin, fatty acids are mobilized to meet energy demand. And cow develop insulin resistance during the late prepartum period to facilitate glucose sparing for the fetus and mammary gland. Insulin resistance is an important adaptation and may be partly responsible for the large increase in plasma nonesterified fatty acid (NEFA) that occurs prior to parturition and coincides with the characteristic decrease in dry matter intake (DMI). Modulation of insulin resistance by administration of thiazolidinedione (TZD) during the periparturient period, likely through the activation of $PPAR{\gamma}$, has substantial potential to minimize energy and immune-related metabolic disorders, and thus may result in increased productivity and improved health of dairy cows.

The effect of dietary ions difference on drinking and eating patterns in dairy goats under high ambient temperature

  • Objective: The present study was carried out to evaluate the effect of high dietary cation and anion difference (DCAD) rations on diurnal variations in eating and meal patterns, water intake and urination patterns in dairy goats fed under high ambient temperature (HTa). Methods: Ten crossbred dairy goats during peri-parturition period were selected and divided into two groups of five animals each. Experimental diets were control DCAD (control, 22.8 mEq/100 g dry matter [DM]) and high DCAD (DCAD, 39.1 mEq/100 g DM). The composition of two diets consisted of 44% corn silage and 56% concentrate. From the 2nd week to 8th week postpartum, goats were fed ad libitum twice daily either with the control or DCAD total mix ration with free access to water. The spontaneous eating and drinking patterns were determined. Results: The environmental conditions in the present experiment indicated that goats were fed under HTa conditions (average peak THI = 85.2) and were in heat stress. In addition to the typical HTa induced tachypnoea in both groups, the respiratory rate in the DCAD group was significantly higher than the control group (p<0.05). Although the goats from both groups showed comparable level of eating, drinking and urination during experiment, the meal pattern and water intake were different. High DCAD apparently increased eating and meal patterns compared with the control. At week 8 postpartum, goats from high DCAD group had significant (p<0.05) bigger meal size and longer meal duration. Moreover, high DCAD appeared to increase night-time water intake (p<0.05). Conclusion: Both meal pattern and night-time drinking effects of DCAD suggested that feeding with high DCAD ration may alleviate the effect of heat stress in dairy goat fed under HTa conditions.

Effects of a Reinforcement Program for Postpartum Care Behavioral Skills of Couples with Their First Baby (산욕기 첫 출산 부부를 위한 산후관리 행동기술 강화 프로그램 효과)

  • Purpose: This study aimed to investigate the effects of a reinforcement program for behavioral skills in postpartum care for couples with their first baby. Methods: The study used a non-equivalent control group and pretest-posttest design. It was conducted from January 14 to April 10, 2016 at a postpartum care center in D city. It analyzed 43 couples (22 in the experimental group and 21 in the control group.) For data analysis, descriptive statistics, test of homogeneity in pretest, independent t-tests, and repeated measures ANOVA were used. Results: For maternal fulfillment of postpartum care and postpartum fatigue, there was no significant difference in the interaction between group and time. In terms of parent-newborns attachment, the interaction between group and time showed a significant difference for mothers (F=13.63, p=.001) and fathers (F=6.51, p=.001). In marital intimacy, the interaction between group and time showed a significant difference for mothers (F=14.40, p<.001) and fathers (F=9.46, p=.004). In parenting stress, the interaction between group and time showed a significant difference for mothers (F=31.8, p<.001) and fathers (F=11.69, p=.001). A significant difference was found for the mothers' postpartum sleeping hours (F=0.14 p=.004). Conclusion: This program for behavioral skills in postpartum care, which is based on the information-motivation-behavioral skills model, improves postpartum care, parent-newborn attachment, marital intimacy, parenting stress, and maternal postpartum sleeping, by reinforcing behavioral skills required for postpartum care.

Improving behavior characteristics and stress indices of gestating sows housed with group housing facility

  • This study was conducted to investigate the effects of group-housing facility (GHF), compared to an individual confinement stall (CON), on the reproductive performance, behavior, and stress hormones of gestating sows. A total of 50 primiparous sows (Landrace × Yorkshire) were randomly allocated into either CON (n = 25) or GHF (n = 25) during the gestation period. One week before parturition, the sows were transferred into conventional farrowing crates, and cross-fostering was conducted within 1 d of delivery. Blood was collected for analyses of stress indices at 75 d of gestation and postpartum. Reproductive performance was estimated during the period of birth to weaning. Behavior patterns were identified at 90 d of gestation. Litter size was not different between the CON and GHF treatments. Weaning to estrus interval, however, tended to be lower in the GHF than in the CON (p < 0.1). Activity, treating, belly nosing, and exploring behaviors were observed only in the GHF group, whereas rubbing was shown only with the CON. Serum cortisol concentration was lower in the GHF than in the CON at 75 d of gestation (p < 0.05). Sows housed in the GHF showed lower epinephrine and norepinephrine concentrations than those housed in the CON at postpartum (p < 0.05). The GHF sows demonstrated more natural behavior characteristics associated with stress relief than the CON sows with no adverse effects on reproductive performance. Therefore, these results suggest that GHF could be applied as an alternative housing facility to improve animal welfare on swine farms.
