  • From crude drug preparation(Soshiho-Tang) ginseng sapogenins were identified by TLC and $ginsenoside-Rb_1$ was determined quantitatively by HPLC. Panaxadiol, pandaxatriol, acid-hydrolysates of ginseng saponin, were identified by TLC with benzene/acetone(4 : 1, v/v). Rf values of which were measured as 0.26 and 0.14, respectively. The content of $ginsenoside-Rb_1$ was determined by HPLC on $Lichrosorb-NH_2$ column with $CH_3CN/H_2O/n-BuOH$(80 : 20 : 10, v/v). Its recovery rate in the extract granules, was as relatively low as $19.8{\pm}1.4%$ compared to the content in raw red ginseng.

  • Background: Although ginsenosides and saponins in Korea red ginseng (KRG) shows various pharmacological roles, their roles in the inflammatory response are little known. This study investigated the anti-inflammatory role of ginsenosides identified from KRG saponin fraction (RGSF) and the potential mechanism in macrophages. Methods: The ginsenoside composition of RGSF was identified by high-performance liquid chromatography (HPLC) analysis. An anti-inflammatory effect of RGSF and its mechanisms were studied using nitric oxide (NO) and prostaglandin E2 (PGE2) production assays, mRNA expression analyses of inflammatory genes and cytokines, luciferase reporter gene assays of transcription factors, and Western blot analyses of inflammatory signaling pathways using the lipopolysaccharide (LPS)-treated RAW264.7 cells. Results: HPLC analysis identified the types and amounts of various panaxadiol ginsenosides in RGSF. RGSF reduced the generation of inflammatory molecules and mRNA levels of inflammatory enzymes and cytokines in LPS-treated RAW264.7 cells. Additionally, RGSF inhibited the signaling pathways of NF-κB and AP-1 by suppressing both transcriptional factors and signaling molecules in LPS-treated RAW264.7 cells. Conclusion: RGSF contains ginsenosides that have anti-inflammatory action via restraining the NF-κB and AP-1 signaling pathways in macrophages during inflammatory responses.

Effect of Different Extraction Procedures on Chemical Composition of Ginseng Extract (추출조건이 인삼엑기스의 화학성분 조성에 미치는 영향)

  • Fresh ginseng roots were extracted by different extraction methods to estimate the amount of extracts, and the content of common constituents and ginsenosides for comparison. The results are as follows: 1 The amount of the extract obtained by water as a solvent was about five times higher than those by ethanol or pressing process. 2. Water extraction at 70-$80^{\circ}C$ gave highest value in saponin yield, which was reduced by half by boiling, 3. The saponin yield by pressing process was shown to be about 52% of total saponin; saponins belonging to protopanaxatriol-ginsenosides being extracted better than those belonging to proto-panaxadiol-ginsenosides . 4. The contents of total sugar, reducing sugar, crude protein and total amino acids in the water extract were revealed to be higher compared to those in an ethanol extract. 5. The extract obtained by press had the highest ash content.

Ginsenoside-Re ameliorates ischemia and reperfusion injury in the heart: a hemodynamics approach

  • Ginsenosides are divided into two groups based on the types of the panaxadiol group (e.g., ginsenoside-Rb1 and -Rc) and the panaxatriol group (e.g., ginsenoside-Rg1 and -Re). Among them, ginsenoside-Re (G-Re) is one of the compounds with the highest content in Panax ginseng and is responsible for pharmacological effects. However, it is not yet well reported if G-Re increases the hemodynamics functions on ischemia (30 min)/reperfusion (120 min) (I/R) induction. Therefore, in the present study, we investigated whether treatment of G-Re facilitated the recovery of hemodynamic parameters (heart rate, perfusion pressure, aortic flow, coronary flow, and cardiac output) and left ventricular developed pressure (${\pm}dp/dt_{max}$). This research is designed to study the effects of G-Re by studying electrocardiographic changes such as QRS interval, QT interval and R-R interval, and inflammatory marker such as tissue necrosis factor-${\alpha}$ (TNF-${\alpha}$) in heart tissue in I/R-induced heart. From the results, I/R induction gave a significant increase in QRS interval, QT interval and R-R interval, but showed decrease in all hemodynamic parameters. I/R induction resulted in increased TNF-${\alpha}$ level. Treatment of G-Re at 30 and $100{\mu}M$ doses before I/R induction significantly prevented the decrease in hemodynamic parameters, ameliorated the electrocardiographic abnormality, and inhibited TNF-${\alpha}$ level. In this study, G-Re at $100{\mu}M$ dose exerted more beneficial effects on cardiac function and preservation of myocardium in I/R injury than $30{\mu}M$. Collectively, these results indicate that G-Re has distinct cardioprotectective effects in I/R induced rat heart.

  • For the information on micellization at each ginsenoside level aqueous solution of purified saponin of Panax ginseng root was dialyzed through dialysis tubing (MW 12,000) or eluted through Bio-Gel P-2 (MW 200-2,000) and analysed for ginsenosides by high performance liquid chromatography. Ginsenosides can be classified into three groups depending upon molecular aggregation pattern and spatial arrangement of hydrophilic parts in molecule. Group I that is large micelle former(aggregation number: above 10) and one side hydrophilic part (HP) includes $ginsenoside\;Rb_1$, $Rb_2$, Rc and Rd (diols). Group II thai is small micelle former (aggregation number:>10-1) and semi-two sales HP includes $Rg_2$, Rf (triol) and $Rg_3$ (diol). Group III that is no micelle former (aggregation number: 1) and two sides HP includes Re and $Rg_1$ (triol).

An Isolation of Crude Saponin from Red-Ginseng Efflux by Diaion HP-20 Resin Adsorption Method (홍삼유출액으로부터 Diaion HP-20 수지 흡착법에 의한 조사포닌의 분리)

  • This study was carried out to isolate saponin compounds from red-ginseng efflux, which was produced during the industrial processing of red-ginseng from fresh ginseng. We isolated crude saponin from the efflux extract (moisture content 35.0%) by using Diaion HP-20 adsorption method. Non-saponin fraction, which was adsorbed on Diaion HP-20 resin, was removed by eluating with $H_{2}O$ and 25% spirit. Then crude saponin was eluated with 95% spirit, continuously. Saponin in the eluated fractions was confirmed by TLC analysis. Crude saponin isolated from red ginseng efflux extract contained 12.10% of saponin. whereas those of white ginseng and red-ginseng were 3.30 and 3.39%, respectively. Ginsenoside contents showed the highest contents kin crude saponin from red ginseng efflux extract. Expacilly, the ginsenoside-$Rb_{1}$ and Re showed the highest contents in red-ginseng efflux extract when compared with those of white ginseng and red ginseng crude saponins. And the other ginsenosides except ginsenoside-$Rb_{1}$ and -Re also showed the highest contents in red ginseng efflux extract. However, the ratio of PD saponin (Panaxadiol saponin: $Rb_{1}+Rb_{2}$+Rc+Rd) to PT saponin (panaxatriol: $Re+Rg_{1}$) showed almost the same level when compared with those of ginseng saponin fractions. Ratio of PD/PT from red ginseng efflux extract was 1.99. Ratios of PD/PT from white ginseng and red ginseng were 1.85 and 1.84, respectively. Saponin purity, which was calculated by ratio percent of total ginsenoside to curde saponin content, was 45.90%. In case of white ginseng and red ginseng, the purities were 35.50 and 36.00%, respectively. However, by PHLC analysis, we confirmed that crude saponin isolated from red ginsengs. It suggested that crude saponin isolated from red ginseng ellux also would be useful component as ginseng saponins.

Effects of Panax ginseng on Galactosamine-induced Cytotoxicity in Primary Cultured Rat Hepatocytes (인삼 분획물이 Galactosamine에 의하여 손상된 일차배양한 흰쥐의 간세포에 미치는 영향)

  • The anti-hepatotoxic activity of Panax ginseng was studied using galactosamine (GalN)-induced cytotoxicity in primary cultured rat hepatocytes. Panax ginseng was fractionated into dammarane glycosides and protein fractions. The dammarane glycosides was further fractionated into panaxadiol and panaxatriol glycosides fractions. The protein fraction was further fractionated into four groups according to the molecular weight; larger than 10,000 dalton, between 5,000 and 10,000 dalton, between 1,000 and 5,000 dalton and between 500 and 1,000 dalton. A significant lowering action on the elevated glutamicpyruvic transaminase (GPT) activity in the culture medium of hepatocytes treated with 1.5 mM GalN was noticed with all four protein fractions studied at the concentration of both $50\;{\mu}g/ml$ and $100\;{\mu}g/ml$. However, the effect of dammarane glycosides fractions was not significant. It was noted that the addition of $100\;{\mu}g/ml$ of protein fractions smaller than 5,000 dalton significantly enhanced the syntheses of protein and RNA in the damaged hepatocytes induced by the treatment of 1.5 mM GalN. Dammarane glycosides fractions significantly enhanced protein synthesis at the concentration of $100\;{\mu}g/ml$ in the damaged hepatocytes by treatment of 1.5 mM GalN.

Influence of Panaxatriol-type Saponin on Secretion of Catecholamines from Isolated Perfused Rabbit Adrenal Gland

  • In the previous observations, it was reported that both total ginseng saponin and panaxadiol revealed the marked secretory effect of catecholamines (CA) from the rabbit adrenal gland and that CA secretion induced by them is due to dual mechanisms, cholinergic action and the direct action. In the present study, an attempt to investigate the effect of panaxatriol-type saponin (PT), which is known as an active component of Korean ginseng, on the secretion of CA from the rabbit adrenal gland was made. PT(200 $\mu$g) administered into adrenal vein evoked significantly secretion of CA from the isolated perfused rabbit adrenal gland. Secretory effect of CA produced by PT was attenuated clearly by treatment with chlorisondamine or adenosine, but was markedly increased by physostigmine. Perfusion of Krebs solution containing PT (200 $\mu$g) for 30 min potentiated greatly secretion of CA induced by acetylcholine. PT-induced CA secretion was weakened considerably by ouabain treatement or perfusion of calcium-free Krebs solution. These experimental data demonstrate that PT releases CA from the isolated perfused rabbit adrenal gland by a calcium-dependentd exocytotic mechanism. It seems that the secretory effect of PT is caused through the release of acetylcholine form cholinergic terminals present in the adrenal gland and a direct action on the chromaffin cell itself.

EFFECTS OF PANAX GINSENG SAPONINS ON CHEMICAL MEDIATOR RELEASE FROM AIRWAY SMOOTH MUSCLE IN ACTIVELY SENSITIZED GUINEA PIG

  • It has been reported that ginseng is effective in the central nervous system, immune system, and the strong inflammatory responses. However, there has been no research report yet about the effect of ginseng on allergic hypersensitivity reactivity. To confirm the ginseng effects on the release of mediators(histamine. leukotrienes etc.) which cause the hypersensitivity reactivity and inflammatory response, we used actively sensitized guinea pig airway tissues by utilizing the superfusion technique. In this procedure. the contractile response and mediators released after antigen stimulation of sensitized tissues, and IgG and IgE antibody products were measured in sera of immunized animals. Then the results of the controll group were compared to those of ginseng pretreatment groups. In the total saponin(TS) and panaxatriol(PT) pretreatment, histamine release decreased by $20\%$ in the tracheal tissues after active sensitization by ovalbumin(OVA, 10mg/kg), but in the lung parenchyma, histamine release decreased by $40\%.$ Panaxadiol(PD) significantly decreased histamine release by $40\%$ in the both tissues after active sensitization. TS, PT and PD of ginseng poorly blocked leukotrienes (LTs) and prostagrandin $D_2(PGD_2)$ release(less than $10\%$). Ginseng TS and PT had no effect on the serum IgG antibody production by ovalbumin, whereas PD significantly increased serum IgG antibody contents(approximately by 2 times). However, $IgG_1$ antibody products in the serum of guinea pig actively sensitized with ovalbumin after PD pretreatment were decreased, compared to that with ovalbumin alone. IgE antibody production by passive cutaneous anaphylaxis(PCA) titer in the TS pretreatment increased 3 times more than in the absence of TS(PCA titer by PT was not detected). These studies show that some ginseng saponins can in part act to inhibit mediator release in antigen - induced airway smooth muscle by inducing the IgG antibody production which has been changed in the specificity.

Protein Concentrate from Ginseng Leaf and its Nutritive Value (인삼엽 단백질의 추출 및 영양가)

  • This study was investigated to evaluate the feasibility of protein concentrate for human food from ginseng leaf. The protein concentrate was prepared from ginseng green leaf by treating with cold acetone , followed by protein extraction with 0.2% NaOH containing 0.5% 2-mercaptoethanol and 0.5% sodium dodecyl sulfate. Proximate composition of the ginseng leaf protein concentrate (LPC) showed that fat and ash was less than 1%, protein was about 75%, total sugar and total saponin was 5% and 1.2%, respectively. As compared to the provisional amino acid pattern reported by FAO/WHO, ginseng LPC was found to be poor in S-containing amino acids, which were the first limiting amino acid. The amino acid score and E/T ratio of ginseng LPC were 43.1 and 3.02, respectively. Digestibility of ginseng LPC by pepsin and trypsin was lower than that of milk casein.
