In this work, a fibrous bed bioreactor with high specific surface area and good adsorption efficacy for S. cerevisiae cells was used as the immobilization matrix in the production of ethanol. In batch fermentation, an optimal ethanol concentration of 91.36 g/l and productivity of 4.57 g $l^{-1}\;h^{-1}$ were obtained at an initial sugar concentration of 200 g/l. The ethanol productivity achieved by the immobilized cells was 41.93% higher than that obtained from free cells. Ethanol production in a 22-cycle repeated batch fermentation demonstrated the enhanced stability of the immobilized yeast cells. Under continuous fermentation in packed-bed reactors, a maximum ethanol concentration of 108.14 g/l and a productivity of 14.71 g $l^{-1}\;h^{-1}$ were attained at $35^{\circ}C$, and a dilution rate of 0.136 $h^{-1}$ with 250 g/l glucose.
Recently, we reported an improved technology for the degradation of organophosphate nerve agents using whole cells of genetically engineered Escherichia coli that anchored and displayed the enzyme organophosphorus hydrolase on the cell surface. In this paper we report the immobilization of these cells on highly porous sintered glass beads and the subsequent application of the immobilized cell in a continuous-flow packed bed bioreactor for the biodetoxification of a widely used insecticide, coumaphos.

Chitosan-alginate capsules were formed by electrostatic interactions and exhibited an appropriate mechanical strength, permeability, and stability for the culture of hepatocytes. Pig hepatocytes were isolated and hepatocyte spheroids formed and immobilized in chitosan-alginate capsules. An encapsulation procedure of 3 min and spheroid formation period of 24 h were the optimum conditions for the best liver functions. Pig hepatocytes with a cell density of $6.0{\tomes}10^6$ cells/ml in the capsules were found to be most suitable for application in a bioartificial liver support system. The encapsulated pig hepatocyte spheroids exhibited stable ammonia removal and urea secretion rates in a bioreactor for 2 weeks. Accordingly, chitosan-alginate encapsulated hepatocyte spheroids in a packed-bed bioreactor would appear to have potential as a bioartificial liver.
A thermostable trehalose synthase (TtTSase) from Thermus thermophilus HJ6 was immobilized on chitosan activated with glutaraldehyde. The yield of immobilization was evaluated as 39.68%. The optimum pH of the immobilized enzyme was similar to that of the free enzyme. However, the optimal temperature ranges were shifted by about $4^{\circ}C$ owing to better thermal stability after immobilization. The half-life of heat inactivation for free and immobilized enzymes was 5.7 and 6.3 days at $70^{\circ}C$, respectively, thus showing a lager thermostability of the immobilized enzyme. When tested in batch reaction, the immobilized enzyme retained its relative activity of 53% after 30 reuses of reaction within 12 days, and still retained 82% of its initial activity even after 150 days at $4^{\circ}C$. A packed-bed bioreactor with immobilized enzyme showed a maximum yield of 56% trehalose from 100 mM maltose in a continuous recycling system (bed volume: 10 ml) under conditions of pH 7.0 and $70^{\circ}C$.
It has known that the intermediates formed in acid reactions occuring during anaerobic reactions for instance, acetic acid, propionic acid and butyric acid have significant effects or the formations of biogases(e. g. mainly CH$_{4}$ and CO$_{2}$). A study on the effects of these intermediates for the formations of biogases, however, is still on initial stage due to the type and structural problems of reactor. The primary objective of this reserch program is to provide a funadmantal mechanism of involved reactions using a modified downflow multistage pecked bed bioreactor. As a first stage of this reaserch program, the following theoretical principles was applied 1. Principle of electroneutrality where the molar concentrations of cation and anion in solution are the same. 2. Relationship between the concentration of bicarbonate anion and pH as follows [HCO$_{3}^{-}$]= $K_{H}P_{T}Y_{CO_{2}}10^{pH-K_{1}}$ Based upon the above two principles, a series of experimental works was conducted to elucidated the relationship between the concentration of CO$_{2}$ and the pH related to the concentrations of cations and anions.
Aiming at the treatment of large volumes of gas with a low concentration of poorly water soluble VOC(Volatile Organic Compound), a new system is proposed: the combination absorption tower/bioreactor. In the scrubber part of the bioscrubbing system, the contaminating compounds are absorbed in a aqueous phase. The contaminated scrubbing liquid is transported to the bioreactor, where the compounds are biodegraded by aerobic microorganisms (mainly to carbon dioxide, water, and biomass). In this study, separation of a volatile organic compound(VOC) out of a waste gas stream has been carried out using a re-cyclable high boiling point extrant(HBE). The liquid stream containing a high boiling point entrant(HBE) scrubs the gas stream in a direct gas-liquid countercurrent contacting operation in a packed tower for the removal of said component from the gaseous stream. A packed-bed column using Pall Ring was set up in order to simulate practical conditions for the scrubbing tower. The liquid stream transported to the bioreactor is recovered and recycled to the scrubber. The model gas, which contained 400 mg/$\textrm{m}^3$ of toluene, at a rate of 100 L/min, flowed into the packed column where the scrubbing liquid trickled over the packing in countercurrent to the rising gas at 10~15L/min. The bioscrubber designed for large volume air streams containing VOCs showed removal efficiency up to 80% in an optimum operating conditions during the tests fer removing toluene from an air stream by scrubbing the air stream with HBE.
Two mesophilic trickling bed bioreactors filled with two different types of media, hydrophilic- and hydrophobic-cubes, were designed and conducted for hydrogen production under the anaerobic fermentation of sucrose. Each bioreactor consisted of the column packed with polymeric cubes and inoculated with heat-treated sludge obtained from anaerobic digestion tank. A defined medium containing sucrose was fed by the different hydraulic retention time(HRT), and recycle rate. Hydrogen concentrations in gas-phase were constant, averaging 40% of biogas throughout the operation. Hydrogen production rate was increased till $10.5\;L{\cdot}h^{-1}{\cdot}L^{-1}$ of bioreactor when influent sucrose concentrations and recycle rates were varied. At the same time, the hydrogen production rate with hydrophobic media application was higher than its hydrophilic media application. No methane was detected when the reactor was under a normal operation. The major fermentation by-products in the liquid effluent of the both trickling biofilters were acetate, butyrate and lactate. In order to run in the long term operation of both reactor filled with hydrophilic and hydrophobic media, biofilm accumulation on hydrophilic media and biogas produced should be controlled through some process such as periodical backwashing or gas-purging. Four sample were collected from each reactor on the opposite hydrogen production rate, and their bacterial communities were compared by terminal restriction fragment length polymorphism (T-RFLP) analysis of PCR products generated using bacterial 16s rRNA gene primers (8f and 926r). It was expressed a marked difference in bacterial communities of both reactors. The trickling bed bioreactor with hydrophobic media demonstrates the feasibility of the process to produce hydrogen gas. A likely application of this reactor technology can be hydrogen gas recovery from pre-treatment of high carbohydrate-containing wastewaters.
To treat fulminant hepatic failure (FHF) patients, various extracorporeal bioartificial liver (BAL) systems have been developed. Several requirements should be met for the development of BAL systems: hepatocytes should be cultured in a sufficiently high density; their metabolic functions should be of a sufficiently high level and duration; and the BAL systems module should permit scaling-up and aseptic handling. Several investigators have found that freshly isolated primary hepatocytes can be cultured into three dimensional, tightly packed, freely suspended, multicellular aggregates, or spheroids. These specialized cell structures exhibited enhanced liver specific functions and a prolonged differentiated state compared to cells maintained in a monolayer culture. Cells in spheroids appear to mimic the morphology and ultrastructure of the in vivo liver lobule. The ability of hepatocytes to organize into three-dimensional structures was hypothesized to contribute to their enhanced liver-specific activities. In this study, the ammonia removal rate and urea secretion rate of pig hepatocytes spheroids encapsulated in Ca-alginate bead were determined. A packed-bed bioreactor with encapsulated pig hepatocytes was devised as BAL support system. The efficacy of the system was evaluated in vitro.

A liquid culture of yeast "Candida tropicalis" was used in a fluidized bioreactor to achieve high removal efficiencies of volatile organic compounds (VOCs). In this study, granular activated carbon (GAC) was used as a fluidized material to improve adsorptive capacity as well as mass transfer of gaseous toluene, the model VOC. The GAC fluidized bioreactor demonstrated toluene removal efficiencies ranging from 50 to 80%, when inlet toluene loading varied in a range between 13.1 and 37.4 g/m$^3$-hr. The maximum elimination capacity determined in the GAC fluidized bioreactor was 172 g/m$^3$-hr at a toluene loading of 291 g/m$^3$-hr. Transient loading experiments revealed that the removal efficiency was remained unchanged during an increased loading period, and toluene introduced to the bioreactor was first absorbed to GAC and then slowly desorbed and became available to the yeast culture. Hence the fluidized GAC helped to achieve an improved mass transfer between the gas and liquid phases, resulting in high toluene removal capacity. Consequently, the GAC fluidized bioreactor using C. tropicalis can be successfully applied for the removal of VOCs, and is a feasible alternative over conventional processes such as packed-bed biofilters.

Epidemiological studies showed that high trans-fat consumption is closely associated with getting the risks of cardiovascular disease. The objective of this study was to produce trans-free fat through lipase-catalyzed interesterification, as a substitute for the cream margarine commonly used in industry. Optimum conditions for interesterification in a packed bed enzyme bioreactor (PBEB) were determined using response surface methodology (RSM) based on central composite design. Three kinds of reaction variables were chosen, such as substrate flow rate (0.4-1.2 mL/min), reaction temperature (60-70$^{\circ}C$), and ratio of fully hydrogenated canola oil (FHCO, 35-45%) to evaluate their effects on the degree of interesterification. Optimum conditions from the standpoint of solid fat content (SFC) were found to be as follows: 0.4 mL/min flow rate, 64.7$^{\circ}C$ reaction temperate, and 42.8% (w/w) ratio of FHCO, respectively. The half-life of immobilized lipase in PBEB with two stages at 60$^{\circ}C$ ($1^{st}$ stage) and 55$^{\circ}C$ ($2^{nd}$ stage) was about more than 30 days as estimated by extrapolating the incubation time course of tristearoyl glycerol (TS) conversion, whereas the half-life of the enzyme in PBEB with single stage at 65$^{\circ}C$ was only about 15 days. Finally, the results from SFC analysis suggest that trans-free fat produced in this study seems to be a suitable substitute for the cream margarine commonly used in industry.
