Effect of pH on Swelling Property of Hyaluronic Acid Hydrogels for Smart Drug Delivery Systems

  • Hyaluronic acid(HA) hydrogels were synthesized by immersing HA microbeads in phosphate buffered saline solutions having different pH levels to assess the effect of pH on the swelling ratio of HA hydrogels for smart drug delivery systems. No beads were formed when the HA solution(below pH 9) was crosslinked with divinyl sulfone(DVS) because DVS is a basic solution. The variation regarding the size of the microbead was not significant, suggesting that the bead size is not a function of pH(10 ~ 14). However, the pore size of the microbeads decreased with increasing pH from 10 to 14, leading to the surface smoothness and dense network as a result of higher crosslinking. The swelling ratio of hydrogels increased when the pH rose from 2(acidic) to 6(neutral). Afterwards, it decreased with further increasing pH(basic). The lower swelling ratio may be due to the lack of ionization of the carboxyl groups. On the other hand, a higher swelling ratio is likely due to the increased electrostatic repulsions between negatively charged carboxyl groups on different chains. Experimental results suggested that pH-responsive HA hydrogels can be applicable to the controlled drug delivery systems.

Partially Hydrolyzed Crosslinked Alginate-graft-Polymethacrylamide as a Novel Biopolymer-Based Superabsorbent Hydrogel Having pH - Responsive Properties

  • In this study, a series of highly swelling hydrogels based on sodium alginate (NaAlg) and polymethacryl­amide (PMAM) was prepared through free radical polymerization. The graft copolymerization reaction was performed in a homogeneous medium and in the presence of ammonium persulfate (APS) as an initiator and N,N'-methylenebis­acrylamide (MBA) as a crosslinker. The crosslinked graft copolymer, alginate-graft-polymethacrylamide (Alg-g­PMAM), was then partially hydrolyzed by NaOH solution to yield a hydrogel, hydrolyzed alginate-graft-poly­methacrylamide (H-Alg-g-PMAM). During alkaline hydrolysis, the carboxamide groups of Alg-g-PMAM were converted into hydrophilic carboxylate anions. Either the Alg-g-PMAM or the H-Alg-g-PMAM was characterized by FTIR spectroscopy. The effects of the grafting variables (i.e., concentration of MBA, MAM, and APS) and the alkaline hydrolysis conditions (i.e., NaOH concentration, hydrolysis time, and temperature) were optimized systematically to achieve a hydrogel having the maximum swelling capacity. Measurements of the absorbency in various aqueous salt solutions indicated that the swelling capacity decreased upon increasing the ionic strength of the swelling medium. This behavior could be attributed to a charge screening effect for monovalent cations, as well as ionic cross-linking for multivalent cations. Because of the high swelling capacity in salt solutions, however, the hydrogels might be considered as anti-salt superabsorbents. The swelling behavior of the superabsorbing hydrogels was also measured in solutions having values of pH ranging from 1 to 13. Furthermore, the pH reversibility and on/off switching behavior, measured at pH 2.0 and 8.0, suggested that the synthesized hydrogels were excellent candidates for the controlled delivery of bioactive agents. Finally, we performed preliminary investigations of the swelling kinetics of the synthesized hydrogels at various particle sizes.

  • The swelling behavior of hyddrogels has been interested in many applications of drug carriers. These gels show reversible swelling changes in response to pH, electric currcnt, and temperature. Among others, the temperature-responsive behavior of poly(N-isopropylacrylanxide) (p(NIPAAm)) was studied, because a lower critical solution temperature(LCST) is in the vicinity of 32$\circ$C, and remarkable temperature-response can be obtained. We propose a novel composite membrane, which is appropriate for transporting drug ingredients above the transition temperature. Our object was to design a high permeation system above the shrinking temperature of p(NIPAAm). The membrane was composed of a matrix polymer and thermosensitive p(NIPAAm) hydrogel. The flux pattern of 4-acctamidophen through membrane in response of temperature was opposite to that of p(NIPAAm) membrane.

Dual Responsive Pectin Hydrogels and Their Silver Nanocomposites: Swelling Studies, Controlled Drug Delivery and Antimicrobial Applications

  • Novel dual responsive pectin hydrogels composed from poly(acrylamidoglycolic acid-co-vinylcaprolactam)/Pectin (PAV-PC) and also PAV-PC hydrogels are used as templates for the production of silver nanoparticles. 5-Fluorouracil is an anticancer drug and has been loaded in situ into PAV-PC hydrogels. Structure and morphology characterization of PAV-PC hydrogels were investigated by fourier transform infrared spectroscopy, differential scanning calorimetry, thermo gravimetric analysis, X-ray diffraction studies, scanning electron microscopy and transmission electron microscopy. The results revealed a molecular level dispersion of the drug in PAV-PC hydrogels. In vitro release of 5-fluorouracil from the PAV-PC hydrogels has been carried out in GIT fluids as well as in various temperatures. 5-Fluorouracil released from PAV-PC hydrogels was 50% at pH 1.2, and 85% at pH 7.4 within 24 h. The release profile was characterized with PAV-PC hydrogels and initial burst effect was significantly reduced in two buffer media (1.2 and 7.4), followed by a continuous and controlled release phase, the drug release mechanism from polymer was due to Fickian diffusion. In situ fabrication of silver nanoparticles inside the hydrogel network via the reduction of sodium borohydrate by PAV-PC chains led to hydrogel nanocomposites. The diameter of the nanocomposites was about 50-100 nm, suitable for uptake within the gastrointestinal tract due to their nanosize range and mucoadhesive properties. These nanocomposite PAV-PC hydrogels showed strong antimicrobial activity towards Bacillus subtilis (G+ve) and Escherichia coli (G-ve).

글루코스를 함유한 음이온 하이드로젤의 pH 감응성 동적 팽윤거동 (pH-Sensitive Dynamic Swelling Behavior of Glucose-containing Anionic Hydrogels)

  • 음이온 하이드로젤은 그들이 가지고 있는 pH 감응성 팽윤거동 때문에 단백질 약물의 경구투여용 전달물질로써 많은 주목을 받고 있다. 본 연구에서는 음이온 하이드로젤의 pH 변화에 따른 용매의 침투 메커니즘을 규명하기 위하여 methacrylic acid와 2-methacryloxyehtyl glucoside를 공중합하여 P(MAA-co-MEG) 하이드로젤을 합성한 후 pH 변화에 따른 하이드로젤의 동적 팽윤거동을 관찰하였다. 용매의 침투 메커니즘이 Fickian 또는 non-Fickian 인지를 설명할 수 있는 특성지수 n을 $M_t/M_{\infty}=kt^n$ 관계식으로부터 계산하였다. 하이드로젤에 대한 용매의 침투 메커니즘은 주위 pH의 영향을 많이 받았으며, 젤의 $pK_a$ 보다 높은 pH인 7.0에서는 침투 메커니즘이 상대적으로 고분자사슬의 이완에 의한 지배를 많이 받는다는 것을 알 수 있었다. 한편, pH 7.0에서 고분자 이완에 의한 용매의 침투 메커니즘은 하이드로젤에 존재하는 카르복실산의 이온화에 기인한 것임을 ATR-FTIR 분광분석을 이용하여 확인하였다.

  • 생체적합하고 생분해성을 갖는 폴리아미노산의 하나인 폴리아스팔트산은 팹타이드 결합으로 이루어진 수용성 합성고분자로서 의료용 소재 및 다양한 응용분야의 연구가 흥미있게 진행되어 왔다. 본 연구에서는 아미노산인 GABA(${\gamma}$-aminobutylic acid)와 ${\beta}$-alanine을 곁사슬에 도입한 폴리아스팔트산 유도체 고분자를 제조하고, 이들을 서로 다른 함량의 ethylene glycol diglycidyl ether(EGDE)를 사용하여 가교시켜 고흡수성 하이드로젤을 제조하였다. 각 하이드로젤에 대해 증류수와 PBS 용액에서 기본적인 팽윤물성, 젤의 가수분해 거동과 모폴로지를 조사하였다. 제조한 하이드로젤은 수용액에서 pH와 염(salt) 농도에 의존하는 민감성 팽윤거동을 보였으며, 또한 비교적 빠른 가수분해 거동을 나타내었다.

Stimuli-Sensitive Poly(NIPA-co-APA) Hydrogels for the Controlled Release of Keterolac Tromethamine

  • The pH sensitive hydrogels composed of N-isopropylacrylamide (NIPA) and acryloyl phenylalanine (APA) were prepared by redox polymerization using N,N'-methylenebisacrylamide (MBA) as a crosslinker. Anti-inflammatory and analgesic agent, Keterolac Tromethamine (KT), was loaded successfully into poly(NIPA-co-APA) copolymeric hydrogels by swelling equilibrium method. To understand the nature of drug in the polymeric matrix, the newly synthesized drug loaded poly(NIPA-co-APA) copolymeric hydrogels were characterized by using differential scanning calorimetry (DSC) and X-ray diffraction (XRD) techniques. The scanning electron microscopy (SEM) technique result indicates the spherical smooth surface of the hydrogels. The drug (KT) releasing nature of the poly(NIPA-co-APA) hydrogels was studied in pH 1.2 and 7.4. Effects of drug loading, crosslinking agent, pH and the ionic strength of the external medium on swelling of hydrogels were also investigated.

Alginate/Carboxymethyl Scleroglucan Hydrogels for Controlled Release of Protein Drugs

  • Alginate/carboxymethyl scleroglucan (CMSG) hydrogels were suggested as a novel carrier for the controlled release of protein drugs. The drug release characteristics of alginate hydrogels were improved by CMSG addition. Scleroglucan (Sclg) was carboxymethylated using monochloroacetic acid in aqueous alkaline medium. Alginate/CMSG hydrogels were prepared by dropping the mixture solution of alginate/CMSG into calcium chloride solution. The swelling behaviors and drug release characteristics of the hydrogels were investigated in the buffers of pH 1.2 or 7.4. As the CMSG content increased in the hydrogels, the swelling ratio of the alginate/CMSG hydrogel increased rapidly in the buffer of pH 7.4. At pH 1.2, however, the swelling ratio significantly decreased compared to that at pH 7.4. According to in vitro release tests, only 15% of ovalbumin, investigated as a model protein drug, was released from the alginate/CMSG hydrogels at pH 1.2 within 6 h. At pH 7.4, however, the drug release significantly increased due to the rapid swelling of the hydrogels. The release and swelling behaviors of the hydrogels could be controlled by changing the CMSG content in the hydrogels. These results supported the use of alginate/CMSG hydrogels as a suitable carrier for the controlled release of protein drugs in a pH responsive manner.
