Effect of Horizontal Pitch-to-Diameter Ratio on the Natural-Convection Heat Transfer of Two Staggered Cylinders (엇갈리게 배열된 두 개의 수평관에서 수평 피치-직경비에 따른 자연대류 열전달 영향)

  • This study measured the natural-convection heat transfer of two vertically staggered cylinders with varying vertical pitch-to-diameter ($P_v$/D) and horizontal pitch-to-diameter ($P_h$/D) ratios. The measured heat-transfer rates for the lower cylinder agreed well with the existing heat-transfer correlations for a single cylinder. At the smallest $P_v$/D, the rising plume from the lower cylinder provides the upper cylinder with a preheated flow, and the heat-transfer rates of the upper cylinder decrease, but increase very sensitively with $P_h$/D. However, at the largest $P_v$/D, the velocity effect dominates, and the heat-transfer rates of the upper cylinder are larger than that of a single cylinder, and decrease less sensitively with $P_h$/D. Even if $P_h$/D is increased, the heat-transfer rate of the upper cylinder is higher than that of the lower cylinder because of the chimney and side flow effects. This work expanded the flow ranges to turbulent flows. The cupric acid-copper sulfate ($H_2SO_4-CuSO_4$) electroplating system was adopted for the measurements of the mass-transfer rates instead of the heat-transfer experiments based on the analogy concept. The measurements were made by varying $P_v$/D (1.02-5) and $P_h$/D (0-2) in both laminar and turbulent flows. The Rayleigh number ranged from $1.5{\times}10^8$ to $2.5{\times}10^{10}$, and the Prandtl number was 2,014.

Continuous Production of Fructooligosaccharides from Sucrose by a Dual Immobilized Enzyme System of Fructosyltransferase and Glucose Isomerase (과당전이효소와 포도당 이성화 효소의 고정화 혼합효소계에 의한 설탕으로부터 프락토올리고당의 연속생산)

  • Continuous production of fructooligosaccharides from sucrose by a dual immobilized enzyme system of fructosyltransferase and glucose isomerase was studied in a column reactor. The optimal temperature and pH of the immobilized fructosyltransferase were $65^{\circ}C$ and 5.5, respectively. The activity of glucose isomerase was favorable as temperature and pH were increased within the ranges examined. However, both the immobilized enzymes were thermally unstable over $5^{\circ}C$, suggesting that long-term operation of the dual immobilized enzyme column should be carried out below $50^{\circ}C$. The optimum packing ratio of fructosyltransferase to glucose isomerase was found to be around 5/3. Under the optimized reaction conditions, the dual enzyme column was successfully operated for 40 days without any loss of initial enzyme activities, yielding 66% of fructooligosaccharides. Furthermore, the relative sweetness of fructooligosaccharides produced by a dual emzyme system was enhanced by 6% compared with that of fructosyltransferase alone.

  • Tests for the physical and chemical properties on the dispersive soils from the 3 selected sites in the northeast region of Thailand were carried out in the field and laboratory. In oder to find out the relationship between c1ay contents and dispersivity of the soil, some specimens are artificially mixed with a certain amount of clay in the laboratory and tested by means of pinhole tests. The artificial soils are dispersed in pinhole tests regardless of their clay contents. Chemical properties on the soil saturation extract and reservoir water are also determined by means of sodium adsorption ratio (SA,R), exchangeable sodium percentage (ESP) and pH. The dispersive soils in the region have ranges of 1.8~6.7 SAR, 1.4~7.9 ESP and 5.4~9.0 pH value. Namely, the dispersive soils studied have somewhat higher SAR, ESP and pH value than those of ordinary soil.

Determination of Optimum Coagulants (Ferric Chloride and Alum) for Arsenic and Turbidity Removal by Coagulation

  • The Raw water from Deer Creek (DC) reservoir and Little Cottonwood Creek (LCC) reservoir in the Utah, USA were collected for jar test experiments. This study examined the removal of arsenic and turbidity by means of coagulation and flocculation processes using of aluminum sulfate and ferric chloride as coagulants for 13 jar tests. The jar tests were performed to determine the optimal pH range, alum concentration, ferric chloride concentration and polymer concentration for arsenic and turbidity removal. The results showed that a comparison was made between alum and ferric chloride as coagulant. Removal efficiency of arsenic and turbidity for alum (16 mg/L) of up to 79.6% and 90.3% at pH 6.5 respectively were observed. Removal efficiency of arsenic and turbidity for ferric chloride (8 mg/L) of up to 59.5% at pH 8 and 90.6% at pH 8 respectively were observed. Optimum arsenic and turbidity removal for alum dosages were achieved with a 25 mg/L and 16 mg/L respectively. Optimum arsenic and turbidity removal for ferric chloride dosages were achieved with a 20 mg/Land 8 mg/L respectively. In terms of minimizing the arsenic and turbidity levels, the optimum pH ranges were 6.5 and 8for alum and ferric chloride respectively. When a dosage of 2 mg/L of potassium permanganate and 8 mg/L of ferric chloride were employed, potassium permanganate can improve arsenic removal, but not turbidity removal.

  • Solvent extraction experiments have been performed to separate Nd and Pr from chloride leaching solution of monazite sand using single Cyanex272 and mixed extractants as PC88A+Cyanex272 and PC88A+TBP. For this purpose, the effect of the concentration of extractants on the extraction and separation of the two metals were studied by varying the pH of aqueous solution. In the experimental ranges conducted in this study, the distribution coefficients of Nd were higher than those of Pr. In Cyanex272 system, our results indicated that concentration of extractant and initial pH did not affect distribution coefficients, but separation factor was increased with increasing initial pH. In binary extractant system, distribution coefficients were lower than those of single PC88A system, whereas separation factor was similar in both mixed and single extractant system.

Effect of Incubation Time, Temperature and pH on the Production of Conidia and Chlamydospore of Cylindrocarpon destrutans (Zinssm.) Scholten Causing Root Rot of Panax ginseng (인삼 근부병균 Cylindrocarpon destructans (Zinssm.) Scholten의 포자 생성에 미치는 배양기간, 온도, pH의 영향)

  • The effects of media, incubation time, temperature and pH on production of conidia and chlamydospore of Cylindrocarpon destructans (Zinssm.) Scholen causing root rot of Panax ginseng were studied. Microconidia of the pathogen were abundantly produced on V-8 juice agar as a solid substrate with 5.73(log conidia/mm2) and in V-8 broth as a liquid substrate with 6.65 (log conidia/ml) among media tested. No difference was observed on the length of microconidia produced from the media with a range of 9.50∼11.38 $\mu\textrm{m}$. However, tryptic soy agar produced the broadest microconidia (average 5.00 $\mu\textrm{m}$) among the media tested. All the media produced chlamydospores In a range of 1.06∼4.37 (log chlamydospores/mm2) without a significant difference in number, while V-8 juice agar produced the bigger one (18.39 $\mu\textrm{m}$ in diameter) as compared to the tested media. The fungus began to sporulate conidia after three days of incubation and reached maximum at the 8th day. It seemed to be in a stationary phase until 30 days of incubation but was decreased thereafter. Chlamydospore was produced at 4th day after incubation. Maximum production was observed at 8th day and the number seemed to be maintained during the observation period. Both conidia and chlamydospore of the pathogen were able to be spoluated at 10∼25$^{\circ}C$. However, optimum temperatures of conidia and chlamydospore formation were 15∼25$^{\circ}C$ and 10∼20$^{\circ}C$, respectively. C. destrmtans produced conida with an wide range of pH from 3.3 to 8.0 and chlamydospore from 2.8 to 8.0. Number of conidia was increased with an increase of pH up to 4.0. There was no significant difference in the number between 4.0 to 8.0. It seemed to have two optimum pH ranges, 3.3∼4.0 and 7.1∼8.0 for the chlamydospore formation.

Antimicrobial Activities of Extracts from Several Native and Exotic Plants in Korea (수 종의 한국자생식물과 귀화식물 추출액이 토양미생물에 미치는 항균활성)

  • The soil pH favored by several native plants in Korea ranges 5.33∼7.20, while a more acidic range of pH 3.95∼6.10 is acceptable to exotic plants. Ethanol extracts of native and exotic plants in Korea were investigated for antimicrobial activity against Bacillus sphiaericus 2362, Bacillus thuringiensis var. subtilis and Bacillus thuringiensis var. cereus and Actinomycetes. Higher antimicrobial activity was observed from the extract of exotic plants than those of native plants. The ethanol extract of Ambrosia artemisiifolia var. elatior was observed to have the highest antimicrobial activity against 4 species of soil microbes. Especially, antimicrobial activity of Ambrosia artemisiifolia var. elatior showed the largest clear zone of 48mm in Actinomycetes. Larger clear zone was formed in the order of caffeic acid, benzoic acid and ρ -coumaric acid among the nine chemical compounds. Accordingly, the antimicrobial activity of Ambrosia artemisiifolia var elatior against Actinomycetes was found to be due to the synergetic effect of chemical compounds.

Properties of Glucoamylase Isozymes Produced by Aspergillus sp. (Aspergillus sp.가 생산하는 Glucoamylase Isozymes의 성질)

  • Glucoamylase (EC 3.2.1.3) of Aspergillus sp. isolated from soil was partially purified by Sephacryl S-200 gel filtration and DEAE-Sephacel ion exchange chromatography, The glucoamylase activity was separated into two isozymes after DEAE-Sephacel ion exchange chromatrography. The optimum pH and temperature for both glucoamylase isozymes (GI, GII) were identical; pH 4.5 and temperature, $65^{\circ}C$. The molecular weights of GI and GII Isozymes were estimated to be 105,000, which were measured by gel filtration on Sephacryl S-200. Both isozymes were stable at pH ranges of 2 to 7, and up to 6$0^{\circ}C$. Glycerol was effective to stabilize the both isozymes. The activation energies of GI and GII isozymes were 10.63 and 10.33 kcal/mole, respectively. The enzyme activities of both isozymes were completely inactivated by addition of 0.1% Hg$^{++}$. In kinetic studies, the Km values of GI isozyme for soluble starch, dextrin, and glycogen were 0.62%, 0.32%, and 1.02%, respectively. For GII isozyme, they became 0.66%, 0.23%. and 0.14% for the substrates.

  • A 50 KD metalloprotease of Serratia marcesrens ATCC 21074 was purified by ammoniumsulfate precipitation. DEAE-cellulose ion exchange chromatography, and sephadex ti-100gel filtration. Optimal pH and temperature of enzyme were pH 8.0 and 37"C, respectively.This enzyme was stable in the ranges of 10-37$^{\circ}$C and pH 5.0--11.0. Thermal denaturationwas investigated by differential scanning calorimetry. Onset temperature of denaturationand endothermic peak temperature were 376$^{\circ}$C and 43.2"C. re:,pectively. The denaturationenthalpy was -8.4mJimg. The purified metalloprotease was ri.sistant to autodigestion for24 hr at 30$^{\circ}$C. Metalloprotease in culture supernatant was also resistant to autodigestionin this conditions. Heat-denatured enzyme. however. was rapidly digested by the nativeenzyme. The metalloprotease was stable to proteolytic digestion by mammalian proteasessuch as trypsin. a-chymotrypsin, and elastase. But the enzyme was easily digested bybacterial protease. thermolysin.bacterial protease. thermolysin.

Study on Spatial Characteristics of Physicochemical Components of Spring Water in Mts. Geumjeong and Baekyang Area Using Kriging (크리깅 기법을 이용한 금정산-백양산 일대 용천수의 물리화학적 성분의 공간적 분포 특성 연구)

  • Sping waters at 60 sites and groundwaters at 6 wells in Mts. Geumjeong and Baekyang area were investigated for measuring nine physicochemical components (temperature, pH, Eh, EC, TDS, DO, salinity, alkalinity and discharge rate). The pH of spring waters ranges from 5.06 to 8.38, alkalinity from 7.93 to 102.21 mg/ㅣ, electrical conductance from 9.5 to $270{\;}\mu\textrm{s}/cm$, Eh from 64.2 to 685.9 mV, DO from 4.57 to 12.13 mg/l, and the discharge rate from 4.26 to 182.2 ml/s. General statistics was carried out to analyze statistical characteristics of those components. To compare the components with one another, regression analyses were carried out. And the components of spring waters were compared to those of groundwaters. Kriging was used to estimate the spatial variation of DO, pH, Eh, EC, alkalinity and discharge rate in the study area. The kriged isopleth maps were made using normalized kriged values to find anomalies of the physicochemical components and to compare anomalies of different components.
