To determine application rate of elemental sulfur to adjust pH of alkaline soil, buffer curve method was investigated. The elemental sulfur required to control pH 8.3 to pH 6.3 by buffer curve calculation was treated in two soils of silty loam and sandy loam, and the sulfur-mixed soils were moistened with 50% of water holding capacity during incubation of 6 weeks at $30^{\circ}C$. Soil pH was lowered with incubation and reached to target point after 4 weeks of incubation, and elemental sulfur was oxidised entirely to sulfate. This means that buffer curve has the accuracy to determine sulfur application rate in alkaline soil. However it is estimated that application rate of sulfur should be carefully determined in the field scale. Excess application of elemental sulfur resulted in extremely low soil pH and caused the hinderance of lettuce growth by nutritional imbalance and ion toxicity. To simplify the determination procedure of sulfur requirement, buffer curve method by addition of 0.1N-HCl solution as unit of mL was developed, it was compared with theroutine methods which diluted $H_2SO_4$ solution and $Ca(OH)_2$ are added as cmolc per kg soil to adjust each pH step. Buffer capacities, cmolc kg $soil^{-1}$$pH^{-1}$, calculated from two buffer curves were not significantly different. The result indicates that buffer curve method by 0.1N-HCl can be used to adjust high pH of alkaline soil.

Heavy metals are extracted from Chonju stream sediment, roadside soils and sediments along Honam expressway, soils and tailings from mining area using partial ectraction in Standard Method, partial ectraction method with maintaining 0.1N of extraction solution and acid digestion. In samples having buffer capacity against acid, 0.1N of extraction solution can not be maintained and pH of extraction solution increases up to 8.0 when partial extraction in Standard Method is used. The averages and ranges of (heavy metals extracted using partial extraction in standard method, HPE)/(heavy metals extracted using partial extraction method with maintaining 0.1N of extraction solution, HPEM) values are 0.506 and 0.145~1.126 in Cd, 0.534~ and 0.078~0.928 in Zn, 0.461 and 0.041~1.715 in Mn, 0.359 and 0.011~0.874 in Cu, 0.195 and 0.018~1.785 in Cr, 0.710 and 0.003~3.075 in Pb, and 0.088 and 1.73$\times$10$^{-5}$ ~0.303 in Fe. These data indicate that the difference between HPE and HPEM is big in the order of Fe, Cr, Cu, Mn, Cd, Zn and Pb. It is quite possible that the partial extraction method in Standard Method of soil in Korea is not adequate for an assessment of contamination in area where buffer capacity of soil will be decreased or lost after a long term exposure of soils to environmental damage.

We investigated the functional properties and in vitro bioactivity of protein isolates (RPIs) recovered from olive flounder Paralichthys olivaceus roes by isoelectric solubilization/precipitation process, according to pH-shift treatments. The buffer capacity of RPIs was shown to be stronger in alkaline pH than in acidic pH. Water holding capacity of RPIs was in range of 4.5-5.2 g/g protein with no significant differences (P>0.05). The foaming capacity and emulsifying activity index of RPIs did not show any significant differences between RPI-1 (pH 11/4.5) and 3 (pH 12/4.5), however they were superior to RPI-2 (pH 11/5.5) and 4 (pH 12/5.5). The 2,2'-azino-bis-3-ethylbenzo-thiazoline-6-sulfonic acid radical scavenging activity of RPI-3 (2.5 mg protein/mL) was 102.4 ㎍/mL, and the angiotensin I converting enzyme inhibitory activity was 30.8%. Among the RPIs, RPI-3 was relatively superior in protein functional properties such as buffer capacity, foaming capacity, emulsification, and anti-oxidative activity. Therefore, we suggest that RPI prepared from olive flounder roes could serve as a potential food resource.
Soil pH is an intensity factor of releasing hydrogen ion which is buffered by aluminum. It depends on pH buffer capacity of Al whether soil pH is governed directly by cations or not. A study was conducted to elucidate the pattern of pH changes by soil EC. Fertilizer and three kinds of organic manures composed of cow and pig and fowl dropping and one kind of rice straw compost were added independently into upland sandy loam soil. This treated soils and four upland soils under plastic film house having different levels in electrical conductivity (EC) were incubated with field capacity at $30^{\circ}C$ for 5, 10, 20 and 40 days. Soil pH varied directly as the cations contained in organic materials according to degree of saturating pH buffer capacity (pBC) of sandy loam soil. pH of the soils under plastic film house was lowered by soil EC due to governing by overplus of cation beyond pBC.

Pesticides from diffuse pollution sources adsorbed in suspended particles flow into surface water and threats to the public health. In this research, dechlorination constants of Atrazine by zero valent iron were measured with addition of buffer solution for simulating buffer capacity of sediment. When initial concentration of Atrazine was 10, 30, and 50 mg/L, their dechlorination was explained using the pseudo-first order reaction. Dechlorination constants $K_{obs}$ were $3.21{\times}10^{-2}/d$ in average.
The optimal pH of the extraction buffer was 7.5 considiering AChE stability and its buffer capacity when AChE was isolated and extracted from the housefly(Musca domesitca L.)and three other insect species with 0.01 M sodium phosphate buffer. Also, the optimal pH of the reaction buffer was 7.5 considering enzyme activity and its buffer capacity when AChE activity was measured with the substrate in 0.1 M sodium phosphate buffer. The Potter Elvehjem type homogenizer with Teflon pestle was used to homgenize the tissues. When preparing a AChE suspension by centrifuging the homogenate, 700 g supernatant of adult head for the housefly, 700 g supernatnat of 5th instar nymphal whole body for the brown planthopper, lipid-eliminated 10,000 g supernatant of 5th instar larval whole body for the diamondback moth, and 700 g supernatant of 4th instar larval head for the tobacco cutworm were considered satisfactory as enzyme sources in view of mass preparation, extraction efficiency and stability of enzyme activity during evaluation. When AChE suspensions of 4 insect species were stored at $-18^{\circ}C$, more than 90% of activity was maintained up to 3 weeks. Km values of AChEs of the housefly, the brown planthopper, and the diamondback moth were 0.042, 0.037 and 0.043 mM, respectively and AChE-specific substrate inhibition was observed at high concentration. Km value of the tobacco cutworm ChE was 1.15 mM and BuChE characteristics was observed, though further study is needed. The optimal substrate concentration for the AChE inhibition tests was 0.5 mM for the housfly, the brown planthopper, and the diamondback moth and 12 mM for the tobacco cutworm.

This study was conducted to find out buffer capacities of forest soils by the treatment of simulated acid rain(SAR) of four forests(Q. spp., P. rigida, P. koraiensis, L. leptolepis) in Kyunghee university's practice forest. All soils of each forest stand were treated by simulated acid rain at the level of pH 3.0, 4.0, 5.0 respectively. The result obtained from this study can be summarized as follows: Soil pH was measured by soil depth of each forest stand. The deeper soil depth was, the higher soil pH was. Also it was appeared that base saturation of sample soils was the highest as 17.42% in P. rigida stand and cation exchange capacity(C.E.C) was the highest as 29.87 me/100 g in Q. spp. stand. for responses of soil leachates to acidification treatment with pH 3.0 simulated acid rain(SAR), as simulated acid rain(SAR)-input was increased, pH value of soil leachates appeared high temporarily, but soon pH value of soil leachates had been low gradually. At the rest of pH 4.0, pH 5.0 treatment, pH value of soil leachates was high proportionably. The amounts of TBC of primary stage had a difference as pH level of simulated acid rain and forest stands. But as simulated acid rain(SAR)-input was increased. Amount of TBC was diminished. Also the amounts of TBC of primary stage in acidification treatment with pH 3.0, 4.0 simulated acid rain(SAR) was higher that of acidification treatment with pH 5.0 simulated acid rain(SAR). These trend showed obvious difference at low soil acidity and high TBC. The amounts of activity Al of primary stage appeared high as increasing the input acidity of simulated acid rain(SAR). Also, by soil depth, the amounts of Activity Al was different between A layer(0-15 cm) and AB layer(0-30 cm). There was considerable the correlation between simulated acid rain-input and activity Al change. But this was oppositional trend in soil leachates of pH 4.0, 5.0 treatment and total base cations(TBC).
Protein affinity membranes were prepared via coating of chitosan gel on the porous flat and hollow-fiber polysulfone membranes, followed by the immobilization of the reactive dye (Cibacron Blue 3GA) to the chitosan gel. Maximum protein binding capacity of these affinity membranes was about 70 $\mu{g/cm}^2$. Using the affinity flat membrane module, the elution chromatography of human serum albumin (HSA) was performed to determine the optimum condition of eluent buffer. The optimum condition of eluent was the universal buffer solution of 0.06 M concentration containing 1 M KCl at pH 10. For the frontal chromatography of HSA using the flat module, the dynamic protein binding capacity was rapidly decreased from the equilibrium values with increasing flow rate and HSA concentration of the loading solution. However, in the case of hollow-fiber module, the dynamic binding capacity was maintained an equilibrium value without depending on the operating conditions. These results showed that the hollow-fiber module was more effective than the flat module as an affinity chromatography column.
The comparative molecular field analysis (CoMFA) method has been employed to correlate the apparent lipophilicity (logkw) and global lipophilicity (logP) for orthopramide derivatives. This study demonstrated that CoMFA is an excellent method in predicting the complex properties of molecules such as apparent lipophilicity (logkw) or lipophilicity (logP). The better predictability of lipophilicity by introducing logkw as an independent descriptor suggests that the HPLC capacity factor measured in a buffer of pH 7.5 (logkw) can be effectively utilized in the prediction of global lipophilicity.

This study was investigated to determine the functional properties of mugbean protein isolates(MPI) from sunhwa-nogdu(SH) and conventional mungbean varieties(C). MPI were prepared from defatted mungbean flour by extraction with 0.1N NaOH, precipitation at pH 4.5, washing of dispersed precipitate with buffer solution and distilled water, and subsequent freeze-drying. Crude protein content of MPI was in the range of $88.7{\sim}91.3%$. The lowest solubility was recorded at $pH\;4{\sim}5$, whereas the best buffering action was in the range of $5.5{\sim}7.5$. On the other hand, gelation of MPI was found to depend on the protein concentration. In the cases of foamability, % volume increase and specific volume were higher for 10 min. with a good whipping ability. And also, the MPI properties of two varieties of SH and C were compared and discussed.
