  • 1.이온토포레시스에 의한 ISP의 경피촙수증가는 단순투과의 약 13배로서 그 증가정도는 전류세기와 약물농도에 비례하였다. 2. 가해주는 $Na^{+}$ 농도가 커질수록 ISP의 flux는 감소하였다. 3. ion-pairing agent률 가하면 ISP의 flux는 감소하는데, 그 감소정도는 TU>SAL>BEN 로서 이는 이 물질들이 ISP와 ion-pair를 형성하는 능력순서와 같았다. 4. ISP용액의 pH증가시 ISP의 flux는 대체적으로 증가하며 그 pattern은 피부의 pKa를 3.5로 가정할 때의 피부해리곡선과 유사하였다. ISP가 광범위한 pH에서 완벽하게 해리된다고 가정할 때 pH증가시 flux증가는 피부해리 증가에 따른 permselectivity 증가에 기인한 것으로 생각되었다.

The Synthesis of p-Dodecylbenzylidene-p'-hexyloxyaniline (p-Dodecylbenzylidene-p'-hexyloxyaniline의 합성)

  • In this study, we synthesized p-hexyloxybenzaldehyde(HBA) by using p-hydroxybenzaldehyde and n-hexylchloride. p-dodecylaniline(DDA) prepared by nitration and reduction of dodecylbenzene. The schiff base, p-dodecylbenzylidene-$p^\prime$-hexyloxyaniline (DBHA) was synthesized by reaction of HBA and DDA. The color of synthetic compound was pale-brown and the yield was 62%. Effects of E-book-based Flipped Learning Education on Critical Thinking Disposition, Academic Self-Efficacy, and Major Satisfaction of Nursing Students (E-book 기반 플립드 러닝(Flipped Learning) 수업이 간호대학생의 비판적 사고성향, 학업적 자기효능감 전공만족도에 미치는 효과)

  • This study was conducted to develop and test effects of E-book based Flipped Learning education for nursing students. This study was one-group pretest-posttest design. The data were collected from 54 second-year nursing students in the located Y city and August 28 2017 to October 16 2017. The data were analyzed by descriptive statistics, paired t-test, Pearson's correlation coefficient, and stepwise multiple regression with SPSS 20.0 program. The results showed that the program was effective in increasing the critical thinking disposition (t=-8.62, p<.001), academic self-efficacy (t=-9.62, p<.001) and major satisfaction (t=-8.11, p<.001). The result of the stepwise multiple regression indicates the critical thinking disposition predict 13.4% (F=9.22, p<.001) of major satisfaction. Therefore, strategies for enhancing critical thinking disposition is needed by applying various teaching and learning strategies for nursing students.

Induction of Apoptosis of DK-5-62, a Novel (-)-Catechin Derivative Through MAPKs Signaling Pathway in HCT116 Cells

  • The present study was designed to investigate the molecular mechanisms of DK-5-62, a novel (-)-catechin derivative on HCT116 human colorectal cancer cells. DK-5-62 inhibited the proliferation in dose- and time-dependent manner accompanied by the morphological changes. Effects of DK-5-62 appeared to be mediated by the induction of apoptosis, as manifested through DNA-binding dye Hoechst 33258 staining. Analysis of the mechanism of these events indicated that DK-5-62-treated cells exhibited an increased ratio of Bax/Bcl-2, resulting in the activation of caspase-9, caspase-3, and poly-ADP-ribose polymerase in a dose-dependent manner. Moreover, DK-5-62-induced apoptosis was accompanied by phosphorylation of the mitogen-activated protein kinase family, c-Jun N-terminal kinase, p38, and extracellular signal-regulated kinase. These results suggest that HCT116 cells are moderately sensitive to growth inhibition by DK-5-62 via apoptosis, as evidenced by activation of ERK/p38/Bcl-2 family signaling, as well as alteration in caspase-9 and caspase-3.

  • The ubiquitin-proteasome system (UPS) and autophagy are two major degradative pathways of proteins in eukaryotic cells. As about 30% of newly synthesized proteins are known to be misfolded under normal cell conditions, the precise and timely operation of the UPS and autophagy to remove them as well as their tightly controlled regulation, is so important for proper cell function and survival. In the UPS, target proteins are labeled by small proteins called ubiquitin, which are then transported to the proteasome complex for degradation. Alternatively, many greatly damaged proteins are believed to be delivered to the lysosome for autophagic degradation. Although these autophagy and UPS pathways have not been considered to be directly related, many recent studies proposed their close link and dynamic interconversion. In this review, we'll focus on the several regulatory molecules that function in both UPS and autophagy and their crosstalk. Among the proposed multiple modulators, we will take a closer look at the so-called main connector of UPS-autophagy regulation, p62. Last, the functional role of p62 in the mitophagy and its implication for the pathogenesis of Parkinson's disease, one of the major neurodegenerative diseases, will be briefly reviewed.

Polyubiquitin-Proteasomal Degradation of Leucine-Rich Repeat Kinase 2 Wildtype and G2019S

  • Parkinson disease (PD) is becoming one of the most neurodegenerative disorder worldwide. The deposited aggregates have been connected in the pathophysiology of PD, which are degraded either by ubiquitin-proteasomal system (UPS) or autophagy-lysosomal pathway (ALP). Leucin-rich repeat kinase 2 (LRRK2), one of the neurodegenerative proteins of PD is also stringently controlled by both UPS and ALP degradation as well. However, the polyubiquitination pattern of LRRK2 aggregates is largely unknown. Here, we found that K63-linked polyubiquitinations of G2019S mutant, most familial variant for PD, is highly enhanced compared to those of wild type LRRK2 (WT). In addition, in the presence of overexpressed p62/SQSTM-1, ubiquitination of LRRK2 WT or D1994A was reduced, whereas G2019S mutant was not diminished significantly. Therefore, we propose that degradation of G2019S via UPS is more involved with K63-linked ubiquitination than K48-linked ubiquitination, and overexpressed p62/SQSTM-1 does not enhance degradative effect on G2019S variant.

Effects of bone-specific physical activity on body composition, bone mineral density, and health-related physical fitness in middle-aged women

  • [Purpose] The study aimed to determine the effects of bone-specific physical activity on body composition, bone mineral density (BMD), and health-related physical fitness in middle-aged women. [Methods] One hundred eighty-six middle-aged women aged 31-49 years participated in this study. The subjects were divided into tertile groups according to the level of physical activity (low-score group, n=62; middle-score group, n=62; high-score group, n=62). Bone-specific physical activity participation was assessed using the bone-specific physical activity questionnaire. Body composition and BMD were measured using dual-energy X-ray absorptiometry. Health-related physical fitness test included isometric muscle strength (grip strength), muscular endurance (sit-ups), flexibility (sit and reach), and cardiorespiratory fitness (maximal oxygen uptake [VO2max]). [Results] The high-score group had a significantly higher fat-free mass (p=.045, partial eta-squared value[ηp2]=.033) than the middle- and low-score groups, whereas the high-score group had significantly lower percent body fat (p=.005, ηp2=.056) than the other two groups. Whole-body BMD (p=.034, ηp2=.036) and lumbar BMD (p=.003, ηp2=.060) were significantly higher in the high-score group than in the low-score group. The high-score group performed significantly better for grip strength (p=.0001, ηp2=.101), sit-ups (p=.0001, ηp2=.108), and VO2max (p=.0001, ηp2=.092) than the other two groups. [Conclusion] The present study suggests that bone-specific physical activity could be useful in improving body composition, BMD, and health-related physical fitness in middle-aged women, significantly enhancing their BMD and health conditions.
