흰쥐에 있어 탄수화물의 종류에 따른 단백질의 체내 이용에 관한 연구 (Effect of Glucose, Starch, Sucrose on the Protein Utilization In Weanling Rats)

  • This study was conducted to compare effects of various types of dietary carboh ydrates fed with different levels of protein on the protein utilization in weanling rats. Sixty male Sprague-Dawley rats weighing $60{\pm}1.3grams$ were adapted for 1 week with 77% starch-15% casein diet. Then the animals divided into 12 groups according to body weight and fed each experimental diet for two weeks. Carbodydrates used were glucose, starch, and sucrose and the amount of protein given were 0g, 1g, 3g, 5g casein/day. Protein portion of the diet was fed in two seperate feedings per day while nonprotein portion was fed ad libitum. It seemed that there was no significant difference in the protein utilization by using the different kinds of carbohydrate, but in P.E.R., N.P.U., weights of organs and protein and lipid in total carcass, glucose groups were tended to be slightly lower than starch and sucrose groups. The larger the amount of casein given, the higher were the value of body weight gain, F.E.R., weights of organs, total lipid in carcass and the amount of nitrogen retention. On the while, the larger the amount of casein given, the lower were the value of the intake of non-protein portion, P.E.R., N.P.U, and the percentage of nitrogen retention.

사과박 첨가가 볏짚 사일리지의 품질과 in situ 소실율에 미치는 영향 (The Effect of Addition of Apple Pomace on Quality and in situ Degradability of Rice Straw Silage)

  • The quality of the rice straw silage added with apple pomace was investigated in this study and the amount of apple pomace added in different treatments were 0, 20, 40 and 60%, respectively. Crude protein contents (6.4-7.5%) of rice straw silage added with apple pomace were significantly (P<0.05) higher than that of 100%. rice straw silage (5.3%), however, crude ash contents were lower (P<0.05) in supplementation of apple pomace. The trends of changing chemical composition between raw materials and end products of silages particularly in the contents of crude protein and crude ash were more apparent in the silages added with apple pomace by 40-60%. Values of pH and the contents of lactic acid and total acid in 40-60% apple pomace added silages were 3.9-4.1, 1.0- 1.5% and 2.7-4.5%, respectively which were significantly (P<0.05) higher than those of 4.6, 0.02% and 0.34% in 100% rice straw silage, respectively. In situ dry matter (DM) and neutral detergent fiber (NDF) disappearance rates in the rumen in the treatments of 40- 60% apple pomace added silages were significantly (P<0.05) higher than those of 100% rice straw silage particularly since after 3 and 24 hour incubation on DM and NDF disappearance, respectively. Although quickly degraded fraction (a) among the treatments were not significantly different, 28.4-28.5% of slowly degraded fraction (b) and 27.2-27.4% of effective degradability (ED, k=0.08) for DM were significantly (P<0.05) higher than those of 100% rice straw silage (12.5 and 24.6% respectively). NDF was in the same trend as in DM. 31.6-63.2% of NDF for b fraction and 18.7- 19.4% for ED in 40-60% apple pomace added silages were significantly (P<0.05) higher than those of 100% rice straw silage (12.4 and 17.6% respectively). Rapid Induction of mRNA for Prostaglandin H Synthase in Ovine Meningeal Fibroblasts

  • We examined effects of interleukin $1{\alpha}$ ($IL1{\alpha}$) and phorbol 12, 13 dibutyrate (PDB), an activator of protein kinase C, on mRNA for Prostaglandin H synthase (PGHS) and prostanoid production in cultured ovine meningeal fibroblasts. Immuno- and morphologically-identified fibroblasts were derived from cerebral cortex and white matter from fetal lambs (approximately 120 days gestation) and grown to confluence on glass coverslips in 12 well plates. Levels of prostaglandin $F_{2{\alpha}}$ and the stable hydrolysis product of prostacyclin (i.e., $6-keto-PGF_{1{\alpha}}$) were determined using enzyme immunoassay. Relative amounts of mRNA were determined by in situ hybridization using ovine cDNA for PGHS1. $IL1{\alpha}$ (10 ng/ml) increased mRNA levels over baseline by $62{\pm}19%$ (p<0.05) at 60 min., $37{\pm}12%$ (NS) at 120 min., and $36{\pm}18%$ (NS) at 240 min (n=12). Levels of $6-keto-PGF_{1{\alpha}}$ were $148{\pm}18%$ pg/ml during baseline, $246{\pm}41%$ pg/ml at 60 min., $248{\pm}40%$ pg/ml at 120 min., and $259{\pm}62%$ pg/ml at 240 min (all p<0.05) (n=12). $PGF_{2{\alpha}}$ was increased although it wasn't statistically significant. However, $IL1{\alpha}$ decreased $PGE_2$ level significantly (all p<0.05). PDB $(10^{-6}M)$ increased mRNA levels over baseline by $25{\pm}6%$ after 30 min., $40{\pm}6%$ after 60 min., and $20{\pm}8%$ after 90 min. (n=9) (all p<0.05). Levels of $6-keto-PGF_{1{\alpha}}$ were $200{\pm}43%$ pg/ml during baseline, $202{\pm}43%$ pg/ml after 30 min. (NS), $268{\pm}58%$ pg/ml after 60 min. (p<0.05), and $296{\pm}60%$ pg/ml after 90 min. (p<0.05) (n=9). Levels of $PGF_{2{\alpha}}$ were $178{\pm}26%$ pg/ml during baseline, $300{\pm}30%$ pg/ml after 30 min., $299{\pm}35%$ pg/ml after 60 min., and $355{\pm}32%$ pg/ml after 90 min (all p<0.05) (n=6). Actinomycin-D (1 mg/ml) prevented increases in mRNA, $6-keto-PGF_{1{\alpha}}$, and $PGF_{2{\alpha}}$ at 60 min. for both $IL1{\alpha}$ and PDB. We conclude that cerebral fibroblasts are avid producers of prostanoids, and that enhanced production of PGHS is responsible for augmented $PGF_{2{\alpha}}$ and prostacyclin production in the presence of an activator of protein kinase C and for decreased $PGE_2$ and increased prostacyclin production in the presence of $IL1{\alpha}$.

파인애플과 키위 과피 추출물을 이용한 가축 혈액, 파란, 글루텐 분말의 가수분해 조건 (Hydrolysis of Blood, Egg and Gluten Meals with the Extracts from the Skins of Pineapple and Kiwi)

  • 파인애플과 키위의 과피 추출물의 단백질분석과 이를 이용한 혈분, 파란분말 그리고 글루텐 분말의 가수분해 최적 조건을 조사하였다. 전기영동에 의하여 단백질을 분석한 결과 파인애플 과피 추출물에서는 분자량 22 kd의 단백질이 나타났으며 키위의 과피 추출물에서는 분자량 27 kd, 22.5 kd, 22 kd, 19kd 그리고 14.4 kd인 단백질 6종이 나타났다. 파인애플 과피 추출물에서 나타난 22 kd 단백질은 단백질 분해효소인 bromelain인 것으로 추정되며, 키위 과피 추출물에서 나타난 27 kd 단백질은 단백질 분해효소인 actinidin인 것으로 추정된다. 파인애플과 키위의 과피 추출물을 이용한 혈분, 파란 분말 그리고 글루텐 분말의 가수분해 최적 시간, 온도 및 pH는 각각 6에서 24시간, $60^{\circ}C$ 그리고 pH 4 에서 7의 범위인 것으로 나타났다.

Effects of Different Additives on Fermentation Characteristics and Protein Degradation of Green Tea Grounds Silage

  • This study evaluated the fermentation characteristics and protein degradation dynamics of wet green tea grounds (WGTG) silage. The WGTG was ensiled with distilled water (control), or lactic acid bacteria (LAB), enzyme (E), formic acid (FA) and formaldehyde (FD) prior to ensiling. Three bag silos for each treatment were randomly opened at 0, 3, 7, 14, 28 and 60 days after anaerobic storage. For all the treatments, except for FA, there was a rapid decline in pH during the first 7 days of ensiling. LAB treatment had higher lactic acid content, lower ammonia-N ($NH_3$-N) and free-amino nitrogen (FAA-N) contents than other treatments (p<0.05). E treatment had higher lactic acid, water-soluble carbohydrates (WSC) and non-protein nitrogen (NPN) content than the control (p<0.05). FA treatment had higher $NH_3$-N and FAA-N content than the control (p<0.05). FD treatment had lower NPN and FAA-N content than the control, but it did not significantly inhibit the protein degradation when compared to LAB treatment (p>0.05). Results indicate that LAB treatment had the best effect on the fermentation characteristics and protein degradation of WGTG silage.

열처리 대두에서 분리한 대두 단백질의 기능성 (Functional Properties of Soy Protein Isolate from Heat Treated Soybean)

  • 대두 단백질의 기능성 변형 방법의 하나로 대두를 열처리 함에 의해 대두 단백질의 기능 특성에 어떠한 변화가 있는지 알아보기 위하여 대두를 $60^{\circ}C$에서 30, 60, 90과 120분 침지한 후 대두 분리 단백(SPI)을 제조하여 열처리하지 않은 대조군과 여러 가지 기능성을 비교하였다. 용해도 특성은 pH 4.5의 등전점 부근에서 최소의 용해도를 보였고 등전점 이하와 이상에서는 비교적 90% 넘는 높은 용해도를 나타내었다. 만리와 태광에서 모두 열처리 시간이 증가함에 따라 등전점 이하의 pH에서 용해도가 증가하였다. 수분흡수력도 열처리 시간이 증가함에 따라 증가하였고 만리의 경우 90분 시료에서 가장 높은 수분흡수력을 나타내었다. 유지흡수력은 만리의 경우 열처리 시간이 증가함에 따라 감소하였고 태광의 경우에는 60분 시료에서 최대값을 나타내었다. 유화 특성은 만리의 경우 열처리에 의해 유화활성도, 유화안정성과 유화형성력이 모두 대조군보다 유의하게 증가하였고, 태광의 경우에는 열처리에 의해 유화활성도와 유화형성력이 유의적으로 증가하였다. 만리의 경우 기포 팽창력과 기포안정성이 열처리에 의해 증가하였고, 만리와 태광 모두 기포팽창력이 90분 시료에서 최대값을 보였으며, 태광은 열처리에 의해 매우 불안정한 기포 안정성을 보였다. 겔의 TPA결과, 열처리 시간이 증가함에 따라 경도, 부착성, 탄성, 검성 및 씹힘성이 증가하였고 특히 모든 특성이 대체로 90분 시료에서 최고값을 보였다. 그러나 응집성은 열처리 시간이 증가함에 따라 감소하였다. 이상의 결과로 대두의 열처리에 의해 대두 단백질의 기능성이 변화하는 것을 알 수 있었고. 대두를 첨가물이나 식품가공재료로 사용할 때 이러한 변화를 고려하여 유용하게 활용할 수 있으리라 생각된다.

난백 첨가가 두부 품질에 미치는 영향 (Effects of Egg-White Addition on the Quality of Soybean Curd)

  • 본 연구에서는 단백질 효율이 보다 높은 두부를 만들고저 함황 아미노산이 비교적 많이 함유된 것으로 알려진 난백을 두유에 각각 혼합하여 만든 혼합물에 대한 열응고성과 응고제에 대한 응고성, 성분함량 변화, 수율, 경도 및 관능평가를 실시하였다. 수분과당 및 지방 함량은 두유만으로 제조한 두부보다 난백 첨가량이 증가함에 따라 감소하였고 단백질과 회분의 함량은 난백의 양이 증가함에 따라 증가되었다. 난백의 첨가는 두유의 응고 온도를 하강시키는 효과가 있었다. 무게와 부피 및 경도는 난백의 첨가량이 증가함에 따라서 증가 되었다. $Ca^{2+}$함량은 난백첨가량에 비례하여 감소한데 반하여 $Mg^{2+},\;K^{+},\;Na^{+}$은 증가되었다. 관능평가에서 색감과 풍미는 80%까지 첨가해도 대조구에 대하여 유의차(p<0.01)가 없었으나, 조직감과 식감에서는 난백을 60%이상 첨가한 경우 평점이 낮아지는 유의차(p<0.01)를 나타냈다. 아미노산 조성은 20, 40, 60, 80%(v/v)의 난백첨가로 함황아미노산(cys+met) 함량은 0.63, 1.20, 1.76, 2.36배의 강화효과를 나타냈다.

Genetic Diversity and Clustering of the Rhoptry Associated Protein-1 of Plasmodium knowlesi from Peninsular Malaysia and Malaysian Borneo

  • Human infection with simian malaria Plasmodium knowlesi is a cause for concern in Southeast Asian countries, especially in Malaysia. A previous study on Peninsular Malaysia P. knowlesi rhoptry associated protein-1 (PkRAP1) gene has discovered the existence of dimorphism. In this study, genetic analysis of PkRAP1 in a larger number of P. knowlesi samples from Malaysian Borneo was conducted. The PkRAP1 of these P. knowlesi isolates was PCR-amplified and sequenced. The newly obtained PkRAP1 gene sequences (n=34) were combined with those from the previous study (n=26) and analysed for polymorphism and natural selection. Sequence analysis revealed a higher genetic diversity of PkRAP1 compared to the previous study. Exon II of the gene had higher diversity (π=0.0172) than exon I (π=0.0128). The diversity of the total coding region (π=0.0167) was much higher than those of RAP1 orthologues such as PfRAP-1 (π=0.0041) and PvRAP1 (π=0.00088). Z-test results indicated that the gene was under purifying selection. Phylogenetic tree and haplotype network showed distinct clustering of Peninsular Malaysia and Malaysian Borneo PkRAP1 haplotypes. This geographical-based clustering of PkRAP1 haplotypes provides further evidence of the dimorphism of the gene and possible existence of 2 distinct P. knowlesi lineages in Malaysia.

단백질 섭취 수준에 따른 고양이의 혈액 및 조직의 유리 아미노산 농도의 변화 (Changes in Blood and Tissue Free Amino Acid Concentrations in Cats Adapted to Low-and High-protein Diets)

  • Changes in free amino acid concentrations is blood and various tissues were evaluated in cats adapted to the low-protein diet(20% protein, LPD) or the high-protein diet(60% protein, HPD) for 5 weeks. Cumulative body weigth gain for the 5 week period was 463$\pm$43g, and -128$\pm$40g for cats fed HPD and LPD, respectively. Feeding HPD significantly increased the size of liver and kidney. Cats adapted to HPD for 5 weeks have significantly elevated plasma concrntrations of essential amino acids (branched-chain amino acides, threonine, trytophan, phenylalanine and methoionine), whereas plasma levels of non-essential amino acids(alanine, asparagine, glycine, glutamine and serine) were significantly reduced in animals adapted to HPD(p<0.01, or p<0.001) compared to the values for the cats fed LPD. Changes in free amino acid concentratioks in whole blood induced by the variations in dietary level of protein closely reflect the pattern seen in plasma. Amino acids such as branched-chain amino acids, proline and threonine were most difficult to maintain homeostasis and consistantly elevated in lever, kidney, skeletal muscle and brain, as well as in blood of cats adapted to HPD(p<0.01 or p<0.001). All of the free amino acids in jejunum, excluding taurine and ornithine, were significantly elevated in animals adapted to HPD, most probably due to the rapid absorption of large amount of amino acids across the epithelium of small intestine.
