Ginsenoside (20S)Rg3 Ameliorates Synaptic and Memory Deficits in an Animal Model of Alzheimer's Disease

  • The amyloid ${\beta}$-peptide ($A{\beta}$), which originates from the proteolytic cleavage of amyloid precursor protein (APP), plays a central role in the pathogenesis of Alzheimer's disease (AD). Mounting evidence indicates that different species of $A{\beta}$, such as $A{\beta}$ oligomers and fibrils, may contribute to AD pathogenesis via distinct mechanisms at different stages of the disease. Importantly, elevation and accumulation of soluble $A{\beta}$ oligomers closely correlate with cognitive decline and/or disease progression in animal models of AD. In agreement with these studies, oligomers of $A{\beta}$ have been shown to directly affect synaptic plasticity, a neuronal process that is known to be essential for memory formation. Our previous studies showed that $A{\beta}$ induces the breakdown of phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2), a phospholipid that regulates key aspects of neuronal function. PI(4,5)P2 breakdown was found to be a key step toward synaptic and memory dysfunction in a mouse model of AD. To this end, we seek to identify small molecules that could elevate the levels of PI(4,5)P2 and subsequently block $A{\beta}$ oligomer-induced breakdown of PI(4,5)P2 and synaptic dysfunction.. We found that (20S)Rg3, an active triterpene glycoside from heat-processed ginseng, serves as an agonist for phosphatidylinositol 4-kinase IIalpha (PI4KIIalpha), which is a lipid kinase that mediates a rate-limiting step in PI(4,5)P2 synthesis. Consequently, (20S)Rg3 stimulates PI(4,5)P2 synthesis by directly stimulating the activity of PI4KIIalpha. Interestingly, treatment of a mouse model of AD with (20S)Rg3 leads to reversal of memory deficits. Our data suggest that the PI(4,5)P2-promoting effects of (20S)Rg3 may help mitigate the cognitive symptoms associated with AD.

Effects of Antiseptics on the Protein Change of Plasma Membrane in various Fungal Cells (진균류의 원형질막 단백질 변화에 미치는 식품보존제의 영향)

  • Plasma membrane from the fungal cells (Aspergillus phoenicis, Rhizopus acidus, Candida albicans) treated with sodium benzoate (S.B), potassium sorbate (P.S) and calcium propionate (C.P) during the cultivation were separated. The contents and patterns of plasma membrane proteins compared with those of the control. The growth of A. phoenicis was decreased by the average 64.0% in the S.B treatment. That of R. acidus was inhibited by the average 69.0% in the P.S treatment. Also, That of C. albicans was showed the deminution of the average 59.5% in the S.B treatment. The contents of protein involved in the plasma membrane of the each fungal cells were inhibited the average 41.0%, 41.7% and 59.5% in the S.B treatment, respectively. In case of A. phoenicis, the changes in the protein pattern involved in the plasma membraneshowed the aspect similar to the control on the 1st day and 2nd day of cultivation in the treatment group, but $116\;KD{\sim}97\;KD$ band almost disappeared in the 5th day of cultivation, and $45\;KD{\sim}29\;KD$ band was uncleared through the cultivation. In S.B treatment group R. acidus was showed the loss of $116\;KD{\sim}97\;KD$ band from the middle stage of cultivation and P.S, and C.P treatment group were started the loss at the early stage and completely lost at the 36 hours of cultivation. In C. albicans, $116\;KD{\sim}97\;KD$ band were started the loss at the early stage to compare with the control and $66\;KD{\sim}45\;KD$ band were dimmed at the 96 hours of cultivation. Especially, the C.P treatment group were perfectly lost at the 96 hours of cultivation.

  • Eight new compounds were prepared by the addition reaction of thioglycolic acid to ${\beta}$-nitrostyrene and its derivatives. s-[2-Nitro-1-phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-methyl)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-methoxy)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-chloro)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-bromo)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-nitro)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(3-methoxy-4-ethoxy)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(3,4,5-trimethoxy)phenylethyl]-thioglycolic acid; The structure of these compound were identified by elemental analysis, UV, IR and NMR spectral data.

Effects of Culture Condition on Solubilization of Coal by Microorganisms (배양 조건의 변화가 미생물에 의한 석탄의 액화에 미치는 영향)

  • Biosolubilization of an Australian lignite was investigated by using Streptomyces viridosporus and Poria cocos. In order to solubilize coals effectively they were pretreated by nitric acid both in surface and liquid cultures. The optimum growth pH was 7.5 for S. viridosporus and 4.5 for P. cocos. The effects of various carbon, nitrogen and metal sources on overall solubilization were also studied. Solubility increased with the addition of urea for S. viridosporus, and peptone and tryptone for P. cocos. However carbon and metal sources had little or negative effects on solubilization. Maximum amount of coal solubilized was 85%(w/w) in a batch fermentation culture. Extracellular materials produced by micro-organism were found to be responsible for the coal solubilization. Approximately 70 to 80% of coal solubilization was determined to be the result of non-enzymatic reactions, and the rest to be the result of enzymatic reactions. Characteristics of the solubilized coal were compared with those of original coal and pretreated coal by the approximate and ultimate composition analysis, and IR-spectrum analysis. The spectroscopic results showed that the mechanism of coal solubilization was caused by continuous oxidation.

  • This study is to analyze factors affecting the school nurse's activities. The survey was undertaken during Sept. 1-Nov. 30, 1986. Variations of pH, EC and anion of stemflow and throughfall in Quercus mongolica and Q. variabilis (신갈나무와 굴참나무 수간류, 임내우의 pH, 전기전도도 및 음이온 변화)

  • The pH, EC and anion of stemflow in Quercus mongolica and Q. variabilis were surveyed and analyzed in order to examine the relationship between watershed conservation function and flood control function of forest in quality and quantity in the Experiment Forests. College of Forest Sciences, Kangwon National University. The results were as follows: 1. pH values of rainfall ranged from 4.47 to 6.55(average: 5.39), and pH values of throughfall ranged from 4.07 to 6.25(average 5.45) for Q. mongolica and from 4.34 to 6.57(average : 5.62) for Q. variabilis, and thus pH values were not different between these two species. Also, pH values of stemflow from Q. mongolica ranged from 4.08 to 6.13(average 5.17) and those of stemflow from Q. variabilis ranged from 3.62 to 6.11(average : 4.68), and pH values of rainfall gave little influence on pH values of stemflow. But, pH values of stemflow in Q. mongolica and Q. variabilis appeard significantly lower in spring and than those in summer and autumn. 2. EC of rainfall was $3.0{\sim}62.6{\mu}s/cm$(average: $18.8{\mu}s/cm$), and EC of throughfall was $5.4{\sim}85.0{\mu}s/cm$(average : $25.1{\mu}s/cm$) for Q. mongolica and $5.0{\sim}253.0{\mu}s/cm$(average : $31.2{\mu}s/cm$) for Q. variabilis. Also, EC of stemflow from Q. mongolica ranged from 9.5 to $500.0{\mu}s/cm$(average : $81.8{\mu}s/cm$) and that of stemflow from Q. variabilis ranged from 11.5 to $534.5{\mu}s/cm$(average : $80.2{\mu}s/cm$). Seasonal EC of rainfall had little variation in the range of 20 to $30{\mu}s/cm$: EC of stemflow showed more than $100{\mu}s/cm$ from March to April and about $30{\mu}s/cm$ in summer period. Seasonal EC of stemflow varied so much and appeared high again from October to November. 3. $Cl^-$, $NO_3{^-}$ and $SO_4{^{2- }}$ concentrations of rainfall and throughfall were from 1 to 15ppm. and $PO_4{^{2- }}$ concentrations showed 0.57ppm and 0.23ppm in rainfall, 0.08ppm in Q. mongolica and 0.14ppm, 0.12ppm and 1.19ppm in Q. variabilis. Also, $Cl^-$, $NO_3{^-}$ and $SO_4{^{2-}}$ concentrations of stemflow were relatively higher than rainfall, and showed differences among seasons. $PO_4{^{2-}}$ concentration of rainfall and throughfall were not possible to observe, but $PO_4{^{2-}}$ concentrations of stemflow ranged from 0.08 to 31.99ppm(average : 3.22ppm) for Q. mongolica and that of stemflow ranged from 0.06 to 12.28ppm(average : 1.93ppm) for Q. variabilis.

Antimutagenic and Antibacterial Activities of Korean and American Propolis (한국산과 미국산 프로폴리스의 항돌연변이 및 항균효과)

  • The antimutagenic activities of ethanol extracts of Korean and American propolis were tested using Salmonella Typhimurium TA98 with two indirect mutagens of 3-amino-1,4-dimethyl-5H-pyrido [4,3-b]indole (Trp-P-1) and 2-aminoanthracene (2-AA) with S9 mix. Additionally, their antimicrobial activities against acne-related pathogenic strains of Propionibacterium acnes, Staphylococcus Epidermidis, Staphylococcus aureus and Pseudomonas aeruginosa were evaluated using both paper disk method and agar dilution method. Ethanol extracts of Korean and American propolis showed strong inhibitory effects, in a dose dependant manner, against the mutagenicities induced by Trp-P-1 and 2-AA. The antimutagenic effect of ethanol extracts of Korean propolis showed significantly higher protective activity than that of American propolis against the Trp-P-1 induced mutagenicity of S. Typhimurium TA98 at the lower concentration ($1-10\;{\mu}g$), but significantly lower protective activity at the higher concentration ($50-200\;{\mu}g$). The antimutagenic effect of ethanol extract of Korean propolis showed significantly higher protective activity than that of American propolis against the 2-AA induced mutagenicity at the concentration of $1\;{\mu}g$, but significantly lower protective activity than that of the American at the higher concentration ($50-200\;{\mu}g$). Both extracts showed strong antimicrobial activities against all the acne-related pathogens tested, with minimal inhibitory concentration (MIC) values in the range $1,500-5,000\;{\mu}g/mL$.

Characterization of an Antibacterial Substance Produced by Bacillus subtilis HS 25 Isolated from Fermented Soybean Foods (대두 발효식품으로부터 분리한 Bacillus subtilis HS 25가 생산한 항균물질의 특성)

  • We investigated an antibacterial substance produced by Bacillus subtilis HS 25. Antibacterial activity was relatively heat-stable, with no effect caused by heating at $100^{\circ}C$ for 10 min, but a gradual decrease in activity after 15 min at $100^{\circ}C$. The antibacterial substance was more stable at pH 7.42-12 than at pH 4.5-5.0. There was strong antibacterial activity against E. coli and P. mirabilis at pH 12. The minimum inhibition concentration (MIC) of the substance was 5 mg/mL for E. coli 7.5 mg/mL for P. mirabilis and S. aureus and 15 mg/mL for S. enteritidis, K. pneunoniae, and V. parahaemolyticus. When the substance was added to cultures of E. coli, S. enteritidis, and P. mirabilis, the bacterial surfaces became irregular and deeply changed. The substance produced from B. subtilis HS 25 was not degraded by Papain but was degraded by a protease from Aspergillus orzae, pancreatin, and pepsin.

Effect of Phytate on the Solubility and Digestibility of Rapeseed Protein Isolate (분리 유채단백의 용해도와 소화율에 미치는 Phytate의 영향)

  • This study was undertaken to investigate the effects of added phytate and pH on the solubility and in vitro digestibility of low-phytate rapeseed protein isolate. Phytate content of low-phytate rapeseed protein isolate was 1.5%, as a result of 66% removal from defatted rapeseed flour and the protein: phytate ratio was 58:1. Solubility of rapeseed protein isolate at pH 2.0 and pH 11.5 was much higher than near the isoelectric point, pH 5.0. It's solubility was lowered by adding an increased amount of phytate especially at pH 2.0. The inhibitory effect of phytate toward pepsin digestibility of rapeseed protein isolate decreased by the increasing amount of phytate added. It is suggested that the production of low-phytate rapeseed protein isolate is necessary to improve the functionality and nutritional value in order to utilize it as food material.

Study on Consumption Patterns and Degree of Checking Food-Nutrition Labeling of Convenience Foods at Convenience Stores by Young Adult Workers in Busan (부산 거주 20대·30대 직장인의 편의점 편의식 이용 및 식품영양표시 확인 정도)

  • The purpose of this study was to investigate convenience foods at convenience stores, by examining consumption patterns, as well as the degree of checking food-nutrition labeling. In this study, 208 workers in Busan in their 20s or 30s were surveyed using 1:1 interview questionnaires. The weekly usage frequencies of cup-ramyeon, gimbap, ramyeon, and sandwiches were 41.0%, 39.4%, 32.7%, and 19.0% respectively. The usage frequency of convenience foods was 12.0% for 'over 5 times/week', 23.0% for '3~4 times/week', 31.7% for '1~2 times/week', 29.8% for '1~2 times/month', and 3.4% for zero usage. Workers in their 20s showed a significantly (P<0.05) higher rate in the '3~4 times/week' as well as '1~2 times/week' intervals than workers in their 30s. Single people showed a significantly (P<0.01) higher rate than married people. Workers in their 20s showed a significantly higher score for the '3~4 times/week' as well as '1~2 times/week' intervals (P<0.05). The mean score of checking food labeling was 4.02/5.00 for the manufacturer's date, or expiration date, 3.23/5.00 for weight or volume, 2.99/5.00 for cooking method, and 2.85/5.00 for storage method. Females showed significantly (P<0.01) higher rates for checking the manufacturer's date or expiration date, as well as source of the raw material. Single workers showed significantly (P<0.01) higher rates for checking the manufacture's or expiration date as well as storage method. The mean score for checking nutrition labeling was 3.06/5.00 for calories, 2.84/5.00 for trans fat, 2.80/5.00 for sodium, and 2.76/5.00 for cholesterol. Females showed a significantly (P<0.01) higher rate for six of 10 items. Workers in their 20s showed a significantly (P<0.05) higher rate for four of 10 items. Expert officials showed significantly higher rate for six of 10 items (P<0.05) and two of 10 items (P<0.01), respectively.
