Optimal Surface Aeration Rate for Bioethanol Production from the Hydrolysate of Seaweed Sargassum sagamianum Using Pichia stipitis (Pichia stipitis를 이용한 모자반 가수분해물로부터의 bioethanol 생산 시 최적 surface aeration rate)

  • We investigated the optimal surface aeration rate during bioethanol production from the hydrolysate of seaweed Sargassum sagamianum using Pichia stipitis. It was observed that, when the working volume was 880 mL in 2.5-L lab-fermentor, the surface aeration rates of 30 to 100 mL/min were the optimal values for bioethanol production, in which this surface aeration rate corresponded to less than 0.05 (1/min) as the oxygen transfer rate coefficient ($k_La$). In addition, during repeated-batch operation was carried out, we examined whether those surface aeration rates were the optimal for bioethanol production. It was also observed that the surface aeration rates of 30 to 100 mL/min in the working volume of 880 mL were the optimal values in terms of the cumulative bioethanol producrion and bioethanol yield. On the basis of the oxygen transfer rate coefficient it is probable that those surface aeration rates will be applied to the large-scale bioethanol production from the hydrolysate of seaweed Sargassum sagamianum.

  • Ignition of hydrogen and oxygen in the "third limit" is theoretically investigated in the stagnation point flow with activation energy asymptotics. With the steady-state approximations of H, OH, O and HO$_2$, a two-step reduced kinetic mechanism is derived for the regime lower than the crossover temperature T$_{c}$ at which the rates of production and consumption of all radicals are equal. Appropriate scaling of Damkohler number successfully provides the explicit relationship between pressure, temperature and strain rate at ignition. It is shown that, compared with those for the counterflow, ignition temperatures for the stagnation point flow are considerably increased with increasing the system pressure. This is because ignition in the "third limit" is characterized by the production of reduction of $H_2O$$_2$, which is reduced by wall effect. Strain rate substantially affects ignition temperature because key reaction rates of $H_2O$$_2$ are comparably with its transport rate, while the mixture temperature and the hydrogen composition do not significantly affect ignition temperature.e.

  • The purpose of this paper is to investigate the reforming characteristics and maximum operating condition for the hydrogen production by methane reforming using the compression ignition engine induced partial oxidation. An dedicated compression engine used for methane reforming was decided operating range. The partial oxidation reforming was investigated with oxygen enrichment which can improve hydrogen production, compared to general reforming. Parametric screening studies were achieved as $O_2/CH_4$ ratio, total flow rate, and intake temperature. When the variations of $O_2/CH_4$ ratio, total flow rate, and intake temperature were 1.24, 208.4 L/min, and $400^{\circ}C$, respectively, the maximum operating conditions were produced hydrogen and carbon monoxide. Under the condition mentioned above, synthetic gas were $H_2\;22.77{\sim}29.22%,\;CO\;21.11{\sim}23.59%$.

Citric Acid Production Using Encapsulated Aspergillus niger (Aspergillus niger 고정화 캡슐을 이용한 구연산 생산 특성)

  • The encapsulatpd A. niger grew up inside the capsule and mycelia penetrated through the pore of the capsule membrane. The mycelia on the capsule wall became loose when the carbon source and oxygen were deficient in the medium. On the contrary, the production rate increased and mycelia made a lump tightly when the carbon source and oxygen were sufficient. Namely, number of proper capsule of unit volume in the medium was existed. The phenomenon which was swelled of capsule membrane in cultivation could prevented by adding CaCl2 into the medium. According to the time adding CaCl2 into the medium, the production rate of citric acid was influenced. In case of adding CaCl2 into the medium at 7th day cultivation, the production yield of citric acid was increased about 40 percent higher than that of adding CaCl2 initially. The production yield of citric acid using encapsulated A. niger of flask culture was influenced with oxygen supply. The production yield of citric acid ($\Delta$p/$\Delta$s) of the flask culture was increased 3.88 time by using T-flask instead of parafilm sealed flask. Therefore, the productivity and consumption rate concerning production which was taken carbon source were increased when oxygen supply was sufficient. The production of citric acid using encapsulated A. niger was increased average 30 percent higher than that of bead in between 6th and 13th day cultivation.

  • Numerical Study with detailed chemistry has been conducted to investigate the flame structure and NOx formation characteristics in oxygen -enhanced(CH$_4$/O$_2$-$N_2$) and oxygen-enhanced-EGR(CH$_4$/O$_2$-$CO_2$) counter diffusion flame with various strain rates. A small amount of $N_2$is included in oxygen-enhanced-EGR combustion, in order to consider the inevitable $N_2$contamination by $O_2$production process or air infiltration. The results are as follows : In CH$_4$/O$_2$-$CO_2$flame it is very important to adopt a radiation effect precisely because the effect of radiation changes flame structure significantly. In CH$_4$/O$_2$-$N_2$flame special strategy to minimize NO emission is needed because it is very sensitive to a small amount of $N_2$. Special attention is needed on CO emission by flame quenching, because of increased CO concentration. Spatial NO production rate of oxygen-enhanced combustion is different from that of air and oxygen-enhanced-EGR combustion in that thermal mechanism plays a role of destruction as well as production. In case CH$_4$/O$_2$-$CO_2$flame contains more than 40% $CO_2$it is possible to maintain the same EINO as that of CH$_4$/Air flame with accomplishing higher temperature than that of CH$_4$/Air flame. EINO decreases with increasing strain rate, and those effects are augmented in CH$_4$/O$_2$flame.

  • A dense mixed ionic and electronic conducting ceramic membrane is one of the most promising materials because it can be used for separation of oxygen from the mixture gas. The $ABO_3$ perovskite structure shows high chemical stability at high temperatures under reduction and oxidation atmospheres. $BaCo_{1-x-y}Fe_xZr_yO_{3-{\delta}}$ (BCFZ) was well-known material as high mechanical strength, low thermal conductivity and stability in the high valence state. Glycine Nitrate Process (GNP) is rapid and effective method for powder synthesis using glycine as a fuel and show higher product crystallinity compared to solid state reaction and citrate-EDTA method. BCFZ was fabricated by modified glycine nitrate process. In order to control the burn-up reaction, $NH_4NO_3$ was used as extra nitrate. According to X-Ray Diffraction (XRD) results, BCFZ was single phase regardless of Zr dopants from y=0.1 to 0.3 on B sites. The green compacts were sintered at $1200^{\circ}C$ for 2 hours. Oxygen permeability, methane partial oxidation rate and hydrogen production ability of the membranes were characterized by using Micro Gas Chromatography (Micro GC) under various condition. The high oxygen permeation flux of BCFZ 1-451 was about $1ml{\cdot}cm^{-2}s^{-1}$. Using the humidified Argon gas, BCFZ 1-433 produced hydrogen about $1ml{\cdot}cm^{-2}s^{-1}$.

Exopolysaccharide Production by Aureobasidium pullulans - Appearance of Melanin Pigment - (Aureobasidium pullulans 에 의한 Exopolysaccharide 생산 - 멜라닌 색소의 출현에 관한 연구 -)

  • In exopolysaccharide fermentation by Aureobasidium pulluans, the effects culture conditions (concentration of nitrogen, potassium phosphate, dissolved oxygen, and initial pH) on the production of exopolysaccharide and the appearance of melanin pigment were investigated. The results are as follows. (1) The specific growth rate and the specific production rate of exopolysaccharide were inhibited by substrate when the carbon source concentration higher than $50g\;/\;{\ell}$ and the cell growth increased with increases of nitrogen source but exopolysaccharide production decreased with the nitrogen concentration when it become greater than $1\;g\;/\;{\ell}$. (2) The maximum cell growth and the maximum exopolysaccharide production were obtained at initial pH values of 3.0 and 7.5 respectively. As the initial pH increased, the yeast-like cells increased and the increased of dissolved oxygen increased the cell growth and exopolysaccharide production. (3) As the concentration of dissolved oxygen is increased or the concentration of nitrogen source is decreased, the period of melanin pigment appearance becomes shorter and the melanin pigment never appeared when the initial pH was less than 3.0 or the potassium phosphate was not added.

Opitimization of culture conditions involved in dissolved oxygen for production of pullulan by Aureosidium pullulan

  • Effect of carbon source and culture conditions involved in the concentration of dissolved oxygen on cell growth and the production of pullulan by A. pullulans HP2001 were investigated. Among those carbon sources, glucose was found to be the best carbon source for the production of pullulan by A. pullulans HP2001. Maximal production of pullulan by A. pullulans HP2001 was 26.6 g/ f when concentrations of glucose and yeast extract were 8% (w/v) and 0.25% (w/v), respectively. It was found that aeration rate, agitation speed and inner pressure of a bioreactor, which were some of physiological factors involved in the dissolved oxygen in the medium may affect cell growth and the production of pullulan by A. pullulans HP2001.

A study on the inflammatory response induced by LPS of the Arthrospira platensis ethanol extract

  • Arthrospira platensis has been reported to contain a variety of substances such as phycocyanin, ${\beta}$-carotene, vitamin E and other carotenoids. In this study, zebrafish were treated with indoor cultivation spirulina ethanol extracts(ICAE) to determine toxicity(coagulation rate, hatching rate, heart rate). We used the DCFH-DA staining method to detect the effect of reactive oxygen species(ROS) generation on lipopolysaccharide(LPS)-induced zebrafish embryos ROS various concentrations(0.01, 0.05, 0.1, 0.5mg/ml) of ICAE. Cell toxicity was measured by WST-1 assay on RAW 264.7 macrophage cell line. Also, measured the inhibitory effect of nitric oxide(NO) and prostaglandin $E_2$ ($PGE_2$) production in RAW264.7 macrophages induced by LPS at various concentrations of ICAE. The results of embryo coagulation rate, hatching rate, heart rate of zebrafish at various(0.01, 0.05, 0.1mg/ml) of ICAE was no toxicity. The ICAE treated group had an inhibitory effect on NO and $PGE_2$ production compared and decreased with concentration. The results of this study ethanol extract of Arthrospira platensis has an anti-inflammatory effect and suggest that is worthy of use cosmetics for skin protection.

  • Sedimentary organic matter, exposed to continental surficial environment, reacts with oxygen supplied from the atmosphee and forms carbon-containing oxidation products. Knowledge of the rate and mechanisms of sedimentary organic matter weathering is important because it is one of the major controls on atmospheric oxygen level through geologic time. Under the abiological conditions, the oxidation rate of coal organic matter by molecular oxygen is enhanced by the increase of oxygen concentration and temperature. At ambient temperature and pressure, aqueous coal oxidation results in the formation of dissolved $CO_2$ dissolved organic carbon and solid oxidation products which are all quantitatively significant reaction products. The effects of pH, ultraviolet light, and microbial activity on the weathering of sedimentary organic matter are poorly contrained. Based on the results of geochmical and environmental studies, it is believed that the photochemical reaction should play an important role in the decomposition and oxidation of sedimentary organic matter removed from the weathering profile. At higher pH conditions, the production rate of DOC can be accelerated due to base catalysis. These high molecular weight oranic matter can react with man-made pollutants such as heavy metal ions via adsorption/desorption or ion exchange reactions. The effect of microbial activity on the oxidative weathering of sedimentary organic matter is poorly understood and remains to be studied.
