Inhibitory Effects on Melanogensis of Scrophularia koraiensis Nakai in Melanocytes

  • Scrophularia koraiensis Nakai (S. koraiensis) has used its roots as traditional herbal medicine. Some research is reported to be effective in allergic inflammation and osteoporosis. In a present study, we conducted to investigate the bioactivity of the ethanol extract of S. koraiensis (ESK) on the inhibition of melanogenesis and the apoptosis of melanocytes. We analyzed Harpagoside of ESK by using LCMS and HPLC-PDA and investigated the regulation of ESK on reactive oxygen species. Also, the expressions of melanin synthesis-related factors and apoptosis-related factors were confirmed. As a result, the quantification results of quercetin and rutin in ESK were 77.2 and 7.4 mg/g. IC50 on DPPH and ABTS radical scavenging activity is 33.1 and 9.5 ug/mL. ESK attenuated not only the expression of tyrosinase, TYRP-1, TYRP-2, and MITF in melanogenesis. It is thought that ESK may be effective in the inhibition of melanogenesis through MAPK cell signaling pathway in melanocytes. These study results suggest that ESK has the ability to inhibit melanin production and induce apoptosis.

Protective Effects of Antoxidant Enzymes of Candida albicans against Oxidative Killing by Macrophages

  • Protective roles of antioxidant enzymes, copper-zinc superoxide dismutase (CuZnSOD), manganese superoxide dismutase (MnSOD), and catalase of Candida albicans against exogenous reactive oxygens and oxidative killing by macrophages were investigated. The initial growth of C. albicans was inhibited by reactive, oxygen-producing chemicals such as hydrogen peroxide, pyrogallol, and paraquat, but it was restored as the production of antioxidant enzymes were increased. The growth inhibition of C. albicans by reactive, oxygen-producing chemicals was reduced by treating the purified candidal SOD and catalase. Also, in the presence of SOD and catalase, the oxidative killing of C. albicans by macrophages was significantly inhibited. These results suggest that antioxidant enzymes, CuZnSOD, MnSOD, and catalase of C. albicans may play important roles in the protection of C. albicans not only from exogenous oxidative stress but also from oxidative killing by macrophages.

Effect of Trolox C on Hypoxia/Reoxygenation-Induced Injury in Isolated Perfused Rat Liver

  • Livers isolated from 18 hours fasted rats were subjected to N$_{2}$ hypoxia (for 45 min) followed by reoxygenation (for 45 min). The perfusion medium used was Krebs-Henseleit bicarbonate buffer (KHBB, pH 7.4). Lactate and alanine were added as gluconeogenic and ureagenic substrates and Trolox C was also added to perfusate. Oxygen consumption, lactate dehydrogenase (LDH), alanine transaminase (ALT), total glutathione, oxidized glutathione, bile flow, glucose and urea were measured. After hypoxia oxygen consumption significantly dropped but Trolox C had no influence on this decrease. ALT and LDH were significantly increased by hypoxia/reoxygenation. This increase was markedly attenuated in the presence of Trolox C. The total glutathione and oxidized glutathione efflux increased following hypoxia, which were prevented by the treatment of Trolox C. Bile flow rate decreased following hypoxia/reoxygenation but did not continue to decrease in the reoxygenation phase by Trolox C. Following hypoxia/reoxygenation glucose and urea releases decreased. Trolox C had no influence on inhibition of glucose and urea production. These results suggest that Trolox C protected the liver cells against hypoxia/reoxygenation injury, yielding further evidence for a causative role of oxidative stress in this model.

팔물탕의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과 (Antioxidant and Protective Effects of Palmul-tang on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes)

  • Objective : In this paper, we investigated the anti-oxidative capacities and protective effects of water extract of palmul-tang (PMT) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). Method : To evaluate the anti-oxidative activities of PMT, we measured scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of PMT. To give an oxidative stress to HaCaT cells, UVB was irradiated with $40mJ/cm^2$ to HaCaT cells. To detect the protective effects of PMT against UVB, we measured cell viability, apoptotic bodies and reactive oxygen species(ROS) generation. Results : PMT showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also PMT showed high reducing values. The UVB-induced oxidative conditions led to the cell apoptosis. However, treatment with PMT reduced oxidative stress conditions, including inhibition of cell apoptosis and expression of ROS. Conclusion : PMT had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. PMT would be useful for the development of cosmetics treating UVB-induced skin aging.

해조환(海藻丸)이 갑상선(甲狀腺) 기능항진증(機能亢進症)에서 항산화(抗酸化) 효과(效果)에 미치는 영향(影響) (Underlying mechanism of antioxidant action of Haejohwan in thyroxine-induced hyperthyroid rats)

  • This study was carried out to examine if Haejohwan (HJ) inhibits oxidant-induced lipid peroxidation and therby produces protective effect against thyroxine-induced hyperthyroid rats. Triiodothyronine $(T_3)$, thyroxine $(T_4)$, lipid peroxidation, xathine oxidase activities and type conversion ratio were increased in thyroxine treated group. However, they were decreased in HJ extract's pre-applied group. Glutathione level, activities of glutathione peroxidase, glutathione Stransferase and glutathione reductase were decreased in thyroxine treated group. But, they were increased in HJ extract's pre-applied group. These results suggest that in thyroxine-induced hyperthyroid rats HJhas an increase in the activities of oxygen free radical scavenging enzymes and inhibition of xanthine oxidase activities, and prevents lipid peroxidation.

천연자원의 라디칼 소거능과 최종당화산물의 생성저해활성 검색 (Screening of Natural Resources with Inhibitory Activity on Free Radicals and Advanced Glycation end Products (AGEs) Formation)

  • Reactive oxygen species (ROS) and formation of advanced glycation end products (AGEs) play an important role in the pathogenesis of diabetic nephropathy by hyperglycemia. To find natural agents improving diabetic nephropathy, 63 natural resources which used to the treatment of diabetes mellitus in a folk remedy were investigated with an in vitro system employing radical scavenging activity and inhibitory activity of AGEs formation. In results, the extracts of Aspalathus linearis, Rubus coreanus, Rosa rugosa, and Epimedium koreanum significantly inhibited the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical with $IC_{50}$ values less than $10{\mu}g/ml$. The extracts of Zea mays, Cucurbita moschata, Cudrania tricuspidata, and Aspalathus linearis effectively reduced the formation of AGEs compared with the positive control $N-acetyl-_L-cystenine$ (NAC) and aminoguanidine (AG). In addition, the extracts of Aspalathus linearis, Commelina communis, Cornus officinalis, and Lespodeza cuneata showed the all inhibitory activity against DPPH radical and AGEs formation. Also, these resources definitely showed the radical scavenging activity against peroxynitrite $(ONOO^-)$ and hydroxyl radical $({\cdot}OH)$ relating to high glucose-induced ROS production. Thus, these results suggest that some natural resources may regulate the initiation and progression of diabetic nephropathy through inhibition of ROS production and AGEs formation.

Anti-Inflammatory, Anti-Angiogenic and Anti-Nociceptive Activities of 4-Hydroxybenzaldehyde

  • The current work was designed to assess novel pharmacological activities of 4-hydroxybenzaldehyde (HD), a major phenolic constituent of various natural products of plant origin, such as Gastrodia elata Blume. HD exhibited a significant inhibition in the chick chorioallantoic membrane (CAM) angiogenesis. HD also displayed an inhibitory effect in acetic acid-induced permeability in mice. Anti-nociceptive activity of HD was convinced using the acetic acid-induced writhing test in mice. HD was able to suppress production of nitric oxide (NO) and induction of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in the lipopolysaccharide (LPS)-activated RAW264.7 macrophages. HD also diminished the reactive oxygen species (ROS) level elevated in the LPS-activated macrophages. In brief, HD exhibits anti-angiogenic, anti-inflammatory and anti-nociceptive activities possibly via down-regulating iNOS and/or COX-2, which may be partly responsible for pharmacological efficacies of various natural products.

  • Hydrogen sulfide is a colorless, and malodorous 'rotten eggs' gas that results from the decay of organic material. It is a byproduct of industry and agriculture. The mechanism of its toxicity is primarily related to inhibition of oxidative phosphorylation, which causes a decrease in available cellular energy. Because there is no rapid method of detection that is of clinical diagnostic use, management decisions must be made based on history, clinical presentation, and diagnostic tests that imply hydrogen sulfide's presence. Although there is some anecdotal evidence to suggest that the early use of hyperbaric oxygen is beneficial, supportive care remains the mainstay of therapy. We describe an occupational exposure to hydrogen sulfide gas in 51-year-old man. While cleaning the sewage of pigs. he became unconscious. When he arrived in the emergency department, he had irritability and confused mentality. The typical smell of rotten eggs on clothing and exhaled air were enough to be considered to be exposed to hydrogen sulfide. Hyperbaric oxygen therapy was performed. He had a recovery to normal function.

MERCURY-INDUCED ALTERATIONS OF CHLOROPHYLL a FLUORESCENCE KINETICS IN ISOLATED BARLEY (Hordeum vulgare L. cv. ALBORI) CHLOROPLASTS

  • Effects of HgCl$_2$-treatment on electron transport, chlorophyll a fluorescence and its quenching were studied using isolated barley (Hordeum vulgare L. cv. Albori) chloroplasts. Depending on the concentration of HgCI$_2$, photosynthetic oxygen-evolving activities of photosystem II (PS II) were greatly inhibited, whereas those of photosystem I (PS I) were slightly decreased. The inhibitory effects of HgCl$_2$ on the oxygen-evolving activity was partially restored by the addition of hydroxyamine, suggesting the primary inhibition site by HgCl$_2$2-treatment is close to the oxidizing site of PS tl associated with water-splitting complex. Addition of 50 $\mu$M HgCI$_2$ decreased both photochemical and nonphotochemical quenching of chlorophyll fluorescence. Especially, energy dependent quenching (qE) was completely disappeared by HgCl$_2$-treatment as observed by NH$_4$CI treatment. In the presence of HgCI$_2$, F'o level during illumination was also increased. These results suggest that pH gradient across thylakoid membrane can not be formed in the presence of 0 $\mu$M HgCl$_2$. In addition, antenna pigment composition might be altered by HgCl$_2$-treatment.

Effect of Phosphorus on the Cadmium Transfer and ROS-scavenging Capacity of Rice Seedlings

  • Environmental toxicity due to cadmium (Cd) pollution in croplands causes critical problems worldwide. Rice (Oryza sativa L.) is an important crop in Asia, including South Korea, and numerous studies have evaluated the relationship between Cd and antioxidants to alleviate Cd uptake from the soil into plants. However, information about the relationship between phosphorus (P) and antioxidants in rice seedlings is still limited with regard to Cd phytotoxicity. We therefore investigated the physiological responses of rice (Oryza sativa L. cv 'Dongjin') seedlings to Cd toxicity and the effect of P application on reactive oxygen species (ROS) and antioxidant changes. The exposure of rice seedlings to $30{\mu}M$ Cd inhibited plant growth; increased the contents of superoxide, hydrogen peroxide, and malondialdehyde; and induced Cd uptake by the roots and leaves. Application of P to Cd-exposed seedlings decreased Cd-induced oxidative stress by enhancing the capacity of ascorbate (AsA) production and ROS-scavenging, and decreased Cd transfer from the roots to the leaves. These results suggest that P application alleviated Cd-induced growth inhibition and oxidative damage by restricting Cd translocation from the roots to the leaves and maintaining sufficient levels of AsA.
