High-temperature Oxidation of CrZrN Films in Air (CrZrN 박막의 대기 중 고온산화)

  • Films of CrN, $Cr_{40}Zr_9N$, and $Cr_{31}Zr_{16}N$ were deposited on a steel substrate by closed field unbalanced magnetron sputtering, and their oxidation behaviors at $700^{\circ}C$ and $800^{\circ}C$ for up to 60h in air were investigated. All the deposited films were composed of the CrN phase. Zirconium atoms in $Cr_{40}Zr_9N$ and $Cr_{31}Zr_{16}N$ films partially dissolved in the CrN phase. They advantageously refined the columnar structure, reduced the surface roughness, and increased the microhardness. The CrN film displayed relatively good oxidation resistance, owing to the formation of the highly protective $Cr_2O_3$ on its surface. The $Cr_{40}Zr_9N$ and $Cr_{31}Zr_{16}N$ films oxidized to $Cr_2O_3$ as the major phase and ${\alpha}-ZrO_2$ as the minor one. They oxidized primarily by the inward transport of oxygen. The addition of Zr could not increase the oxidation resistance of the CrN film, because the formed $ZrO_2$ that was intermixed in the $Cr_2O_3$-rich oxide layer was oxygen permeable, and developed the compressive stress in the oxide scale owing to the volume expansion during its formation.

Effects of Combined Preparation (DWP715) Containing Alaska pollack Extract, Maltol, Ascorbic Acid and Nicotinamide on Decreasing of Blood Alcohol Concentration, Anti- fatigue and Anti-oxidation (북어엑스 및 말톨 함유 복합 조성물(DWP715)의 혈중 알콜농도 저하, 항피로 및 항산화 효과)

  • Effect of combined preparation (DWP715) containing Alaska pollack extract, maltol, ascorbic acid and nicotinamide on decreasing of blood alcohol was evaluated in human blood. Treatment of DWP715 prior to administration of 25% alcohol (100 mL) decreased alcohol concentration in blood and showed significant difference after 2 hours. The pharmacokinetic parameters such as area under the concentration-time curve (AUC), $C_{max},\;T_{max}\;and\;T_{1/2}$ were also decreased and delayed when compared with control values. Effects of DWP715 on anti-fatigue and anti-oxidation activities were also studied in the restraint stress model using various parameters (GOT, GPT, LDH values and organ weights) on mild condition and examined through the content of lipid peroxide induced by 2% $CCl_4$ in mouse livers. While GPT level, thymus and adrenal weight were not influenced by DWP715 dosing, LDH, GOT level and spleen weight used as a parameter against fatigue and stress states were recovered almost to the nomal level. Furthermore, lipid peroxidation due to $CCl_4$ was significantly inhibited by DWP715 treatment. These results suggest that DWP715 seems to metabolize the blood alcohol rapidly and to restore the damaged liver and fatigue conditions which was caused by alcohol metabolism to normal condition.

Effects of quercetin derivatives from mulberry leaves: Improved gene expression related hepatic lipid and glucose metabolism in short-term high-fat fed mice

  • BACKGROUND/OBJECTIVES: Mulberry leaves contain quercetin derivatives, which have the effects of reducing obesity and improving lipid and glucose metabolism in mice with obesity. It is not clear whether or not mulberry leaves can directly affect metabolic disorders, in the presence of obesity, because of the interaction between obesity and metabolic disorders. The aim of the current study was to assess the direct action of quercetin derivatives on metabolic disorders in non-obese conditions in short-term high-fat diet fed mice. MATERIALS/METHODS: C57BL/6N mice were fed a high-fat diet, supplemented with either 0% (control), 1%, or 3% mulberry leaf powder (Mul) or 1% catechin powder for five days. Anthropometric parameters and blood biochemistry were determined, and hepatic gene expression associated with lipid and glucose metabolism was analyzed. RESULTS: Body and white fat weights did not differ among the four groups. Plasma triglycerides, total cholesterol, and free fatty acids in the 1%, 3% Mul and catechin groups did not differ significantly from those of the controls, however, plasma glucose and 8-isoprostane levels were significantly reduced. Liver gene expression of gp91phox, a main component of NADPH oxidase, was significantly down-regulated, and PPAR-${\alpha}$, related to ${\beta}$-oxidation, was significantly up-regulated. FAS and GPAT, involved in lipid metabolism, were significantly down-regulated, and Ehhadh was significantly up-regulated. Glucose-metabolism related genes, L-PK and G6Pase, were significantly down-regulated, while GK was significantly up-regulated in the two Mul groups compared to the control group. CONCLUSIONS: Our results suggest that the Mul quercetin derivatives can directly improve lipid and glucose metabolism by reducing oxidative stress and enhancing ${\beta}$-oxidation. The 1% Mul and 1% catechin groups had similar levels of polyphenol compound intake ($0.4{\times}10^{-5}$ vs $0.4{\times}10^{-5}$ mole/5 days) and exhibited similar effects, but neither showed dose-dependent effects on lipid and glucose metabolism or oxidative stress.

Effect of Curcuma longa Hot Water Extract on Activity of Neuronal Cells Related to Oxidative Stress (산화적 스트레스와 관련하여 신경세포의 활성에 미치는 강황 열수추출물의 영향에 대한 연구)

  • The aim of this study is to screen a therapeutic agent with a cognitive function. The inhibitory effect of $Curcuma$ $longa$ hot water extract (CLWE) on the angiotension-converting enzyme and acetylcholinesterase derived from rabbit lungs and neural cells (PC12), as well as its antioxidant effect, was investigated in this study. Thus, for the first time, the direct scavenging effect of CLWE on DPPH radicals, superoxide anions, hydroxyl radicals, lipid peroxidation, reducing power, and the protective effect of DNA oxidation related to oxidative stress was evaluated in vitro. In addition, it was observed that CLWE especially exhibited a scavenging effect on reducing power and superoxide anions in this study. CLWE showed a protective effect on DNA oxidation produced by hydroxyl radicals. Furthermore, CLWE inhibited the activity of angiotensin-converting enzymes above 0.25%. Additionally, the extract inhibited oxidative stress and inducible nitric oxide in neuronal cells. Therefore, these results demonstrated that CLWE has antioxidant activity and neuronal cell protective effects, suggesting that it may have great potential as a natural source for human health.

The Analysis of the Physiologic Activities of the Jeju Teas according to the Fermentational Degree (제주산 차의 발효 정도에 따른 생리활성 기능에 관한 연구)

  • In this present study, we investigated the anti-oxidant activity, the inhibition ability of lipid peroxidation, and the protective effect of cow pulmonary epithelium (CPAE) cells under oxidative stress using green tea and 3 types of fermented teas of Jeju Island. To compare the physiological activity of non-fermented and 3 types of fermented teas, the fermented time was controlled with 0 hr. (non fermented tea, G), 12 hrs. (20% fermented tea, F20), 17 hrs. (50% fermented tea, F50) and 24 hrs. (80% fermented tea, F80), respectively. Scavenging ability on DPPH radicals of 80 ${\mu}g/mL$ concentration of F20 was similar to that of 50 ${\mu}M$ epigallocatechin gallate (EGCG) but it was stronger than those of G, F50 and F80. All extracts tested inhibited LDL oxidation but G and F20 inhibited LDL oxidation 25~30% more than F50 and F80 at 40 ${\mu}g/mL$ concentration which was similar to that of 50 ${\mu}M$ EGCG. We observed that the CPAE cells treated with the tea extracts had a significant increase in cell viability, especially the cells under oxidative stress with 1 mM $H_2O_2$ as compared with the control group (no treatment with tea extracts). These findings suggested that all tea extracts containing fermented tea had a protective effect on oxidative stressed CPAE cells through their free radical scavenging activity. It can be concluded that F20 extracted from 20% fermented tea has the most significant antioxidative effects that inhibit lipid peroxidation and protect the CPAE cells under oxidative stress.

Electrorheology of conducting polyaniline-$BaTiO_3$ composite

  • Organic-inorganic composite of polyaniline and barium titanate (PANI-$BaTiO_3$) was synthesized via an in-situ oxidation polymerization of aniline in the presence of barium titanate ($BaTiO_3$) nanoparticles dispersed in an acidic medium. Barium titanate has large electric resistance and relatively high dielectric constant which is one of the essential properties for its electrorheological (ER) applications. The microstructure and composition of the obtained PANI/$BaTiO_3$ composite were characterized by SEM, FT-IR and XRD. In addition, we also employed a rotational rheometer to investigate the rheological performance of the ER fluids based on both pure PANI particle and PANI/$BaTiO_3$ composite. It was found that the composite materials possess much higher yield stresses than the pristine PANI due to unique dielectric properties of the inorganic $BaTiO_3$ particles. Finally, we also examined dynamic yield stress by analyzing its extrapolated yield stress data as a function of electric field strengths. Using the critical electric field strengths deduced, we further found that the universal yield stress equation collapses their data onto a single curve.

Inhibitory Effect of Spermidine with Antioxidant Activity on Oxidative Stress in Human Dermal Fibroblasts (사람피부섬유아세포에서 산화적 스트레스에 대한 항산화 활성을 가진 spermidine의 억제효과)

  • Spermidine is a ubiquitous polycation that is synthesized from putrescine, which serves as a precursor of spermine. In recent years, spermidine was found to be a polyamine that plays an important role in longevity. Reactive oxygen species (ROS) such as hydroxyl radical, superoxide and hydrogen peroxide have been shown to be involved in various pathogenic processes as well as aging. The direct scavenging effect of spermidine on DPPH radical, $H_2O_2$ and hydroxyl radical, and its protective effect against DNA oxidation related to oxidative stress were evaluated in vitro. It was observed that spermidine exhibits scavenging activities on DPPH radical and H2O2 above 500 ${\mu}M$. Spermidine was especially effective in exerting a scavenging activity on hydroxyl radical. In addition, spermidine at 1000 ${\mu}M$ showed a clear protective effect against DNA oxidation. Furthermore, the expression level of anti-oxidant enzymes such as superoxide dismutase in humam dermal fibroblasts increased in the presence of spermidine compared with blank group. These results suggest that spermidine can be used as an antioxidant to prevent ROS-related diseases including inflammation, cancer and aging.

The Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Alcoholic Liver Damages by Applying Proteomics (청간해주탕(淸肝解酒湯)이 알코올 유발 간섬유화와 단백질 발현에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the effects of Chungganhaeju-tang(Qingganjiejiu-tang) on alcoholic liver damaged by applying proteomics. Materials and Methods : Sprague-Dawley rats were used in this experiment the rats were divided into the normal group, the control group(alcohol) and the sample group(CGHJT +alcohol). The ethanol was orally administered twice a day for 6 weeks in the control and sample groups. Water instead of ethanol was orally administered twice a day for 6 weeks in the normal group. CGHJT extract was orally administered once a day for 6 weeks in the sample group. The livers of each group were processed and assessed by histology, Western Blot, $Oxyblot^{TM}$, CBB and 2-dimensional electrophoresis. Results : In the histological findings of the liver, CGHJT inhibited hepatic fibrogenesis induced by alcohol. TIMP-1 decreased in the sample group assessed by western blot and statistical significance was noted by dot blotting(p<0.05). In the $Oxyblot^{TM}$, protein oxidation induced by alcohol treatment decreased with CGHJT. In the 2-dimensional electrophoresis finding, increased proteins alcohol such as HSP 60, 60kDa heat shock protein, 3-mercaptopyruvate sulfurtransferase were normalized by CGHJT. CGHJT was considered to normalize the anti-oxidation activity elevated by alcohol. In the 2-dimensional electrophoresis finding, increased oxidized proteins such as actin, prolyl 4-hydroxylase beta polypeptide, 94kDa glucose regulated protein(GRP94), heat shock protein 90-alpha(HSC86), calreticulin precursor(CRP55), ATP synthase beta chain mitochondrial precursor, caspase-8 precursor, and dihydrolipoamide succinyltransferase(E2) decreased with CGHJT. CGHJT was considered to reduce the oxidative stress of alcohol. Conclusion : Chungganhaeju-tang(Qingganjiejiu-tang) exerts an inhibitory effect against the fibrosis and protein oxidation induced by alcohol treatment of rat liver. CGHJT was considered to normalize the elevated anti-oxidation activity by alcohol and to reduce the level of oxidative stress due to alcohol.

  • Electrochemical surface treatment of PAN-based carbon fibers in acidic electrolyte has been studied in increasing the surface functional groups on fiber surfaces for the improvement of fiber-matrix adhesion of the resulting composites. According to the FT-IR and XPS measurements, it reveals that the oxygen functional groups on fibers are largely influence on the composite mechanical behaviors, whereas the nitrogen functional groups are not affected in the system. In this work, a good correlation between surface functionality and mechanical properties is established.
