  • This research was performed to study the actual behavior of 1-2W type pipe greenhouse under the influence of typhoon by measuring the vairous strains instructural materials. These results can eventually be utilized in the desgin criteria as well as in the modification of conventional equaltion for calcu more realistic wind loads. Tehfirst data under the influence of Typhoon Olga arrived in Jinju on Aug. 1999. Were obtained by strain gage with 10 sensor points. According to the data obtained, the typical variation ofstrain depending on wind patter could be observed. The strains in structural frame were fluctuated very sensitively depending on the direction and magnitude of wind velocity. But some of the data were lost or missed by system's failure. A kind of inherent vibration pattern of greenhouse pipe frame was observed from the plotted data, but this phenomenon is not so clear as to be separated from the overall fluctuation so far. This experimental research is expected to be continued as a long term project to measure and analyze the strain pattern of structural frame depending on the various locations and section characteristics by wasy of adopting more efficientg instrument with sufficient number of measuring points and accuracy.

  • FK506 hyper-yielding mutant, called the TCM8594 strain, was made from Streptomyces tsukubaensis NRRL 18488 by mutagenesis using N-methyl-N'-nitro-N-nitrosoguanidine, ultraviolet irradiation, and FK506 sequential resistance selection. FK506 production by the TCM8594 strain improved 45.1-fold ($505.4{\mu}g/mL$) compared to that of S. tsukubaensis NRRL 18488 ($11.2{\mu}g/mL$). Among the five substrates, wheat bran was selected as the best solid substrate to produce optimum quantities of FK506 ($382.7{\mu}g/g$ substrate) under solid-state fermentation, and the process parameters affecting FK506 production were optimized. Maximum FK506 yield ($897.4{\mu}g/g$ substrate) was achieved by optimizing process parameters, such as wheat bran with 5 % (w/w) dextrin and yeast extract as additional nutrients, 70 % (v/w) initial solid substrate moisture content, initial medium pH of 7.2, $30^{\circ}C$ incubation temperature, inoculum level that was 10 % (v/w) of the cell mass equivalent, and a 10 day incubation. The results showed an overall 234 % increase in FK506 production after optimizing the process parameters.

  • The metabolic pathway of eugenol degradation by thermophilic Geobacillus sp. AY 946034 strain was analyzed based on the lack of data about eugenol degradation by thermophiles. TLC, GC-MS, and biotransformation with resting cells showed that eugenol was oxidized through coniferyl alcohol, and ferulic and vanillic acids to protocatechuic acid before the aromatic ring was cleaved. The cell-free extract of Geobacillus sp. AY 946034 strain grown on eugenol showed a high activity of eugenol hydroxylase, feruloyl-CoA synthetase, vanillate-O-demethylase, and protocatechuate 3,4-dioxygenase. The key enzyme, protocatechuate 3,4-dioxygenase, which plays a crucial role in the degradation of various aromatic compounds, was purified 135-fold to homogeneity with a 34% overall recovery from Geobacillus sp. AY 946034. The relative molecular mass of the native enzyme was about $450{\pm}10$ kDa and was composed of the non-identical subunits. The pH and temperature optima for enzyme activity were 8 and $60^{\circ}C$, respectively. The half-life of protocatechuate 3,4-dioxygenase at the optimum temperature was 50 min.

  • The effects of slenderness ratio, eccentricity and column slope on the load-carrying capacities and failure modes of variable and uniform concrete filled steel tubular (CFST) latticed columns under axial and eccentric compression were investigated and compared in this study. The results clearly show that all the CFST latticed columns with variable cross section exhibit an overall failure, which is similar to that of CFST latticed columns with a uniform cross section. The load-carrying capacity decreases with the increase of the slenderness ratio or the eccentricity. For 2-m specimens with a slenderness ratio of 9, the ultimate load-carrying capacity is increased by 3% and 5% for variable CFST latticed columns with a slope of 1:40 and 1:20 as compared with that of uniform CFST latticed columns, respectively. For the eccentrically compressed variable CFST latticed columns, the strain of the columns at the loading side, as well as the difference in the strain, increases from the bottom to the cap, and a more significant increase in strain is observed in the cross section closer to the column cap.

  • The immunological role of the Pasteurella multocida toxin (PMT) in mice was examined using a PMT mutant strain. After a nasal inoculation, the mutant strain failed to induce interstitial pneumonia. Moreover, PMT had no significant effect on the populations of CD4+, CD8+, CD3+, and CD19+ immunocytes in blood or on the populations of CD4+ and CD8+ splenocytes (P<0.01). However, there was a significant increase in the total number of cells in the BAL samples obtained from the wild-type P. multocida-inoculated mice. On the other hand, the level of IL-l expression decreased when the macrophages from the bronchio-alveolar lavage were stimulated with PMT. Overall, PMT appears to play some role (stimulating and/or inhibiting) in the immunological responses but further studies will be required to confirm this.

  • Connections play a significant role in strength of structures against earthquake-induced loads. According to the post-seismic reports, connection failure is a cause of overall failure in reinforced concrete (RC) structures. Connection failure results in a sudden increase in inter-story drift, followed by early and progressive failure across the entire structure. This article investigated the cyclic performance and behavioral improvement of shape-memory alloy-based connections (SMA-based connections). The novelty of the present work is focused on the effect of shape memory alloy bars is damage reduction, strain recoverability, and cracking distribution of the stated material in RC moment frames under seismic loads using 3D nonlinear static analyses. The present numerical study was verified using two experimental connections. Then, the performance of connections was studied using 14 models with different reinforcement details on a scale of 3:4. The response parameters under study included moment-rotation, secant stiffness, energy dissipation, strain of bar, and moment-curvature of the connection. The connections were simulated using LS-DYNA environment. The models with longitudinal SMA-based bars, as the main bars, could eliminate residual plastic rotations and thus reduce the demand for post-earthquake structural repairs. The flag-shaped stress-strain curve of SMA-based materials resulted in a very slight residual drift in such connections.

  • Test equipment for strain and vibration measurement was designed for educational purposes. Widely available and affordable materials were put into making this device. Three strain gauges placed on an iron ruler made cantilevered beam were used to measure values according to external load. An electromagnet triggered excitation and a function generator created vibration of the beam. We present three different tests conducted with this equipment regarding production of scales, measurement of resonant frequency, and calculation of the difference between excitation frequency and measured frequency. Overall, this paper presents a piece of simple yet inexpensive test equipment and its corresponding portfolio with expectations of being applied to the educational field for efficient measurement of load and vibration.

  • The relationship among the flame radiation, NOx emissions, residence time, and global strain rate are examined for turbulent non-premixed jet flames with wide variations in coaxial air conditions. Measurements of NOx emission, flame geometry and flame radiation were made to explain the NOx emission scaling based on global parameters such as flame residence time, global strain rate, and radiant fraction. The overall 1/2-power scaling is observed in coaxial air flames, irrespective of coaxial air conditions, but the degree of deviation from the l/2-slope curve in each case differs from one another. From the comparison between the results of pure hydrogen flames and those of helium diluted hydrogen flames, it is observed that flame radiation plays a significant role in pure hydrogen flames with coaxial air and the deviation from 1/2-power scaling may be explained in two reasons: the difference in the flame radiation and the difference in jet similarity in coaxial air flames. From the radiation measurements, more detailed explanations on these deviations were suggested.
