Effects of Osmotic and Non-Osmotic Priming Methods for Enhancing Germination of Pepper Seed

  • This study was conducted to investigate the effects of osmotic priming, liquid smoke (LS), sonication and modified drum priming treatments to improve the germination speed and uniformity of pepper seeds for high quality export seeds. Seeds were treated in 0.5, 1.0, 5.0, 10% LS solution only or with 100mM $K_2SO_4$ solution for 6days at $15^{\circ}C$. Sonication treatment was performed for 5, 10, and 20 minutes at an intensity of 5.2, 10.4, 15.7, 21.0, 26.1kHz in water at $15^{\circ}C$. After sonication treatment, seeds were primed with water or 100mM $K_2SO_4$ for 4days. 40, 50 and 60% seed moisture content (SMC) of hydrated seeds were incubated for 84, 96 and 108h in a container with a relative humidity of 99% at 26rpm for a modified drum priming treatments. 0.5% LS treatment with water or 100mM $K_2SO_4$ priming showed significant effects with 71% GP (9.0days MGT) and 66% GP (7.4days MGT), respective, while untreated seeds resulted in only 61% GP and 10days MGT. The healthy seed % was increased by 24% by $K_2SO_4$ priming with LS treatment compared to untreated seeds. However, sonication treatment showed less germination or no difference compared to untreated seed, regardless of intensity and treatment time. The modified drum priming treatment (108h incubation after 50% SMC hydration) significantly improved the GP (on the $4^{th}$days after sowing), germination rate (GR), and MGT to 65%, $28%{\cdot}day^{-1}$, and 3.6days, whereas untreated seed resulted only 7% GP, $17%{\cdot}day^{-1}$ GR and 5.9days MGT. Thus, modified drum priming treatment effectively improved seed germination and is considered as an industrially promising treatment methods considering the shortening of the treatment period and environment-friendly aspects.

열탕처리와 염수선 처리가 콩과 녹비작물 발아에 미치는 영향 (Effects of Hot Water and Specific Gravity Treatment on Germination of Legumes and Green Manure Crops)

  • 본 연구는 해마다 수입을 많이 해오고 있는 콩과 녹비작물 종자의 발아율을 높이기 위해서 연구되었다. 열탕처리는 콩과 식물에서처럼 두꺼운 종피로 인해 휴면상태에 빠지는 종자의 발아율 향상 및 발아속도 증진을 위해 사용하는 방법이다. 클로버류와 자운영은 열탕처리가 무처리 보다 발아일수와 발아속도가 다소 높았다. 하지만 발아율과 발아세의 경우 무처리가 열탕처리보다 다소 높았지만 유의미한 차이를 보이지 않아 열탕처리가 발아율에 효과가 있다고 판단되지는 않는다. 네마장황의 경우 발아율과 발아속도를 높이는데 있어서는 염수선 처리가 가장 효과가 좋았고, 열탕처리는 초기 발아속도를 높일수는 있지만 발아율을 높이는데는 적합하지 않은 것으로 보였다. 헤어리베치의 경우 수입산 헤어리베치와 국내산 헤어리베치(토익과 청풍보라)에서 열탕처리와 염수선처리는 발아율에는 효과가 없었으나 발아세, 발아속도, 평균발아일수에는 다소 효과가 있다고 판단된다. 따라서 콩과 녹비작물과 같이 두꺼운 종피로 인하여 휴면상태에 빠지는 종자의 경우 발아율 향상과 발아속도를 향상시키기 위해 열탕처리나 염수선처리를 하는 것이 좋을것이라 사료된다.

Repetitive Somatic Embryogenesis in Cacao and Optimisation of Gene Expression by Particle Bombardment

  • In order to achieve repetitive somatic embryogenesis in cacao (Theobroma cacao L.), callus derived from floral tissues were continuously cultured in a medium containing 2,4-D. In 5% of the explants, repetitive somatic embryogenesis was observed after 8 weeks and maintained in a globular stage for several weeks. This is the first report showing repetitive somatic embryogenesis in cacao. The calli were bombarded with a plasmid containing $\beta$-glucuronidase (gus) as reporter gene. Two week old calli showed the high average number of cells expressing the us gene. The effect of osmotic agents (mannitol, sorbitol and sucrose) on gene expression was evaluated. Pre-treatment during 16 h with 0.25 M mannitol revealed an improvement in gene expression. The potential utilization of the repetitive embryogenesis, combined with osmotic treatment, is discussed as an alternative to achieve stable transgenic cacao plants.

Aquaporin 4 water channel 인산화에 의한 수분 투과도의 조절 (Phosphorylation of AQP4 Water Channel Regulates Water Permeability)

  • Aquaperin 4 (AQP4) is the mercurial water channel expressed abundantly in brain, especially the region related with cerebrospinal fluid reabsorption and osmoregulation. The primary structure of AQP4 water channel was elucidated but the molecular mechanism of AQP4 channel regulation is still unknown. To investigate the possible regulation of AQP4 water channel by phosphorylation via various protein kinases, osmotic water permeability of AQP4 expressed in Xenopus oocytes was measured by videomicroscopy technique. Forskolin (10 $\mu$M) did not affect osmotic water permeability of oocytes injected with AQP4 cRNA, excluding the regulation of AQP4 water cnannel by protein kinase A. Osmotic water permeability (P아래첨자) of AQP4-expressed oocytes was ingibited by the pretreatmeat of BAPTA/AM (up to 500$\mu$M), an intracellular Ca윗첨자 chelator, and calmidazolium (100$\mu$M), a specific Ca윗첨자/calmodulin antagonist, in a dose-dependent manner. The inhibition of osmotic water permeability (P아래첨자) by the calmidazolium treatment was completely reversed by the addition of calyculin A (0.1$\mu$M), a nonspecific phosphatase inhibitor. Phorbol 12-myristate 13-acetate (PMA), a protein kinase C activator, had biphasic effects on osmotic water permeability in AQP4 cRNA injected oocytes depending on its concentration; 21% increase by 100 nM PMA, 35% decrease by 1$\mu$M PMA. These effects were reversed with 2$\mu$M staurosporine, a nonspecific PKC inhibitor. These results suggest that phosphorylation of AQP4 water channel by Ca윗첨자/calmodulin kinase and protein kinase C might regulate the osmotic water permeability.

한국잔디 종자의 발아특성과 발아율 향상을 위한 다양한 전처리 방법의 비교 (Comparison of Germination Characteristics and Various Pre-treatment Methods for Enhancing Germination on Zoysiagrass)

  • 들잔디 종자의 발아를 향상시키기 위해서, 적정 종자 전처리 방법을 조사하였다. 종자들은 종피 연화 및 4가지 프라이밍 처리, 즉, 종피에 KOH 처리 유무에 따른 Hydro 프라이밍(증류수), Osmotic 프라이밍(폴리에틸렌 글리콜 -0.5MPa, -1.0MPa, -2.0MPa), Halo 프라이밍(질산칼륨 100 mM, 200 mM, 400 mM)과 Solid matrix 프라이밍(종자:microcel-E:증류수 = 4:1:8) 방법을 조합하였다. 종피 연화는 발아율을 향상시키고 발아소요시간을 단축시켰다. 종피 연화 처리 후 Osmotic 프라이밍(PEG -0.5 MPa)에서 발아율이 유의적으로 증가하였고, MGT, $T_{50}$이 감소하였다. 종자 발아율은 자연상태와 비교하여 60% 이상 증가시켰다. 결론적으로, 종피연화와 프라이밍 처리한 잔디종자는 더 빠르고 균일하게 발아를 하였다. 현 연구를 통해 KOH처리와 PEG 프라이밍 처리는 파종 시 발아율을 향상 시켜 초기 잔디 조성을 빠르게 할 수 있을 것으로 기대된다.

  • Pfu DNA polymerase from Pyrococcus furiosus was expressed in the E. coli periplasm, and the fully active polymerase was partially purified by applying osmotic shock, ammonium sulfate precipitation, and heat treatment. This method represents a new way of expressing and purifying functional Pfu DNA polymerase without the use of chromatography.

Modeling of flux enhancement in presence of concentration polarization by pressure pulsation during laminar cross flow ultrafiltration

  • A theoretical study for the flux enhancement by pulsation of transmembrane pressure is presented for osmotic pressure controlled ultrafiltration under laminar flow regime. The transient velocity profile is solved analytically using Green's function method. Time dependent convective diffusive equation is solved to quantify the membrane surface concentration and the permeate flux, numerically. The effects of the amplitude and frequency of pulsation on flux, surface concentration and observed retention are studied.

The effect of osmotic potentials on the temperature response for germination of pepper seed

  • Primed seeds germinate faster and uniformly, since these have already started imbibition and metabolism requiring to complete germination. Several factors such as solution composition, osmotic potential, and treatment duration affect seed priming response. Water potentials of priming solution and germination response of primed seed were investigated to clarify the effects different inorganic salt types and the duration. Pepper seeds were primed in osmotic solutions that were osmotic potential ranged -1.54 to -0.45 MPa in an aerated solution of polyethylene glycol 8000 (PEG; 17%, 22%, 27%), and inorganic salt solution of $KNO_3$, $K_2SO_4$, and $Na_2SO_4$ (100, 200, 300mM) in aerated condition. The seeds were treated at $20^{\circ}C$ for 4, 6 day (D) and stepwise duration combined concentration of 100mM (2 or 4 days) and 300mM(2 or 4days). After soaking treatment, seeds were washed with distilled water and then were dried to approximately 6% moisture content in dry chamber at $25^{\circ}C$. The germination of seed was characterized by ISAT rules at $20^{\circ}C/30^{\circ}C$ and $15^{\circ}C$. Total germination percentage (GP), mean germination time (MGT), germination uniformity (GU), germination rate (GR), and health seedling percent (HS) were calculated on the germinated seed in a 14-day period. Seed water potential (${\psi}$) was correlated with water potential of priming solution ($r^2=0.84$). The effect of seed priming on germination varies with inorganic salt and organic agents (PEG). Germination percentage (GP, 77 and 73%), GR (21.3 and $19.4 %{\cdot}day^{-1}$), and HS (58 and 52 %) was greatest and lowest MGT (4.2 and 4.8 day) when they were primed in 100mM $K_2SO_4$ (${\psi}=-0.45MPa$), and 100mM $KNO_3$ (${\psi}=-0.52MPa$) for 6 days compared to untreated control (67% GP, $19.7%{\cdot}day^{-1}\;GR$, 18% HS, and 10.8 day MGT) in $15^{\circ}C$ chamber. Water potential less then -1.2MPa of osmotic solution resulted lower GP, GR and HS and in $K_2SO_4$ and $KNO_3$. However, stepwise treatment of water potential that changed solution as 100mM for 4 D + 300mM for 2 D or 300mM for 4 D + 100mM for 2 D) were not better than single concentration alone in germination characteristics.

알팔파 캘러스로부터 삼투압 스트레스 처리에 의한 효율적인 식물체 재분화 (Efficient Plant Regeneration from Alfalfa Callus by Osmotic Stress Treatment)

  • 알팔파의 최적 조직배양 조건을 확립하기 위하여 자엽으로부터 배발생 캘러스 유도 및 캘러스로부터 식물체 재분화에 미치는 배지첨가물질과 삼투압 스트레스 처리의 영향을 조사하였다. 배발생 캘러스 유도시에 첨가되는 식물생장조절물질로는 5mg/L 2,4-D와 0.2mg/L kinetin이 혼용 첨가된 SH 배지에서 배발생 캘러스가 가장 높은 빈도로 유도되었다. 배발생 캘러스를 1mg/L 2,4-D와 2mg/L BA가 첨가된 SH 배지에서 배양했을 때 체세포 배가 형성되었다. 재분화 배지에 첨가되는 질소원으로 5mM L-proline과 1g/L casein hydrolysate를 동시에 첨가해주었을 때, 식물체 재분화율이 증가되었다. 배발생 캘러스를 0.7M 농도의 삼투압 스트레스 조건에서 12 ${\sim}$ 18시간 처리한 후, 재분화 배지에서 배양한 결과 재분화율이 현저히 증가되었다. 가장 높은 식물체 재분화율은 0.7M sucrose가 첨가된 배지에서 18시간 처리한 후, 재분화 배지에서 배양했을 때 30.7%의 효율을 나타내었다. 본 연구를 통하여 확립된 알팔파의 효율적인 배발생 캘러스의 유도 및 식물체 재분화체계는 분자육종을 통한 신품종 알팔파의 개발에 유용하게 이용되어질 수 있을 것이다.

한발저항성 정도가 다른 보리 품종들의 한발처리에 따른 생리적 특성변화 (Changes in Physiological Characteristics of Barley Genotypes under Drought Stress)

  • 이 논문은 한발저항성이 다른 6개 보리 품종의 한발에 따른 생장, 잎의 수분포텐셜(leaf water potential, LWP), 상대함수량(leaf relative water content, RWC), 삼투압(leaf osmotic potential, OP), 삼투조정(osmotic adjustment, OA), 팽압(leaf turgor pressure, LTP), 순광합섬, 기공전도도, 엽육전도도, 엽록소형광 등의 변화를 조사하여 비교한 결과를 요약하면 다음과 같다. 1 한발 처리시 토양수분포텐셜은 -0.05㎫이었고. 종료시에는 -0.29㎫로 저하하였다. Dicktoo-S 동보리 1호, Dicktoo-L, Dicktoo-T, 수원쌀보리 365호, 탑골보리 품종의 한발처리구 건물중은 각각 대조구(처리기간종 -0.05㎫ 유지)에 비하여 68%, 69%, 70%, 86%, 55%, 37%를 나타내어 Dicktoo 계통과 동보리1호의 한발저항성이 강하였고, 수원쌀보리 365호와 탑골보리는 한발저항성이 약하였다. 2. 한발저항성이 강한 품종은 삼투조정능력이 커서 한발처리에 따른 RWC와 LWP의 저하가 작았고 팽압유지능력이 컸다. 3. 한발처리에 따라 순광합성이 저하하였고 그 저하정도는 한발저항성이 큰 품종이 작았는데, 이는 한발저항성이 큰 품종이 기공전도도, 엽육전도도 및 PSII 최대양자수율(Fv/Fm)의 저하가 적었기 때문이었다. 4. 결론적으로 저항성이 큰 품종은 삼투조정에 의한 수분유지능력이 크고 이에 따라 광합성저하가 적어 상대적으로 생장의 감소가 적은 것으로 판단되었다.
