Lignification in Relation to the Influence of Water-deficit Stress in Brassica napus

  • To investigate lignification process and its physiological significance under water-deficit condition, the responses of peroxidases, polyphenol oxidase (PPO) and phenylalanine ammonia-lyase (PAL) in relation to leaf water status to the short term of water deficit treatment in the leaves with different maturities in forage rape were measured. The significant decrease in relative water content (RWC) and leaf osmotic potential (${\Psi}{\pi}$) were apparent after 5 d of water-deficit treatment. The activity of guaiacol peroxidase (GPOD), ascorbate peroxidase (APOD), coniferyl alcohol peroxidase (CPOD), and syringaldazine peroxidase (SPOD) was depressed especially in middle and old leaves when compared with that of control leaves. On the other hand, in young leaves, a significant increase in CPOD (+34%) and SPOD (+24%) activity as affected by water-deficit treatment was apparent. The activation of PAL and PPO was observed in middle and old leaves for PAL and in young and middle leaves for PPO. These results suggest that peroxidases in middle and old leaves did not involve in lignification under mild water-deficit stress, whereas CPOD and SPOD in young leaves participate in lignification by a coordination with PAL and PPO to incorporate phenol and lignin into the cell walls.

수분(水分) 및 양료(養料) 처리(處理)에 따른 참나무류와 물푸레나무의 생장 및 생리 반응 (Growth Performances and Physiological Responses of Quercus spp. and Fraxinus rhynchophylla Subjected to Different Soil Moisture Regimes and Nutrition Levels)

  • 장기간의 수분 및 양료 stress를 받는 다섯 가지 주요 수종의 생리적인 대사 반응을 알아보기 위하여 상이한 토양수분 및 양료 조건의 pot에서 기른 실생묘의 시기적인 생육, 엽록소 함량 및 수분특성의 변화를 조사하였다. 이를 위해 상수리나무, 굴참나무, 신갈나무, 졸참나무 및 물푸레나무 실생묘를 내경 16cm, 깊이 16cm의 프라스틱 용기에 번식시켰다. 이들 묘목은 5월~9월까지 비닐 온실 내에서 건, 습 두가지 토양수분 조건 및 N+P+K 시비, 무시비의 두가지 시비 조건으로 조합 처리하였다. 환경 stress에 대한 반응을 분석하기 위해 5월, 7월, 9월에 묘고, 근원경, 엽록소 함량, P-V curve 모수들을 반복하여 측정했다. 수분 및 양료 부족에 따른 환경 stress는 묘목의 생육에 있어 수종, 생육시기 및 묘고와 근원경 간에 각기 다른 영향을 미쳤다. 상수리나무의 생육은 stress에 보다 예민하게 반응하여 감퇴되었지만 신갈나무는 나머지 수종에 비해 stress에 의한 생육부진 영향이 적었다. 엽록소의 함량은 생육기간 중 일반적으로 물푸레나무에 비해 참나무류에서 더 높은 값을 보였다. 엽록소 중 chlorophyll a의 함량은 0.14~1.96mg/g dry wt., chlorophyll b는 0.16~1.79mg/g dry wt. 내에서 생육기간 중 수종 및 처리 별로 변화했다. 그러나 엽록소 함량은 지속적인 환경 stress와 잎의 노화에 따라 점차 감소되었다. 최대포수상태와 위조점에서의 osmotic potential(${\Psi}{{\pi}o}$, (${\Psi}{{\pi}p}$)은 일부 예외는 있지만 각각 5월의 -7.0~-12.4bars에서 9월의 -10.2~-17.5bars로 3~5bars까지, 또한 5월의 -7.6~-14.2bars에서 9월의 -12.9~-20.4bars로 5~6bars까지 시간이 지나면서 감소했다. ${\Psi}{{\pi}p}$의 값은 일반적으로 5월 및 7월에 물푸레나무에서 높았으나 9월에는 졸참나무에서 높은 값을 보였다. 위조점에서의 상대수분함량 (RWCp)은 일반적으로 물푸레나무에서 높았지만 그 값의 시기적인 변화는 수종이나 처리 조건에 따라 상당한 차이를 보였다.

이종 심장 판막 이식편에서 효과적인 탈세포화 방법에 관한 연구; 저장성 용액(hypotonic solution)의 삼투압 처치법 효과 (A Study on an Effective Decellularization Technique for a Xenograft Cardiac Valve: the Effect of Osmotic Treatment with Hypotonic Solution)

  • 배경: 조직판막에 남아있는 세포물질은 환자의 면역반응과 관련이 있고 석회화의 원인이 된다고 알려져 있으며 탈세포화 된 판막조직의 세포외 기질은 숙주세포(host cell)가 부착되고 내피화(endothelialization)되어 조직으로 재구성되게 하는 생물학적 지자체로도 사용될 수 있다. 그러므로 탈세포화는 조직판막의 면역반응 최소화와 지자체의 형성에 가장 중요한 부분이다. 탈세포화를 위해 많은 방법과 약제들이 개발 되어 왔다. 그러나 화학적 혹은 효소적 탈세포화 방법에 앞서 삼투압(osmolality)을 이용한 저장성 용액 처치에 대한 효과를 보여준 논문은 드물다. 본 연구는 탈세포화에 있어서 저장성 용액 처치의 효과를 밝히고 아울러 탈세포화 시 적절한 온도, 처치시간, sodium dodecylsulfate (SDS)의 농도를 밝히고자 시행되었다. 대상 및 방법: 돼지 판막을 여러 조건 즉 SDS농도(0.25%, 0.5%), 저장성용액 처치시간(6, 12, 24시간), 온도($4^{\circ}C$, $20^{\circ}C$)를 달리하여 일종의 이온세정 용매인 SDS를 이용하여 탈세포화를 하였다. 저장성 용액에 노출시키지 않은 군을 B군으로 하고 동 수의 돼지 판막을 같은 조건에서 SDS에 처치하기 전에 저장성용액에 노출 시켰다(A군). 저장성 용액에의 노출은 4, 7, 14시간, $4^{\circ}C$$20^{\circ}C$로 구분하여 각각 탈세포화의 정도를 조사하였다. 탈세포화와 세포외 기질의 보존 정도는 hematokylin-eosin (H-E) 염색으로 표본을 관찰하였다. 결과: SDS의 두 농도, 0.25%, 0.5%에서 탈세포화의 정도의 차이는 발견할 수 없었다. $4^{\circ}C$ 환경에서 24시간 SDS로 처치한 것이 A, B군 모두에서 가장 양호한 탈세포화를 소견을 보였고 $20^{\circ}C$에서는 탈세포화 정도에 차이가 없었다. 저장성 용액으로 처치한 A군 모두에서 같은 조건의 처치하지 않은 B군에 비래 더 양호한 탈세포화 소견을 보였다. $4^{\circ}C$, 14시간 저장성용액에 노출된 판막이 가장 좋은 탈세포화의 소견을 보였고, $20^{\circ}C$의 저장성 용액에 노출된 판막은 양호한 탈세포의 소견을 보였으나 심한 세포외 기질의 파괴를 관찰할 수 있었다. 결론: 판막조직의 탈세포화에 있어서 삼투압처리를 이용한 저장성 용액 처치는 매우 효과적인 방법으로서, 돼지 판막조직의 탈세포화에 꼭 적용되어야 할 방법으로 생각된다 저장성 용액에의 노출이 탈세포화에 필요한 화학적 세정제(chemical detergent)의 농도를 줄이고 이러한 세정제의 농도나 노출시간을 줄일 수 있는지 그리고 세포외 기질의 변화에 어느 정도 영향을 주는지에 대해 추가적인 연구가 필요할 것으로 생각된다.

홍조류 꼬시래기(Agarophyton vermiculophyllum)의 과포자방출 유도 방법에 대한 연구 (A Study on Carpospore Release Induction Method of Agarophyton vermiculophyllum)

  • The aim of this study was to examine optimal induction method for carpospore release from Agarophyton vermiculophyllum cystocarps for seedling production. We tested the effects of environmental factors on carpospore release by using five different induction methods; spontaneous, desiccation, low temperature, desiccation+low temperature, and osmotic shock. Also, carpospores release was estimated at three temperatures (20, 25, and 30℃), and then under combinations of three day lengths (8, 12, and 16h) and two irradiances (30 and 60 μmol photons m-2 s-1), after pretreatment at desiccation+low temperature for 2 hr. The number of carpospores released was between 113 ~ 682 spores /cystocarp/day and it was maximal in the desiccation+low temperature treatment. Optimal environmental conditions for carpospore release of A. vermiculophyllum were 25℃, 16 h, and 60 μmol photons m-2 s-1. The present results suggest that massive carpospores for seedling production of A. vermiculophyllum could be obtained under a combination of 25℃, 16 h, and 60 μmol photons m-2 s-1 after pretreatment in the desiccation+low temperature.

Gliocladium virens 와 Trichoderma harzianum의 속간(屬間) 원형질체융합(原形質體融合) (Intergeneric protoplast fusion between Gliocladium virens and Trichoderma harzianum)

  • The protoplast formation and intergeneric protoplast fusion between Gliocladium virens and Trichoderma harzianum were attempted to obtain fusants. Protoplast formation was the most effective when the strains were treated with concentration of 5 mg/ml of Novozyme 234 and Cellulase at $25^{\circ}C$ for 3 hours in phosphate buffer, pH 6.5, supplemented with 0.6 M sorbitol as osmotic stabilizer. Auxotrophic mutants of G. virens G88 did not grow in minimal medium and benomyl resistant T. harzianum T95 from wild types, however, was selected by treatment with UV light as genetic marker to isolate fusants. When the intergeneric protoplast fusion between G. virens G88 and T. harzianum T95 was carried out using 30% PEG 4000 containing 10 mM $CaCl_{2}$, and 50 mM glycine (pH 8.5) as fusogenic agent at $25^{\circ}C$ for 10-15 min, the fusion frequency was $0.8{\times}10^{-4}$. Fusants obtained from intergeneric protoplast fusion were spontaneously segregated into va rious strains by continous culture on complete medium. Several intergeneric hybrids were classified into three types: parent-like hybrids, segregants, and recombinants.

주요식량작물 종자세 개선에 관한 연구 (Studies on Practical Methods to Control Seed Vigour in Several Food Crops)

  • 발아의 신속성과 균일성을 높이기 위하여 벼, 보리, 콩 종자에 대하여 몇가지 화학물질처리와 종자의 흡습건조처리를 통하여 종자퇴화지연효과를 검토하였다. 처리한 생장촉진제 및 노화지연제간에 경미한 차이는 있었으나 GA$_3$가 벼의 발아율을 높인것 외에는 뚜렷한 종자세 개선의 효과를 보이지 않았다. 노화종자를 PEG용액에 흡습건조처리할 경우 용액농도가 높을수록 평균발아일수가 길어지고 발아율이 낮아지는 경향이나 발아율 유지의 효과나 발아균일성향상의 효과는 없었다. 저온, 과습 및 삼투조건 등 불량한 환경에 대한 발아반응도 미미하였다. 어느 처리도 종자세 개선에 뚜렷한 효과를 보이지는 않았으나 GA$_3$ 처리가 전체적으로 발아반응이 그 중 좋은 편이었다.

Organic fouling in forward osmosis (FO): Membrane flux behavior and foulant quantification

  • Forward osmosis (FO) is an emerging membrane technology with potential applications in desalination and wastewater reclamation. The osmotic pressure gradient across the FO membrane is used to generate water flux. In this study, flux performance and foulant deposition on the FO membrane were systematically investigated with a co-current cross-flow membrane system. Sodium alginate (SA), bovine serum albumin (BSA) and tannic acid (TA) were used as model foulants. Organics adsorbed on the membrane were peeled off via oscillation and characterized by Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). When an initial flux of $8.42L/m^2h$ was applied, both flux reduction and foulant deposition were slight for the feed solution containing BSA and TA. In comparison, flux reduction and foulant deposition were much more severe for the feed solution containing SA, as a distinct SA cake-layer was formed on the membrane surface and played a crucial role in membrane fouling. In addition, as the initial SA concentration increased in FS, the thickness of the cake-layer increased remarkably, and the membrane fouling became more severe.

Optimal Conditions for the Distribution of Cryoprotectant into the Intact Fish Muscle of Oncorhynchus mykiss during Freeze/Thaw Cycling

  • Conditions for sufficient and rapid distribution of a cryoprotectant (sorbitol solution) into intact fish muscle (Oncorhynchus mykiss) were studied as changing in the residual Ca2+ ATPase activity during freeze/thaw cycling. Chunks of the fish muscle were immersed in 4 concentrations of sorbitol solutions ($20\%$, $30\%$, $45\%$, and $60\%$) by a shaker mechanism at 5$^${circ}C. Whole immersion samples (W) showed a higher value of the residual Ca2+ ATPase activity than those in the untreated controls (C), except in the treated controls (TC), while less effect of immersion concentration could be found. Comparing the extent of penetration of sorbitol into the surface layer to inner layer of immersed fish chunks, outer portion samples achieved excellent cryoprotection with $100\%$ of the residual ATPase activity values or more. For the inner portion samples, $30\%$ and $45\%$ sorbitol solution treatments indicated a higher ATPase activity than $60\%$ treatment. At high concentrations, mass transfer rates during osmotic dehydration might berapid and it causes faster surface drying by dewatering at surface solute layer. Periodically immersed and relaxed samples, W (5-3-1), led to good cryoprotection effect: W (5-3-1) indicated high residual Ca2+ ATPase activity values and the residual ATPase activity values excess $100\%$ in immersion of $30\%$ and $45\%$ sorbitol solutions.

Membrane behavior of bentonite-amended compacted clay towards Zn(II) and Pb(II)

  • Zinc and lead pollution are public environmental issues that have attracted lots of attention for a long time. Landfill leachate contains heavy metals, such as Zn(II) and Pb(II), which are usually related to the pollution of groundwater, especially in developing countries. Bentonite has been proven to be effective in enhancing the membrane property of clay, by which landfill liners can have better barrier performance towards the migration of contaminants. In this study, 5% sodium bentonite amended with locally available Fukakusa clay was utilized to evaluate the membrane behavior towards the heavy metals zinc and lead. The chemico-osmotic efficiency coefficient, ${\omega}$, was obtained through Zn(II) and Pb(II) solutions with different concentrations of 0.5, 1, 5, 10, and 50 mM. According to the results, ${\omega}$ continually decreased as the Zn(II) and Pb(II) concentrations increased, which is consistent with the Gouy-Chapman theory. Compared to normal inorganic ions, the membrane behavior towards heavy metal ions was lower. The migration of heavy metal ions was not observed based on experimental results, which can be attributed to the adsorption or ion exchange reaction. The mechanisms of the membrane performance change were discussed with the assistance of XRD patterns, free swelling results, XRF results, and SEM images.

Increased Expression of Epithelial Cell Adhesion Molecule (EpCAM) in Rat Hepatic Tumors Induced by Diethylnitrosamine

  • The epithelial cell adhesion molecule (EpCAM) is a pan-epithelial differentiation antigen that is expressed on almost all carcinomas. However, a role in rat liver carcinogenesis has never been reported previously. Thus, its expression was investigated herein in rat liver tumors induced by diethylnitrosamine (DEN). Twenty male 5-week-old F344 rats were used in this experiment. Mini-osmotic pumps containing doses of 47.5 mg of DEN were inserted into the abdominal cavity of each animal to initiate liver carcinogenesis. All animals were sacrificed at 26 weeks after DEN treatment. At necropsy, hepatic masses were processed for histopathological examination, which revealed forty-four hepatocellular adenomas (HCAs) and twenty hepatocellular carcinomas (HCC). Tumors were immunohistochemically analyzed for EpCAM, proliferating cell nuclear antigen (PCNA) and co-localization of the two. EpCAM expression was mainly detected in hepatic tumor cells, showing a cytoplasmic staining pattern. However, expression was also slightly observed in normally-appearing surrounding hepatic cells. PCNA expression was highly detected in tumor cells, showing nuclear staining. Double staining of EpCAM and PCNA in tumors showed many cells with co-localization. Taken together, EpCAM and PCNA expression were increased in DEN-induced tumors and many tumor cells showed co-expression. It is suggested that EpCAM may increase during DEN-induced tumors, possibly associated with cell proliferation.
