Seeds of chicory were collected and selected with good germination, emergence and productivity. To develop chicory as a new forage crop seeds were primed with polyethylene glycol (PEG 6000) and coated with some chemicals, and tested in laboratory and field. The results obtained are summarized as follows: 1. Among the seed lines collected, PUlgAH, PU21TH, PU37CH and PU30TK were germinated more quickly and took less time for germination than the others. 2. PUI8AH was the best in emergence and growth although PU21TH showed superior germinative ability among the lines collected. 3. Chicory seeds primed with 20~25% PEG solution for three days at $10^{\circ}C$, or the seeds treated with 25% PEG solution at $15^{\circ}C$ regardless the treatment period showed good germination performances. 4. Osmotic priming with PEG accelerated the germination of chicory, but not in the field. 5. Coated seeds geminated more slowly in Petridishes but showed superior emergence and growth to those of primed or untreated seeds. Treatment 3 showed the best performances in pot experiment. 6. Coated seeds showed remarkable increase in establishment and dry matter yield especially in sod sowing compared to the other sowing method. 7. Most nutrient contents except iron and manganese of chicory were much higher than those of mixed pasture .
Forward osmosis (FO) is an emerging membrane technology with potential applications in desalination and wastewater reclamation. The osmotic pressure gradient across the FO membrane is used to generate water flux. In this study, flux performance and foulant deposition on the FO membrane were systematically investigated with a co-current cross-flow membrane system. Sodium alginate (SA), bovine serum albumin (BSA) and tannic acid (TA) were used as model foulants. Organics adsorbed on the membrane were peeled off via oscillation and characterized by Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). When an initial flux of $8.42L/m^2h$ was applied, both flux reduction and foulant deposition were slight for the feed solution containing BSA and TA. In comparison, flux reduction and foulant deposition were much more severe for the feed solution containing SA, as a distinct SA cake-layer was formed on the membrane surface and played a crucial role in membrane fouling. In addition, as the initial SA concentration increased in FS, the thickness of the cake-layer increased remarkably, and the membrane fouling became more severe.
Conditions for isolation of protoplasts from spores and mycelia of Rhizopus nigricans were studied. Larger amount of protoplasts was obtained from swollen spores in liquid medium contained with 5% of 2-deoxy-D-glucose for 4 hours than from mycelia. Enzyme mixture of Novozym 234(2%) and ${\beta}-glucuronidase(5000\;unit/ml)$ was most effective for the isolation of protoplasts from swollen spores and from mycelia. The solution of 0.6 M $MgSO_4$ or mannitol and pH 6.0 showed good results as the osmotic stabilizer and the optimal condition of pH of the enzyme solution for the isolation of protoplast from the swollen spores, respectively. At this condition, $8.0{\times}10^6\;cells/ml$ of protoplasts was obtained from swollen spores by digestion with lytic enzyme mixture for 2 hours.
Conditions for sufficient and rapid distribution of a cryoprotectant (sorbitol solution) into intact fish muscle (Oncorhynchus mykiss) were studied as changing in the residual Ca2+ ATPase activity during freeze/thaw cycling. Chunks of the fish muscle were immersed in 4 concentrations of sorbitol solutions ($20\%$, $30\%$, $45\%$, and $60\%$) by a shaker mechanism at 5$^${circ}C. Whole immersion samples (W) showed a higher value of the residual Ca2+ ATPase activity than those in the untreated controls (C), except in the treated controls (TC), while less effect of immersion concentration could be found. Comparing the extent of penetration of sorbitol into the surface layer to inner layer of immersed fish chunks, outer portion samples achieved excellent cryoprotection with $100\%$ of the residual ATPase activity values or more. For the inner portion samples, $30\%$ and $45\%$ sorbitol solution treatments indicated a higher ATPase activity than $60\%$ treatment. At high concentrations, mass transfer rates during osmotic dehydration might berapid and it causes faster surface drying by dewatering at surface solute layer. Periodically immersed and relaxed samples, W (5-3-1), led to good cryoprotection effect: W (5-3-1) indicated high residual Ca2+ ATPase activity values and the residual ATPase activity values excess $100\%$ in immersion of $30\%$ and $45\%$ sorbitol solutions.
The effects of Gypsophila saponin, sodium dodecylsulfate (SDS) and Triton X-100 on volume changes and fragility of red blood cells were compared to ginseng saponin to elucidate whether there are any difference in their action on membrane lipid. Cell volume was decreased to about 38% in 1M NaCl and increased to about 20% in 1/10M NaCl. Hematocrit value was decreased by Gypsophila saponin, SDS, and Triton X-100 which caused hemolysis in isotonic NaCl solution. These detergents also inhibited increase of cell volume and accelerated hemolysis in hypotonic solution. However, ginseng saponin did not influence to osmotic volume changes and hemolysis of red blood cells. These results suggest that the disruptive effect of plant saponin on membrane barriers induced by removing membrane lipid is different from their source and ginseng saponin has very low affinity to membrane lipid.
Many researchers have been studied with guard cell protoplasts and detached epidermis as they think that properly stabilized protoplasts and detached epidermis retain many of the properties of intact guard cells. However, some studies have shown that stomata in detached epidermis behave differently, both quantitatively and qualitatively, from those in the intact leaf. Stomata in the intact leaf are very sensitive to environmental factors such as light, $CO_2$ and osmotic stress, but stomata in detached epidermis are less sensitive to these factors than those in the intact leaf. The clearest evidence to suggest the different response between detached epidermis and intact leaf obtained from the experiments with heavy metal, cadmium. 3-weeks old Commelina. communis was transferred to and grown in Hoagland solution in the presence or absence of 5 mM $Cd^{2+}$ for 4 days. The application of $Cd^{2+}$ showed about 70% inhibition of stomatal conductance when measured at various light intensity (100-1,000 $\mu$mole $m^{-2}s^{-1}). However, stomata in detached epidermis floated on an incubation medium containing 100 $\mu$M $Cd^{2+}$ opened to a degree of about 8.38 fm, but the stomata treated with no cadmium opened to 3.74 ${\mu}{\textrm}{m}$. These results were unexpected as the intact leaf grown in a Hoagland solution containing cadmium showed very negative physiological responses. These results showed that stomata in detached epidermis and in the intact leaf could respond reversely. Therefore, it is possible that we now misunderstand how stomata open in real natural condition.
We employed the whole-cell patch clamp technique to investigate the effects of arachidonic acid (AA) on barium inward current through the L-type calcium channels ($I_{Ba}$) and on osmotic stretch-induced increase of $I_{Ba}$ in guinea-pig antral gastric myocytes. Under isosmotic condition, AA inhibited $I_{Ba}$ in a dose-dependent manner to $91.1{\pm}1.4,\;72.0{\pm}3.2,\;46.0{\pm}1.8,\;and\;20.3{\pm}2.3%$ at 1, 5, 10, 30 mM, respectively. The inhibitory effect of AA was not affected by 10 ${\mu}M$ indomethacin, a cyclooxygenase inhibitor. Other unsaturated fatty acids, linoleic acid (LA) and oleic acid (OA) were also found to suppress $I_{Ba}$ but stearic acid (SA), a saturated fatty acid, had no inhibitory effect on $I_{Ba}$. The potency sequence of these inhibitory effects was AA ($79.7{\pm}2.3%$) > LA ($43.1{\pm}2.7%$) > OA ($14.2{\pm}1.1%$) at 30 ${\mu}M$. On superfusing the myocyte with hyposmotic solution (214 mOsm) the amplitude of $I_{Ba}$ at 0 mV increased ($38.0{\pm}5.5%$); this increase was completely blocked by pretreatment with 30 mM AA, but not significantly inhibited by lower concentrations of AA (1, 5 and 10 ${\mu}M$) (P>0.05). Unsaturated fatty acids shifted the steady-state inactivation curves of $I_{Ba}$ to the left; the extent of shift caused by AA was greater than that caused by LA. The activation curve was not affected by AA or LA. The results suggest that AA and other unsaturated fatty acids directly modulate L-type calcium channels and AA might modulate the hyposmotic stretch- induced increase of L-type calcium channel current in guinea-pig gastric smooth muscle.
This study investigated the effects of osmotic dehydration (OD) and vacuum impregnation (VI) on the quality of dried apple products. Weight reduction and water loss increased during OD, but these decreased in the apples during VI. In particular, VI's effect on increasing solid gain was superb. For apples in 40% sucrose solution, OD and VI were followed by hot-air drying at 50$^{\circ}C$. The experimental data were fitted successfully using the modified Page model. OD and VI increased drying time and decreased the drying rate constant of these apples as compared to the control. Shrinkage and rehydration capacity greatly decreased in the apples dried by OD and increased in the apples dried by VI as compared to the control. OD also decreased titratable acidity and ascorbic acid content considerably. Sensory evaluations of the products indicated that the apples prepared by OD had higher palatability in their rehydrated form in yoghurt, and the apple products prepared by VI had higher palatability in their dried form.
In order to develop the protoplast fusion method of the strains of Fusarium, the interspecific protoplast fusion was attempted between Fusarium poae and F. sporotrichioides. Various auxotrophic mutants were isolated by the treatment of N-Methyl-N'-Nitro-N-Nitrosoguanidine. The optimal conditions for the formation and regeneration of protoplasts were examined and the characteristics of a fusant were studied. As a results, protoplasts were readily obtained from 18 hours cultured mycelia by the treatment of driselase for 3 hours and 0.6 M KCl as a best osmotic stabilizer at pH 6.0 for the formation of protoplast. Sucrose was the most suitable for the regeneration. Polyetylene glycol (M.W. 8,000) in $CaCl_{2}$-glycine solution was used to induce the protoplast fusion. The interspecific fusion frequency between protoplasts among the auxotrophic mutants of the two strains ranged from $2.7*10^{-2}$ to $5.7*10^{-3}$ . DNA content and cellulase activity were rather increased in the interspecific fusant. The lag phase of growth curve was slightly elongated in the fusant.
Tobacco (Nicotiana tabacum L. cv KF109) seeds were artificially aged in a controlled environment of 45$^{\circ}C$ and 80% relative humidity condition for different duration up to 14 days before priming in polyethylene glycol 6000 solution of equivalent osmotic potential of -0.8 MPa for 8 days at $25^{\circ}C$. The seeds aged only and primed after aging were germinated at 15$^{\circ}C$ and $25^{\circ}C$ to observe the priming effects on the germination of aged seeds at different temperature. The germination percentage of the aged seeds was rapidly dropped starting from 8 days of aging and mean germination time (T$_{50}$) was greatly increased, particularly in germination at 15$^{\circ}C$. The germination capacity was greatly restored in the primed seeds after aging, particularly in the seeds of longer aging and germinating at 15$^{\circ}C$.>.
